유향 물 추출물의 HL-60 혈액암세포에서 세포사멸 유도효과 (The Water Extract of Boswellia carterii Induces Apoptosis in Human Leukemia HL-60 Cells)

  • The possible mechanism of the antiproliferative and apoptotic effects of Boswellia carterri water extract were studied in HL-60 human leukemia cells. The cytotoxicity of HL-60 cells after the treatment of Boswellia carterii water extract showed dose- and time-dependent manner. The apoptotic effect of 300 $\mu$g/ml Boswellia carterii water extract was demonstrated by DNA laddering. The activity of caspase 3-1ike protease was markedly increased in HL-60 cells treated with Boswellia carterii water extract. Furthermore, the level of Bcl-2 was time-dependently reduced, whereas Bax protein level was enhanced by Boswellia carterii water extract treatment. In conclusion, our results suggest that apoptotic effect of Boswellia carterii water extract may partly mediated through activations of caspase-3 activity and Bax expression, and inhibition of Bcl-2 expression.

  • In activated macrophage, large amounts of nitric oxide (NO) are generated by inducible nitric oxide synthase (iNOS), resulting in acute or chronic inflammatory disorders. In Raw 264.7 cells stimulated with lipopolysaccharide (LPS) to mimic inflammation, water extracts from the fruit of Chaenomeles sinensis, the root of Polygonum cuspidatum and Boswellia carterii inhibited the LPS-induced NO production in a parallel dose-dependent manner. To investigate the mechanism by which those extracts inhibits NO production, we examined the expression of iNOS and COX-2 in both mRNA and protein levels. We observed a significant change in the iNOS expression between LPS alone and LPS plus those extracts-treated cells. However, water extracts from Chaenomeles sinensis, Polygonum cuspidatum and Boswellia carterii did not inhibit COX-2 expression which was induced by LPS treatment in Raw 264.7 cells. These data suggest that water extracts from Chaenomeles sinensis, Polygonum cuspidatum and Boswellia carterii can modulate anti-inflammatory immune response, which may be in part associated with the regulation of NO synthesis through the regulation of iNOS expression in mouse macrophage cells.

  • Matrix metalloproteinase-9 (MMP-9) is considered to be an important component in the progression of inflammation. Monocytes/macrophages are prominent at inflammation sites, and activation of these cells by stimulants such as lipopolysaccharide (LPS) leads to the production of significant amounts of MMP-9. Here, we show that LPS-induced MMP-9 production and activation was inhibited by the water extract from the fruit of Chaenomeles sinensis (CS), the root of Polygonum cuspidatum (PC), but increased by the extract from Boswellia carterii (BC). To investigate the mechanism by which those extracts inhibits MMP-9 activation, we examined the level of MMP-9 mRNA expression. We observed a significant change in the MMP-9 expression between LPS alone and LPS plus Chaenomeles sinensis and Polygonum cuspidatum extracts-treated cells. In addition, LPS significantly up-regulated MMP-9 promoter activity in Raw 264.7 cells, which was attenuated by the CS and PS extracts. However, water extracts from Boswellia carterii increased MMP-9 expression and MMP-9 promoter activity which were induced by LPS treatment in Raw 264.7 cells. These data suggest that water extracts from Chaenomeles sinensis and Polygonum cuspidatum can modulate anti-inflammatory immune response, which may be in part associated with the regulation of MMP-9 production and/or activation through the regulation of MMP-9 expression in mouse macrophage cells.

오향(五香) 성분의 살균 및 살충효과 (Antifungal and Insecticidal Activity of Ohyang (Five Medicinal Plants))

  • 정향, 유향, 곽향, 청목향, 그리고 침향의 에탄올 및 휘발성 추출물의 항균력 및 살충력 실험결과, 정향의 에탄올 및 휘발성 추출물은 실험균주 4종(Mucor hiemalis, Aspergillus niger, Penicillium funiculosum, Trichoderma viride)에 대해서 강한 항균력을 나타냈으며, 유향의 휘발성 추출물은 실험균주 M. hiemalis, A. niger, P. funiculosum에서 강한 항균력을 보였다. 공시충 흰개미에 대해서 정향 에탄올 추출물이 접종 후 48 시간 뒤에 $100\%$ 살충력을 보였고, 청목향 에탄올 추출물은 72 시간 뒤에 개체를 $32\%$ 감소시키는 것을 확인하였다. 휘발성 추출물에 대한 살충력 실험은 흰개미에 대해서 정향 추출물이 48시간 내에 $100\%$의 사충력을 보였다. 넓적나무좀에 대한 살충력은 정향이 접종 후 7일 내에 $19\%$의 사충력을 보였다. 정향의 휘발성 추출물에서 9 개의 휘발성 성분을 확인하였으며, 정향의 휘발성 향기성분 중 가장 큰 농도$(92\%)$를 차지하고 있는 eugenol은 항균활성 물질로써 확인되었다.

Arginase II Inhibitory Activity from Crude Drugs

  • Arginase competitively inhibits nitric oxide synthase (NOS) via use of the common substrate L-arginine. Arginase II has recently reported as a novel therapeutic target for the treatment of cardiovascular diseases such as atherosclerosis. In our experiment, the EtOH extracts of four-hundreds extracts drugs were investigated for the arginase inhibitory activity. Among them, four extracts exhibited over 50% inhibition of arginase II activity compared to control at a concentration of 150${\mu}g/ml$. In particular, the seed of Arctium lappa, gum-resin of Boswellia carterii, aerial part of Artemisia apiacea and rhizome of Cyperus rotundus inhibited arginase II activity, with $IC_{50}$ values of 118.4, 135.4, 123.9 and 86.7${\mu}g/ml$, respectively. In addition, four plant extracts showed less than 20% inhibition of arginase I activity at 150${\mu}g/ml$. These plants might be the potential candidate materials in the development of the novel atherosclerosis drug.

Boswellic Acid Improves Cognitive Function in a Rat Model Through Its Antioxidant Activity - Neuroprotective effect of Boswellic acid -

  • Objectives: Boswellic acid (BA), a compound isolated from the gum-resin of Boswellia carterii, is a pentacyclic terpenoid that is active against many inflammatory diseases, including cancer, arthritis, chronic colitis, ulcerative colitis, Crohn's disease, and memory impairment, but the mechanism is poorly understood. This study investigated the effects of boswellic acid on spatial learning and memory impairment induced by trimethyltin (TMT) in Wistar rats. Methods: Forty male Wistar rats were randomly divided into 5 groups: Normal group, TMT-administrated rats (8.0 mg/kg, Intraperitoneally, i.p.) and TMT + BA (40, 80 and 160 mg/kg, i.p.)-administrated rats. BA was used daily for 21 days. To evaluate the cognitive improving of BA, we performed the Morris water maze test. Moreover, to investigate the neuroprotective effect of BA, we determined the acetylcholinesterase (AchE) activity, the malondialdehyde (MDA) level as a marker of lipid peroxidation, and the glutathione (GSH) content in the cerebral cortex. Results: Treatment with TMT impaired learning and memory, and treatment with BA at a dose of 160 mg/kg produced a significant improvement in learning and memory abilities in the water maze tasks. Consistent with behavioral data, the activity of AChE was significantly increased in the TMT-injected rats compared to the control group (P < 0.01) whereas all groups treated with BA presented a more significant inhibitory effect against AChE than the TMT-injected animals. In addition, TMT reduced the GSH content and increased the MDA level in the cerebral cortex as compared to the control group) P < 0.01). On the other hand, treatment with BA at 160 mg/kg slightly increased the GSH content and reduced the MDA level in comparison to the TMT-administered group (P < 0.01). Conclusion: The above results suggest that the effect of BA in improving the cognitive function may be mediated through its antioxidant activity.

痔瘡과 痔瘻에 對한 文獻的 考察 (A Literatural study on the hemorrhoids and hemorrhoids complicated by anal fistula)

  • A Literatural study on the etiological factors, classification, prescription of hemorrhoids and hemorrhoids complicated by anal fistula following results were obtained. 1. The cause of hemorrhoids are long time sit, long time gate, overfatigue, overeating, imbalance of stool( constipation or diarrhea), pregnant fertility(overfatigue after childbirth, insufficiency of middle warmer energy), uncontrol sexual excess, pathgenic factors of wetness, heat, wind, dry, genetic cause, excess of anxiety, pile up of heat poison, weakness of entrails and viscera. The cause of hemorrhoid complicated by anal fistula are attack of external wind, heatness, dry, fire, wetness(pathgenic factors), inapporiate treatment and chronic disease, greasy diet, excess of anxiety, constipation, uncontrol sexual excess, obstacle of circulation of vital energy and blood on anal site. 2. Classification of hemorrhoids are female hemorrhoids, male hemorrhoids, pulse hemorrhoids, intestines hemorrhoids, vital energy hemorrhoids, wine hemorrhoids, blood hemonhoids, flowing hemorrhoids. Classification with other method are external hemorrhoids, internal hemorrhoids, mixed hemorrhoids, excrescence hemorrhoids, nipple homorrhoids. External hemorrhoids is classified of varicosis of hemorrhoidal vein, connective tissue form, thrombus form. Classification of hemorrhoid complicated by anal fistula are simple lower hemorrhoid, lower mixed hemorrhoid, deep hemorrhoid, outer of one hole hemorrhoid, a horseshoe hemorrhoids. Once more classificated of four are space of sphincter muscle form, penetration sphincter muscle form, upper of sphincter muscle form, outer of sphincter muscle form. 3. Therapy method of hermorrhoid and hemorrhoid complicated by anal fistula are internal method, fumigation method method, ointment, method of close with medicine, necrotizing method, hot medicated compress( gxternal method), injection, insertion, bind, (operation) and acupuncture therapy (the others method) 4. Herb medicine for many used of internal method are Scutellaria baikalensis George(黃芩), Coptis japonia Makino(黃連), Rehmania giutinosa Liboschitz ex Fischer & Meyer(生地黃), Poncirus trifoliata Refinesque(枳殼), Sanguisorba officinalis Linne(地楡), Sophora japonica L.(槐花), Cnidium officinale Makino (川芎), Astragalus membranaceus Bunge(황기), Angelica gigas Nakai (當歸). 5. Herb medicine for many used of fumigation are Schlechtendalia Chinesis J. Bell (五倍子), Artemisia Vulgaris L. var indica Maxim(艾葉), Poncirus trifoliata Refinesque (枳殼), Nepeta japonica Maximowicy(荊芥), And herb medicine for many used of ointment are Calomelas(輕粉), Alum(白礬), Boswellia carterii Birdwood(乳香), Os Draconis Fossilia Ossis Mastodi(龍骨).

발모효과를 가지는 한방복합처방단 (Medicinal Herbal Complex Extract with Potential for Hair Growth-Promoting Activity)

  • 탈모방지 및 발모효과를 가지는 소재를 개발하기 위해 한의학에서 전통적으로 사용되는 23가지 한방소재를 선정하여 한방복합처방단을 개발하였다. 한방복합처방단을 구성하는 한방소재들은 예로부터 전통적으로 발모 및 탈모방지, 흰머리방지, 염증 치료 및 혈액순환개선 효과를 가진 것으로 알려져 있는 당귀, 보골지, 측백엽, 한련초, 구기자, 복분자, 상백피, 숙지황, 여정실, 적하수오, 흑지마, 고삼, 백지, 익모초, 단삼, 도인, 몰약, 감국, 유향, 인삼, 천궁, 합환피, 현호색 등이다. 또한, 한방복합처방단의 발모효과를 확인하기 위해 in vitro와 in vivo 평가모델을 이용하여 모발성장 및 촉진에 미치는 영향을 실험하였다. In vitro 상에서는 모유두세포, 각질형성세포 및 섬유아세포의 증식을 확인하였다. 또한, 흰머리방지 효과와 관련하여 멜라노마 세포에서의 멜라닌 합성능력을 확인하였다. 한방복합처방단의 in vitro 상에서의 육모효과는 C57BL/6 마우스를 이용한 in vivo 상에서도 확인하였다. 연구 결과 한방복합처방단은 $50{\mu}g/mL$의 농도에서 모유두 세포의 증식을 175 %까지, 섬유아세포인 NIH3T3 세포의 증식은 120 %까지 증가시켰으며, $20{\mu}g/mL$의 농도에서 각질형성세포인 HaCaT 세포의 증식을 133 %까지 증가시켰다. 멜라닌 합성의 경우, $50{\mu}g/mL$의 농도에서 154 %까지 증가시켰다. 또한, C57BL/6 마우스를 이용한 육모효과에 있어서는 한방복합 처방단 처리 4주 후 98 % 이상의 육모효과를 나타내는 것을 확인하였다. 이상의 결과로 볼 때 본 연구에서 개발한 한방복합처방단은 모발의 성장 촉진에 유용하게 활용될 수 있는 처방으로 사료된다.
