A Comparison of three Enrichment Media for Isolating Salmonella (Salmonella균(菌) 분리용(分離用) 증균배지(增菌培地)의 비교실험(比較實驗))

  • The practical significance of using a selective enrichment procedure for detecion and enumeration of salmonella is well recognized. There are still various selective enrichment media has been communly used. Early years selenite broth was recomnended as an enrichment media for the isolating of salmonella. Hajna introduced a modified tetrathionate broth and demonstrated the greater efficiency to compare with the previous enrichment media. Raj also described that the new medium called dulcitol selenite enrichment and has been found to be very satisfactory, especially general implication in food poisoning. Authors tried to compare these 3 enrichment media for isolating salmonella. 1. When salmonella strains were inoculated $1{\sim}10^6$ cells per tube to these 3 enrichment media, mostly similar results were obtained between selenite broth and DS broth. In these 2 enrichment broth were showed $10^7/ml-10^8/ml$ cells of all tested salmonella strains. But in the case of TT broth it was found that the growth was $10^3/ml{\sim}10^4/ml$ cells for tested strain. 2. When E. coli, Proteus, Citrobacter were inoculate $10{\sim}10^6$ cells per tube to these 3 enrichment media. It was suggested that DS broth was showed more inhibitory action than that of selenite broth. TT broth showed high inhibition to these 3 organisms tested. 3. It was generally known that the incubation time is influenced to the frequency of salmonella detection. For this tendency, DS broth and selenite broth were showed similar results within 24 hrs to 48hrs incubation to the test. But DS broth showed more inhibitory action to E. coli and Proteus than that of selenite broth. 4. When $1{\sim}10$ cells were inoculated(per tube) to these 3 enrichment media, DS broth was found to be more sensitive than that of selenite broth.

Comparison of Enrichment Media of Shigella sonnei (쉬겔라 증균배지의 성능 비교)

  • The object of this study was to compare the performance of commercial enrichment media used for Shigella spp. A total of four enrichment media, Gram negative (GN) broth, Shigella broth (SB), selenite-F (SF) broth, and selenite cystine (SC) broth, were tested. When S. sonnei was inoculated into each enrichment broth at 10 cfu/mL of concentration, the highest growth was observed in Shigella broth. Morganella spp., which was not differentiated in selective agar of Shigella spp. thus can be counted as false positive, did not grow in Shigella broth in enrichment step. When S. sonnei was artificially inoculated into pork, it was mostly recovered through an enrichment process with GN broth and SF broth. However, in the case of beef, S. sonnei was mostly recovered with GN broth but largely failed with Shigella broth. Therefore, enrichment media for Shigella spp. should be selected by considering the food matrix in order to increase the chance of isolating it from foods.

Determination of best enrichment media for growth of Salmonella injured from cold temperature during process and storage (저온저장으로 인해 손상된 살모넬라를 배양하기 위한 최적의 배지 선정에 관한 연구)

  • This purpose of this study was to determine the best enrichment medium for rejuvenating and recovering Salmonella placed in cold temperature prior to the employment of the gold biosensor combined with a light microscopic imaging system. A mixture of nalidixic-resistant Salmonella Typhimurium and Enteritidis were inoculated onto chicken (1,000 CFU/chicken). After cold injury at $4^{\circ}C$ for 24 hr, Salmonella on chicken was enriched for 6 hr with six non-selective media including buffered peptone water broth, lactose broth, brain heart infusion broth (BHI), universal pre-enrichment broth, nutrient broth, and tryptic soy broth, and five selective media including brilliant green broth (BG), rappaport-vassiliadis R10 broth, selenite cystine broth, selenite broth, and tetrathionate brilliant green broth (TBG) for the comparison of Salmonella growth. Various concentrations of Salmonella (10, 50, 100, 500, and 1,000 CFU/chicken) were then enriched for 6 hr in both BHI and BG media to select the best media. BHI was selected as the most effective non-selective enrichment medium, while BG was selected as the most effective selective enrichment medium. Finally, BHI medium was selected as the most efficient enrichment medium for Salmonella growth injured from cold temperature during processing or storage.

Comparative Studies on Streptomycin Producing Strains and Media (스트렙토마이신 생성균주들과 배지들에 대한 비교연구)

  • When various strains of Streptomyces griseus and S. galbus were examined for the ability on the production of streptomycin in tryptic soy(TS) broth, S. griseus ATCC 27001 was found to be the best. S. griseus ATCC 12475 and ATCC 23345 showed also good growth and favorable production of streptomycin. Examination of various complex media reported in fermentation literatures for the industrial production of streptomycin indicated that glucose-soybean media-sodium chloride (GSS) broth and K (Chucken) broth gave higher yields of streptomycin than others studied by us. Examination of the ingredients of media producing streptomycin in high yield indicated that some components in soybean activated the production of streptomycin. Addition of meat extract enhanced the yield of streptomycin but it could be substituted with distillers solubles without much effect on the yield. Addition of corn steep liquor decreased the production of streptomycin.

Evaluation of enrichment broth and selective media for the detection of non-O157 enterohemorrhagic Escherichia coli (Non-O157 장출혈성대장균 검출을 위한 증균배지 및 선택배지 성능 평가)

  • In this study, specific and rapid enrichment and growth conditions for the most important, classic non-O157 enterohemorrhagic Escherichia coli (EHEC) serogroups were assessed. Three enrichment broth types, namely, EC medium with MUG broth, BRILA broth, and mTSB broth with novobiocin, were compared to identify the optimum enrichment broth for EHEC isolation. Four kinds of selective media, namely, ENDO agar, Chromocult agar, TBX agar, and CHROMagar$^{TM}$ STEC medium, were compared to identify the optimum one for non-O157 EHEC isolation. The results suggested that incubation in EC medium with MUG broth at $42^{\circ}C$ for 20 h is optimum for the enrichment of non-O157 EHEC. TBX agar was identified to have the highest specificity among the tested media. Consequently, a combination of complementary selective media according to serotype must be considered for comprehensive isolation of specific EHEC.

  • In this study, we selected suitable broth media for mass production of Phytophthora capsici oospore, investigated oospore germination and secured $F_1$ progeny. Carrot broth and V8C broth were determined most effective for oospore formation by calculating and comparing oospore concentration produced from 8 different liquid media. Eleven strains were selected from P. capsici (CapA)/P. tropicalis (CapB) and 9 crosses were formed. Oospore progeny were produced, isolated and germinated from A1 and A2 combinations of P. capsici (CapA) with P tropicalis (CapB). This resulted in a total number of 129 $F_1$ isolates of P. capsici/P. tropicalis with a 0.64-4.0% (mean 1.85%) oospore germination.

Patulin Producing Capacity in Broth Culture Media of Penicillium crustosum Isolated from Korean Apple (국내산 사과로부터 분리된 Penicillium crustosum의 액상배지에서의 Patulin 생성능 평가)

  • The patulin producing capicity of Penicillium crustosum, an isolate from Korean apple, in various broth culture media, was investigated, and compared with patulin production by the standard strain P. griseofulvum(ATCC 46037). The maximal patulin production capacity of the P. griseofulvum ATCC 46037 was 2,029-2,829 ppm in 5-GYEP, SY and MEB broth media. The patulin-producing capacity of the isolated fungus(P. crustosum) attained 2,794 ppm in a 5-GYEP broth medium, but was only 324 and 11 ppm in SY and MEB media, respectively. There were no significant correlations between mycelial growth levels and patulin-producing ability in either P. crustosum or P. griseofulvum. The patulin production of P. griseofulvum was induced in the wide pH range of pH 3.0-11.0, while that of P. crustosum was induced in the acidic pH range pH 3.0-5.0. Patulin production levels were dependent on the carbon sources in the media and maximal patulin production by P. griseofulvum and P. crustosum was observed in media containing glycerol and fructose, respectively.

  • Various methods for the isolation of salmonellae were compared by examining mesenteric lymph nodes and faeces of 395 pigs at slaughter. The conclusions were as follows. (i) Tetrathionate broth was significantly superior to selenite broth for the examination of mesenteric lymph nodes. (ii) There was no significant difference between tetrathionate broth and selenite broth for the examination of faeces. The use of both tetrathionate broth and selenite broth as enrichment media for faeces produced far better results than the use of any single enrichment medium. (iii) Brilliant green agar was significantly superior to SS agar for the examination of mesenteric lymph nodes. (iv) There was no significant difference between brilliant green agar and SS agar when faeces were examined. The use of both media produced far better results than the use of either singly. (v) Brilliant green MacConkey broth was much inferior to tetrathionate broth as an enrichment broth. Direct culture of faeces on brilliant green MacConkey agar yielded less isolates than prior enrichment in tetrathionate or selenite broth. (vi) The optimum incubation period of enrichment was 24 hours for faeces and 48 hours for mesenteric lymph nodes when either tetrathionate broth or selenite broth was used as enrichment media.

A Study on Industrial Media for Production of Lactic acid in Batch and Continuous Fermentations (회분식 및 연속배양에 있어서 고농도 젖산의 생산을 위한 공업용 배지연구)

  • We have investigated industrial media for lactic acid fermentation to reduce the cost of nitrogen sources. Corn steep liquor (CSL) was successfully used at 5% (v/v) in batch fermentations. Use of soluble CSL improved the productivity about 20% with an advantage of clearer fermentation broth. Yeast extract-complemented CSL improved the productivity about 20% with an advantage of clearer fermentation broth. Yeast extract-complemented CSL media further increased the increased the productivity. It was found that 3.1 g/L yeast extract and 5% CSL could be an effective substitute for 15 g/L yeast extract in 10% glucose medium. Brewing yeast was also used as a sole nitrogen source equivalent to 5% CSL. A continuous culture coupled with cell-recycle by microfiltration at the dilution rate of 0.05-0.065 h-1 led to the highest lactic acid productivity. Lactic acid was recovered by electrodialysis from the cell free broth. Depleted cell free broth supplemented with 5-10 g/L of yeast extract performed reasonably in batch and continuous cultures. Reuse of the fermentation broth may reduce the cost of raw materials as well as minimize the fermentation wastes.

Comparison of Three Antibiotic Susceptibility Tests for Viridans Group Streptococci

  • Oral viridans streptococci are recognized as one of the etiological agents of a variety of infectious diseases such as dental caries and infective endocarditis. Although antimicrobial susceptibility tests for these fastidious bacterial species are now established and standardized, a comparison between the broth microdilution and broth macrodilution tests has not previously been performed. This comparison was performed in the present study using the tests adopted by the Clinical and Laboratory Standards Institute (CLSI) and seven clinical isolates of oral viridans streptococcal strains. A modified broth macrodilution susceptibility test method was also included in this analysis, in which the media was not supplemented with horse blood. The susceptibility interpretation category agreements were measured at 83% (broth microdilution versus broth macrodilution) and 71% (broth microdilution versus modified broth macrodilution). The interpretation category agreement between the broth macrodilution and modified broth macrodilution tests was also 83%. These data indicate that the interpretation of antibiotic susceptibility test results for oral viridans streptococci are influenced by the methods used.
