항방사선 인삼분획의 butanol 추출물과 수용성 성분이 세포 생존율에 미치는 영향 (Effects of Butanol Extract and Water-Soluble Constituent of Radioprotective Ginseng Fractioil on Cell Survival)

  • Radiation protective fraction was Isolated and partially purified from Korean white ginseng. The effect of the fraction was studied on the cell survival of W-damaged CHO-Kl cells. As a result, it was found that the fraction increased the survival rate of damaged cells significantly within the dose range of which cytotoxicity did not appear This fraction was separated into two parts by adding butanol, namely the precipitated protein component and the butanol extract. Damaged cells were treated with each of these components and their survival rates were measured. The protein component demonstrated significant increase in the survival rates, while the butanol extract showed no such increment. These results suggest that the radiation protective effect of the ginseng fraction is originated from the butanol-precipitated protein component, not from the butanol-soluble compounds.

뽕잎 추출물의 항산화 효과와 항산화 성분 분리 및 동정에 관한 연구 (The Antioxidative Effects and Isolation and Characterization of the Extracts from Morus alba L.)

  • This study identified the formula of the antioxidant substance separated from the ethyl acetate and the butanol extract and tested the antioxidant properties with the electron donating ability(EDA). Each phase with the fractionated methanol extract from mulberry leaf was screened in advance for the antioxidant substance with EDA. As the result, activity appeared in the ethyl acetate and butanol phase and the antioxidant component was separated. As the consequence, 2 components from the ethyl acetate phase and 1 from the butanol phase were separated, among which the structures of the components from ethyl acetate were determined by wogonin and linarin, whereas the structure of the component from the butanol phase was determined by pectolinarin. In the screening of antioxidant activity by the scavenging effect of the DPPH radical, the wogonin and linarin components from ethyl acetate phase showed more powerful antioxidant property than the component from butanol. The results from this study indicate that the chemical compound separated from the ethyl acetate extract has more powerful antioxidant property than the one separated from the butanol extract. The components separated from the ethyl acetate extract were wogonin and linarin, which are flavonoids, whereas the component from butanol was pectolinarin. Therefore, this study suggested that the feasibility of mulberry leaf as a functional food additive and its value as a natural antioxidant is very high.

소목 부탄올 추출물이 B16/F10 흑색종세포의 멜라닌 합성에 미치는 효과 (Butyl Alcohol Extract from Caesalpinia sappan L. Regulates Melanogenesis in B16/F10 Melanoma Cells)

  • Caesalpinia sappan L. has long been commonly used as emmenagogue, analgesic, and a cure for contusion and sprain as well as a remedy for thrombosis in the Oriental medicine. The main constituent of C. sappan is brazilein, which is an antioxidative substance that has a flavonoid structure. In this study, we examined the effect of butanol extract of C. sappan on proliferation and melanogenesis in B16/F10 melanoma cells. After 48h treatment of cells with various concentrations of butanol extract, the cells exhibited a dose-dependent inhibition in their proliferation without apotosis. Therefore, the growth retardation by the extract may be due to the cell arrest, not due to the cell death induced by cytotoxicity. We also estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in melanogenesis of B16/F10 melanoma cells. Our result showed that the melanin contents and tyrosinase activity were decreased in butanol extract-treated cells in a dose dependent manner compared to control group. In conclusion, it was observed that butanol extract of C. sappan inhibited melanization of these cells and therefore butanol extract could be developed as skin whitening components of cosmetics.

Effect of Butanol Fraction of Panax ginseng Head on Gastric Lesion and Ulcer

  • From our previous result that Panax ginseng head extract had inhibition of gastric damages, the extract was fractionated. Among the hexane, chloroform, butanol and water fractions, butanol fraction Showed the most potent inhibition of HCl.ethanol-induced gastric lesion, aspirin-induced gastric ulcer, acetic acid-induced ulcer and Shay ulcer. Butanol fraction showed significant increase in mucin secretion, and inhibited malondialdehyde (MDA) and $H^{+}/K^{+}ATPase$ activity in the stomach. This results indicate that the effectiveness of the fraction on gastric damages might be related to inhibition of acid secretion, increment of mucin secretion and antioxidant property.

오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향 (Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours. Comparative antidiabetic activity of different fractions of Enicostemma littorale Blume in streptozotocin induced NIDDM rats

  • Aqueous extract of Enicostemma littorale is reported to have antidiabetic activity. In the present investigation, we studied the effect of aqueous extract of E. littorale and its different fractions i.e., toluene, chloroform, ethyl acetate, n-butanol fractions and remaining residual fraction in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in NIDDM were significantly (P<0.05) higher than control rats and they were significantly decreased by treatment with aqueous extract of E. littorale and its n-butanol and ethyl acetate fractions. Results of oral glucose tolerance test (OGTT) showed that aqueous extract and its n-butanol and ethyl acetate fractions significantly (P<0.05) decrease both $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM treated groups. Insulin sensitivity $(K_{ITT})$ index of NIDDM control was significantly lower as compared to normal control and this was significantly (P<0.05) increased after treatment with aqueous extract, its n-butanol and ethyl acetate fractions. Treatment with aqueous extract of E. littorale and its n-butanol and ethyl acetate fractions lowered the elevated cholesterol and triglyceride levels observed in NIDDM rats. Treatment with aqueous extract of E. littorale and its n-butanol fraction showed significant decrease in creatinine, urea, SGPT and SGOT levels as compared to NIDDM control rats. However ethyl acetate fraction showed significant changes only in creatinine and SGOT levels, and not in the levels of urea, and SGPT as compared to NIDDM control rats. Treatment with toluene, chloroform and residual fractions of E. littorale did not produce any effect on glucose, insulin, triglyceride, cholesterol, creatinine, urea, SGPT or SGOT levels as compared to NIDDM control rats. Our data suggest that n-butanol and ethyl acetate fractions contain the active compounds which may be responsible for the above activity and associated complications in NIDDM diabetes mellitus.

  • Periodontitis is a chronic inflammatory disease that is known to have the characteristics of destructed periodontal tissue. Anti-oxidant and anti-inflammatory effects of mulberry leaves in periodontal tissue is not well known until now. We investigated the effects of n-butanol extract of mulberry leaves on the lipopolysaccharide (LPS)-induced proinflammatory cytokines, such as $IL-1{\beta}$, IL-6, IL-8 and modulates osteogenic differentiation in periodontal ligament cells. The expression levels of Runx2, ALP and mRNA were increased by n-butanol extract of mulberry leaves at the concentration of $100{\mu}g/ml$ in periodontal ligament cells. n-Butanol extract of mulberry leaves extract reduced the range of pathophysiological processes, such as inflammation and increase in the level of osteogenic-related genes. These findings suggest that n-butanol extract of mulberry leaves has therapeutic effects on periodontitis and periodontal tissue regeneartion.

한국산 삽주의 기능성 (Functional Properties of Korean Atractylodes japonica Koidz)

  • 생삽주의 기능성을 분석한 결과, 추출 수율은 ethanol 추출물에 14.8%와 water 분획물에 17.7%를 나타내었다. Acetone 추출물과 butanol 분획물의 수소공여 능은 각각 72.9%와 74.2%로 높게 나타났으며, methanol 추출물과 butanol 분획물의 아질산염 소거 효과는 각각 95.0%와 79.2%로 가장 높게 나타났다. 생삽주의 용매별 추출물 중 methanol 추출물이 높은 항균 활성을 나타내었는데, Bacillus subtilis에 대해 20 mm clear zone을 나타내었으며, Pseu. aeruginosa에 대해 19 mm clear zone을 나타내었다. 항균 활성이 가장 높은 methanol 추출물을 계통분획하여 항균 활성을 확인한 결과, butanol 분획물이 높은 항균 활성을 나타내었는데, Bac. subtilis, V. parahaemolyticus에 대해 각각 18 mm clear zone을 나타냈다. Methanol 추출물과 butanol 분획물을 이용하여 최소저해농도를 확인한 결과 대체로 gram(+)균에 대해 2 mg/disc와 4 mg/disc에서 나타났구 gram(-)균에 대해서는 6 mg/disc에서 저해활성을 나타내었다.

Investigation of n-Butanol and Ethyl Acetate Extracts from Thermal Treatment Yam (Dioscorea batatas DECNE.) for their Antioxidant Activities

  • n-Butanol and ethyl acetate extracts of thermal treatment yam (Dioscorea batatas $D_{ECNE.}$) belonging to the family Dioscoreaceae were measured for their radical scavenging activity and lipid peroxidation inhibition ability. In this study, ethyl acetate extract showed the most potent antioxidant activity evaluated by ferrous ion chelating activity and NO radical scavenging activity. Nevertheless, n-butanol extract was more effective in inhibiting linoleic acid peroxidation. A significant difference between n-butanol extract and ethyl acetate extract in nitrite scavenging activity ${\beta}$-carotene bleaching assays could not be found. Also, the results of this study showed that thermal treatment yam could be used as easily accessible source of natural antioxidants and as a possible food supplement.

포공영 추출물이 흰쥐간 Xanthine Oxidase 활성에 미치는 영향 (Effect of Taraxacum herba Extract on the Hepatic Xanthine Oxidase Activity in Rats)

  • This study was undertaken to investigate the effect of Taraxacum herba extract on the hepatic xanthine oxidase activity as a oxygen free radical generating enzyme in vitro and in vivo. It was observed that partial purified hepatic xanthine oxidase (type O) activity was strongly inhibited by the addition of Taraxacum herba n-butanol extract in vitro. The Km value of xanthine oxidase without affecting the Vmax value for xanthine was significantly increased by the addition of ta-dase (type O) activity was significantly inhibited by the treatment of Taraxacum gerba n-butanol ex-tract for 5days(over 40mg/kg, i.p), whereas, xanthine oxidase (type D) activity was not changed by the injection of Taracacum herba n-butanol extract. Meanwhile, liver weight / body weight(%), serum alanine aminotransferase activity and hepatic lipid peroxide content in Taraxacum herba n-buta-nol extract-treated rat were not changed. These findings led us to conclude that Taraxacum herba n-butanol extract may regulate the hepatic xanthine oxidase type O activity to prevent toxic effect of oxidative stress by the oxygen free radicals.
