페레니얼 라이그라스의 품종에 따른 성숙종자 유래의 캘러스로부터 식물체 재분화 (Plant Regeneration Capacity of Calluses Derived from Mature Seed of Perennial Ryegrass Cultivars)

  • 페레니얼 라이그라스의 5가지 품종을 이용하여 성숙종자로부터 배발생 캘러스 유도 및 캘러스로부터의 식물체 재분화 효율을 조사하였다. 성숙종자로부터 4주 동안 배양한 캘러스 유도율은 'Accent', 'Topgun', 'Renenge GLX', 'Bison', 'Tetrellite' 순으로 높은 캘러스 유도율을 보였고 암상태에서 배양한 캘러스가 광상태에서 배양한 것보다 7~9% 정도 높은 효율을 나타내었다. 식물체로의 재분화에 적합한 배지는 MS 배지보다 N6 배지에서 식물체 재분화율이 우수하였다. 품종간의 재분화율은 'Bison', 'Accent', 'Topgun', 'Renenge GLX', 'Tetrellite' 순으로 높게 나타났으며 광상태에서 유식물로 분화되는 개체가 1~7% 정도 많이 관찰되었다. 본 연구를 통하여 밝혀진 캘러스 유도율과 식물체 재분화율이 우수한 품종은 분자육종을 통한 신품종 페레니얼 라이그라스의 개발에 유용하게 이용될 수 있을 것이다.

  • A combined technology of transvaginal ovum pick-up(OPU) system with in vitro-oocyte manipulation technique can be used for improving reproductive efficiency in the cattle. The objective of this study was to establish a newly-conceived breeding program using OPU in the pregnant cows. The OPU trial was performed in pregnant cows every 10 days from 40 through 90 days of artificial insemination (Al), and number of follicles in ovary, number of retrieved oocytes and embryo development following in vitro-fertilization, were evaluated. Reduced number of follicles in the ovaries of pregnant cows was firstly detected from 70 days after A' and a significant (P<0.05) decrease in the follicle number (5.4 follicles /donor) was found at 90 days than at 40, 50, 60 and 80 days after Al (8.0~9.2). A similar pattern was also observed in the number of oocytes retrieved by OPU apparatus during experimental period. When retrieved oocytes were matured and inseminated in vitro with frozen bull semen, development of the oocytes to the blastocyst stage was not significantly affected by the retrieval time. Four embryos (morula or blastocyst stage) derived from oocytes retrieved from pregnant cows were nonsurgically transferred to four recipient cows on day 7 of estrus cycle. For the first time in Korea, three of four transferred embryos developed to live calves with normal physiological parameters. In conclusion, an effective breeding program employing pregnant cow can be developed by use of OPU trial and in vitro culture techniques of oocytes ; OPU system could be repeated in pregnant cows with no risk of abortion and viable offsprings were borne after transfer to the recipients.

The Membrane-Bound Protein, MoAfo1, Is Involved in Sensing Diverse Signals from Different Surfaces in the Rice Blast Fungus

  • To establish an infection, fungal pathogens must recognize diverse signals from host surfaces. The rice blast fungus, Magnaporthe oryzae, is one of the best models studying host-pathogen interactions. This fungus recognizes physical or chemical signals from the host surfaces and initiates the development of an infection structure called appressorium. Here, we found that protein MoAfo1(appressorium formation, MGG_10422) was involved in sensing signal molecules such as cutin monomers and long chain primary alcohols required for appressorium formation. The knockout mutant (ΔMoafo1) formed a few abnormal appressoria on the onion and rice sheath surfaces. However, it produced normal appressoria on the surface of rice leaves. MoAfo1 localized to the membranes of the cytoplasm and vacuole-like organelles in conidia and appressoria. Additionally, the ΔMoafo1 mutant showed defects in appressorium morphology, appressorium penetration, invasive growth, and pathogenicity. These multiple defects might be partially due to failure to respond properly to oxidative stress. These findings broaden our understanding of the fungal mechanisms at play in the recognition of the host surface during rice blast infection.

사육 밀도와 개체 크기에 따른 북방전복(Haliotis discus hannai) 치패의 성장과 이동 특성 (Analysis of the Effects of Stocking Density and Individual Size on the Growth and Movement Characteristics of Juvenile Abalone Haliotis discus hannai)

  • Behavior serves as an important indicator of the effects of environmental stressors on organisms. In this study, one-year-old abalone Haliotis discus hannai (15.1±0.8 mm) were used and each 0.34 m2 tank was stocked with 100, 150, 200, 250, and 300 juvenile abalone. After 328 days, we found that the low stocking density treatment showed significantly higher growth, whereas the high stocking density treatment showed significantly lower growth. In addition, abalone showed a lower rate of movement, distance moved, and velocity at higher stocking densities, but only velocity differed significantly. In contrast, In the Supplied the feed tanks significant difference between the moving distance and velocity. When the two groups of different sizes were mixed in tank, the rate of movement, distance moned, and velocity showed significant differences between differently-sized indivisuals. Our study indicates that breeding abalone at high densities has a negative effect on their growth and survival, which is thought to primarily be caused by spatial competition, and secondarily by differences in body size, ultimately affecting growth and productivity.

Genetic (Co)variance Components for Body Weight and Body Measurements in Makooei Sheep

  • The aim of this paper was to estimate genetic parameters for body weight and five body measurements for an experimental population of Iranian Makooei sheep maintained at the Makooei Sheep Breeding Station at Makoo, Iran. To do this, yearling live weight (YW), and five body measurements, i.e., body length (BL), heart girth (HG), height at withers (HW), height at back (HB) and scrotal circumference (SC), were analyzed in a multi-trait animal model using the DXMUX program of DFREML software package. Heritability estimates were $0.22{\pm}0.08$, $0.11{\pm}0.06$, $0.21{\pm}0.07$, $0.17{\pm}0.06$, $0.17{\pm}0.06$ and $0.32{\pm}0.10$ for YW, BL, HG, HW, HB and SC, respectively. These estimates indicate that selection in Makooei sheep would generate moderate genetic progress in body weight and body measurements. Scrotal circumference, as an indicator of reproductive potential, exhibited the highest heritability. This trait, therefore, could successfully be used to increase productivity of males and, indirectly, female fertility. Genetic correlations between traits studied were all positive and ranged from 0.15 (YW/HB) to 0.99 (HW/HB). Phenotypic correlations were also positive and ranged from moderate (0.32, HW/SC) to high (0.94, HW/HB). Positive genetic and phenotypic correlations indicate that improvement in body measurements both at the genetic and phenotypic levels is expected through selection on body weight and vice versa.

RAPD법을 이용한 고구마 품종간 유연관계 평가 (Evaluation of Genetic Relationship among Sweetpotato Cultivars Using Randomly Amplified Polymorphic DNA (RAPD) Analysis)

  • 본 실험은 RAPD (random amplified polymorphic DNA)를 이용하여 국내에서 육성된 13개 품종의 고구마 (Ipomoea batatas)를 대상으로 유연관계분류 및 품종구분 가능성을 탐색하였다. RAPD를 이용하여 고구마 품종을 비가중산술법(UPGMA)으로 3개의 그룹표로 분류할 수 있었는데 그룹 I은 '충승 100호'로, 그룹 II는 '은미', '생미', '수원147호'와 '율미', 그룹 III는 '홍미', '진미', '관동95', '선미', '원미', '신율미', '증미', '풍미'로 나뉘어졌다. RAPD를 이용한 분류 결과는 대체로 육성모부본의 유전자형과 일치함을 나타내고 있고, 상이한 점은 영양계의 변이에 의한 것으로 추측된다, 앞으로 이러한 marker system을 이용하여 육종시 조기에 원하는 형질을 갖는 계통을 선별할 수 있을 것이며 이에 따라 다양한 고구마 품종의 육종프로그램과 품종판별에 유용하게 이용될 수 있을 것으로 사료된다.

고품질 무핵 포도 품종 '흑이슬' 육성 (Breeding of a Seedless Table Grape Cultivar 'Heukisul' (Vitis sp.) with High Quality)

  • 무핵 포도 품종을 육성하고자 2000년에 'Kyoho'와 'Thompson seedless' 교잡으로부터 '흑이슬'이 육성되었으며, 2004년도에 1차 선발되었다. 2004년부터 2005년까지 두 곳에서 KTS014의 이름으로 지역적응시험을 실시하여 2006년에 최종 선발하였다. '흑이슬'은 'King Dela'에 비하여 화방의 과실 착과가 우수하여 화진현상이 낮았다. 발아기는 4월 23일경, 만개기는 6월 9일경이며 수확기는 8월 29일경으로 중생종 품종으로 생각된다. 평균 과립무게는 4.0g이고 당도는 $21.9^{\circ}Brix$이다. 과피색은 과분이 많은 자흑색이고 과육은 단단하다. 착립밀도가 높아 알솎기가 필요 없다. 또한 열과율이 대단히 낮은 특성을 가지고 있다.

Construction of an Integrated Pepper Map Using RFLP, SSR, CAPS, AFLP, WRKY, rRAMP, and BAC End Sequences

  • Map-based cloning to find genes of interest, marker-assisted selection (MAS), and marker-assisted breeding (MAB) all require good genetic maps with high reproducible markers. For map construction as well as chromosome assignment, development of single copy PCR-based markers and map integration process are necessary. In this study, the 132 markers (57 STS from BAC-end sequences, 13 STS from RFLP, and 62 SSR) were newly developed as single copy type PCR-based markers. They were used together with 1830 markers previously developed in our lab to construct an integrated map with the Joinmap 3.0 program. This integrated map contained 169 SSR, 354 RFLP, 23 STS from BAC-end sequences, 6 STS from RFLP, 152 AFLP, 51 WRKY, and 99 rRAMP markers on 12 chromosomes. The integrated map contained four genetic maps of two interspecific (Capsicum annuum 'TF68' and C. chinense 'Habanero') and two intraspecific (C. annuum 'CM334' and C. annuum 'Chilsungcho') populations of peppers. This constructed integrated map consisted of 805 markers (map distance of 1858 cM) in interspecific populations and 745 markers (map distance of 1892 cM) in intraspecific populations. The used pepper STS were first developed from end sequences of BAC clones from Capsicum annuum 'CM334'. This integrated map will provide useful information for construction of future pepper genetic maps and for assignment of linkage groups to pepper chromosomes.

Genetic Analysis of Pod Dehiscence in Soybean

  • Pod dehiscence (PD), defined as the opening of pods along both the dorsal and ventral sutures, causes the seed to shatter in the field before harvesting and results in loss of seed yields. However, breeding for resistance to PD is difficult due to the complicated genetic behavior and environmental interaction. The objective of the present research was to analyze the genetic behavior of PD for improving the breeding efficiency of resistance to PD in soybean. PD after oven-drying the sampled pod at $40^{\circ}C$ for 24 hours was the most reliable to predict the degree of PD tested in the field. Keunolkong, a dehiscent parent, was crossed with non-dehiscent parents, Sinpaldalkong and Iksan 10. Using their $F_1\;and\;F_2$ seeds, PD was measured after oven drying the pod at $40^{\circ}C$ for 24 hours. The gene conferring PD behaved in different manners depending on the genetic populations. In the Keunolkong$\times$Sinpaldalkong population, PD seemed to be governed by single major recessive gene and minor genes, while several genes were probably involved in the resistance to pod dehiscence in the Keunolkong$\times$Iksan 10 population. Heritability for PD estimated in F2 population showed over $90\%$ in the two populations. High heritability of PD indicated that selection for resistance to PD should be effective in a breeding program. In addition, genetic mapping of quantitative locus (QTL) for PD in both populations may reveal that genes conferring PD are population-specific.

고온성 백색 양송이 신품종 '하담' 육성 (Breeding a new white button mushroom cultivar 'Hadam' to produce mushrooms at high temperature)

  • 기후변화에 대비하여 고온성 양송이 백색 품종을 육성한 결과는 다음과 같다. 모본은 KMCC00540, KMCC00591, KMCC00643을 선발하였고, ISSR과 SSR마커로 각각 동핵균주를 선발하여 교잡하였다. 선발 교잡주를 고온 20~25℃, 습도 80% 이상에서 3반복으로 재배하여 농업적 형질과 자실체 특성의 변이가 가장 적은 것은 Abs4-2016-121 계통이었다. 이 계통은 KMCC00000와 KMCC0000를 교잡한 계통이며, 경주와 부여 농가에서 현장실증시험결과 Abs4-2016-121 계통이 자실체 경도가 높으며 고온에서 갓개열이 지연되어 고온성 품종 '하담'으로 육성하였다.
