This experiment was conducted to evaluate the effects of dietary wild-ginseng adventitious root meal (WGM) on growth performance, blood profiles, relative organ weight and meat quality of broiler chickens. A total of 480, 2-day-old male broiler chicks (BW = $42.8{\pm}1.38\;g$) were randomly allocated to 1 of 4 dietary treatments (6 cages with 20 broilers per cage). Dietary treatments were: i) CON (basal diet), ii) WGM0.1 (basal diet+0.1% WGM), iii) WGM0.2 (basal diet+0.2% WGM) and iv) WGM0.3 (basal diet+ 0.3% WGM). Birds fed WGM0.3 diet (p<0.05) had a higher feed intake (FI) than those fed the CON diet during weeks 1 to 3. During weeks 3 to 5, dietary WGM0.1 treatment led to a higher (p<0.05) BW gain (BWG) and feed intake (FI) compared with the CON and WGM0.3 treatments. Overall, birds fed WGM0.1 improved BWG and FI compared with those fed the CON treatment. A greater lymphocyte count was observed (p<0.05) in WGM0.2 and WGM0.3 treatments compared with the CON treatment; dietary WGM decreased (p<0.05) the total cholesterol concentration compared with the CON group. The inclusion of WGM increased the relative weight of spleen and bursa of fabricius (p<0.05) compared with CON, while less abdominal fat was observed in the WGM0.3 treatment (p<0.05) compared with CON. The 2-thiobarbituric acid reactive substances (TBARS) of breast muscle were decreased (p<0.05) by WGM supplementation. Overall, our results indicated that the use of WGM at the 0.1% level could enhance growth performance in broilers. The supplementation of WGM could induce a decreased TBARS, abdominal fat and serum cholesterol in broiler chickens.
This study was conducted to explore the effects of squid ink on growth performance, immune functions and antioxidant ability of broiler chickens during a period of six weeks. Either sex Arbor Acres broilers were equally allotted to 4 groups with 3 replicates of 20 chickens each. Broilers diets for the 4 test groups were prepared separately with starter and finisher phases. Control chickens were fed with basal diet and birds of group Exp 2, Exp 4 and Exp 6 were fed with the basal diet supplemented with 2%, 4% and 6% of squid ink, respectively. Broilers were sacrificed to investigate antioxidant parameters of sera, indices of thymus, spleen and bursa of fabricius and spleen lymphocyte proliferation, as well as growth performance on the $21^{th}$ and $42^{th}$ day. The results revealed that, i) squid ink promoted growth performance of broilers during days 22 to 42 and the whole trial period (p<0.05 or p<0.01); ii) squid ink elevated relative weight of the three immune organs during the starter phase and spleen lymphocyte proliferation throughout the experiment (p<0.05); iii) squid ink increased SOD activity and decreased MDA level in sera from broilers during the whole period (p<0.05). The above results suggest that squid ink could improve growth performance, antioxidant ability and immune functions of growing broiler chickens and be employed in the development of feed additives for animals.
This study was performed to produce monoclonal antibodies (mAb) specifically reacting with chicken leukocyte surface antigens. Popliteal lymph node cells of BALB/c mice previously immunized through foot-pad with peripheral blood mononuclear cells (PBMC) of chickens separated by Ficoll-Histopaque method. They were fused with P3X63Ag14 mouse myeloma cells. A total of 34 hybridomas secreted antibodies specifically binding to the PBMC. According to the reactivity patterns with PBMC, the mAbs were divided into 4 groups. Group 1 mAbs (IIB3, IIB10, IIE10) specifically reacted with non-adherent lymphocytes but not with adherent cells which were mainly composed of thrombocytes and monocytes in PBMC culture. These mAbs were reactive with 25-59% of thymus cells and 42-64% of spleen cells of chickens. They did not show any significant reactivity with cells in the bursa of Fabricius, T-cell (MDCC-MSB1) and B-cell (LSCC-1104B1) lines. These results indicate that Group I mAbs specifically reacted with T-lymphocyte subpopulation. Monoclonal antibodies in Group II (IC6, IG2-2 and IID9) showed specific reactivity with monocytes but not with thrombocytes or non-adherent cells in PBMC culture. These mAbs, though not reacted with the chicken macrophage cell line, HD11, also bound to macrophages of the spleen and lung in immunohistochemical staining. Five mAbs in Group III showed characteristics of binding to lymphocytes and monocytes, but not to thrombocytes. Twenty-three mAbs in Group IV showed specific reactivity to lymphocytes, monocytes, and thrombocytes. Two mAbs (IC3 and IE9) in Group IV reacted with most of PBMC.
The aim of this study was to evaluate the potential of fermented red ginseng extract (FRGE) as feed additive in broilers and laying hens. In broilers, 480 Arbor Acre male broilers were randomly allotted to 4 treatments with 6 replications per treatment and 20 chicks per pen. The experiment lasted 5 weeks and dietary treatments were as follows: i) CON, basal diet; ii) FRGE1, basal diet+1 g/kg fermented red ginseng extract; iii) FRGE2, basal diet+2 g/kg fermented red ginseng extract and iv) FRGE3 basal diet+4 g/kg fermented red ginseng extract. Throughout the experiment, no effects were observed (p>0.05) in performance in response to FRGE. At the end of the experiment, FRGE administration improved (p<0.05) the lymphocyte level compared with CON. The relative weight of bursa of fabricius and spleen were increased (p<0.05) by the inclusion of FRGE3. Besides, redness ($a^*$) value for the breast meat was higher (p<0.05) in FRGE1 and FRGE3 treatments than that in CON. In laying hens, 240 ISA brown layers at 35 weeks of age were used in this 8-week trial. Dietary treatments were the same as in the broilers trial with 10 replicates per treatment and 6 layers per replicate. During the entire experiment, there were no significant differences (p>0.05) in performance or egg quality among all the treatments. However, the layers fed diets supplemented with FRGE had higher lymphocyte level (p<0.05) compared with those fed CON. In conclusion, the dietary supplementation with FRGE did not influence performance but improved the lymphocyte level in both broilers and laying hens.
The immune response against Salmonella gallinarum infection was investigated in immunosuppresed chickens. Newly hatched chickens were treated with cyclophosphamide at the first and second day after birth and were challenged intraperitoneally with S gallinarum ($1{\times}10^7CFU/m{\ell}$) on day 6. Group 1, Immunosuppresed and Challenged group, was treated with cyclophaiphamide and challenged with S gallinarum; group 2, Immunosuppressed group, was only treated with cyclophsphamide; group 3, Challenged group, was only challenged with S gallinarum; group 4, Control group. In each group, the localization of lymphocytes of the lymphoid organs and intestine was immunohistochemically compared using a variety of monoclonal antiboies ($CD4^+$, $CD8^+$, and B lymphocyte). Also, S gallinarum were assessed by Maccallum-Goodpasture stain and immunohistochemical analysis in the paraffin-embedded intestinal tissues. In S gallinarum challenged chickens, $CD4^+$ and $CD8^+$ lymphocytes of the intestinal organs such as duodenum, jejunum, ileum and colon were increased. However, in cyclophophamide treated chickens, $CD4^+$ and $CD8^+$ lymphocytes and especially B lymphocytes of the lymphoid organs such as thymus, spleen, and bursa of Fabricius were dramatically decreased. These results suggest that cyclophsophamide is an immunosuppressive agent that especially causes depletion of B lymphocytes, suppress humoral immunity and eventually suppresses avian immune responses. Its protection against S gallinarum infection is mainly dependent on both cell-mediated mechanism and the humoral immune response.
Infectious bursal disease (IBD) caused by infectious bursal disease virus (IBDV) is a highly contagious viral disease in chicken. It causes heavy economic loss by immune suppression and high mortality. The IBDV, designated Avibirnavirus in the Family Birnaviridae, has a double-stranded RNA genome formed by two segments, segment A and segment B. Segment A encodes a 108 KDa polypeptide that is self-cleaved to produce pVP2, VP3 and VP4, and later pVP2 is cleaved to VP2. The VP2 contains the antigenic regions responsible for elicitation of neutralizing antibodies and VP3 is a major immunogenic protein of IBDV. In this study, monoclonal antibodies (MAbs) specific for IBDV were produced and characterized. All 15 MAbs were specific for IBDV and did not react with other viruses used in this study. The protein specificity of MAbs was determined by comparing the reactivity patterns of each MAb with IBDV VP2 and VP234 recombinant baculoviruses and Western blot analysis. As a result, 7 MAbs (1F5, 2C8, 2F4, 3C7, 4C3, 6F11, 6G5) and 5 MAbs (2A4, 2G2, 3F5, 3G2, 4F10) were specific for VP2 and VP3, respectively. The protein specificity of 3 MAbs (2B8, 3F7, 3F8) were not determined. Five (2C8, 2F4, 4C3, 6F11, 6G5) of the VP2-specific MAbs had a neutralizing activity against IBDV. Some MAbs reacted with IBDV-infected bursa of Fabricius by indirect fluorescence antibody (IFA) and immunohistochemistry (IHC) assay. The MAbs produced in this study would be used for diagnostic reagents for the detection of IBDV infection.
This experiment was conducted to study the effects of medicinal plant extract on performance in broiler chickens. A total of eight hundred forty, 1-d-old broiler chicks(Ross) were randomly allocated to 7 treatments with 4 replicates of 30 birds per replicate. The treatments were negative control (NC, diets without antibiotics), positive control (PC, diets with-containing antibiotics), Schisandrae fructus (T1), Scutellariae radix (T2), Camellia sinensis (T3), Artemisia capillaris (T4) and mixed medicinal plants (T5) extracts added to drinking water. The final body weight, body weight gain and feed conversion rate were significantly improved in all treatments compared to NC (P<0.05). No significant differences were observed in the relative weights of liver, spleen, pancreas, bursa of Fabricius and intestinal tract among the groups. The relative lengths of duodenum, jejunum and ileum were significantly decreased in PC compared with other groups (P<0.05). No significant differences were observed in the cecal microflora. The contents of triacylglycerol (TG), blood urea nitrogen (BUN), glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) in blood serum were not significantly influenced. The concentration of cholesterol in blood serum was significantly decreased in all treated groups as compared to those of the control (P<0.05). There was no difference in the components of leukocytes and erythrocytes among the groups.
Low pathogenic avian influenza (LPAI; H9N2) and Newcastle disease (ND) are economically important poultry diseases in Korea. In this study, we investigated pathological features and virus distribution in the lymphoid tissues of chicks experimentally infected with H9N2 and/or ND virus. Six-weeks-old SPF chickens were divided into 4 groups, Control (C), H9N2 (E1), NDV (E2), and H9N2+NDV (E3). E1 group was challenged with 0.1 ml A/Kr/Ck/01310/01 (H9N2) $10^{5.6}$$EID_{50}$ intranasally, E2 group was challenged with 0.5 ml KJW (NDV) $10^{5.0}{\sim}10^{6.0}$$ELD_{50}$ intramuscularly, and E3 group was challenged with H9N2, followed 7 days later by NDV. In histopathological examination, E1 group showed depletion and necrosis in bursa of Fabricius, thymus, cecal tonsil, and spleen, whereas E2 and E3 groups were noted severe lymphocyte depletion and necrosis with destruction of lymphoid organs structures. In TUNEL assay, apoptotic bodies were detected in lymphoid organs of all experimental groups, which was most severe in E3 group. H9N2 and ND viruses were predominantly detected in cecal tonsil of E1, E2, and E3 groups by PCR and immunohistochemistry (ICH). In conclusion, co-infection of H9N2 with NDV caused severe pathologic lesions and apoptosis in lymphoid tissues compared to single infections.
가시오갈피와 두충은 강력한 항산화 작용을 가진 약용 식물로 알려져 있다. 본 연구에서는 상업용 브로일러를 대상으로 가시오갈피 잎과 두충 잎의 첨가 급여(0.5% 및 1%)가 개체의 생산 능력, 장기 무게, 혈장 생화학적 성분 및 텔로미어 함량에 미치는 영향을 알아보고자 하였다. 시험 전체 기간($4{\sim}35$일) 동안 체중, 증체량 및 사료 섭취량에서는 처리구 간 유의적 차이를 보이지 않았으나, 사료 요구율에 있어서는 가시오갈피 첨가 급여구가 대조구 및 두충 첨가구에 비해 다소 저조한 결과를 나타내었다. 장기 무게에 있어서 간, 비장 및 흉선의 무게는 처리구 간 차이가 없었으나, F낭(bursa of Fabricius)의 무게는 가시오갈피 및 두충 첨가 급여가 대조구에 비해 유의적으로 높게 나타났다. 혈액 생화적 성분 분석에서는 혈장 내 albumin, AST, ALT 및 total protein은 대조구와 처리구 간에 거의 비슷한 수치를 나타내었으나 triglycerides, cholesterol 및 gluocose 수준은 생리활성 물질 첨가 급여구가 대조구에 비해 높게 나타났다. 가시오갈피 및 두충 급여에 따른 각 조직 별 텔로미어 함량 분석에 있어 백혈구 내 텔로미어 함유율은 모든 첨가 급여구가 대조구에 비해 현저히 높은 함량을 보이나 간, 심장, 정소 조직 간에는 처리구별 차이가 없었다. 이상의 결과로부터 브로일러에 가시오갈피와 두충의 첨가 급여는 생산의 능력 저하 없이 개체의 면역 활성을 증진시키고, 텔로미어 길이의 감축을 상대적으로 완화시키는 것으로 판단된다.
본 연구는 닭에서 사육밀도[(대조군(SSD) $495cm^2$/수 vs. 고밀도사육군(HSD) $245cm^2$/수] 및 LPS 투여(5 mg/kg BW)가 다른 각종 생리적 스트레스 지표에 미치는 영향을 조사하기 위해 실시되었다. 대조군, HSD군 및 LPS군(5 mg/kg BW)에서 체중 및 장기 무게를 조사한 결과, SSD군과 HSD군간 비교 시 체중은 고밀도사육 시 현저히(p<0.05) 감소하였으나, 간, 비장 및 흉선 무게는 차이가 없었다. LPS 투여군에서는 대조군에 비해 흉선과 F-낭 무게가 유의적(p<0.05)으로 증가하였다. 혈액 생화학적 성분에서 AST, ALT 및 BUN 수준은 SSD군과 HSD군 간에는 차이가 없었지만, LPS군에서 증가되었다(p<0.05). 혈중 Ca, P, CK, uric acid 수준 모두 LPS군에서유의하게(p<0.05) 높았다. 혈중 corticosterone 수준은 LPS군에서 SSD군과 HSD군에 비해 유의적(p<0.05)으로 증가되었으나, SSD와 HSD군 간에는 차이가 없었다. 친염증 사이토카인 mRNA 발현을 조사한 결과, 간 조직의 $IL-1{\beta}$, IL-6 및 iNOS mRNA 발현은 LPS군에서 SSD군에 비해 증가되었으나(p<0.05), SSD군과 HSD군 간에는 차이가 없었다. 흉선 $IL-1{\beta}$, IL-6, IL-18 mRNA 유전자 역시 LPS군에서 다른 군들에 비해 유의하게(p<0.05) 증가되었다. HSD군에서 흉선의 $IL-1{\beta}$는 SSD군에 비해 현저히(p<0.05) 증가되었다. F-낭의 $IL-1{\beta}$ 발현은 LPS군에서 SSD군에 비해 증가되었으나(p<0.05), 다른 사이토카인은 차이가 없었다. 이상의 결과, 고밀도사육에 따른 만성스트레스 요인은 닭의 환경적응성에 따라 생산성을 제외한 다른 지표들의 변화는 적었으나, LPS 투여는 혈액생화학성분 및 친염증 사이토카인 mRNA 발현을 현저하게 변화시켰다.
