Osteoclasts are bone-resorbing giant cells that differentiate from hematopoietic cells of the monocyte/macrophages. Excessive osteoclast differentiation leads to gradual loss of bone mass causing fracture of the skeleton. The aim of this study was to develop a drug candidates for the treatment of osteoporosis. RANKL-induced osteoclast differentiation was dose-dependently inhibited by myricetin. Myricetin inhibited the expression of c-Fos, NFATc1, and TRAP in BMMs treated with RANKL. Myricetin disrupted the structure of actin ring and suppressed osteoclastic bone resorption. Also, myricetin induced apoptosis in mature osteoclasts. Myricetin inhibited the phosphorylation of ERK in mature osteoclasts treated with M-CSF. The activation of caspase-9 and caspase-3 was increased by myricetin treatment. Our results suggest that myricetin may be an effective agent to prevent bone diseases such as osteoporosis.

This study examined the effect of cancellous bone microstructure on stress distribution within a premolar tooth and a dental implant against mastication force by the micro-finite element method (FEM). The mandibular specimen including a premolar was obtained from a cadaver and scanned with micro-CT to obtain CT images. FE models were reconstructed from CT images at mid-sagittal plane of the tooth. Six models were generated and analyzed for different structure and density in cancellous bone. Stress distributions fur each implant (or tooth) and the surrounding bone were compared. The study indicated that the microstructure of cancellous bone should be considered in finite element analysis to produce reasonable results and thus implant systems with high success rate.
Trabecular bone can be accurately represented using image-based finite element modeling and analysis of these bone models is widely used to predict their mechanical properties. However, the choice of thresholding technique, a necessary step in converting grayscale images to finite element models which can thus significantly influence the structure of the resulting finite element model, is often overlooked. Therefore, we investigated the effects of thresholding techniques on micro-computed tomography (micro-CT) based finite element models of trabecular bone. Three types of thresholding techniques were applied to micro-CT images of trabecular bone which resulted in three unique finite element models for each specimen. Bone volume fractions and apparent moduli were predicted for each model and compared to experimental results. Our findings suggest that predictions of apparent properties agree well with experimental measurements regardless of the choice of thresholding technique in micro CT images of trabecular bone.
Composites of hydroxyapatite (HAp) and chitosan (CS) have been successfully used in bone healing in humans and animals. However, the characteristics of HAp and CS are different. Therefore, the effects of HAp/CS composites on canine bone formation could differ according to their ratio. This study investigated the therapeutic effects of different contents ratios (100, 80:20, 60:40 wt%) on bone defects in a canine model. Thirty intrabony cylindrical defects were created in the humeruses and femurs of 5 beagle dogs, and then the defects were implanted with different composites. The evaluations were performed using radiographs obtained at 10 weeks post-surgery and by histological findings. In radiographic evaluation including the grades of bone filling, periosteal and endosteal reactions, pure hydroxyapatite composite had a significant effect on bone filling, and chitosan containing the composites showed vigorous responses at the periosteum and endosteum. In histological findings, the defect implanted with pure hydroxyapatite had healed completely into mature bony tissue with an obvious osteon structure, and the defect implanted with chitosan containing the composites had the amount of fibrous connective tissue increased significantly within the cortical bone tissue. The results indicate that hydroxyapatite/chitosan composites are therapeutically useful, promoting effective bone healing in defects when the ratio of hydroxyapatite is high and enhanced fibrous connective tissue formation at the periosteum as more chitosan is added.
Bone properties are one of the key components when constructing models that can simulate the mechanical behavior of a mandible. Due to the complexity of the structure, the tooth, ligaments, different bones etc., some simplifications are often considered and bone properties are one of them. The objective of this study is to understand if a simplification of the problem is possible and assess its influence on mandible behavior. A cadaveric toothless mandible was used to build three computational models from CT scan information: a full cortical bone model; a cortical and cancellous bone model, and a model where the Young's modulus was obtained as function of the pixel value in a CT scan. Twelve muscle forces were applied on the mandible. Results showed that although all the models presented the same type of global behavior and proximity in some locations, the influence of cancellous bone can be seen in strain distribution. The different Young's modulus defined by the CT scan gray scale influenced the maximum and minimum strains. For modeling general behavior, a full cortical bone model can be effective. However, when cancellous bone is included, maximum values in thin regions increase the strain distribution. Results revealed that when properties are assigned to the gray scale some peaks could occur which did not represent the real situation.

Objectives: We compared changes in fractal dimension (FD) and grayscale value (GSV) of peri-implant alveolar bone on digital panoramic radiography (DPR) and cone-beam computed tomography (CBCT) immediately after implant surgery and 12 months postoperative. Materials and Methods: In this retrospective study, 16 patients who received posterior mandibular area dental implants with CBCT scans taken about 2 weeks after implantation and one year after implantation were analyzed. A region of interest was selected for each patient. FDs and GSVs were evaluated immediately after implant surgery and at 12-month follow-up to examine the functional loading of the implants. Results: There were no significant differences between DPR and CBCT measurements of FD values (P>0.05). No significant differences were observed between FD values and GSVs calculated after implant surgery and at the 12-month follow-up (P>0.05). GSVs were not correlated with FD values (P>0.05). Conclusion: The DPR and reconstructed panoramic CBCT images exhibit similar image quality for the assessment of FD. There were no changes in FD values or GSVs of the peri-implant trabecular bone structure at the 12-month postoperative evaluation of the functional loading of the implant in comparison to values immediately after implantation. GSVs representing bone mass do not align with FD values that predict bone microstructural parameters. Therefore, GSVs and FDs should be considered different parameters for assessing bone quality.
Dental implant may be immediately placed in postextraction socket which has alveolar bone defect. The purpose of this study was to compare the bone regeneration and bone quality around defects adjacent to implants that were placed into extraction sockets according to EFEB, GTAM barrier and GTAM barrier with DFDB. Mandibular P2, P3 and P4 were extracted bilaterally in dogs, and buccal defects were created about 4mm in depth and 3.3mm in width. Screwed pure titanium implants, 3.8mm in diameter and 10mm in length, were placed into the extraction sockets. The experimental groups were divided into four groups : the G group was covered with a GTAM barrier on the defective area, the D+G group was filled with DFEB and covered with a GTAM barrier, the D group was filled with DFDB only and the control group was sutured without any special treatment on the defective area. The experimental animals were killed after 12 weeks and specimens were prepared for light microscopic evaluation and fluorescent dyes were administered daily for 2 weeks after implantation, and injected on the 4th and 11th week for fluorescent microscopic examination to observe new bone formation and bone remodeling. The new Bone height of the buccal defect was measured and compared with the another for bone gain and the removal torque for the implant was measured for the comparison of bone density and bone-implant osseointegration. Results obtained were as follows : 1. Experimental groups showed bone regeneration in oder from D+G, G, D group and control. D+G and G group was significantly from D group and control(P<0.01). 2. In the defective area of control the regenerated alveolar bone showed poorly developed lamellated structure and fibrous tissue intervention into the bone-implant interface but the others showed well developed lamellated structure and osseointegration. 3. All implant groups showed no significaant difference in the removal torque for implant(P>0.05) These results suggest that immediate implants placed in defective sockets were successfully osseointegrated and utilizing placed in defective sockets were successfully osseointegrated and utilizing not only the combination of GTAM and DFDB but also only the GTAM was favorable for the predictable regeneration of the defective area.

Nanoporous bioactive glass(NBG) ceramic with well interconnected pore structures were fabricated bytriblock copolymer templating and sol-gel techniques. Hierarchically porous BGbeads were also successfully synthesized by controlling the condition of solvent.The beads have hierarchically nano- and macro-pore structure with a sizesbetween several tens nanometers and several hundred micrometers. Both NBG andBG beads show superior bone-forming bioactivity and good in vitrobiodegradability. Biocompatibility both in vitro and in vivo were examed andwas revealed that it largely relies on the pore morphology as well ascomposition. Our synthetic process can be adapted for the purpose of preparingvarious bioceramics, which have excellent potential applications in the fieldof biomaterials such as tissue engineering and drug storage.

Graft copolymerized polycarboxylate(PC)-type superplasticizers(PCs) which have carboxylic acid with $\pi$ bond among the molecular structure and polyethyleneglycol methyl ether methacrylate(PMEM) were synthesized by free radical reaction and investigated the chemical structure, polymerization condition, and physical and chemical properties. Also, the effects of PCs in the dispersion, adsorption and hydration of cement were evaluated. As the molecular weight of graft chain decreases, the adsorption amount on cement particles increased. It was advantageous for the flow to reduce molar ratio, the lower the side bone molecular weight, and increase the molar ratio, the larger the side bone molecular weight. The hydration reaction speed was highly delayed at day 1, due to increase in molar ratio and reduction in side bone molecular weight, but it was recovered in the days after.
Many of the problems which are faced to the edentulous patients are related to a minimal amount of available mandibular bone volume and height. Most of the patients with mild atrophy of the mandible are treated using endosseous implant prosthodontics. TMI(Trans-mandibular Implant) can be used in case of severe mandibular atrophy, poor bone quality, atrophy of the mandible with exposed of inferior alveolar nerve, osteoporosis and the fracture of the atrophic mandible. Also it can eliminate the need for bone grafting and vestibuloplasty. The TMI is a rigid box frame structure which controls and distribute the masticatory force over the severely resorbed mandible. The box frame structure consist of a superstructure, baseplate, 4 transosseous posts, and 5 cortical screws. This is a case report that also describes about the transmandibular implant reconstruction system.
