Effect of Dietary Oil Containing $\gamma$-Linolenic Acid on the Plasma Lipid Levels and Thrombotic Activity in Rats (감마-리놀렌산을 함유하는 식이지방이 흰쥐의 혈전작용 및 혈액지질 수준에 미치는 영향)

  • This study was carried out to determine the effect of dietary ${\gamma}$-linolenic acid on decreasing the plasma lipid levels and the thrombotic activity in rats. Sprague-Dawley male rats (B.W 120 g) were fed a experimental diet containing 5% lard (46.05% saturated fatty acids) , corn oil (51.36% linoleic acid) , evening primrose oil (EPO,72.80% linoleic acid and 9.16% ${\gamma}$-linolenic acid) or borage oil (BO,40.29% linoleic acid and 24.25% ${\gamma}$-liolenic acid) for 30 days. Although there were no significant differences in the food intake among the groups, the body weight gain of the BO group was significantly lower than that of the other groups. The bleeding time of the BO group was significantly longer than that of the other groups. There were significantly differences in the whole blood clotting time among the groups except for the EPO and corn oil groups, where the whole blood clotting time of the BO group was the highest among the groups, and that of the lard group was the lowest. The plasma triacyglyceride (TAG) , total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) concentrations were the lowest in the BO group, but highest in the lard group, and there were significant differences among the groups. The plasma HDL-C concentrations were in the following order: BO, EPO, corn oil and lard groups and there were significant differences among the groups. The excretions of fecal neutial steroids and acidic steroids of the BO group were the highest among the groups, and there were significant differences compared to the other groups. The results suggest that dietary EPO and BO containing ${\gamma}$-linolenic acid has an antithrombotic activity, and inhibits the increasing of plasma TAG, TC and LDL-C concentrations compared to lard, which contains saturated fatty acids, or corn oil, which contains linoleic acid.

Relationship Between Plasma, Erythrocyte Membrane, and Dietary Intake Levels of $\omega$-3 Fatty Acids in Young Korean Females : Effect of Diet Survey for Two Months (한국 성인 여성의 혈장 및 적혈구막의 $\omega$-3 지방산함량과 2개월간의 식이섭취와의 관계)

  • This study was conducted to asses the $\omega$-3 fatty acid intake levels and to evaluate the relationship between the levels of $\omega$-3 fatty acids in plasma and erythrocyte membrane and the dietary intake of these fatty acids over the period of two months in 56 young Korean females. Dietary survey was conducted to obtain 7-day weiged records and six 24-hour recalls. Fasting blood sample was collected from each subject after the dietary survey period. Mean daily intakes of energy, fat, and cholesterol were 1,569㎉, 41.8g, and 217mg, respectively. Fat suppled 24% of total energy intake. Mean daily intake levels of eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA), and total $\omega$-3 fatty acids were 0.04g, 0.06g, and 0.48g, respectively. Plasma cholesterol levels of most of the subjects were within normal range, and there was no significant correlation between plasma cholesterol levels and intake levels of any specific fatty acid. Levels of EPA, DHA, EPA+DHA in plasma and erythrocyte membrane, but not the levels of $\alpha$-linolenic acid(LNA), were significantly correlated with dietary in take of respective fatty acids. Such a correlation, however, was not observed in a previous study where dietary intake was assessed for 3 days. The results of this study show that dietary intakes of $\omega$-3 fatty acids are low in the subject and that about two-month period is required to assess dietary intake levels of $\omega$-3 fatty acids with a reasonable accuracy.

Plasma LDL Particle Sizes Affect the Blood Lipid Profile and Dietary Intakes among Korean Adults (한국 성인에서 혈중 LDL 입자의 크기에 영향을 미치는 관련 인자 연구)

  • The purpose of this research was to examine the relationship between the plasma LDL particle size and blood lipid profile, dietary factors and anthropometric values (body mass index, waist circumference and waist/hip ratio). The subjects were 173 adults aged 23 to 81 years, selected from the Outpatient Clinic and Cardiovascular Department of the Seoul Municipal Hospital. Dietary data were obtained using a 3-day food record and analyzed using Korean and US nutrient databases. The subjects were divided into three groups by LDL particle size : type A (large buoyant LDL, > 25.5 nm, n=96), type I (Intermediate LDL,$25.2\leq-\leq25.5$ nm, n=18), and type B (small dense LDL, < 25.2 nm, n=59) groups. The type B group had higher age, waist circumference, and waist/hip ratio (WHR) than the type A and type I groups. Serum concentration of triglyceride, Apo B, LDL/HDL cholesterol ratio and atherogenic index were significantly higher in the type B group as compared to those in the other two groups. HDL cholesterol level and Apo A-I/Apo B ratio were significantly lower in the type B group than the other two groups. The plasma LDL particle size was highly correlated with triglyceride (r= -0.450), Apo B (r= -0.402) and HDL cholesterol (r= 0.418). However, there was no correlation between plasma LDL particle size and dietary intakes. This study showed that small dense LDL was an important biochemical risk factor that was associated with other risk factors.

Effect of hCG on TeBG (hCG가 TeBG에 미치는 영향)

  • In the previous experiment, authors have shown that during the latter half of estrous cycle there was an increase in plasma testosterone level in the rats stimulated with hCG. To determine the physiologic significance of elevated plasma testosterone, changes of the plasma concentrations of TeBG and testosterone following hCG stimulation were analyzed in the rats having a regular 5 day cycle. The rats were divided into three groups; the control, the rats stimulated with single hCG on the day of proestrus and stimulated with hCG throughout the entire cycle. Blood samples were obtained once a day for an estrous cycle and analyzed for the binding capacity of TeBG using ammonium sulphate precipitation method and testosterone concentration by means of radioimmunoassay. Followings were the results; 1) There was no significant variation in the binding capacity of TeBG in peripheral blood during the estrous cycle of the control rats. 2) No cyclic variation in the binding capacity of TeBG was observed in the rats stimulated with single hCG on proestrus. although the levels tended to be higher in the rats with stimulation than in the control rats. 3) Continual stimulation of hCG produced a marked increase in the binding capacity of TeBG especially on the day of metaestrus. 4) The changes in the plasma level of testosterone followed the same basic pattern seen in the TeBG binding capacity. 5) From above results, the followings were suggested. a. hCG related increase of the binding capacity of TeBG is probably secondary to a modest increase in estrogen as well. b. hCG related increase of plasma testosterone in female rats is not entirely due to excess production rather in part due to decreased metabolism induced by the rise in TeBG. c. It seems likely that most of elevated testosterone shown in the rat stimulated with hCG is bound to TeBG and only small portion is unbound form which influence cellular activity. It is rather possible that an increase in TeBG could augment estrogen activity.

Stability in Plasma and Intracellular Uptake of Thermally Denatured Protein-coated anionic Liposomes (열변성 단백질이 결합된 음이온성 리포솜의 혈장 내 안정성 및 세포 내 이입 평가)

  • Liposomes have been used as one of the efficient carriers for drug delivery. In this study, anionic liposomes of which surface was modified by using both electrostaic interaction between anionic liposomes and cationically charged BSA molecules at lower pH than isoelectric point (pI) of BSA and denaturation of the BSA-coated liposomes by thermal treatment. The thermally denatured BSA-coated liposomes (DBAL) had mean particle diameter of 125.2${\pm}$1.7 nm and zeta potential value of -22.4${\pm}$4.5 mV. Loading efficiency of model drug, doxorubicin (DOX), into liposomes was 83.0${\pm}$2.6%. Results of in vitro stability study of DBAL in blood plasma showed that the mean particle diameter of DBAL 400 did not increase in blood plasma and adsorption of plasma protein was much less than plain or anionic liposomes. Intracellular uptake of DBAL 400 evaluated by confocal microscopy observation was higher than that of PEG liposomes.

  • Ovulation of maturing female yellow puffer, Takifugu obscrus, was induced by using single injection of human chorionic gonadotropin (HCG) or gonadotropin releasing hormone-analogue (GnRH-A) $des-Gly^{10}[D-Ala^6]$ GnRH-ethylamide plus pimozide. The response was evaluated using the fertilization and embryo-formation rate after insemination and the gonadotropin (GTH) level in blood plasma using radioimmunoassay. In the fertilization and embryo-formation, maximal effects were recorded by using 1,000 IU/kg HCG or $10\;{mu}g/kg$ GnRH-A plus 5 mg/kr pimozide. Pimozide (1, 5 mg/kg) or GnRH-A treatment alone was not effective in elevation of GTH level, however combinations of these treatments were particularly effective. Injection of dopamine blocked the rapid elevation of plasma GTH levels of blood. These data suggest that yellow puffer secrete GnRH and gonadotropin-releasing-inhibiting factor during the spawning or the other period.

Systemic Expression of Vascular Endothelial Growth Factor in Patients with Cerebral Cavernous Malformation Treated by Stereotactic Radiosurgery

  • Objective : Increased expression of angiogenic factors, such as vascular endothelial growth factor (VEGF), is associated with the pathogenesis of cerebral cavernous malformations (CCMs). The purpose of this study was to investigate plasma levels of VEGF in normal subjects and in patients with CCM and to evaluate change in these levels following stereotactic radiosurgery (SRS). Methods : Peripheral venous blood was collected from 6 patients with CCM before SRS using Gamma Knife and at the 1 week, 1 month, 3month, and 6 month follow-up visits. Plasma VEGF levels were measured using commercially available enzyme-linked immunosorbent assay kits. Peripheral blood samples were obtained from 10 healthy volunteers as controls. Results : Mean plasma VEGF level of 41.9 pg/mL (range, 11.7-114.9 pg/mL) in patients with CCM at baseline was higher than that of the healthy controls (29.3 pg/mL, range, 9.2-64.3 pg/mL), without significant differences between CCM patients and controls (p=0.828). Plasma VEGF level following SRS dropped to 24.6 pg/mL after 1 week, and decreased to 18.5 pg/mL after 1 month, then increased to 24.3 pg/mL after 3 months, and 32.6 pg/mL after 6 months. Two patients suffering from rebleeding after SRS showed a higher level of VEGF at 6 months after SRS than their pretreatment level. Conclusion : Plasma VEGF levels in patients with CCM were elevated over controls at baseline, and decreased from baseline to 1 month after SRS and increased further for up to 6 months. Theses results indicated that anti-angiogenic effect of SRS might play a role in the treatment of CCMs.

Selenium Status of Healthy Women Consuming Different Diets (식이에 따른 건강한 성인 여자의 셀레늄의 영양 상태)

  • Human blood selenicum(Se) level depends on the Se content and bioavailablilty of foods. In countries where the soil has low Se content, the differences of Se intake and blood Se concentration are shown according to the type of diet. In this study, Se status of women eating an average German diet(175 German healthy women) and wholesome nutrition group(243 women) were studied. There were significant differences in wholeblood and plasma Se levels between the two groups. In average German diet group, mean wholeblood Se concentration was 84.97ug/l and plasma Se concentration was 75.02ug/l. In wholesome nutrition group mean wholeblood Se concentration and plasma Se concentration with Se levels, correlation analysis was conducted between Se concentration and BMI, age and daily food intake. BMI and age did not correlate significantly with wholeblood and plasma Se levels. In wholesome nutrition group, serum and wholeblood Se levels had significant correlation with fish intake.

Effects of Tissue Cultured Ginseng on Blood Glucose and Lipids in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유도 당뇨모델을 이용한 배양인삼 분말의 혈당 강하 및 지질대사의 개선효과)

  • Hypoglycemic and hypolipidemic efficacies of cultured ginseng (CG) in streptozotocin (STZ)-induced diabetic rats were investigated. Male Sprague-Dawley rats were divided into normal and three diabetic groups. The diabetic groups were fed CG-free control diets supplemented with 5 and 10% of CG for 2 weeks. CG-supplemented groups showed significantly lower blood glucose, triglyceride, and total cholesterol content compared to the diabetic control group. CG supplementation at 5% significantly increased plasma high density lipoprotein (HDL)-cholesterol content compared to the diabetic control group. CG significantly increased plasma HDL-cholesterol/total cholesterol ratio and reduced atherogenic index, whereas, it did not affect plasma total lipid content in diabetic rats. The 5% CG reduced plasma AST and ALT activities in diabetic rats and inhibited the reduction of plasma creatinine level caused by STZ-treatment in rats. These data suggest that tissue-cultured ginseng has hypoglycemic, hypolipidemic, and anti-atherogenic effects on STZ-diabetic rats and can be useful as a dietary supplement for the treatment of diabetes.

Comparison of Plasma Proteome Expression between the Young and Mature Adult Pigs

  • Here, we present an approach of blood plasma proteome profiling and their comparisons between the young and the adult pigs as prerequisite for the identification of bio-markers related to the health conditions, growth performance and meat quality. To profile the proteome in porcine plasma, blood samples were collected from 19 young piglets and 20 adult male barrows and the plasma was retrieved. Then, protein profiling was initiated using one and two-dimensional electrophoresis. Proteins were spotted and then identified by MALDI-TOF-TOF and LC-MS-MS. In the results, more than thirty-six and twenty eight protein spots were selected in young piglets and adult pigs, respectively and twenty three proteins were identified. The proteome profile images were compared between those ones using Image Master Version 7.0. The image of expressed proteome showed that most of proteins from plasma of young piglet separated clearly and concentrated in 2DE display compared to ones from adult. Image analysis in detail was carried out to look for the specific proteins related to age progression. It demonstrated that the characteristics of proteome expression could be distinct to their age stages. Further investigations needed to proceed to understand the age dependent change of protein conformation and biological meaning of those differences in proteome expression between young and mature adult pigs.
