  • 급성 심근경색 진단용 DNA 컴퓨팅 시스템 모듈로서, 트로포닌 I (troponin I, Tnl). 트로포닌 T (troponin T, TnT). 미오글로빈 (myoglobin), C-반응 단백질 (C-reactive protein, CRP) 과 각각 결합할 수 있는 네 가지 종류의 앱타머틀 선정하고, 이의 개발을 시도하여, 그 중 첫 번째로 C-반응 단백질-결합 앱타머를 SELEX 기법을 이용하여 선별해내었다. 또한, 선별된 앱타머 염기서열에 기초하여 각각 10-mer 길이의 FDNA 와 QDNA 를 제작하고, 표적 단백질 (CRP) 과 혼합시켜 형광발현 변화의 추이를 살펴보았다. 앱타머 및 FDNA. QDNA 가 결합할 경우에는 형광감쇄효과가 발생하므로, 형광감쇄효과가 일어나지 않은 경우에 비하여 현저하게 형광측정값이 저조하게 나타나는 현상을 확인할 수 있었다. 향후 연구로, 나머지 세 가지 종류의 앱타머를 SELEX기법을 이용하여 선별해내고. 기확보된 C-반응 단백질-결합 앱타머 모듈과 함께 논리회로를 구성하는 DNA 컴퓨팅 칩을 제작할 예정이다.

Hydrolysis of Isolate Soybean Protein Using Subcritical Water (아임계수를 이용한 분리대두단백질의 가수분해)

  • Hydrolysis of isolate soybean protein (ISP) using subcritical water (SCW) was conducted to study the feasibility for producing protein hydrolyzate. SCW hydrolysis of SPI suspension (5-15%) was conducted in an electrically heated batch reactor (2 L). The effects of temperature (230 to $270^{\circ}C$) and holding time (10 to 50 min) on the degree of hydrolysis (DH) and the production of amino acids were studied by surface response method. The DH was determined by derivatizing the hydrolyzates with ortho-phthalaldehyde (OPA) solution. It was confirmed that reaction temperature and holding time affected the hydrothermolysis of soybean protein. However, the holding time was less effective on amino acid yield when the temperature was higher than $230^{\circ}C$. In order to achieve optimal yields of amino acids exceeding 43%, the temperature should be within the range between 256 and $268^{\circ}C$ with holding time from 29 to 41 min, respectively. A maximum estimated amino acid yield of 43.5% was obtained at $268^{\circ}C$ for 35 min.

  • 본 연구에서는 산성전리수와 환원전리수가 생체물질인 DNA와 단백질의 분해에 미치는 영향을 조사하였다. 산성전리수에서 DNA는 반응 1일 후 약 $40\%$가 분해되었고 1주일 후에는 거의 완전히 분해되었다. 이에 비해 환원전리수에서 DNA는 총 10일간의 반응시간동안 거의 분해가 되지 않았다. 산성전리수에 크기가 약 40 kDa인 효소단백질을 반응시켰을 때 반응시점부터 효소단백질이 분해되기 시작하여 26 kDa와 35 kDa의 단백질 조각이 잘라져 나왔다. 증류수에 반응시킨 효소단백질도 산성전리수에서와 동일한 크기의 단백질 조각을 생성하면서 분해되었다. 산성전리수와 증류수에 의한 단백질의 분해는 $4^{\circ}C$ 보다 $25^{\circ}C$에서 더 심화되었다.

  • Monoclonal antibodies were prepared against Bombyx densonucleosis virus type-II(Yamanashi isolate). Four hybridoma clones, named C4, Fl, H2, M9 were only reacted with the DNV-II, but those were not reacted with Bombyx densonucleosis virus type-I(Ina isolate) and infectious flacherie virus(IFV) by double diffusion test in 0.8% agarose gel. C4, Fl and M9 of them were reacted with 53KDa polypeptide of DNV-II, and H2 was reacted with 46. 5KDa polypeptide of the virus.

  • To analyse the antigen specificity of patients sera from 24 confirmed neurocysticercosis and a monoclonal antibody, SDS-PAGE using 10~15% linear gradient gel and EITB were done. Cystic fluid, saline extracts of scolex and of whole worm of C. cellulosae, saline extracts of sparganum, hydatid cyst fluid, saline extracts of Fasciola, Clonorchis and Paragonimus were used as antigen. Of protein bands in cystic fluid of C. cellulosae, patient sera reacted frequently to bands of 152, 94, 64, 48, 24, 15, 10 and 7kDa proteins. To saline extracts of scolex and whole worm of C. cellulosae, patients sera reacted frequently to 94, 64, 52, 39, 34, 15 and 10kDa bands. Two bands in sparganum extract (130 and 64kDa) and two bands in hydatid cyst fluid (52 and 27kDa) were cross-reacting bands with sera from cysticercosis patients. Saline extracts of Fasciola, ClonorchiJ and Paragonimus did 'not exhibit cross-reacting bands. Monoclonal antibody to cystic fluid of C. cellulosae was found to react with low molecular weight proteins of 15, 10 and 7kDa.

  • Effects of pH, amino acids, hydrolyzed protein and potassium phosphate on caramelization were investigated for improvement of its reaction rate. The caramelization was performed with starch syrup at $110^{\circ}C$ and the different color functions-metric saturation(Suv), 5-hydroxymethylfurfural (HMF) contents and absorbance at 420 nm were measured. As the pH was raised from 4 to 10, the reaction rate (Suv/hr) was increased by 31.9% along with significant increase in HMF content and absorbances at 420 nm. Among the several amino acids, arginine and glycine were very effective for improvement of caramelization, which may be due to Maillard reaction. When $K_2HPO_4$ were added in different ratio with arginine, glycine, HVP or HAP, the effects of arginine and HAP on thee rate were markedly enhanced while the effects of glycine and HVP were rather reduced.

  • The contribution of lipid to thermal flayer generation from glucose-protein reaction was accomplished by isolating flavor compounds from casein-glucose (CG)and casein-glucose-coin oil (CGL) which were stored for 2 and 4 weeks at 6$0^{\circ}C$ and then reacted at 16$0^{\circ}C$ for 1hr. The volatiles from the reactant mixtures were isolated by a solvent extraction method with methylene chloride and analyzed by gas chromatography and gas chromatography-mass spectrometry. Pyrazine, methylpyrazine, 2,5-dimethylpyrazine, 2-dimethylpyrazine ,2-ethy-5- methyIpyrazine and 2-acetylpyrrole originated from interaction of thermal degradation of casein and lipid oxidation were identified in the CGL samples. It was also found that 3-methyl-1-butanol, 2-cyclopene-1,4-diode, heptanal, nonanal, and 2-heptanone were derived from lipid source. Two additional fatty acids, heptanoic acid and octanoic acid were also identified in the CGL samples. 5-Hydroxymethyl-2-furfural, the most abundant volatile, was responsible for the formation of sugar degradation product. The results suggested that the presence of lipid in the samples had more effect on the contribution of volatile formation of glucose-protein thermal reaction than the absence of lipid in the samples.

Process Optimization of Peptides Production from Protein of Sea Cucumber and Its Antioxidant Capacity Analysis (해삼 단백질로부터 펩타이드 제조 최적공정 확립 및 항산화 특성)

  • Protein hydrolysates derived from plants and animals having antioxidant, suppression of hypertension, immunodulatory, alleviation of pain, and antimicrobial activity has been known as playing important role like hormone. This study was performed to optimize the hydrolysis of protein of sea of cucumber by a flavourzyme. The ranges of processes were the reaction temperature of 40 to $60^{\circ}C$, pH 6 to 8, and enzyme concentration 0.5 to 1.5%(w/v). As a result, the optimization of process was determined at temperature of $48-50^{\circ}C$, pH of 7.0-7.2, and enzyme concentration of 1.0-1.1%(w/v), and degree of hydrolysis was 43-45 at above conditions. The molecular weight of hydrolysate was distributed to 500-3,500 Da and showed typical peptides. Inhibition concentration ($IC_{50}$) of peptides of DPPH radical scavenging activity, Superoxide anion radical scavenging activity, Hydroxy radical scavenging activity, $Fe^{2+}$ cheating activity was 1.25, 3.40, 10.3, and 22.11 mg/mL, respectively. Therefore, we expect that those products are useful as functional food ingredients.

  • When cystic fluid of Taenia solium metacestodes (CF) was filtrated through Sephacryl S-300 Superfine, major proteins were in fractions III add IV Major protein in fraction III was Band C protein of 150 kDa and that in fraction IV was Band N protein (Choi et of., 1990). When CF was electrophoresed in 0.9% agarose gel and reacted with anti-CF rabbit serum (RACF), two main bands, a long outer and a short inner band, were precipitated, together with 8 minor bands. RACF reacted with fraction III forming the long outer band whereas RACF formed the short infer band with fraction IV in immunoelectrophoresis (IEP) The long outer precipitin band of CF fraction III was similar to antigen B in hydatid fluid (HF) of Oriol et at. (1971), while the short inner band of CF fraction IV was similar to HF antigen 5 of Caption et at. (1967) . When HF was reacted with RACF, the short inner band was immunoprecipitated without forming the long outer band. Common antigenicity between CF and HF seemed to exist in fraction IV rather than in fraction III of CF. Patient sera of neurocysticercosis reacted more frequently with fraction III than with fraction IV.

  • Probiotic 활성이 높은 L. acidophilus 30SC의 생존성을 증진시키기 위한 기초자료를 얻고자, 열처리 동안 새로이 발현되는 단백질을 일차원 및 이차원 전기영동을 이용하여 확인하였으며, 세포 모양을 주사전자현미경을 사용하여 관찰하였다. $55^{\circ}C$의 heat shock에는 L. acidphilus 30SC의 사멸이 있는 것으로 나타났다. 나머지 처리구는 $37^{\circ}C$에서 계속 배양한 것과 별다른 차이를 나타내지 않았다. $45^{\circ}C$로 heat shock을 준 경우 $37^{\circ}C$에서 배양한 것과 거의 동일하였다. $55^{\circ}C$에서 15분 heat shock을 준 경우 약 22kDa와 25kDa의 단백질들이 새로이 발현된 것으로 나타났으나, 24 kDa와 27kDa로 추정되는 단백질의 발현정도는 낮았음을 확인하였다. 이차원 전기영동을 실시한 결과, $37^{\circ}C$와 비교할 때 $55^{\circ}C$로 heat shock을 준 경우 새로이 5개의 protein spot을 발견할 수 있었다. 그러나 6개의 단백질 spot은 $55^{\circ}C$ heat shock에서 소실된 것으로 확인되어 추가적인 단백질의 분석이 필요한 것으로 생각되었다.
