Membrane Proteins and Their Antigenicity of Toxoplasma gondii (yoxoplusmg leondii의 세포막 단백 성분과 그 항원성)

  • Surface membrane proteins of virulent RH strain and tissue cyst-forming Fukaya strain of Toxoplasma gondii were analysed by SDS-polyacrylamide gel electrophoresis after LPO-catalyzed surface iodination and lectin blotting, then identified the zoite-specific antigens. Prior to the analyses, purification of RH tachyzoites from mouse peritoneal exudate and of Fukaya bradyzoites from mouse brain tissues were performed by centrifugation - on the discontinuous Percoll density-gradient. Ta- chysoites were obtained at the interface of 50U and 60% Percoll solution and brain cysts were harvested at the interfaces of 40-50% and 50-60%, then bradyzoites were obtained by treating the cysts with hypertonic solution. The LPO-catalyzed iodination detected 15 KDa and 14 KDa proteins o( brady- zoites and 30 KDa protein of tachysoites as major bands with several other minor bands. But Con A blotting revealed some bands of 200 K∼50 KDa glycoproteins of bradyzoites and 52 KDa band as major and minor bands of 33 K∼20 KDa of tachyzoites. Phytohemagglutinin did not detect any band in the two forms. EITB with anti- Fukaya antibody and anti-RH antibody revealed cross-reactivities between the two forms. Despite the cross-reactivity, anti-Fukaya antibody reacted with 15 KDa band of bradyzoites specifically and, anti-RH antibody with 52 KDa, 30 KDa, and 25 KDa bands of tachyzoites, respectively. It was identified that 15 KDa protein in bradyzoite, which was not a glycoprotein, was a major membrane protein with sufficient antigenicity, and in the case of tacky- zoite, 52 KDa surface glycoprotein (gp52) with specific antigenicity might be added to the major surface protein, p30.

  • Toxoplasmic encephalitis is caused by reactivation of bradyzoites to rapidly dividing tachyzoites of the apicomplexan parasite Toxoplasma gondii in immunocompromised hosts. Diagnosis of this life-threatening disease is problematic, because it is difficult to discriminate between these 2 stages. Toxoplasma PCR assays using gDNA as a template have been unable to discriminate between an increase or decrease in SAG1 and BAG1 expression between the active tachyzoite stage and the latent bradyzoite stage. In the present study, real-time RT-PCR assay was used to detect the expression of bradyzoite (BAG1)- and tachyzoite-specific genes (SAG1) during bradyzoite/tachyzoite stage conversion in mice infected with T. gondii Tehran strain after dexamethasone sodium phosphate (DXM) administration. The conversion reaction was observed in the lungs and brain tissues of experimental mice, indicated by SAG1 expression at day 6 after DXM administration, and continued until day 14. Bradyzoites were also detected in both organs throughout the study; however, it decreased at day 14 significantly. It is suggested that during the reactivation period, bradyzoites not only escape from the cysts and reinvade neighboring cells as tachyzoites, but also converted to new bradyzoites. In summary, the real-time RT-PCR assay provided a reliable, fast, and quantitative way of detecting T. gondii reactivation in an animal model. Thus, this method may be useful for diagnosing stage conversion in clinical specimens of immunocompromised patients (HIV or transplant patients) for early identification of tachyzoite-bradyzoite stage conversion.

Histopathologic Observation of the Aborted Fetus from Pregnant Dairy Cows Naturally Infected with Neospora caninum (Neospora caninum에 자연 감염된 임신우로부터 유산된 태아의 병리조직학적 관찰)

  • This study examined the histopathologic and electron microscopic findings of aborted fetuses from pregnant dairy cows naturally infected with Neospora caninum (N. caninum) at four farms in Gongju city and Yeonki gun of Choongnam province. Systemic subcutaneous edema was observed in the aborted fetuses. The necropsy revealed considerable serosanguinous fluid in the body cavity of the aborted fetuses. Light microscopy showed the infiltration of many inflammatory cells consisting of macrophages, lymphocytes and mononuclear cells, accompanied by congestion, hemorrhage and necrosis of myocardiac cells and hepatocytes in the liver and heart of the aborted fetuses. In the liver, clusters of tachyzoites were formed in the cytoplasm of hepatocytes and the interstitial tissue. In the brain, many tissue cysts of various sizes were observed in the nerve cells and their adjacent areas. Tissue cysts had a round shape and contained a large amount of bradyzoite. In addition, there was diffuse gliosis accompanied by congestion and hemorrhage and focal necrosis in the brain. Infiltration of microglial cells were observed at the periphery of the focal necrosis and perivascular area in the brain. Electron microscopy showed that the tissue cyst wall had a thickness of approximately 1 ${\mu}m$ with an irregular shape. On the interior side, more than 100 bradyzoites with lengths of 2-5 ${\mu}m$ and widths of 1-2 ${\mu}m$ were observed. The nucleus of in the bradyzoites was located approximately 1-1.5 ${\mu}m$ anterior to the posterior tip of the zoite. In the cytoplasm between the nucleus and the posterior tip, there were many amylopectin granules, electron-dense small-sized and electron-thin large-sized round granules, homogeneously electron-dense rhoptries and micronemes oriented perpendicularly to the zoite pellicle. To summarize, tissue cysts were identified on electron microscopy from the aborted fetus from N. caninum seropositive pregnant cow by the ELISA. This led to the confirmed presence of N. caninum.

  • An in uipo culturing to examine the cyst stage of ToxopLQsma gondii (ME49 strain) was Investigated using murine peritoneal macrophages, and we also examined the effect of CAMP or DHFR Inhibitors on the growth of bradyzoltes. For experiments ICR mice were Injected 1.p. with 1,500 brain cysts. At 1, 3, 5 and 7 days, peritoneal exudates were Isolated and then adherent peritoneal macrophages were cultured for 1,3,5 and 10 days. Growth pattern of bradyzoltes was measured by (3H)-uracil uptake assay and morphological pattern of pseudocysts formed in macrophages was observed Uth Glemsa stain. Mostly bradyzoites were observed In the macrophages extracted at 3 and S days post Infection. After 3 days in vitro, a number of pseudocysts were formed in the macrophages and the size of pseudocysts was increased during further 5 and 10 days in vitro culture. CAMP stimulated the growth of bradyzoltes when in uiuo 3 and 5 days and then in vitro 5 and 10 days conditions were applied. In case of.DHFR Inhibitors, pynmethamlne produced a linearly decremental effect with a cont.-dependent mode but methotrexate was not effective against Intracellular bradyzoltes or pseudocysts In this system. It was suggested that cyst-forming strain of T gondii (ME49 strain) could be maintained and cultivated in uitro by use of murine peritoneal macrophages. In uivo 3 and 5 days and then in uiko 5 and 10 days conditions appeared to be suitable for culturing of bradyzoltes. CAMP and pyrimethamine had an effect of stimulation and inhibition on the growth of bradyzolte, respectively.

  • 고양이를 제외한 포유동물 및 조류는 oocyst 또는 cyst의 섭취로 감염되고 무성생식을 한다. 급성기에는 tachyzoite ($2~3{\times}5~7$\;\mu\textrm{m}$)가 여러 장기에서, 숙주 변역이 활성화된 이후인 만성기에는 100~1,000 개의 bradyzoite를 내장하며 주변에 염증반응이 없는 cyst (직경 $20~50;\mu\textrm{m}$)가 근육 및 뇌에서 관찰된다. Tachyzoite는 태반을 통해 태아 감염을 일으킨다. 고양이는 oocyst 또는 cyst의 섭취로 감염되고, 기타 동물에서 보이는 감염양식 외에 소장에서 coccidiosis양의 증식이 이루어짐으로 oocyst (Isospora 형, $10{\times}12;\mu\textrm{m}$)를 배출한다.

  • For the comparison of surface fine structures in the proliferative forms of two major protoroan parasites, Toxoplasma gondii and Sarcocystis species in mammalian hosts, isolated from the artificially infected mice and from the naturally infected cattle, respectively, an SEM(Hitachi S-570) was applied to the fixed, dried and coated with gold ion on the microslide glasses. The tachyzoites of T gondii from the peritoneal cavity of the mouse showed the crescent-like feature and measured as $5.57{\mu}m$ in length and $2.33{\mu}m$ in width, while the bradyzoites of Sarcocystis species from the heart muscle of slaughtered cattle was banana-shaped and measured as $14.18{\mu}m$ in length and $2.85{\mu}m$ in width. On the surface of Sarcocystis species bradyzoite, a distinct elliptical micropore was identified in the high magnification observation of 60,000X, and it measured as $0.35{\mu}m$ in length and $0.18{\mu}m$ in width.

A Survey of Sarcocystis Infections in the Slaughtered Cattle and Identification of Sarcocystis cruzi (도축우의 심장근육내 주육포자충 감염실태조사와 Sarcocystis cruzi의 동정)

  • 330 Samples of the slaughtered cattle heart muscle were collected from the abattoirs of five regions in Kangwon - do to reveal the frequency of sarcocystis infections during January through December in 1991. The samples were inspected for bradyzoites by the trypsin digestion technique and the possitive samples were fed to dogs and cats for the detection of sporocysts shed in the feces. The results obtained were summarized as follows : 1. The infection rate of bovine Sarcocystis investigated from 330 samples was 43.6%. 2. It revealed that the infection rate of Sarrocystis increased gradully with the advance in the age, 14.5% in below two years, 26.1% in the three years, 30% in four years, 54.7% in five years, 74.4% in six years, 90% in seven years and 100% in older than eight years. 3. The cyst walls detected out from the heart muscles were less than l${\mu}{\textrm}{m}$ in thickness and the size of bradyzoites were $11.8{\times}2.8{\mu}m$ in average. 4. The size of sporocysts shed in the feces of dogs were $15.8{\times}9.8{\mu}m$ in average and the prepatent periods ranged from 12 to 16days. 5. Sarcorystis found in the bovine heart muscles were identified as Sascocystis cruzi ( Hasselman, 1923) , wenyon, 1926.

Evaluation of Recombinant SAG1, SAG2, and SAG3 Antigens for Serodiagnosis of Toxoplasmosis

  • Serologic tests are widely accepted for diagnosing Toxoplasma gondii but purification and standardization of antigen needs to be improved. Recently, surface tachyzoite and bradyzoite antigens have become more attractive for this purpose. In this study, diagnostic usefulness of 3 recombinant antigens (SAG1, SAG2, and SAG3) were evaluated, and their efficacy was compared with the available commercial ELISA. The recombinant plasmids were transformed to JM109 strain of Escherichia coli, and the recombinants were expressed and purified. Recombinant SAG1, SAG2, and SAG3 antigens were evaluated using different groups of sera in an ELISA system, and the results were compared to those of a commercial IgG and IgM ELISA kit. The sensitivity and specificity of recombinant surface antigens for detection of anti-Toxoplasma IgG in comparison with commercially available ELISA were as follows: SAG1 (93.6% and 92.9%), SAG2 (100.0% and 89.4%), and SAG3 (95.4% and 91.2%), respectively. A high degree of agreement (96.9%) was observed between recombinant SAG2 and commercial ELISA in terms of detecting IgG anti-Toxoplasma antibodies. P22 had the best performance in detecting anti-Toxoplasma IgM in comparison with the other 2 recombinant antigens. Recombinant SAG1, SAG2, and SAG3 could all be used for diagnosis of IgG-specific antibodies against T. gondii.

Sarcocystis infection and identification of Sarcocystis species in pigs in Korea (국내(國內) 돼지의 Sarcocystis 감염과 Sarcocystis종의 동정에 관한 연구)

  • Five hundred and forty-eight samples of pig heart muscle were collected from the abattoirs of many regions in Korea to reveal the frequency of Sarcocystis infections and to identify the species from June 1988 to April 1989. Heart muscle of the pigs was inspected for sarcocysts by the direct detection technique and for bradyzoites by the trypsin digestion technique. For examination of development of the parasites in the final host, 5 cross bred mature dogs, 5 puppies and 5 kittens were fed 100g, 50g and 50g of the infected meat respectively, four times in 2 days. Of 402 fattened and 146 older culled breeding pigs, 3 fattened pigs and 39 culled pigs were positive for Sarcocystis. Sarcocystis cysts from heart muscle measured an average of $425{\times}169{\mu}m$ and bradyzoites an average of $15.6{\times}3.5{\mu}m$. Of 15 animals, only 2 puppies were infected with Sarcocystis. The prepatent period was 11 to 12 days and patent period was not examined since the puppies were infected with some another infections and one died on day 11 and another died on day 12 after ingestion of the meat. The sporulated oocysts were detected 11 days after ingestion of the meat and sporocysts 12 days from the puppy feces. The sporulated oocysts measured an average of $16.5{\times}11.5{\mu}m$ and sporocysts an average of $12.6{\times}7.9{\mu}m$. On scraping examination of the intestinal mucosa, fully sporulated oocysts were detected in the tip of the intestinal villi. Considering above all descriptions, Sarcocystis in pig heart muscle in Korea was identified with Sarcocystis suicanis.

Toxoplasmic meningoencephalitis in a stray cat in Korea

  • A dead stray cat was necropsied for zoonotic feline disease monitoring. Grossly, there were no specific lesions. Major microscopic lesions included lymphocytic meningoencephalitis, malacia, and tissue cysts in the cerebral and cerebellar cortex. The size and shape of tissue cysts were identical to those of Apicomplexa including Toxoplasma (T.) gondii. Bradyzoites in the tissue cyst were strongly positive for T. gondii by immunohistochemistry. Electron microscopy revealed that bradyzoites within the tissue cyst were similar to the morphological features of T. gondii. Fresh tissue samples were examined by a polymerase chain reaction assay and resulted in a specific band of T. gondii only in the brain. Based on the results, this case was diagnosed as toxoplasmosis. This is the first case of toxoplasmic meningoencephalitis in a cat in Korea.
