한우에서 모색관련 유전자 변이에 관한 연구 (Investigation of Coat Color Candidate Genes in Korean Cattle(Hanwoo))

  • 본 연구는 한우의 모색발현에 정확히 어떤 유전자가 어떤 유전기작에 의해 관여하고 있는가를 규명하고자 황갈색의 모색을 지닌 한우와 검정모색을 지닌 홀스타인과의 교배를 통해 만든 F2집단의 DNA를 이용하여, MC1R, ASIP 및 TYRP1 유전자형과 한우모색 발현양상을 연관분석 하였으며, 또 한우 집단내에서의 이들 유전자형 빈도를 조사하여 황갈색 한우모색 다양성과 후보유전자 변이의 연관성연구에 필요한 정보를 제공하고자 하였다. MC1R 유전자의 경우 황갈색을 지닌 3두의 유전자형은 모두 e/e 형으로 밝혀졌으며, 검정모색을 지니고 태어난 나머지 3두의 두 좌위에서의 유전자형은 ED/e임을 확인하였는데 황갈색과 검정모색의 비율이 1:1로 나온다는 것은 MC1R 단일유전자가 한우의 모색에 중요한 영향을 미치는 것으로 사료된다. MC1R 이외의 모색발현에 영향을 줄 수 있는 ASIP와 TYRP1 유전자들은 F2 집단에서 염기서열을 분석한 결과 이들 유전자들이 한우 황갈색모색에 주된 영향을 미치지 않는 것으로 나타났다. 하지만 TYRP1 유전자에서 발견된 329번 (Glu329Lys) 아미노산 변이는 TYRP1 단백질의 구조와 화학적 성질에 영향을 줄 수 있는 것으로 사료되어 한우집단에서 황갈색바탕의 모색변이에 영향을 줄 수 있는지에 대한 추가적인 연구가 이루어져야 할 것이다.

DNA 분석법에 의한 한우고기 판별 (Identification of Hanwoo Meat by DNA Analysis)

  • 본 연구는 개발된 품종 특이적인 DNA 표지인자를 이용하여 한우와 비한우의 고기를 판별하고자 하였다. 한우와 유럽종 고기소들의 유전적 차이를 Random amplified polymorphic DNA(RAPD) 분석으로 조사한 바, 홀스타인, 헤어포드, 에버딘엥거스, 리모진 및 시멘탈과 같은 한우가 아닌 유럽 종들의 RAPD 패턴들은 동일하였으나, 한우는 이들과 다른 패턴을 보였다. 한우는 유럽종들에서 모두 검출된 특정의 밴드가 발견되지 않았다. 1.4Kbp인 밴드는 한우와 수입소 고기를 판별해 낼 수 있는 유용한 DNA 표지인자로 인정되었다. 실제로, 한우 673 마리, 홀스타인 141 마리의 혈액시료 및 수입육 115의 고기시료를 가지고 시행한 실험에서 그 DNA 표지인자는 비한우에서는 256두 중 245두에서 검출되었으나, 한우에서는 673두 중 644두의 시료에서 검출되지 않아서, 비한우의 검출율은 95%, 한우의 비검출율은 96%를 보였다. 이러한 결과는 그 DNA 표지인자를 이용하여 한우와 비한우 고기를 판별할 수 있음을 시사하였다. 그러나 두 품종 사이의 교잡종은 한우나 비한우의 RAPD 패턴 중에서 한 가지를 무원칙하게 선택하여 나타내고 있으므로, 이러한 DNA 표지인자만으로서는 판별하기가 어려웠다.

Prediction of Retail Beef Yield Using Parameters Based on Korean Beef Carcass Grading Standards

  • Two sets of data on carcass traits and beef cut parameters were used to investigate the relationships between carcass and beef cut measurements, which can be used to make predictions of retail cut percentages. One set had a total of 1,141 measurements of Hanwoo cattle of three different sex origins, which were slaughtered in an abattoir located at the National Institute of Animal Science, RDA, Korea from 1996 to 2008. To develop prediction models for retail cut percentage with higher accuracies than the current model, another set consisting of a total of 13,389 records of carcass and beef cut traits were collected from 30 abattoirs and butcheries in Korea from 2008 to 2009. Bulls yielded heavier and leaner carcasses than steers. High correlation coefficients were estimated between amount of body fat and percent retail cut (-0.82) as well as between back fat thickness (BF) and percent retail cut (-0.62). The amount of retail cut, however, was highly correlated with body weight before slaughter (BW, 0.95) or with cold carcass weight (CWT, 0.94). Relationships between percent retail cut and measurable beef yield traits, BF, loin eye area (LEA) or CWT varied by sex class, which must be considered for development of a prediction model with high accuracy. Models of data for all breeds and sexes fit the effects of breed, sex, and interaction of abattoir by butchers, whereas models of data for each breed and sex fit the effect of interaction of abattoir by butcher only. Due to possible future changes in back fat control, we performed a log transformation of BF. Our new models fit better than the currently used model.

한우, 칡소 및 제주 흑우 Calpain-Calpastatin 유전자 다양성 (Diversity of Calpain-Calpastatin gene frequencies in Brown, Brindle and Jeju Black Hanwoo)

  • The aim of study was to investigate genetic diversity for the calpain/calpastatin gene in three Hanwoo breeds [(Brown (n=62), Brindle (n=81) and Jeju Black (n=30)]. Random samples from three breeds of Hanwoo were selected and genotyped for the 7 SNPs of calpain/calpastatin using TaqMan method. Allele frequencies were investigated for CAPN1/CAST gene. Allele frequency of CAST2 SNP was 0.75, 0.59 and 0.22 for Brown, Brindle and Jeju black, respectively. The CAST3 revealed allele frequency of 0.59 and 0.57 in Brown and Jeju Black, while it showed very low allele frequency (0.07) in Brindle. In particular, favorable allele (G allele) for the CAPN1-2 SNP which was shown a strong association with tenderness in Taurine and Indicine cattle revealed 16% and 17% higher allele frequency in Brown Hanwoo (0.82) comparing Brindle (0.66) and Jeju Black Hanwoo (0.65). AMOVA demonstrated that among population variance occupied only 10% of total variance and among individual variance was 0%, while within individual variance was 90% of total variance. This result showed that population effect contributed very small portion of genetic to these three Hanwoo breeds, while within individual variance contributed large portion of genetic diversity within these Hanwoo breeds. In conclusion, three Hanwoo breeds (Brown, Brindle and Jeju black) showed a genetically homogeneous based on the 7 SNPs of CAPN1/CAST gene and it came from same ancestor to form modern Hanwoo breed.

The impact of different diets and genders on fecal microbiota in Hanwoo cattle

  • Bovine fecal microbiota is important for host health and its composition can be affected by various factors, such as diet, age, species, breed, regions, and environments. The objective of this study was to evaluate the impact of diet and gender on fecal microbiota in Korean native Hanwoo cattle. The 16S rRNA gene amplicon sequencing of fecal microbiota was conducted from 44 Hanwoo cattle divided into four groups: (1) 11 heifers fed an oat hay plus total mixed ration (TMR) diet for breeding (HOTB), (2) 11 heifers fed an early fattening TMR diet (HEFT), (3) 11 steers fed the early fattening TMR diet (SEFT), and (4) 11 steers fed the late fattening TMR diet (SLFT). Firmicutes and Bacteroidota were the first and second most dominant phyla in all the samples, respectively. The Firmicutes/Bacteroidota (F/B) ratio associated with feed efficiency was significantly greater in the SLFT group than in the other groups. At the genus level, Romboutsia, Paeniclostridium, and Turicibacter were the most abundant in the SLFT while Akkermansia, Bacteroides, and Monoglobus were the most abundant in the HOTB group. Although the same early fattening TMR diet was fed to Hanwoo heifers and steers, Marvinbryantia and Coprococcus were the most abundant in the HEFT group while Alistipes and Ruminococcus were the most abundant in the SEFT group. Shannon and Simpson diversity indices were significantly lower in the SLFT group than in the other groups. Distribution of fecal microbiota and functional genetic profiles were significantly different among the four treatment groups. The present study demonstrates that different diets and genders can affect fecal microbiota and the F/B ratio may be associated with feed efficiency in Hanwoo cattle. Our results may help develop strategies to improve gut health and productivity through manipulation of fecal microbiota using the appropriate diet considering Hanwoo cattle gender.

체내 또는 체외에서 생산된 한우 수정란을 젖소 수란우에 이식한 결과 (Results of Embryo Transfer with Hanwoo Embryos Produced In-Vivo or In-Vitro to Holstein Cows as Recipients)

  • This study was performed to investigate the result that in-vivo or in-vitro embryos of Hanwoo cows were transferred to Holstein cows. Seventeen Hanwoo cows were used as donors for production of in-vivo embryos and fresh hanwoo in-vivo embryos were transferred to 1,150 Holsteins. And 2 embryos were transferred to 188 Holstein recipients to produce twin calves. Diagnosis on pregnancy was performed by rectal palpation at $60\sim90$ days after transfer. The pregnancy rate of Holstein recipients was 55.8% after transfer with Hanwoo in-vivo embryos and 38.2% after transfer with Hanwoo in-vitro embryos. The delivery rate of pregnant Holstein recipients was 88.4% after transfer with Hanwoo in-vivo embryos and 75.6% after transfer with Hanwoo in-vitro embryos. The rate of delivery of Holstein recipients transferred with two Hanwoo embryos was 36.2% and the rate of twin production was 25.9%. The rate of twin production by embryo transfer with in-vivo embryos was 30.4%, whereas the fate with in-vitro embryos was 15.6%. The pregnancy rate according to the grade of corpus luteum of Holstein recipients transferred with Hanwoo in-vitro embryos was 41.5 and 36.0% for A and B grade, respectively. The pregnancy rate according to the transfer in site in the uterine lumen of recipients was 40.9 and 32.7% for anterior and middle site, respectively. The pregnancy rate according to day of embryo transfer after estrus of recipients was 45.5, 38.8 and 39.7% for day 6, day 7 and day 8, respectively. There was difference of pregnancy rate according embryo transfer technician ($30.5\sim45.8%$) individual dairy farm ($21.1\sim51.0%$). These results are supposed to indicate that the rate of pregnancy after transfer with Hanwoo embryos to Holstein recipients was similar to that within the same breed, and consequently that this method would be beneficial to enhance the productivity in Hanwoo reproduction.

Linkage Disequilibrium and Effective Population Size in Hanwoo Korean Cattle

  • This study presents a linkage disequilibrium (LD) analysis and effective population size ($N_e$) for the entire Hanwoo Korean cattle genome, which is the first LD map and effective population size estimate ever calculated for this breed. A panel of 4,525 markers was used in the final LD analysis. The pairwise $r^2$ statistic of SNPs up to 50 Mb apart across the genome was estimated. A mean value of $r^2$ = 0.23 was observed in pairwise distances of <25 kb and dropped to 0.1 at 40 to 60 kb, which is similar to the average intermarker distance used in this study. The proportion of SNPs in useful LD ($r^2{\geq}0.25$) was 20% for the distance of 10 and 20 kb between SNPs. Analyses of past effective population size estimates based on direct estimates of recombination rates from SNP data demonstrated that a decline in effective population size to $N_e$ = 98.1 occurred up to three generations ago.

한우 사육실태 및 육종개량에 대한 농가인식도 조사 분석 - 경북지역 중심으로 - (A survey study of farmers' recognition on reality of Hanwoo raising and improving quality : Focused on Gyeongsangbuk-Do)

  • 경북지역의 한우농가를 대상으로 한우 사육실태와 육종개량에 대한 농가들의 인식도를 조사 분석하여, 앞으로 사육 기술지도와 한우개량 방향을 개선하는데 기초자료로 이용하고자 설문조사하여 분석한 결과는 다음과 같다. 후보번식우 선발은 '외모, 체형, 혈통등록우' (39.0%)를 고려하여 선발하였으며, 번식우의 종부방법은 '계획교배를 통한 인공수정' (38.6%)을 실시하고 있어 가장 높게 나타났다. 비육밑소 구입시 외관상 가장 관심을 두는 부분은 '체장' (40.7%)부분으로 나타났고, 비육 밑소의 거세는 '생후 6~7개월'이 가장 많이 실시하였고, 한우고급육 생산을 위하여 '고급육생산 프로그램에 준하여 80% 이상 맞춰준다' (51.7%)고 하였다. 그러나 농가들은 비육출하 월령은 '생후 28개월' (35.2%)이 가장 경제적일 것이라고 생각하고 있었다. 이러한 결과는 조사항목에 따라 다소 차이가 있긴 하지만 대체로 조사대상자의 일반적 특성에 따라 학력, 연령, 직업, 가족 노동력에 따라 유의수준 0.05에서 통계적으로 유의한 차이가 있었다. 그러나 비육 후 출하 시에 육질 1+등급 이상 출현율은 '출하두수의 30% 정도' (22.1%)라고 응답하여 가장 높았지만, 조사대상자의 알반적 특성 간에는 유의한 차이가 나타나지 않았다.

근내지방도가 높은 수입 등심육과 한우육의 육질특성 비교 (Meat Quality of Highly Marbled Imported Beef with Reference to Hanwoo Beef)

  • 본 연구는 근내지방도가 높은 냉장 수입육과 한우 1등급 및 3등급육의 육질 비교를 위해 실시하였다. 쇠고기 전문점에서 화우(Wagyu) 4두, 엥거스(Angus) 4두, 한우 1등급 4두와 한우 3등급 3두의 등심육을 구입하여 시료로 이용하였다. 화우, 엥거스, 한우 1등급과 3등급의 근애지방 함량은 각각 22‘ 8, 13 및 4%였고, 숙성기간은 31, 71, 14 및 14일 이였다. 한우 등급간 또는 수입욱간의 육질 차이는 없었다. 수입육은 한우보다 유의적으로(P<0.05) 낮은 전단력을 보였다. 하지만 관능평가의 연도와 다즙성에는 위 내가지 육질사이에 차이점이 관찰되지 않았다. 한편, 한우가 향미에서 유의적으로(P<0.05) 높은 점수를 받아, 요리된 고기의 향이 더 우수한 것으로 평가되었다. 축종, 사양방법, 숙성일 차이 중 어떤 요인이 본 실험결과에 영향을 주었는가에 대한 의문이 남아있다. 하지만 이 결과는 높은 근내지방을 함유한 수입육 보다 한우고기가 더 우수한 관능 특성을 가졌다는 것을 단편적으로 보여주고 있다. 판별력 분석에서 수입육과 한우육을 분별하는 지방산은 C18:0, C18:ln-7, C18:ln-9, C20:3n-6, C20:4n-6 and n-3으로 나타났다. 하지만 수입육과 한우육에서 이 지방산들의 차이는 C18:0에서만 관찰되었다. 선별된 지방산들과 향미의 상호관계를 관찰하기 위해 주요인 분석을 실시한 결과 C18:0(P<0.05)와 C18:ln-7(P<0.1)가 향미와 음의 상관관계를 보였다. 본 연구의 결과는 한우고기가 더 높은 근내지방을 함우한 냉장 수입육보다 더 우수하다는 것을 시사하고, 낮은 양의 C18:0가 이와 관련된 후보 지방산이 될 수 있다는 것을 시사한다. 한우고기 차별화를 위해 본 결과가 축종, 사양방법 또는 도축 후 처리의 어떤 요인들과 관련되어 있는가에 대한 연구가 요구도된다.

Characterization of BoLA-DRB3.2 Alleles in Hanwoo (Korean cattle) by Sequence Based Typing (SBT)

  • A study was conducted with 70 Hanwoo (Korean cattle) for genotyping bovine leukocyte antigen (BoLA)-DRB3.2 gene by using the polymerase chain reaction (PCR) and sequence based typing (SBT). Two-step PCR was carried out for amplifying a 284 bp fragment of the target gene and the PCR products were digested with three restriction enzymes namely RsaI, BstYI and HaeIII. Seventeen alleles were detected with frequencies ranging from 1.43 to 18.57% and one (x'aa) of these alleles was identified as a new allele that has not been reported before. The frequency of the new x'aa allele identified in this breed was 12.86%. In addition, the seven most frequently observed alleles (DRB3.2 *10, *15, *16, *26, *27, *54 and x'aa) accounted for 74.28% of the alleles in this population. The phylogenetic tree showed that the BoLA-DRB3.2 allele sequences of Hanwoo were shared with other Bos taurus breeds and no specific clade for Hanwoo was identified. It indicates high heterogeneity of the BoLA-DRB3 gene in this population and may give some ideas for breeding animals having better disease resistance.
