Effect of Low Intensity Pulsed Ultrasound with Adipose-Derived Stem Cells on Bone Healing around a Titanium Implant in Tibia of Osteoporosis-Induced Rats (골다공증 유도 백서 경골에 티타늄 임플랜트 매식 시 지방조직유래 줄기세포 주입과 저출력 초음파 적용이 골치유에 미치는 영향)

  • Purpose: Osteoporosis, is a major health problem for the elderly and post-menopausal women and shown to alter the properties of bone as well as impair bone healing around titanium implants in both human and animals. The objective of this study was to examine the effect of LIPUS with adipose-derived stem cells on the healing process around a titanium implant in rats with osteoporosis. Methods: Sixteen osteoporosis-induced rats were divided into two groups: an adipose-derived stem cell injected with Low Intensity Pulsed Ultrasound (LIPUS) application group and a control group. Titanium screw implants (diameter, 2.0 mm: length, 3.5 mm, Cowell Medi, Korea) were placed into both tibia of 16 rats, on 8 rats as the control group and the other 8 rats as the experimental group. Rats were sacrificed at different intervals from 1, 2, 4 and 8 weeks after implantation for histopathologic and immunohistochemical analyses. Results: Histopathological analysis revealed newly formed bone in experimental group earlier than that in control group. Especially at 1 week after implantation, more amounts of new bone matrix and collagen around the implant in the experimental group were seen compared with the control group. Immunohistochemical analysis showed that the levels of osteoprotegerin (OPG) expression in the experimental group were increased at early stages compared with that of control group until 2 weeks after implantation. But after 2 weeks, the expression level of OPG similar in both groups. The expression levels of receptor activator of nuclear factor kB ligand (RANKL) were stronger in the experimental group than the control group until 2 weeks after implantation. After 4 weeks, expression of RANKL in experimental group was similar to the control group. Conclusion: The results of this study suggest that LIPUS with Adipose-Derived Stem Cells in implantation could promote bone healing around titanium implants in rats with osteoporosis.

Usefulness of Atelo-collgen sponge (Teruplug$^{(R)}$) for Treatment of Mandibular Angle Fractures with Third Molar Extraction (하악각 골절에서 제 3 대구치 발치 후 아테로-콜라겐 스펀지의 유용성)

  • Purpose: Fibrillar collagens like type I collagen, are the major constituent of the extracellular matrix and structural protein of bone. Also, it can be a scaffold for osteoblast migration. The purpose of this study is to estimate the effects of absorbable atelo-collagen sponge (Teruplug$^{(R)}$, Terumo biomaterials Co., Tokyo, Japan) insertion in tooth extraction sites on periodontal healing of the second molar, healing of the fractured mandibular bone and new bone formation of third molar socket after the extraction of the impacted third molar with mandibular angle fracture. Methods: In our study of six cases of mandibular angle fractures, all of them underwent the extraction of the third molar tooth & absorbable atelo-collagen sponge insertion in tooth extraction site. Three of them had a intraoral infection & oral opening to fracture site, two of the six had dental caries, and only one had reduction problem due to third molar position. Six consecutive patients with noncomminuted fractures of the mandibular angle were treated by open reduction and internal fixation using one noncompression miniplates and screws placed through a transoral incision. Results: All of the patients have showed good postoperative functions and have not experienced complications requiring second surgical intervention. There was well healing of the mandibular bone and the most new bone formation of third molar socket after the extraction of the impacted third molar with mandibular angle fracture. Conclusion: The results of this study suggest that absorbable atelo-collagen sponge is relatively favorable bone void filler with prevention of tissue collapse, food packing, and enhance periodontal healing. Thus, the use of atelo-collagen sponge and one noncompression miniplate seems to be relatively easy, safe, and effective for the treatment of fractures of the mandibular angle and third molar extraction.

NONLINEAR CONTRAST ENHANCEMENT ON SUBTRACTION IMAGES (계수공정영상의 비선형 대조도 증강효과에 관한 연구)

  • This study was performed to demonstrate the effect of linear or nonlinear contrast enhancement on subtraction images. Three different textures were radiograped on dental film. The first radiograph was taken without the presence of an object. the second, which showed trabucular bone, was taken of the molar area of a human. the third radiograph was taken of the coronal part of molars. Each film was digitized into a 1312 x 1024 pixel x 8 bit depth matrix by means of a Nikon 35 mm film scanner(LS-3510AF, Japan) with fixed gain and internal dark current correction to maintain constant illumination. The scanner was interfaced to a Macintosh Le ill computer(Apple Computer, Charlotte, N.C) This resulted in three pairs of images, including different textures-plain, bone and enamel. Digital regular, linearly and nonlinearly enhanced subtraction was performed. Computer software was ,used to simulate lesions in the shape of a 2D-Gaussian curve on each of a pair of images. The each subtraction images were presented in a random sequence to two groups of 10 observers(students and dentists). ROC analysis was used to compare observer performance. The following results were obtained ; 1. All of LCE subtraction, equalized subtraction and regular subtraction images of plain texture were diagnosed the best by far. 2. The data revealed a siginificant LCE effect in both the student group and the expert group. 3. Clinical expertise was a helphul factor for the observers in this study.

Biocompatible Hydroxyapatite Ceramics Prepared from Natural Bones and Synthetic Materials (천연 및 인공원료로부터 제조한 생체친화형 하이드록시아파타이트 세라믹스)

  • Hydroxyapatite (HA) is well known as a biocompatible and bioactive material. HA has been practically applied as bone graft materials in a range of medical and dental fields. In this study, two types of dense hydroxyapatite ceramics were prepared from natural bones and synthetic materials. The biocompatibility of HA ceramics for supporting osteoblast cell growth and cytotoxicity using an in vitro MG-63 cell line model were respectively evaluated. Artificial hydroxyapatite shows relative density of 93% with 1-2 ${\mu}m$ after sintering, but a hydroxyapatite compact derived from bovine bone has low sintered density of 85% with a small content of MgO. Irrespective of the starting raw materials, both types of sintered hydroxyapatite displayed similar biocompatibility in the tests. FE-SEM observations showed that most MG-63 cells had a stellar shape and formed an intercellular matrix containing fibers on sintered HA. The cells were well attached and grown over the HA surface, indicating that there was no toxicity.

  • The ginseng plant (Panax ginseng Meyer) has a large number of active ingredients including steroidal saponins with a dammarane skeleton as well as protopanaxadiol and protopanaxatriol, commonly known as ginsenosides, which have antioxidant, anticancer, antidiabetic, anti-adipocyte, and sexual enhancing effects. Though several discoveries have demonstrated that ginseng saponins (ginsenosides) as the most important therapeutic agent for the treatment of osteoporosis, yet the molecular mechanism of its active metabolites is unknown. In this review, we summarize the evidence supporting the therapeutic properties of ginsenosides both in vivo and in vitro, with an emphasis on the different molecular agents comprising receptor activator of nuclear factor kappa-B ligand, receptor activator of nuclear factor kappa-B, and matrix metallopeptidase-9, as well as the bone morphogenetic protein-2 and Smad signaling pathways.

A MICROSCOPIC STUDY OF THE EFFECTS OF EXPERIMENTAL RICKETS ON THE DENTAL STRUCTURE OF RATS (실험적구루병에서 백서 치아조직 변화에 관한 현미경학적 연구)

  • Rickets is not the deposite of minerals in the skeletal tissue and the retardation of skeletal growth in growing in growing animals. This study was undertaken to investigate the histologic effects of experimental rickets on the dental structure of the albino rats, and to show the relationship between the histological effects and the pulpal disease which induced premature loss of the primary teeth. This study was based on material obtained from 40 white rats that were placed on a rachitogenic diet for a period 1 to 56 days after weaning (at 24 days). In addition, a study was made of 25 litter mates, 24 to 80 days, that were fed a normal diet. The following results were obtained: 1. Enamel formation and calcification showed no significant changes and no hypoplasia. 2. Dentin formation and calcification was retarded and disturbed. In the experimental group, predentin/calcified dentin was remarkablly increased. 3. Newly formed dentin showed interglobular texture (less homogenous calcification) and the predentin was significantly wider and thicker, and there was an irregular wave in the basal portion of the rat's incisors. 4. In cementum, Matrix formed at almost a normal rate but calcification was defective. So cementoid tissue was increasesd. 5. The formation of the alveolar bone was at almost a normal rate but calcification was retarded. The trabecular bone was filled with osteoid tissue and thicker than in normal groups.

Nano-Hydroxyapatite Modified by Grafting Polylactide and its Tissue Engineering Application

  • PLLA grafted Hydroxyapatite / polylactide (g-HA/PLA) composites were prepared by three grafting methods. The modified particles (p-HA) were dispersed more uniformly in the PLLA matrix than pure n-HA. The p-HA/PLLA composites exhibited better mechanical properties and thermal stability than the n-HA/PLLA composites. The composites also demonstrated improved cell compatibility due to the good biocompatibility of the HAP nanoparticles and the more uniform distribution of the PLLA-grafted HAP nanoparticles on the film surface. All of these results indicated that the p-HAP/PLLA nano-composites might have a promising medical application in bone repair and in bone tissue-engineering.

  • For the purpose of investigating the response of the periodontal tissue in relation to the experimental tooth movement, the rats were inhibited from collagen formation by adminstration of aminoacetonitrile. Findings were as follows: 1) In experimental group, the principal fibers of the periodontal ligament came to run parallel along the alveolar bone or root surface on the pressure side, while the decrease in density, irregular arrangement, and partial loss of principal fibers were observed on the tension side. 2) Sharpey's fibers at the alveolar bone decreased in number, and as the aminoacetonitrile administration continued, the capability of matrix formation decreased on the tension side, the narrowing of the alveolar septum and poor-bony trabeculation appeared on the pressure side. 3) In cementum, Sharpey's fibers were distributed irregularly. The formation of acellura cementum was decreased on the tension side, while the formative capability of cellular cementum was increased. 4) The degree of staining by Herovici technique decreased in the periodontal membrane. By PAS and ninhydrine -Schiff reaction it was appeared weakly positive in the region where the Sharpey's fibers existed.

Effect of Cynandione A of Cynanchi Wilfordii Radix in RANKL and Lipopolysaccharide-induced on Osteoclastogeneis in RAW 264.7 Cells (백하수오(Cynanchi Wilfordii Radix)의 Cynandione A가 RAW 264.7 세포에서 RANKL과 LPS로 유도된 파골세포형성에 대한 영향)

  • Cynanchi wilfordii Radix roots have been utilized as traditional medicine for variety of diseases including diabetes mellitus, aging progression and scavenging free radicals, enhancing immunity, reducing high serum cholesterol, and anti-tumor activity. However, the mechanisms underlying this effect remain poorly understood. The principal objective of this study was to determine the effect of cynandione A on osteoclast cells. Thus, we was isolated cynandione A from Cynanchi wilfordii Radix roots and evaluated the effect of cynandione A on receptor activator of nuclear factor-${\kappa}B$ ligand (RANKL)-induced osteoclast differentiation. We found that cynandione A significantly inhibited osteoclast differentiation stimulated-RANKL in RAW 264.7 cells. Cynandione A conspicuously inhibited the mRNA and protein expression of matrix metallopeptidase 9 (MMP-9), tartrate-resistant acid phosphatase (TRAP) in cynandione A treated with RANKL. Taken together, our results demonstrated that Cynanchi Wilfordii Radix may be useful treatment option of bone-related disease such as osteoporosis leads to fracture of bone and rheumatoid arthritis.

Ecklonia cava Extract Containing Dieckol Suppresses RANKL-Induced Osteoclastogenesis via MAP Kinase/NF-κB Pathway Inhibition and Heme Oxygenase-1 Induction

  • Ecklonia cava, an edible marine brown alga (Laminariaceae), is a rich source of bioactive compounds such as fucoidan and phlorotannins. Ecklonia cava extract (ECE) was prepared using 70% ethanol extraction and ECE contained 67% and 10.6% of total phlorotannins and dieckol, respectively. ECE treatment significantly inhibited receptor activator of nuclear $factor-{\kappa}B$ ligand (RANKL)-induced osteoclast differentiation of RAW 264.7 cells and pit formation in bone resorption assay (p <0.05). Moreover, it suppressed RANKL-induced $NF-{\kappa}B$ and mitogen-activated protein kinase signaling in a dose dependent manner. Downregulated osteoclast-specific gene (tartrate-resistant acid phosphatase, cathepsin K, and matrix metalloproteinase-9) expression and osteoclast proliferative transcriptional factors (nuclear factor of activated T cells-1 and c-fos) confirmed ECE-mediated suppression of osteoclastogenesis. ECE treatment ($100{\mu}g/ml$) increased heme oxygenase-1 expression by 2.5-fold and decreased intercellular reactive oxygen species production during osteoclastogenesis. The effective inhibition of RANKL-stimulated osteoclast differentiation and oxidative stress by ECE suggest that ECE has therapeutic potential in alleviating osteoclast-associated disorders.
