Purification and Properties of Laccase of the White-rot Basidiomycete Coriolus hirsutus

  • Laccase produced by Coriolus hirsutus was purified to electrophoretic homogeneity by acetone precipitation, Sephacryl S-2000 HR chromatography, DEAE Sepharose CL-6B chromatography, and Mono Q HR 5/5 chromatography. The purification of laccase was 46.6-fold with an overall yield of 23.7%. Laccase from this fungus was a monomeric glycoprotein with 16% carbohydrate content, and has an isoelectric point of 4.2, and molecular mass of 78 kDa, respectively. The N-terminal amino acid sequence of the enzyme showed significant homology to hoste of laccases from Coriolus versicolor, Pycnoporus cinnabarius, and an unidentified basidiomycete, PM1. The highest rate of 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS) oxidation by laccase was reached at 45$^{\circ}C$, and te pH optima of the enzyme varied depending on the substrate in the range of 2.0 to 4.5. The enzyme was stable at 60$^{\circ}C$ for 5 h and lost 80% activity at 80$^{\circ}C$ in 30 min. The enzyme oxidized a variety of usual laccase substrates including lignin-related phenol, and had the highest affinity toward ABTS. Under standard assay conditions, the apparent Km value of the enzyme toward ABTS was 8.1 ${\mu}$M. The enzyme was completely inhibited by L-cysteine and sodium azide, but not by potassium cyanide, SDS, ad thiourea.

  • Application of sludge wastes into the field may help soil fertility with physical, chemical and biological properties. Efficient use of sludge waste, however, requires an individual assessment of the waste products. A lot of experiment into the organic waste-treated soils has been done for decade. However, studies have not been carried out on the assessment of agricultural soil by Basidiomycota. This study was assessed the influence of sludge application on soil healthy in agricultural upland soils. The organic wastes selected for long-term application experiment in this study were municipal sewage sludge (MSS), industrial sewage sludge (ISS), leather processing sludge (LS), alcohol fermentation processing sludge (FS), and pig manure compost (PMC). To develop the soil healthy assesment method, soil samples were diluted by 20X with distilled water. After shaking at 200rpm for 30 minutes, the shaked sample was mixed on PDA(Potato Dextrose Agar). And sterilized at $121^{\circ}C$ for 20 minutes. Coriolus hirsutus (MKACC 50560) was inoculated on petri-dish including PDA mixed sample. After the media was incubated at $25^{\circ}C$ for five days, the mycelial growth of C. hirsutus was measured. When the mycelial growth on sample media was compared with growth on media contained PDA only, well grown media contained sample soil was assesed as healthy soil. The results suggest that the simple method by Coriolus hirsutus is a handy way to assess the healthy of waste sludge-applied upland soils.

  • The occurrence of Anagyrus kamali, a solitary endoparasite of the mealybug, Maconellicoccus hirsutus has been reported for the first time from India. The parasitoid was fecund to parasitizing the field population of mealybug to the tune of 10.37 to 42.70% in different months. A comprehensive study on the development of the parasitoid on different stages of mealybug indicated that the parasitoid was able to complete its development in all the stages. Higher parasitism (67.48-78.08%) and more female progeny were observed when 3rd instar nymphs and adult female of the mealybug were exposed to the parasitoid. The biology of A. kamali studied at 25${\pm}1^{\circ}C$ temperature and 60${\pm}$$\pm$1.12 days. The duration of egg, larva (3 instars) and pupa were 2.67${\pm}$${\pm}$${\pm}$${\pm}$

  • Seven white rot fungi (Irpex lactenis, Coriolus hirsutus, Lopharia mirabilis, Schizopora paradoxa, Ganoderma lucidum, Pleurotus ostreatus and Pycnoporus coccineus) native to Korea and two famous exotic lignin degradable white rot fungi (Coriolus versicolor and Phanerochaete chrysosporium) were investigated to clarify their physiological and physicochemical characteristics on white-rotted wood blocks. G. lucidum degraded wood blocks more seriously than those by exotic lignin-degrading fungi, C. versicolor and P. chrysosporium, but only slightly decreasecl the strength of wood which was compared to the weight loss, persumably on the account of its small use of cellulose when attacking wood. It is quite interesting to note that the holocellulose degradation rate of G. lucidum was also higher than any of the other tested fungi. The order of fungi, according to the lignin-decomposing rates, was G. lucidum>P. coccineus>C. versicolor>S. paradoxa>P. chrysosporium>L. mirabilis>P. ostreatus>C. hirsutus>I. lactenis. The lignin degradation of G. lucidum and P. coccineus which were collected in Korea was greater than that of C. versicolor and P. chrysosporium. If holocellulose degradation is not considered. G. lcidum has the merit of actual application in biomass conversion due to linin removal.

  • Extracellular laccase excreted from Lentinus edodes ATCC 48085 was purified through a series of DEAF, Sephadex A-50. Con A-Sepharosc and Sephadex G-150 chromatography. Extracellular enzyme. which consists of a single polypeptide, has a n~olecular mass of 87.000 daltons and contains 12.0'%, carbohydrate. The N-terminal amino acid sequence (I5 residues) of the puritied enzyme was similar to that of laccases of PIeurotus ostreatus and Coriolus hirsutus. The enzyme showed optimal activity at near pH 4.8 and $40^{\circ}C$. The enzyme was stable at pH 7-9 and below $30^{\circ}C$. $K_{M}$ and $k_{cat}$ values for syringaldazine were estimated to be $0.4\mu\textrm{M}$ and 77 sec, respectively. The developed patterns of reaction products of thevenzyme on thin layer chromatography were similar to those of laccase of Pleurotus ostreatus.

Frequency and Wood Decaying Ability of Butt-rot Fungig Isolated form Larch (Larix leptolepis) (낙엽송근주심재부후병균(根株心材腐朽病菌)의 분포빈도(分布貧道)와 목재열화능력(木材劣化能力))

  • Butt-rot fungi were isolated from decayed portion in butt rotted larch trees[Larix leptolepis(Sieb. et Zucc.) Gord.]. From a total of 216 affected trees in 38 forests of five provinces, Sparassis crispa(Wulf. ex Fr.) Fr. was isolated with the highest frequency, i.e.. 28% of the total trees. Laetiporus sulphureus(Fr.) Bond. et Sing, with 19%, and Phaeolus schweinitzii(Fr.) Pat. with 8%. However, none of these fungi was isolated from 45% of the trees. Wood decaying abilities and interaction of four basidiomycetous and three non-basidiomyo:etous fungi isolated from butt rotted trees were investigated. The weight loss of larch wood blocks, when exposed to the culture of a fungus for six months, was 19.6% for P. schweinitzii, 10% for S. crispa, 8.2% for L. sulphurous, 5.1% for Coriolus hirsutus, and less than 4.4% for three non-basidiomycetous fungi. In incubation with combinations of the fungi, the wood decay activity of S. crispa, L. sulphurous and C. hirsutus was increased by the non-basidiomycetous fungi of Geotrichum candidacy, G. sp. and Verticillium sp., but that of P. schweinitzii was not.

Effect of Medicinal Plant Extracts on Mealy Bugs (Maconellicoccus hirsutus Green) Affecting Mulberry

  • Efficacy of aqueous leaf extracts of medicinal plants viz., Eucalyptus globulus, Ocimum sanctum and Piper betle were evaluated against the hatching of eggs, mortality of nymphs and mortality and fecundity of adult mealy bugs (Maconellicoccus hirsutus Green) under in vitro conditions. Totally seven concentrations 1, 5, 10, 20, 50, 70 and 100 percent were tested against mealy bugs. The results revealed that with the increase in the concentration of extracts, there was a corresponding decrease in hatching, nymphal & adult mortality and fecundity in all the extracts tested. The hatching of eggs was minimum at 100 percent concentration in E. globulus (20.00%) followed by O. sanctum (30.42%) and P. betle (36.06%) over control (96.73%). The growth and development of nymphs and adults were adversely affected after the treatment. The nymphs did not attain the adult stage and there was total mortality of nymphs in different treatments. The duration of nymphal stage was reduced by 2-10 days in E. globulus, 1-7 days in O. sanctum and 1-2days P. betle at higher concentrations. However, at lower concentrations it was prolonged by 2-4 days in all the three extracts. At lower concentrations the mortality of adults was on par with the control. Similarly the adult duration was also reduced by 3-4 days at 100 percent concentration and prolonged by 5-6 days at lower concentrations in all the extracts. With the increase in concentrations of the extracts there was decrease in the fecundity of eggs. Thus, the leaf extracts of E. globulus was found to be most effective followed by O. sanctum and P. betle against mealy bugs.

Characterization of Laccase Purified from Korean Trametes hirsuta S1 (한국산 흰구름버섯(Trametes hirsuta S1)으로부터 정제된 Laccase의 특성)

  • Laccase produced by Trametes hirsuta S1 isolated from Korea was partially purified and characterized using ultrafiltration, anion exchange chromatography and affinity chromatography. The laccase was produced as the predominant extracellular enzyme during primary metabolism. Neither lignin peroxidase nor veratryl alcohol oxidase (VAO) were detected in the culture fluid. Addition of 2,5-xylidine enhanced 4-fold laccase production. Purified laccase was a single polypeptide having a molecular mass of approximately 66 kDa, as determined by SDS-polyacrylamide gel electrophoresis, and carbohydrate content of 12%. $K_{m}\;and\;V_{max}$ values for laccase with ABTS [2,2-azinobis (3-ethylbenzthiazoline 6-sulfonic acid)] as a substrate (Lineweaver-Burk plot) was determined to $51.2\;{\mu}M\;and\;56.8\;{\mu}mole$, respectively. The optimal pH for laccase activity was found to be 3.0. The enzyme was very stable for 1 hour at $50^{\circ}C$. Half-life ($t_{1/2}$) of the enzyme was about 20 min at $70^{\circ}C$. Spectroscopic analysis of purified enzyme indicated that the enzyme was typical of copper-containing protein. Substrate specificity and inhibitor studies for laccase also indicated to be a typical fungal laccase. The N-terminal amino acid sequence of the T. hirsuta S1 laccase showed 100% of homology to those of laccase from C. hirsutus.

Fungal flora of Ullung Island (III) -on some polyporoid fungi- (울릉도의 균류상 (III) -수종의 구멍장이버섯류에 관하여-)

  • Fresh fungi were collected during field trips to Ullung Island from August, 1990, to September, 1991. Among them, some polyporoid fungi were recognized to the species. Thirty three species were identified, among which nine fungi were confirmed new in Korea and registered here with descriptions. These fungi are Auriporia aurulenta, Ceriporiopsis gilvescens, Ceriporiopsis subvermispora, Gloeophyllum trabeum, Junghuhnia separabilima, Oligoporus stipticus, Oxyporus similis, Phellinus lavigatus, and Polyporus melanopus.
