Modification of Sweet Potato $\beta$-Amylase with Periodate-Oxidized Soluble Starch (과요소산 산화 전분에 의한 고구마 $\beta$-아밀라제의 수식)

  • Sweet potato $\beta$-amylase is a tetrameric enzyme consisting of four identical polypeptide chains with a molecular weight of 5.6$\times$104, though most of the other $\beta$-amylases are monomeric enzymes. But, the relationship between subunit structure and catalytic function of the enzyme is not known. This study was done to know what the function of the subunit structure of the enzyme is. We obtained the monomer from the enzyme by the treatment of SDS, alkali pH buffer and urea. But the monomer had not activity. We tried to prepare the active monomer from the enzyme by the modification with periodate-oxidized soluble starch , In the result, we succeeded in isolating an active monomer as an oxidized soluble starch-conjugated form The active monomer had 57% of the original activity, 13.2% of the sugar and the molecular weight was estimated to be 5.4$\times$104. This results suggest that the tetrameric form of the enzyme is a most stable one and exists in nature, and the subunit structure of the enzyme Plays an important role in stabilization but not catalytic function.

Effects of Kammaegdaejotang on the hemodynamic function in the isolated perfused rat heart (감맥대조탕(甘麥大棗湯)이 적출 흰쥐 심장의 혈역학적(血力學的) 기능(機能)에 미치는 영향(影響))

  • Objectives : In order to verify the cardiovascular hemodynamic function of Kammaegdaejo-tang, the experiment was performed in the rats. Methods : Twelve hearts removed from male Sparague-Dawley rats weighing between 250g and 300g were perfused by the Langendorff technique with modified 37 Krebs-Henseleit's buffer solution at a constant perfusion pressure. They were randomly assigned to one of two groups, supplied with either normal saline or Kammaegdaejotang administration. Heart rate, left ventricular pressure, +dp/dt maximum, -dp/dt maximum, and -dp/dt/ +dp/dt ratio were evaluated at baseline after the administration of either normal saline or Kammaegdaejotang. Results : Kammaegdaejotang made the heart rate increasing significantly (p<0.05). Kammaegdaejotang did not effectively work on left ventricular pressure of the isolated heart(p=0.11, no significance). The significant effects of Kammaegdaejotang were observed on +dp/dt max and -dp/dt max(p<0.05). Kammaegdaejotang did not effectively work on -dp/dt/ +dp/dt ratio(p=0.24, no significance).

Jitter Tolerances in Digital Transmission Equipment (디지틀 전송 장치의 지터 허용치)

  • In the digital transmission equipment, the input jitter tolerance is a function of input timing recovery circuit characteristics. Especially, in the asynchronous multiplexers, it is also a function of the frame format, the buffer sizes in the synchronizer and desynchronizer, the PLL transfer function, and operating range of VCO in PLL In this paper, a new algorithm for calculating the jitter tolerance of the saynchronous digital transmission equipment is presented. With the new algorithm, we analyzed how the above factors limit the jitter tolerance in the equipment. We also measured the input jitter tolerance for a 45M-140M multiplexing equipment, whose results show the same trend with calculated tolerance.

A Study on Development of Evaluation Method on Riverine Ecobelt (수변 생태벨트 평가방법 개발에 관한 연구)

  • This study aims to develop the diagnostic evaluation method of the riverine ecobelt for construction, conservation, and maintenance of the riverine ecobelt. The value indices in the proposed evaluation method are composed of total 5 fields and 19 elements. The 5 fields are flood control, environmental function, growth of plants, ecobelt function, and restoration potential. Flood control field is composed of total 3 elements such as length, width, and density of green area. Environmental function field is composed of 4 elements such as park use, landscape boundary and edge, microclimate control, non-point pollution control. Growth of plants field is composed of 6 elements such as species composition, forest height, stratum structure, vine plants, plant vitality, and succession of plants. Ecobelt function field is composed of 4 elements such as longitudinal connectivity, lateral connectivity, in-stream forest or habitat, roads on bank top. Restoration potential field is composed of 2 elements such as landform and land use of the immediate vicinity. The score system ranging 1~4 was adopted. The weighting parameters of elements were unified with each other. The final grade system ranging 1~5(1: very good~5: very bad) was adopted, and the final grade was evaluated by the mean values of each field. According to the test application of the diagnostic evaluation method of the riverine ecobelt, the final grades showed effectively the real condition of each site.

Study on Polymerization Condition of Water-based Acrylic Adhesion (수분산성 아크릴계 점착제 중합 조건에 관한 연구)

  • In this study, emulsion polymerizations for synthesizing acrylic pressure-sensitive adhesive(PSA) were carried out using 2-ethylhexyl acrylate(2-EHA), n-butyl acrylate(n-BA), methyl metacrylate(MMA) as fundamental monomers and acrylic acid(AAc) as a functional monomer in the presence of anionic SLS (sodium lauryl sulfate). To obtain the optimized synthetic condition in the polymerization, we analyzed the polymerization variables such as the effect of surfactant concentration and hydrophilic lipophilic values(HLB). At the same time, the final adhesive properties were also analyzed by the function of the initiator concentration and buffer concentration. In the results, the most stable emulsion was obtained at the surfactant concentrations between 3 and 5 wt%. It was also determined the effect of HLB value of nonionic surfactant and the initiator concentrations on the gel content. Stable emulsion is obtained using the surfactant having HLB value of 12.3. The rate of emulsion polymerization was increased at the initiator concentration greater than 1 wt%, but the stability of the emulsion was decreased. Finally, the effect of the buffer concentrations on the pH and the conversion of the acrylic emulsion product were experimentally measured. At the sodium bicarbonate concentration above 0.4 wt%, the buffer infulence was apparent. The buffer effect was fully acceptable at the concentrations between 0.6 and 0.8 wt% regardless of the monomer composition.

A Study of the Functional Improvement for Comfortable Expressway Service Areas - Metropolitan Expressway Service Areas - (고속도로 휴게소 환경개선을 위한 기능 증진 방안 연구 - 수도권 휴게소를 중심으로 -)

  • This study was carried out to suggest improvement plans for problems with the landscape areas of highway service areas. The survey factors were land use, placement of green area and land coverage in terms of spatial and environmental property. Noise and topographic structure in detached green areas were surveyed in terms of impact factor. The status of the planting area around each service area was analysed in regards to planting structure and planting landscape. As a result, the mean of rations of land use were 81.6% building and 18.4% green areas, which were composed of 5.4% landscape area, 9.1% buffer area, etc. Planting areas as usable space accounted for only 0.7%, and the result of noise measurement was different according to the distribution of highway and service area and type of detached green area. The mean of ratio of planting area was 18.4%, which consists of 6.7% landscape planting, 3.4% other green area, 4.6% buffer area, 2.0% buffer and landscape area, 1.1% shade planting and 0.5% landscape and shade planting. Most planting areas aim at landscape appreciation and areas in which visitors can relax and gain recreation were insufficient. The planting structures of service areas were 52.2% canopy layer, 11.8% shrubs and 9.4% canopy and shrub layers, and most of the planting areas were a single layer of green area. Multi-layered planting structures in the landscape and buffer areas were required and a shade planting area was needed to improve amenities as planting canopy layered trees. This study suggested improvement methods for based environments, spatial function and planting function in landscape areas.

Dynamic VBR traffic characterization for video service in ATM network (ATM 망에서 비디오 서비스를 위한 동적 VBR 트래픽 특성화)

  • This paper is focused on the traffic characterization for the efficient transmission of the VBR video source in the ATM network. For the traffic characterization, low traffic monitoring technique is applied and the dynamic VBR characterization method is suggested to satisfy the delay requirement. The dynamic VBR method uses the token bucket algorithm buffering though Cumulative Constraint Function. According to the Cumulative Constraint Function, the packet initially started transferring at the peak rate and the token bucket provided proper amount of buffer for traffic after a certain period of monitoring. It also reduced the network resource bandwidth through renewal of the cumulative frame and changed the rate from the previous frame information. It requires only small amount of monitoring and causes little overhead. In addition, it lowered the complexity of Deterministic Constraint Function to 0(n) and mapped the token rate and token depth to the token bucket. This study shows less network resource consumed than the previous method, comparing and analyzing the result of simulations.

  • This paper categorized the functions of and investigated the characteristics of planting in Pyeonghwa Park near the Seoul World Cup Stadium as a preliminary study for improving tree planting in the park. The functions of planting were deduced based on related theoretical studies and an analysis of the intention of design and environmental factors. The characteristics of planting were analyzed after drawing the distribution and stratification of dominant planting species and setting samples by function. The functions of planting were classified into natural landscape planting, buffer planting, and shade planting. For dominant planting species, pine trees and elm-like trees have been widely observed while various species in addition to pine trees in natural landscape planting areas have been introduced in terms of the function of planting. These trees that were randomly planted before the park was developed have been obstacles in the formation of natural landscapes. For buffer planting and topographical correction, tall trees were planted. However, it seems that comprehensive complementary measures should be taken on the under-story planting.

A new method to predict swelling pressure of compacted bentonites based on diffuse double layer theory

  • Compacted bentonites were chosen as the backfill material and buffer in high level nuclear waste disposal due to its high swelling pressure, high ion adsorption capacity and low permeability. It is essential to estimate the swelling pressure in design and considering the safety of the nuclear repositories. The swelling pressure model of expansive clay colloids was developed based on Gouy-Chapman diffuse double layer theory. However, the diffuse double layer model is effective in predicting low compaction dry density (low swelling pressure) for certain bentonites, and invalidation in simulating high compaction dry density (high swelling pressure). In this paper, the new relationship between nondimensional midplane potential function, u, and nondimensional distance function, Kd, were established based on the Gouy-Chapman theory by considering the variation of void ratio. The new developed model was constructed based on the published literature data of compacted Na-bentonite (MX80) and Ca-bentonite (FoCa) for sodium and calcium bentonite respectively. The proposed models were applied to re-compute swelling pressure of other compacted Na-bentonites (Kunigel-V1, Voclay, Neokunibond and GMZ) and Ca-bentonites (FEBEX, Bavaria bentonite, Bentonite S-2, Montigel bentonite) based on the reported experimental data. Results show that the predicted swelling pressure has a good agreement with the experimental swelling pressure in all cases.

Effects of Myocardial Protection of Verapamil in Serum: Under Langendorff Apparatus in Ischemic Arrest Heart (Verapamil [anticalcium agent]의 심근 보호작용Langendorff씨 장치하의 심근 보호 작용)

  • This study was evaluated the metabolic, physiologic and histologic effects of myocardial protection of verapamil[isoveratril]on isolated rat hearts to 90 minutes of ischemic arrest. Heart was perfused with a modified Kreb’s Henseleit bicarbonate buffer with glucose and arrested with retrograde coronary perfusion by glucose insulin[GI], potassium and verapamil. Mean aortic systolic pressure, heart rate, coronary flows were measured and morphologic changes were examined during working heart perfusion. Perfusion and arrest were controlled four groups subjected 60 isolated rat hearts. Four groups hearts reperfused during 40 minutes after 90 minutes global ischemia for physiologic recovery. 15 hearts of four groups were assayed to histological morphologic changes. GI treated hearts recovered less than 28% of function and changed more than 80% of mitochondria of control group. Verapamil hearts[0.2, 0.1 gm/kg] recovered more than 88% of function and permitted the maintenance of continuous cellular level of Serum Glutamic Oxalaxetate Transaminase[SGOT], but declined 28% of Phosphate Kinase[CP], GI treated heart showed widespread evidence of extensive damage of mitochondria. The damage was that interstitial huge edema are present and there was contraction band formation within the swollen cells. The verapamil and potassium group were not found morphologic change compared with control group. Their functions were shown that metabolic and physiologic action of verapamil-group lasted 20 minutes longer than potassium group.
