Effect of Dietary Protein Level on Ca Efficiency in Bone Mineral Density in Growing Rats (식이단백질량이 성장기 흰쥐가 골밀도에 대한 칼슘효율에 미치는 영향)

  • This study was performed to evaluate the effect of dietary protein level on Ca efficiency in bone mineral density in growing male rats. Twenty male rate were divided into two groups. The rats in one group were fed on casein 20% diet as control group and the others were fed on casein 40% diet as protein group. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. The total body, spine and femur bone mineral density and bone mineral content were measured using dual energy-x ray absorptiometry. Urinary calcium, phosphate, pyridinoline and creatinine, serum calcium, phosphate, total protein, albumin, alkaline phosphatase(ALP) and osteocalcin were measured. Urinary Ca excretion, pyridinoline and crosslinks value and serum ALP content seem to be increased in high protein group. It appears that the growing rats in high protein group had a higher bone resprption and bone formation than those in control group. Animal fad a high protein diet had a siginficantly higher Ca efficiency in BMD, BMC of total body, spine and femur. The results of this show that increasing of dietary protein level (40%) is beneficial of improvement of Ca efficiency during growing period.

Effects of plant-derived natural products on inflammatory bone destructive disease

  • Rheumatoid arthritis, osteoarthritis, and periodontal disease are bone destructive diseases mainly caused by inflammation. Various studies are being conducted to develop treatments for inflammatory bone destructive diseases. Many of these studies involve plant-derived natural compounds. In these studies, cell differentiation, signal transduction pathways, and bone resorption were measured at the cellular level. In disease-induced animal models, the amount of inflammatory mediators or matrix destructive enzymes and serum metabolic markers were measured. This study examined the effects of plant-derived natural compounds, such as flavonoids, on inflammatory bone destructive diseases. In addition, we structurally classified various substances used to maintain bone health and summarized the biological effects and related mechanisms of the components.

The Association between Changes in Food and Nutrient Intakes and Changes in Bone Metabolic Indicators in Postmenopausal Women with Osteopenia after a 12-week Intervention of Nutrition Education and Aerobic Exercise (폐경 후 골감소증 여성에 대한 12주간의 영양교육과 운동 중재 전.후 식품 및 영양소 섭취량 변화와 골밀도 지표 변화와의 관계)

  • Few studies investigated the effects of nutrition education and exercises in women with osteopenia. This study examined the relationship between changes in dietary intakes and changes in indicators related to bone health in postmenopausal women with osteopenia (-2.5 ${\leq}$ T-score ${\leq}$ 1) after a 12-week intervention. Thirty-one postmenopausal women aged > 50 years residing in Seoul were recruited and participated in nutritional education regarding bone health and general nutrition practices and aerobic exercises (three times a week; 60 min per session). Twenty-five subjects completed the study and were eligible for the analysis. Bone mineral density (BMD) at femoral neck was measured by dual energy x-ray absorptiometry. Serum calcium, osteocalcin, and intact parathyroid hormone (PTH) were also measured. Dietary intake was estimated by using a one-day 24 recall by a clinical dietitian. After 12 weeks, meat consumption increased (P = 0.028) but vegetable intake decreased (P = 0.005). Intakes of animal protein (P = 0.024), vitamin B1 (P = 0.012) and vitamin $B_2$ (P = 0.047) increased, and sodium intake decreased (P = 0.033). Intact PTH (P = 0.002) decreased and osteocalcin (P = 0.000) increased, however, BMD decreased (P = 0.000). Changes in mushroom consumption were positively correlated with femoral neck BMD (r = 0.673, P = 0.003). Changes in animal iron intake were negatively correlated with intact PTH (r = -0.488, P = 0.013) but were positively correlated with osteocalcin (r = 0.541, P = 0.005). These results suggested that the association between animal iron intake and biochemical markers of bone turnover may play an important role in bone metabolism. Further studies are needed to shed light on complicated mechanisms of diet, hormonal levels of bone metabolism, and bone density.

  • This mouse calvarial defected model is frequently used for new scaffold development in the bone regeneration. Most experiments are carried out in this way by measuring the bone regeneration of mouse calvaria defected area. As a next step, hematoxylin and eosin staining is analyzed by sacrificing mice On the other hand, the quantitative analysis for bone regeneration is carried out by micro computed tomography. However, there are several drawbacks with the micro computed tomography. That is, it takes a long time and it is quite expensive for bone regeneration quantitative analysis. This study was performed by simply measuring the quantity of bone regeneration in mouse clavaira defected area on two-dimensional digital x-ray images via Image J. Consequentially, this experimental method by using J program might help bio-technologist researcher regarding new bone regeneration by comparing the quantity of bone regeneration quickly and precisely as well.

Association of Sasang Constitutional Type with Bone Mineral Density, Osteopenia, and Osteoporosis (사상체질과 골밀도, 골감소증, 골다공증과의 연관성)

  • Object Although Taeeum and Soyang constitutional types have bigger body shapes and higher body mass index values than those with the Soeum, the relationship between the Sasang constitutional type and bone mass density is controversial and the association of osteopenia and osteoporosis remains unknown. Therefore, we investigated the relationship between bone mineral density, osteopenia, and osteoporosis according to Sasang constitutional type. Methods A total of 2,508 participants were included in this study. Among the study participants, 1,396 had Taeeum type, 276 had Soeum type, and 836 had Soyang type, respectively. The relationships to bone mass density, osteopenia, and osteoporosis in those with Sasang constitutional type were estimated using logistic and linear regression models. Results Bone mass density was significantly higher in the order of Taeeum, Soyang, and Soeum group (p < 0.01). Soeum group in comparison with Taeeum or Soyang group was significantly associated with a high odds ratio for osteopenia and osteoporosis except in the hip and femoral neck in the comparison of Taeeum and Soeum group (p < 0.01). Moreover, the bone mass density of Soeum group decreased more rapidly as the age increased when compared with Taeeum and Soyang group. Conclusions Our findings may contribute to the early prevention and management of high-risk individuals with poor bone mass density, osteopenia, and osteoporosis using Sasang constitution medicine.

Effect of Soybean Intake on Bone Mineral Density and Bone Turnover Markers in Postmenopausal Women (콩 섭취가 폐경 후 여성의 골밀도 및 골대사 지표에 미치는 영향)

  • Objective: The purpose of this study was to evaluate the effects of soybean intake on bone mineral density and bone turnover markers in postmenopausal rural Korean women. Method: This study was carried out during nine months from Oct. 25 2004 to Aug. 31 2005. The subjects of this study were female patients over 50 living in rural areas diagnosed with osteoporosis. There were 18 women in the experimental group and 20 in the control group. In this study, the experimental group received 100 mg of isoflavone (soybean) and calcium 1,500 mg for nine months while the control group received 1,500mg of calcium only. Results: After the soybean intake, the change of bone mineral density between the experimental group and control group was statistically significant. However, the bone turnover markers of osteocalcin and deoxypyridinoline between the experimental group and control group were not significantly different statistically. In the Pearson Correlation between bone mineral density and bone turnover markers, the osteocalcin and deoxypyridinoline of the experimental group had a positive correlation, and osteocalcin and DPD/osteocalcin ratio had anegative correlation. In the control group, osteocalcin and DPD/osteocalcin ratio had a negative correlation. Conclusions: This result showed that soybean intake changed bone mineral density in postmenopausal woman.

Effect of Sodium Chloride Supplementation on Bone Metablism in Rats Consuming a Low Calcium Diet (저칼슘 식이 섭취시 식염첨가가 흰쥐의 골격대사에 미치는 영향)

  • This study was performed to evaluate the effect of sodium cholride supplementation on bone metabolism in female rats consuming a low calcium diet. Twenty five female rats were divided into three dietary groups (control Na : 0.1038%, 1% Na : 1.036%, 2% Na : 2.072%). All experimental diets contained 0.27% Ca and were fed to rats with deionized water for 7 weeks. Bone mineral density(BMD) and bone mineral content(BMC) of total body, spine and femur were measured using energy x-ray absorptiometry(DEXA) by small animal software. Then Ca efficiency was calculated from BMD and BMC. Serum Ca, P, Na and urine Ca, P, Na were determined. Urinary pyridinoline, serum ALP were measured to monitor bone resorption. Following 7 weeks, sodium cholride supplemented groups had higher urinary Ca excreteion, urinary pyridinoline, crosslinks value and serum ALP. There was no significant difference in case of serum Ca among all groups. Sodium chloride supplemnted groups had lower Ca effciency of total, spine and femur BMD and BMC than that of control group. In conclusion high salt intake not only increases urinary Ca excretion as urinary Na excretion does but also increase bone resorption and decrease Ca efficiency of each bone. It is been suggested that high salt intake may be harmful for bone maintenance. Therfore, the decrease of salt intake to the level of recommendation would be desirable.

The Effects of Dietary Ca Levels on Ca and Skeletal Metabolism in Ovariectomized Rats of Different Age (난소를 절제한 나이가 다른 흰쥐에서 식이 칼슘 수준이 골격 대사에 미치는 영향)

  • To investigate the effect of dietary Ca levels on metabolic changes of Ca and skeleton in postmenopausal women, 10-month-old ovariectomized female rats were compared with 2 month old rats. The rats were fed either 0.2% or 1.2% Ca diets for 16 weeks. Food intake and weight gain as higher in rats fed high Ca diets and in ovariectomized rats. Apparent Ca absorption as higher, and Ca balance was lower in the low Ca groups. Vertebrae density was higher in old rats or those fed a high Ca diets. The old rats and ovariectomized rats showed decreased bone formation, increased bone resorption and kidney function deterioration resulting in increased urinary Ca excretion. Contradictory to the above observation, old rats and ovariectomized rats still showed higher bone mass and bone ash content. Therefore aging was not fully onging in 10-month-old rats. Bone weights, mineral contents, and mineral/wt ratio were lower in ovariectomized rats. Dietary Ca level did not affect urinary Ca excretion, urinary protein excretion, GFR, serum alkaline phosphatase, or urinary hydroxyporline excretion. This means that dietary Ca level did not influence kidney function or bone turnover. However Ca content and the ash content of femur, 4th vertebra, and scapula were increased in high Ca groups. Therefore, it is considered that decreased bone formation and accelerated bone resorption may account for the increased osteoporotic risk in women in menopause after middle age. However, Ca metabolism can be improved and bone components can be maintained if Ca is supplemented.

Biodegradable Hydroxyapatite/Chitosan Composites on the Bone Defect of Canine Model

  • Composites of hydroxyapatite (HAp) and chitosan (CS) have been successfully used in bone healing in humans and animals. However, the characteristics of HAp and CS are different. Therefore, the effects of HAp/CS composites on canine bone formation could differ according to their ratio. This study investigated the therapeutic effects of different contents ratios (100, 80:20, 60:40 wt%) on bone defects in a canine model. Thirty intrabony cylindrical defects were created in the humeruses and femurs of 5 beagle dogs, and then the defects were implanted with different composites. The evaluations were performed using radiographs obtained at 10 weeks post-surgery and by histological findings. In radiographic evaluation including the grades of bone filling, periosteal and endosteal reactions, pure hydroxyapatite composite had a significant effect on bone filling, and chitosan containing the composites showed vigorous responses at the periosteum and endosteum. In histological findings, the defect implanted with pure hydroxyapatite had healed completely into mature bony tissue with an obvious osteon structure, and the defect implanted with chitosan containing the composites had the amount of fibrous connective tissue increased significantly within the cortical bone tissue. The results indicate that hydroxyapatite/chitosan composites are therapeutically useful, promoting effective bone healing in defects when the ratio of hydroxyapatite is high and enhanced fibrous connective tissue formation at the periosteum as more chitosan is added.

The effect of fibroblast growth factor receptor inhibition on resistance exercise training-induced adaptation of bone and muscle quality in mice

  • Aging in mammals, including humans, is accompanied by loss of bone and muscular function and mass, characterized by osteoporosis and sarcopenia. Although resistance exercise training (RET) is considered an effective intervention, its effect is blunted in some elderly individuals. Fibroblast growth factor (FGF) and its receptor, FGFR, can modulate bone and muscle quality during aging and physical performance. To elucidate this possibility, the FGFR inhibitor NVP-BGJ398 was administrated to C57BL/6n mice for 8 weeks with or without RET. Treatment with NVPBGJ398 decreased grip strength, muscular endurance, running capacity and bone quality in the mice. FGFR inhibition elevated bone resorption and relevant gene expression, indicating altered bone formation and resorption. RET attenuated tibial bone resorption, accompanied by changes in the expression of relevant genes. However, RET did not overcome the detrimental effect of NVP-BGJ398 on muscular function. Taken together, these findings provide evidence that FGFR signaling may have a potential role in the maintenance of physical performance and quality of bone and muscles.
