Prevalence of Detection of Prohibited Drugs on Doping Tests of Pre- and Post-races in Korea (2002-2013) (국내 경주마 도핑검사에서 금지약물 검출빈도 및 종류(2002-2013))

  • There was no study on the prevalence of doping control of racehorses in Korea. The purpose of this study was to determine prohibited substances in horse races of a drugs testing program. Blood samples were taken from those 298,543 starters prior to racing and the that finished top 3 runners of each race and horses designated by the stewards shall be taken for collection of 91,482 urine samples for the purpose of post-race doping test in Seoul, Busan and Jeju Race Park between 2002 and 2013. Detection and measurement of prohibited substances were carried out by ELISA, GC/MS and LC/MS using standard methods at the Doping Control Center, Korea Racing Authority. Total 0.0030% of pre-races and total 0.0186% of post-races tested positive for prohibited substances. In pre- and post-race, caffeine and ketoprofen were the most detected prohibited substance respectively. We thought that characteristics of pharmacokinetics and pharmacodynamics of drugs resulted in different between plasma and urine. These substances have also been detected with other prohibited drugs suggesting that unintentional feeding or bedding may be the reason and groomers' confusion of candidate horses based by the stewards' research.

  • Bisphenol A (BPA), an environmental endocrine disrupter, enters the human body continuously in food and drink. Young children are likely to be more vulnerable than adults to chemical exposure due to the immaturities of their organ systems, rapid physical development, and higher ventilation, metabolic rates, and activity levels. The direct effect of BPA on peripheral tissue might also be of importance to the development of insulin resistance. However, the influence that BPA has on insulin signaling molecules in skeletal muscle has not been previously investigated. In this study, we examined the effect of BPA on fasting blood glucose (FBG) in post-weaned Wistar rats and on insulin signaling proteins in C2C12 skeletal muscle cells. Subsequently, we investigated the effects of BPA on insulin-mediated Akt phosphorylation in C2C12 myotubes. In rats, BPA treatment (0.1-1,000 ng/mL for 24 hours) resulted in the increase of FBG and plasma insulin levels, and reduced insulin-mediated Akt phosphorylation. Furthermore, the mRNA expression of insulin receptor (IR) was decreased after 24 hours of BPA treatment in C2C12 cells in a dose-dependent manner, whereas the mRNA levels of other insulin signaling proteins, including insulin receptor substrate-1 (IRS-1) and 5'-AMP-dependent protein kinase (AMPK), were unaffected. Treatment with BPA increased GLUT4 expression and protein tyrosine phosphatase 1B (PTP1B) activity in C2C12 myotubes, but not in protein levels. We conclude that exposure to BPA can induce insulin resistance by decreasing IR gene expression, which is followed by a decrease in insulin- mediated Akt activation and increased PTP1B activity.

Methanol extract of Lespedeza maximowiczii var. tricolor Nakai improves glucose metabolism through PPARγ agonist and insulin-mimetic effect in 3T3-L1 adipocytes and db/db mice (삼색싸리 메탄올 추출물의 3T3-L1지방세포와 db/db 마우스에서의 PPARγ 작용제와 인슐린 유사효과를 통한 혈당조절 개선효과)

  • The aim of this study is to investigate the effect of Lespedeza maximowiczii var. tricolor Nakai (LMTN) on glucose metabolism. LMTN extract significantly enhanced the glucose uptake and lipid accumulation in 3T3-L1 adipocytes compared with control. Also, LMTN extract in 3T3-L1 adipocytes significantly increased the protein expression of peroxisome proliferator-activated receptor (PPAR)γ, insulin receptor substrate-1, and glucose transporter (GLUT)4. The regulatory effect on glucose uptake or insulin signal transduction of LMTM extract was lower than troglitazone or pinitol such as the positive control, but increased PPARγ activation. Additionally, LMTM extract has an insulin-mimetic effect. In db/db mice, LMTN extract (250 mg/kg BW) significantly reduced water and food intake, blood glucose, and level of plasma triglyceride and total cholesterol. Furthermore, the expression of PPARã and GLUT4 mRNA in adipose or muscle tissue effectively was increased by oral treatment of LMTN extract. Thus, our results suggest that LMTN extract improves the glucose metabolism through PPARγ and insulin-mimetic effect in 3T3-L1 adipocytes and db/db mice.

Inhibition of Acetylcholinesterase and Butyrylcholinesterase by Phosalone via Bioactivation (Phosalone의 활성화과정을 통한 acetylcholinesterase와 butyrylcholinesterase에 대한 활성 저해)

  • The purpose of this study was to investigate a role of cytochrome $P_{450}$, for the toxicity of the phosalone in in vitro and in vivo bioactivation systems. The bimolecular inhibition rate constants$(k_i)$ of the phosalone to acetylcholinesterase(AChE) and butyrylcholinesterase(BuChE) were approximately $10^2M^{-1}{\cdot}min^{-1}$, respectively, which meant a poor inhibitor. The potency of the phosalone as an inhibitor of AChE and BuChE was increased about 300 and 40 fold, respectively, when the inhibitor and the ChE were incubated with microsomes fortified with NADPH compared with microsome alone. Piperonyl butoxide(PB) addition to these coupled systems greatly reduced the inhibition of both target enzymes by blocking a bioactivation process. The $I_{50}$ value of the Phosalone alone for rat brain AChE was 170 mg/kg. When PB was pretreated, that value was altered to 42.5 mg/kg. PB pretreatment synergized the inhibition of brain AChE with four times. Rat blood erythrocyte AChE and plasma BuChE were similarly inhibited in vivo by the phosalone and PB pretreatment didn't affect significantly the pattern of the inhibition. The in vivo studies showed different results in the role of cytochrome $P_{450}$ from those of the in vitro studies.

Effects of Season and Split-sex Feeding on Performance, Stress and Carcass Quality of Finishing Pigs (계절과 성분리 사육이 비육돈의 생산성, 스트레스 및 도체품질에 미치는 영향)

  • This study was designed to determine the effect of season and split-sex feeding on performance, stress and carcass quality of finishing pigs. Two hundred and seventy cross-bred finishing pigs $(82.1{\pm}0.82kg)$ were randomly divided into six groups in 2 seasons (summer and winter) ${\times}$ 3 split-sex feeding (female only, castrated male and mixed) factorial arrangement of treatments. In winter, feed intake and weight gain of finishing pigs were better than summer (p<0.05). Split-sex feeding was affected the feed conversion, feed conversion of finishing pigs were superior in winter and mixed groups. Plasma concentrations of cortisol as a stress indicators was greater in the mixed groups vs female only and castrated male groups. There were no interactions between the season and split-sex feeding in their effects on these blood variables. The season was affected the carcass characteristics and PSE incidence of the finishing pigs, PSE incidence was low in winter and mixed groups. Meat quality did not show any significant difference due to the difference of season and split-sex feeding of finishing pigs. There was no significant difference in the pH, drip loss and meat color of pork loin between the season and split-sex feeding of finishing pigs. In conclusion, the results of this study suggest that we need more precise on-farm management practice in summer and application of split-sex feeding skills to reduce the stress and improve welfare status of finishing pigs.

Protective Effects of Water Extracts Composed of Adenophora triphylla var. japonica Hara on the Acetaminophen-induced Hepatotoxicity (아세트아미노펜에 의해 유도된 간독성 모델에서 잔대를 주원료로 하는 추출물의 간 보호 효과)

  • The hepatoprotective effects of water extracts composed of Adenophora triphylla var japonica Hara (ATJH) on acetaminophen (APAP)-induced hepatotoxicity were investigated in vivo and in vivo. The effects of ATJH on liver toxicity induced by APAP were assessed by blood biochemical and histopathological analyses. APAP treatment (350 mg/kg) caused severe liver injury in mice as indicated by their significantly elevated plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels. Pretreatment with ATJH for 3 or 7 days attenuated the increases in ALT and AST when challenged with APAP. The reductions in viability caused by high dose of APAP (450 mg/kg) in vivo were reversed by pretreatment with ATJH. These protective effects of ATJH against APAP-induced toxicity were consistent with the results from the histopathological examinations. We next examined the effects of ATJH on the gene expression of glutathione S-transferases (GSTs) that detoxify the metabolic intermediates of APAP in H4IIE cells. The hepatic GST protein levels [$\alpha$ class (GSTA2, GSTA3/5)] were significantly elevated in a dose-dependent manner by ATJH treatment. In summary, ATJH is effective at protecting against APAP-induced hepatotoxicity by GST induction, implying that ATJH should be considered a potential chemopreventive agent.

Radioprotective Effect of Post-treatment with Rutin on γ-Irradiation-induced Cellular Damage in Mice (감마선 조사로 유도된 세포 손상 마우스에서 루틴 투여 후의 방사선 방호 효과)

  • This study aimed to investigate the therapeutic effect of rutin against whole-body ${\gamma}$-irradiation in BALB/c mice. BALB/c mice were randomly divided into four groups and exposed to 6 Gy ${\gamma}$-irradiation. One hour later, mice were orally administered rutin (50 and 100 mg/kg) for seven consecutive days. ${\gamma}$-Irradiation (6 Gy) resulted in cellular damage as manifested by elevated levels of plasma hepatic marker enzymes and lipid peroxidation in liver tissue, accompanied with decreased spleen and thymus indices, and white blood cell count. In addition, ${\gamma}$-irradiation significantly decreased the levels of antioxidant enzymes such as superoxide dismutase, glutathione peroxidase and catalase. Rutin treatment significantly protected against ${\gamma}$-irradiation-induced cellular damage, which was evident by the improvement in the status of most of the investigated parameters. Therefore, rutin has beneficial effects against radiation-induced damage.

Effects of extruded pellet and moist pellet on growth performance, body composition, and hematology of juvenile olive flounder, Paralichthys olivaceus

  • A feeding trial was conducted to evaluate the effects of two different sizes of extruded pellets (EP) ($EP_1$ - 3 mm or $EP_2$ - 5 mm) and a moist pellet (MP) in olive flounder, Paralichthys olivaceus, reared in semi-recirculation system. A total of 450 fish with an average initial weight of $5.0{\pm}0.2g$ (mean ${\pm}$ SD) were fed one of the three experimental diets in triplicate groups. At the end of a 6-week feeding trial, weight gain, specific growth rate, and feed efficiency of fish fed EP diets were significantly higher than those of fish fed MP (P < 0.05). Water quality parameters like turbidity, total ammonia nitrogen, and total phosphorous from tanks of fish fed $EP_1$ and $EP_2$ were significantly lower than those from tanks of fish fed MP. Blood plasma glutamic oxaloacetic transaminase and glucose concentration were significantly higher in fish fed MP diet compared to fish fed EP diets (P < 0.05). Whole body crude protein contents in fish fed EP diets were higher than those from the fish fed MP diet. Whole body amino acid content like threonine, aspartic acid, serine, tyrosine, and cystine were found to be significantly higher in fish fed EP diets than those in fish fed MP diet. In considering overall performance of olive flounder, $EP_2$ diet could be recommended for the successful aquaculture of this important fish species.

A COMPARATIVE STUDY ON THE VITAL SIGN AND BEHAVIOR APPEARANCE DEPENDING ON THE ROUTE OF FLUMAZENIL ADMINISTRATION IN CONSCIOUS SEDATION BY MIDAZOLAM (Midazolam을 이용한 의식진정시 flumazenil의 투여경로에 따른 생징후 및 행동양상의 비교 연구)

  • The purpose of this study was to evaluate the efficacy and safety depending on the route of flumazenil, as an antagonist against midazolam. The subjects of this study were 15 volunteers of $22{\sim}24$ years old. They were sedated with midazolam 0.2mg/Kg intranasal spray, and then 40 minutes after midazolam administration, they were given flumazenil 0.2mg intranasal spray for their reversal. For evaluation of the efficacy and safety of intranasal spray for flumazenil, they were monitored with pulse-oxymeter(Nellcor symphony N-3000, Nellcor Puritan CO. USA) and electric sphygmomanometer (Heartcare 200, National CO. Japan), and were assessed themselves using visual analogue scale(VAS) for tranquilization, sleep, fatigue and attitude. All of these subjects were reduced completely without any undesired situations. The results from this study can be summarized as follows ; 1. Nasaly administered flumazenil using spray device produced much more rapid reduction than intravenously administered flumazenil, but soon after fell in more deep sedated state than intravenously administered flumazenil. 2. There were no considerable side effects or bad influence on vital signs of both nasaly administered flumazenil and intravenously administered flumazenil. These results suggested that the flumazenil administered nasaly using spray device for reversal, we could treat patients safely and effectively under conscious sedation using midazolam administration. But, We will have to research about its optimal dosages for flumazenil, used as intranasal spray for reversal agents against the midazolam by evaluating the blood plasma concentration of midazolam and flumazenil.

Preparation of Cheonggukjang added onion (Allium cepa L.) and its antioxidative activity (양파(Allium cepa L.) 첨가 청국장의 제조 및 항산화능 평가)

  • Yellow and black soybean Cheonggukjangs (YBSC) prepared with an addition of onion (Allium cepa L.) in different contents (0, 5, 10, 20, and 30%, w/w) were manufactured, and the sensory evaluation was carried out. The optimum addition ratio of onion was determined to be 20%. Total phenolic and flavonoid contents of YBSC prepared with an addition of onion were higher than those of the YBSC prepared without an onion. In addition, YBSC prepared with an addition of onion showed higher 1,1-diphenyl-2-picrylhydrazyl radical-scavenging activities than those prepared without an onion. Furthermore, rat blood plasma 1 hour after oral administration of YBSC prepared with an onion was more effective in suppressing the accumulation of cholesteryl ester hydroperoxide than those prepared without onion and control. These results indicated that the consumption of the YBSC prepared with onion may contribute to the antioxidant defense in vivo.
