$[^{123}I]$Idoxifene 합성과 유방암의 세포섭취에 관한 연구 (Study for the Synthesis of $[^{123}I]$Idoxifene and Its Uptake in the Breast Cancer Cell)

  • 목적: 현재 유방암 치료제로서 임상실험 제 2단계에 들어간 idoxifene은 항에스트로겐 의약품으로서 기존의 tamoxifen보다도 많은 장점을 가지고 있는 것으로 연구결과 밝혀졌다. 또한 방사성 동위원소 $[^{123}I]$를 표지한 $[^{123}I]$idoxifene은 SPECT을 이용한 유방암세포를 영상화하여 조기에 진단할 수 있는 진단시약으로서 널리 각광 받고 있다. 따라서 본 연구에서는 idoxifene의 전구체를 합성하고 $[^{123}I]$를 표지하여 세포 내 섭취론 관찰하였다. 대상 및 방법: $[^{123}I]$idoxifene을 위한 전구체는 McCague가 연구 발표한 자료를 바탕으로 (2-chloroethoxy)benzene과 2-phenylbutanoic acid를 출발물질로 하여 합성하였다. 표지는 $[^{123}I]$를 사용하였으며 분리는 Silica Sep-Pak을 사용하였으며 세포 내 섭취실험은 에스트로겐 리셉터를 가진 MCF-7과 대조군으로서 에스트로겐 리셉터가 없는 MDA-MB-468을 이용하였다. 결과 및 결론: Idoxifene의 전구체인 4-stannylated 화합물의 합성수율은 약 30%이었으며, $[^{123}I]$ 표지는 60분 경과에서 $90{\sim}92%$로 최대의 표지수율을 보였으며 방사화학적 순도는 98%이상이었다. 또한 세포 내 섭취실험에서 실험군과 대조군 사이에 섭취비율은 180분에서 1.7:1로 나타나 idoxifence은 항에스트로겐 효과가 아주 높은 것으로 판명되었다. 이를 바탕으로 배양세포와 동물모델을 이용한 추가적인 실험이 필요하며, 유방암 환자에게도 임상이용이 기대된다.

  • Zinc is known to have an inhibitory effect on apoptosis and an antioxidative effect scavenging reactive oxygen species (ROS) under oxidative stress. We studied the influence of zinc on cadmium-induced apoptosis especially associated with ROS in MCF-7 human breast carcinoma cell line. For the determination of appropriate experimental concentration and time, we excecuted MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide] assay and DNA fragmentation assay. Isolation and Properties of Antitumor Antibiotic YS-1649 from Penicillium sp. strain 1649

  • An antitumor antibiotic named YS-1649 was isolated from the culture filtrate of a newly isolated fungus identified as Penicillium sp.. The fermentation yield reached about 40 mg per liter of the broth. YS-1649, a $\gamma$-Iactone - structured antibiotic, has the molecular fomular of $C_7H_6O_4$, Its structure was determined to be patulin by spectral analysis. It is active against some bacteria and showed cytotoxic effect on the proliferation of human breast cancer cell line, MCF-7, at concentrations of more than 0.048 $mu g/ml$. This compound also showed strong cytotoxic effect on the proliferation of human cancer cell lines, A549 and ACHN.

Determination of human breast cancer cells viability by near infrared spectroscopy

  • Near infrared spectroscopy (NIRS) was employed to qualify and quantify on survival, the injury rate and apoptosis of the human breast cancer cell line MCF-7 cells. MCF-7 cells were cultured in RPMI medium supplemented with 10% FCS in a 95% air and 5% CO2 atmosphere at 37$^{\circ}C$. For the viable cells preparation, cells were de-touched by 0.1% of trypsin treatment and washed with RPMI supplemented with 10% FCS medium by centrifugation at 1000 rpm for 3min. For the dead cells preparation, cells were de-touched by a cell scraper. The cells were counted by a hemacytometer, and the viability was estimated by the exclusion method with frypan blue dye. Each viable and dead cells were suspended in PBS (phosphate bufferred saline) or milk at the cell density desired. For the quantitative determination of cell death by measuring the LDH (lactate dehydrogenase) activity liberated from cells with cell membrane injuries, LDH-Cytotoxic Test Wako (Wako, Pure Pharmaceutical Co. Ltd., Japan) was used. We found that NIRS measurement of MCF-7 cells at the density range could evaluate and monitor the different characteristics of living cells and dead cells. The spectral analysis was performed in two wavelength ranges and with 1,4, 10 mm pathlength. Different spectral data pretreatment and chemometrics methods were used. We applied SIMCA classificator on spectral data of living and dead cells and obtained good accuracy when identifying each class. Bigger variation in the spectra of living cells with different concentrations was observed when compared to the same concentrations of dead cells. PLS was used to measure the number of cells in PBS. The best model for measurement of dead cells, as well as living cells, was developed when raw spectra in the 600-1098 nm region and 4 mm pathlength were used. Smoothing and second derivative spectral data pretreatment gave worst results. The analysis of PLS loading explained this result with the scatter effect found in the raw spectra and increased with the number of cells. Calibration for cell count in the 1100-2500 nm region showed to be very inaccurate.

Establishment of In Vitro Test System for the Evaluation of the Estrogenic Activities of Natural Products

  • In order to evaluate estrogenic compounds in natural products, an in vitro detection system was established. For this system, the human breast cancer cell line MCF7 was stably trans-fected using an estrogen responsive chloramphenicol acetyltransferase (CAT) reporter plas-mid yielding MCF7/pDsCAT-ERE119-Ad2MLP cells. To test the estrogenic responsiveness of this in vitro assay system, MCF7/pDsCAT-ERE119-Ad2MLP cells were treated with various concentrations of 17f3-estradiol. Treatments of 10$^{-8}$ to 10$^{-12}$ M 17$\beta$-estradiol revealed significant concentration dependent estrogenic activities compared with ethanol. We used in vitro assay system to detect estrogenic effects in Puerariae radix and Ginseng radix Rubra extracts. Treat-ment of 500 and 50 $\mu\textrm{g}$/ml of Puerariae radix extracts increased the transcriptional activity approximately 4- and 1.5-fold, respectively, compared with the ethanol treatment. Treatment of 500, 50, and 5 $\mu\textrm{g}$/ml of Ginseng radix Rubra extracts increased the transcriptional activity approximately 3.2-,2.7, and 1.4-fold, respectively, compared with the ethanol treatment. These observations suggest that Puerariae radix and Ginseng radix Rubra extracts have effective estrogenic actions and that they could be developed as estrogenic supplements.

감마선 조사된 율무종자의 세포독성 및 다제내성 극복활성 (Cytotoxicity and Multidrug -Resistance Reversing Activity of Extracts from Gamma-Irradiated Coix Zachryma-jobi L. var. ma-yuen Stapf Seed)

  • 율무종자에 0, 1, 4, 8, 16, 32 및 64 Gy 선량의 감마선을 조사한 후 메탄을 추출하였으며 조사 종자 추출물이 인체의 유방암세포주(MCF-7), 폐암세포주(Calu-6), 위암세포주(SNU-601), 그리고 인체 급성골수성백혈암세포주(AML-2/WT)에 대한 세포독성 및 다제내성세포주(AML-2/D100)에 대한 다제내성 극복활성에 미치는 영향을 알아보고자 MTT 방법을 이용하여 분석하였다. 감마선 조사한 율무종자의 추출물은 대조구보다 비교적 낮은 농도에서 암세포 증식 저해 활성을 나타내는 경향을 보였으며 그 활성은 암세포 주와 조사선량별 추출물에 따라 다르게 나타났다. Calu-6의 경우 4, 8 및 16 Gy 추출물에서 암세포 증식 억제 효과를 나타냄을 알 수 있었으며 MCF-7에서 는 유일하게 8 Gy 추출물에서 $IC_{50}$값을 측정할 수 있었다. 그리고 SNU-601의 경우는 Calu-6에서와 같이 4, 8 및 16 Gy 추출물에서 증식저해 활성을 관찰할 수 있었으나 그 활성은 대조구인 0 Gy 추출물 보다 약간 높은 농도에서 관찰되었다. 한편, 선량이 높을수록 즉, 32 Gy 이상 감마선 조사는 암세포 증식억제 활성을 거의 나타내지 않았다. 유의할 만한 결과로는 8 Gy추출물이 3가지 암세포$(Calu-6:\;633\;{\mu}g/mL,\;MCF-7:\;653\;{\mu}g/mL,\;SNU-601:\;683\;{\mu}g/mL)$ 모두에서 증식 억제 활성을 나타냈다. 다제내성 세포에 대한 감수성 세포의 세포독성도를 측정하여 선택독성을 관찰한 결과 4,8 및 16 Gy추출물들은 오히려 감수성 세포에 대한 세포독성이 높게 나타나 교차내성을 나타내지 않았다. 그러나 다제내성 세포에 대하여 항암제인 vincristine과 함께 추출물을 처리한 결과 내성극복도(RF)가 4, 8 및 16 Gy 추출물에서 각각 1.7, 1.8 및 1.6으로서 다제내성 조절 활성을 관찰할 수 있었다. 따라서 감마선 조사기술은 약용식물 종자의 생리활성 증진에 활용 가능성이 매우 높을 것으로 기대된다.

  • Recent industrial society has human widely exposed to PAHs that are comming from the incomplete combustion of organic material as widerspread environmetal contaminants. Biological activities of PAHs are not known although PAHs are considered as carcinogens. PAHs in the mammalian cells affect CYP1A1 gene expression as well as other phase II drug metabolizing enzymes as UDPGT, NMOR etc. The mechanism of action of PAHs has been studied extensively, however it is not clear how PAHs turn on CYP1A1 in human breast cancer. Our labolatory have been studied the effect of PAHs in the human breast cancer cell lind MCF7. In this study, we examined the ZR-75-1 human breast cancer cells as a new system to evaluate bioactivity of PAHs. ZR-75-1 human breast cancer cell line has been estabilished from the breast cnacer patient, has estrogen receptors and progesteron receptors. We have been able to estbilish long term culture system of this cells then used for the study to observe the effect of PAHs. We demonstrate that PAHs induced the transcription of an aryl hydrocarbon-responsive reporter vector containing the CYP1A1 promoter and 7-ethoxyresolufin O-deethylase(EROD) activity of CYP1A1 enzyme in a concentration-dependant manner. RT-PCR analysises indicated that PAHs significantly up-regulate the constitutive level of CYP1A1 mRNA. Apparently, ZR-75-1 cells have Aryl hydrocarbon recetors, therefore it would be good experimental tool to study the cross-talk between PAHs and steroid actions.

적포도의 주 항산화물질, 레스베라트롤의 항암작용: 아폽토시스 의한 인체 암세포 사멸 유도 (Anticarcinogenic Activity of Resveratrol, a Major Antioxidant Presnet in Red Wine : Induction of Apoptosis in Human Cancer Cells)

  • Resveratrol (3,5,4'-trihydroxy-trans-stilbene) has been considered to be as one of major antioxidants present in grapes responsible for beneficial effects of red wine consumption on coronary heart disease. This triphenolic stilbene has been suggested as a potential cancer chemopreventive agent based on its striking inhiitory effects on diverse cellular events associated with tumor initiation, promotion, and progression. The compound has strong antioxidative and anti-inflammatory activities which amy contribute to its chemopreventive/chemoprotective properties. In the present work, we have found that resveratrol reduces viability and DNA synthesis capability of cultured human promyelocytic leukemia (HL-60) cells. Likewise, the viability of human breast cancer cell line, MCF-7 was reduced by resveratrol treatment. The growth inhibitory and antiproliferative properties of resveratrol appear to be associated with its induction of apoptotic cell death as determined by morphological and ultrastructural changes, agarose gel electrphoretic analysis of internucleosomal DNA fragmentation, and in situ terminal end-labeling of fragmented DNA (TUNEL). This compound also inhibited the phorbol ester-induced expression of cyclooxygenase-2 (COX-2) protein in immortalized human mammary epithelial MCF-10A cells. These results suggest that resveratrol has the promising cancer therapeutic/chemopreventive potential.

Estrogenic Activity of Sanguiin H-6 through Activation of Estrogen Receptor α Coactivator-binding Site

  • A popular approach for the study of estrogen receptor ${\alpha}$ inhibition is to investigate the protein-protein interaction between the estrogen receptor (ER) and the coactivator surface. In our study, we investigated phytochemicals from Rubus coreanus that were able to disrupt $ER{\alpha}$ and coactivator interaction with an $ER{\alpha}$ antagonist. The E-screen assay and molecular docking analysis were performed to evaluate the effects of the estrogenic activity of R. coreanus extract and its constituents on the MCF-7 human breast cancer cell line. At $100{\mu}g/mL$, R. coreanus extract significantly stimulated cell proliferation ($574.57{\pm}8.56%$). Sanguiin H6, which was isolated from R. coreanus, demonstrated the strongest affinity for the $ER{\alpha}$ coactivator-binding site in molecular docking analysis, with a binding energy of -250.149. The initial results of the study indicated that sanguiin H6 contributed to the estrogenic activity of R. coreanus through the activation of the $ER{\alpha}$ coactivator-binding site.

Knockdown of vps54 aggravates tamoxifen-induced cytotoxicity in fission yeast

  • Tamoxifen (TAM) is an anticancer drug used to treat estrogen receptor (ER)-positive breast cancer. However, its ER-independent cytotoxic and antifungal activities have prompted debates on its mechanism of action. To achieve a better understanding of the ER-independent antifungal action mechanisms of TAM, we systematically identified TAM-sensitive genes through microarray screening of the heterozygous gene deletion library in fission yeast (Schizosaccharomyces pombe). Secondary confirmation was followed by a spotting assay, finally yielding 13 TAM-sensitive genes under the drug-induced haploinsufficient condition. For these 13 TAM-sensitive genes, we conducted a comparative analysis of their Gene Ontology (GO) 'biological process' terms identified from other genome-wide screenings of the budding yeast deletion library and the MCF7 breast cancer cell line. Several TAM-sensitive genes overlapped between the yeast strains and MCF7 in GO terms including 'cell cycle' (cdc2, rik1, pas1, and leo1), 'signaling' (sck2, oga1, and cki3), and 'vesicle-mediated transport' (SPCC126.08c, vps54, sec72, and tvp15), suggesting their roles in the ER-independent cytotoxic effects of TAM. We recently reported that the cki3 gene with the 'signaling' GO term was related to the ER-independent antifungal action mechanisms of TAM in yeast. In this study, we report that haploinsufficiency of the essential vps54 gene, which encodes the GARP complex subunit, significantly aggravated TAM sensitivity and led to an enlarged vesicle structure in comparison with the SP286 control strain. These results strongly suggest that the vesicle-mediated transport process might be another action mechanism of the ER-independent antifungal or cytotoxic effects of TAM.
