Dietary Tea Catechin Inclusion Changes Plasma Biochemical Parameters, Hormone Concentrations and Glutathione Redox Status in Goats

  • The beneficial effects of tea catechins (TCs) are related not only to their antioxidant potential but also to the improvement of animal meat quality. In this study, we assessed the effects of dietary TC supplementation on plasma biochemical parameters, hormone responses, and glutathione redox status in goats. Forty Liuyang goats were randomly divided into four equal groups (10 animals/group) that were assigned to four experimental diets with TC supplementation at 4 levels (0, 2,000, 3,000 or 4,000 mg TC/kg DM feed). After a 60-day feeding trial, all goats were slaughtered and sampled. Dietary TC treatment had no significant effect on blood biochemical parameters, however, low-density lipoprotein cholesterol (p<0.001), triglyceride (p<0.01), plasma urea nitrogen (p<0.01), and glucose (p<0.001) decreased and total protein (p<0.01) and albumin (p<0.05) increased with the feeding time extension, and day 20 was the turning point for most of changes. Interactions were found in glutathione (p<0.001) and the ratio of reduced and oxidized glutathione (p<0.05) in whole blood between treatment and feeding time. Oxidized glutathione in blood was reduced (p<0.05) by 2,000 mg TC/kg feed supplementation, and a similar result was observed in longissimus dorsi muscle. Though plasma glutathione peroxidase (p<0.01) and glutathione reductase (p<0.05) activities were affected by treatment and feeding time interactions, and glutathione S-transferases activity increased with feeding day extension, no changed values appeared in longissimus dorsi muscle. In conclusion, dietary TC supplementation affected the concentrations of some blood metabolites and accelerated GSH depletion in the blood of goats. In terms of less high-density lipoprotein cholesterol, the highest insulin and IGF-I concentrations, the highest ratio of reduced and oxidized glutathione in plasma, the dosage of 2,000 mg TC/kg feed might be desirable for growing goats to prevent glutathione depletion and keep normal physiological metabolism.

Studies on the Pharmacodynamic Action of Methemoglobin (Methemoglobind의 약력학적(藥力學的) 작용(作用)에 관(關)한 연구(硏究))

  • For the purpose of stydying the pharmacodynamic action of methemoglobin, the author made the following experiments: 1. Preparation of hemoglobin and methemoglobin solutions: Red cell suspension from rabbit blood was hemolysed with distilled water and then divided into two portions. One portion was dialysed through cellophane paper and made isotonic with the proper amount of sodium chloride. The second portion was treated with sodium nitrite to convert hemoglobin to methemoglobin, dialysed through cellophane paper and made isotonic. 2. The concentration of methemoglobin in solution, plasma and urine was determined by Horecker and Brackette's method, and that of hemoglobin by the cyanmethemoglobin method. 3. The concentration of methemoglobin and hemoglobin in the plasma and urine of rabbits was measured at several intervals of time after infusion of the above samples. 4. The blood pressure and respiration of rabbits were recorded on a kymograph, and the effects of the samples on them were observed. 5. The effects of the samples on the movements of the in-situ heart and the isolated intestine of rabbits were studied. 6. The kidneys of rabbits were excised 4 to 5 hours after injection of the samples, and histopathological examinations were made. These experiments revealed the following results: 1. When methemoglobin solution was allowed to stand in room air, there was no decrease in the concentration of methemoglobin. 2. When methemoglobin solution was mixed with whole blood and incubated at $37^{\circ}C$, the concentration of methemoglobin decteased gradually. 3. After the infusion of methemoglobin and hemoglobin solutions, the rate of disappearance of methemoglobin in the plasma was more rapid than that of hemoglobin in the plasma. The higher the initial concentration in the plasma, the larger was the rate of disappearance of methemoglobin. 4. The rate of disappearance of methemoglobin was exceedingly rapid for 30 minutes after the infusion. 5. The urinary excretion of methemoglobin was more rapid than that of hemoglobin. 6. It would seem that the circulating blood contains substances which are promptly mobilized in the plasma to reduce methemoglobin to hemoglobin. 7. Moderate amounts of methemoglobin solution caused some rise in the blood pressure and a transient acceleration of the respiration of the rabbits. These effects of methemoglobin were milder than those of hemoglobin. 8. The movements of the in-situ heart and the isolated intestine of rabbits were accelerated by methemoglobin. These accelerating effects were milder than those of hemoglobin. 9. In the kidneys of rabbits treated with methemoglobin solution, hyperemia of the glomeruli, cloudy swelling and hemoglobin deposit in the tubular epithelium, hemoglobin casts in the tubular lumina of the proximal tubules, and interstitial congestion were constantly observed. There was no definite difference between the histological findings in the rabbit kidneys injected with methemoglobin, and those injected with hemoglobin solutions.

The Effect of BuOH Fraction of Polygonatum odoratum with Selenium on Blood Glucose Level and Lipid Peroxidation in Streptozotocin Induced Diabetic Rats (둥굴레 분획물과 Selenium이 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질과산화에 미치는 영향)

  • The purpose of this study was to investigate the effect of butanol(BuOH) fraction of Polygonatum odoratum with selenium tr-eatment on blood glucose levels and lipid peroxidations in streptozotocin(STZ) induced diabetric rats. Male Sprague-Dawly rats weighing(180-200g) were divided into five groups : normal STZ-control and three expreimental groups(P, odoratum group P, odo-Se group and Se group) Diabetes mellitus was induced by injection STZ in the tail vein at the dose of 45mg/kg B.W The BuOH fraction of Polygonatum odoratum(500mg/kg. B,W) given orally administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na2Seo3(2mg/kg diet). Diabetic rats showed the lower weight gain compared to the normal rats. the plasma glucose levels of the P. odo-Se group were significantly lower than the other experimental groups. The plasma cholesterol levels were higher in STZ-control and Se groups compared toP.odoratum and P. odo-Se groups and HDL-cholesterol levels were increased in the diabetic experimental groups fed on BuOH fraction of P. odoratum with Se supplementation. The liver and muscle glycogen levels were not significantly differ among all groups. The plasma free fatty acid levels were lower in diabetic experimental groups fed on BuOh fraction of P. odoratum or Se sup-plementation than STZ-control and Se groups. Diabetics rats showed the higher levels of triglyceride in plasma andlower levels in liver compared with the normal group. Supplementation with Se decreased significantly the liver triglyceride level. The MDA levels in liver and kidney were significantly reduced in all the experimental groups. In conclusion administration of BuOH fraction of Polygonatuum odoratum with selenium supplementation reduced blood glucose levels and peroxdative tissue damage in STZ induced diabetic rats showing the possibility of preventiave and therapeutic use of the wild edible plant to the diabetes mellitus.

Effects of Antihypertensive Diets Mainly Consisting of Buckwheat, Potato, and Perilla Seed on Blood Pressures and Plasma Lipids in Normotensive and Spontaneously Hypertensive Rats (메밀, 감자, 들깨를 이용한 항고혈압 기능성 식이가 정상혈압쥐 및 본태성고혈압쥐에서 혈압 및 혈장지질에 미치는 영향)

  • The study was carried out to investigate a new type of functional foods with hypotensive effect which is critical in the prevention and treatment of hypertension and related circulatory diseases. The experimental diets(A, B, C) were prepared from plant based ingredients such as buckwheat, potato, perilla seed with different ratios formulated as an edible form appropriate for human consumption according to AIN-77 standard. Control group(D) was fed commercial rat chow. Twety-four 15-week-old SDR(Sprague Dawley rats) and twenty-four 20-week-old SHR(spontaneously hypertensive rats), weighing 200g respectively, were assigned to 4 treatments of 6 rats each in a completely randomized design. Blood pressure was measured at 7 day interval by tail-cuff sphygmomanometer using an IITC cuff pump and amplifier. The growth rates of both SHR and SDR were not statistically different in comparions with the control except those of diet B and C in SDR and SHR, respectively(p<0.05). When exprimental diets were fed, systolic blood pressure of SDR and SHR at day 28 had been lowered by 17-20 mmHg(ave. of 19.6mmHg for both strains) compared with the reference pressure at day 0. The levels of HDL-cholesterol were increased, while the levels of LDL-cholesterol consistently decreased in both strains when experimental diets were fed(p<.05). Plasma total cholesterol levels were not different among treatments. Plasma triglyceride levels were higher in control diet(commercial rat chow) due to two times higher fat content of control diet itself(p<.05). Atherogenic indices were lower compared to those of control when the experimental diets were fed in both strains (p,.05). The results suggest that the antihypertensive diets mainly consisting of plant-based ingredients may possibly effective in relieving hypertension as well as circulatory diseases.

The Relationships between Plasma δ - aminolevulinic acid Concentration and Lead Exposure Indices in Lead Workers (연 노출 근로자들의 혈장 δ - aminolevulinic acid 량과 연 노출 지표들과의 관련성)

  • This study was carried out to investigate relationship between plasma $\delta$ - aminolevulinic acid (ALAP) and lead exposure indices in exposure to lead. The subjects were 218 male workers in 2 storage battery companies and 2 secondary smelting companies. Blood lead(PbB), blood zinc-protoporphyrin( ZPP), urinary $\delta$ - aminolevulinic acid (ALAU), hemoglobin(Hb), and hematocrit(Hct) were measured as lead exposure indices. The results were as follows, 1. The means of blood lead and blood ZPP concentration of subjects were $27.2{\pm}14.0{\mu}g/d{\ell}$ and $55.1{\pm}47.6{\mu}g/d{\ell}$, respectively. The means of plasma $\delta$ - ALA and urinary $\delta$ - ALA concentration were $18.9{\pm}25.1{\mu}g/d{\ell}$ and $2.1{\pm}4.6mg/{\ell}$, respectively. 2. The concentration of ALAP was $11.2{\mu}g/{\ell}$ for below $20{\mu}g/d{\ell}$ PbB, $12.8{\mu}g/{\ell}$ for from $21-40{\mu}g/d{\ell}$ PbB, and $51.2{\mu}g/{\ell}$ for over $40{\mu}g/d{\ell}$ PbB, respectively. 3. ALAP was significantly correlated with ALAU(r=0.829, p<0.01), ZPP(r=0.724, p<0.01) and PbB(r=0.552, p<0.01).

Noninvasive fetal RHD genotyping using cell-free fetal DNA incorporating fetal RASSF1A marker in RhD-negative pregnant women in Korea

  • Purpose: Conventional methods for the prenatal detection of fetal RhD status involve invasive procedures such as fetal blood sampling and amniocentesis. The identification of cell-free fetal DNA (cffDNA) in maternal plasma creates the possibility of determining fetal RhD status by analyzing maternal plasma DNA. However, some technical problems still exist, especially the lack of a positive control marker for the presence of fetal DNA. Therefore, we assessed the feasibility and accuracy of fetal RHD genotyping incorporating the RASSF1A epigenetic fetal DNA marker from cffDNA in the maternal plasma of RhD-negative pregnant women in Korea. Materials and Methods: We analyzed maternal plasma from 41 pregnant women identified as RhD-negative by serological testing. Multiplex real-time PCR was performed by amplifying RHD exons 5 and 7 and the SRY gene, with RASSF1A being used as a gender-independent fetal epigenetic marker. The results were compared with those obtained by postnatal serological analysis of cord blood and gender identification. Results: Among the 41 fetuses, 37 were RhD-positive and 4 were RhD-negative according to the serological analysis of cord blood. There was 100% concordance between fetal RHD genotyping and serological cord blood results. Detection of the RASSF1A gene verified the presence of cffDNA, and the fetal SRY status was correctly detected in all 41 cases. Conclusion: Noninvasive fetal RHD genotyping with cffDNA incorporating RASSF1A is a feasible, reliable, and accurate method of determining fetal RhD status. It is an alternative to amniocentesis for the management of RhD-negative women and reduces the need for unnecessary RhIG prophylaxis.

Relationship Between Nutrient Supply to Muscle and Adipose Tissues and Nitrogen Retention in Growing Wethers on Forage Based Diets Fed with Different Forage Sources

  • Three growing wethers were used to investigate the differences in nitrogen (N) retention, blood plasma metabolite concentration and energy-yielding nutrient supply to muscle and adipose tissue. The wethers were fed one of three diets: timothy hay with concentrate (THD), Italian ryegrass with concentrate (IRD), and rice straw with concentrate (RSD) for 11 days. The experimental diets were adjusted to the animals to provide 100 g of daily gain. The triglyceride (TG) concentration of blood plasma in arterial and portal veins was higher with THD and IRD than with RSD. Conversely, the available amount of TG in tissues was higher with IRD. The daily amount of glucose and non-esterified fatty acids (NEFA) supplied to muscle tissue and adipose tissue was numerically higher with THD than IRD or RSD. Although N retention did not differ among the diets, it was numerically higher with THD than with IRD or RSD. The results suggest that the difference in the amount of glucose and NEFA delivered to muscle tissue may reflect the N retention in response to forage based diets.

Comparisons of Dietary, Living Habits and Blood Parameters in Underweight and Overweight University Students (일부 저체중과 과체중 대학생의 식습관, 생활습관과 혈액 지표의 비교 분석 연구)

  • This study was carried out to compared the characteristics of the underweight university students with those of overweight university students. The surey was carried out by self-questionnaires with 97 underweight and 156 overweight university students. The results are summarized as fellows. Breakfast was skipped in 37.1% of underweight and 46.2% of overweight students and it appeared overweight students eat faster than underweight students. The 16.5% of underweight and 75.0% of overweight students have correct perception about their body image. The percentage of weight control experience were 25.8and 55.8 in the underweight and overweight students, respectively. Blood levels of lipid profiles(triglyceride, LDL cholesterol, and HDL cholesterol), hemoglobin, AST and ALT were anaylzed. Plasma levels of triglyceride, total cholesterol, LDL cholesterol, and HDL cholesterol of underweight students were 96.41, 143.75, 97.53 and 56.27 mg/dl, and those of overweight students were 107.99, 164.31, 100.08 and 53.64 mg/dl, respectively. Blood parameters were in normal range in both group. Plasma triglyceride, total cholesterol and LDL cholesterol levels o( overweight students were higher than those of underweight students. On the other hand, plasma HDL cholesterol levels of overweight students were lower than those of underweight students. Plasma levels of AST and ALT were in normal range in both group. However, AST and ALT levels of overweight students were higher than that of underweight students. Therefore they should have a nutritional education program to improve their dietary and living habits for overweight students' health. And nutritional education program should be organized practically and systematically

Effect of He-Ne laser intravascular irradiation by live blood analysis (생혈구분석을 통한 He-Ne laser 정맥혈관내 조사의 효과)

  • Objectives : The purpose of this study was to investigate the effect of Intravascular Laser Irradiation of Blood(ILlB) by the live blood analysis. Methods : We had analysed the changing forms of the live blood samples with Ultra Darkfield Microscope before and after Intravascular Laser Irradiation of Blood. Results : 1. Somatid did not showed significant change. 2. In the rouleau of red blood cells was decreased significantly. 3. In the morphological change of red blood cells, Burr cell, Ovalocyte and Poikilocyte were decreased significantly, but Acanthocyte and Target cell were increased significantly. 4. In the abnormal matters in plasma, the Cholesterol cristal did not showed significant change, but the Aggregation of platelet, Lipids, Spicule, Leucocyte, Uric acid cristal did showed a little significant decrease. Conclusion : These findings suggest that live blood analysis is useful to judge the effect of treatment and diagnosis in oriental medicine, and with the effect of Intravascular Laser Irradiation of Blood, it had showed significant effect on rouleau of red blood cells, morphological change of red blood cells and abnormal matters in plasma.

Effects of Different Selenium Sources on Performance, Carcass Characteristics, Plasma Glutathione Peroxidase Activity and Selenium Deposition in Finishing Hanwoo Steers

  • This study was conducted to determine effects of different selenium (Se) sources on performance, carcass characteristics, blood measures (whole blood Se concentration and plasma glutathione peroxidase (GSH-Px) activity), and Se concentrations in tissues of finishing Hanwoo steers (Korean native steers). Twenty finishing Hanwoo steers (average body weight=536${\pm}$23.4 kg, average age=approximately 20 months) were allotted to treatments in four groups of five steers per pen for 16 weeks preceding slaughter. Treatments were control (CON), spent mushroom composts from Se-enriched mushrooms (Se-SMC), selenized yeast (Se-Y), and sodium selenite (SS). Dietary Se levels of all treatments except CON were 0.9 mg Se/kg on the dry matter basis. Body weight was measured at the first and final day of trial, and blood samples were collected to analyze whole blood Se concentration and plasma GSH-Px activity at 2, 4, 8, and 16 weeks. At the end of trial, steers were slaughtered to collect muscle and liver samples for their Se analyses, and carcass data were recorded. In terms of dry matter intake, body weight gain and carcass characteristics, no significant differences among treatments were observed. Whole blood Se concentrations were significantly higher (p<0.05) for Se-SMC and Se-Y treatments than for CON at each collection period, with no significant difference between SS and CON. For weeks 2 and 8, there was no significant difference for whole blood Se concentration between Se-SMC and Se-Y, but for weeks 4 and 16, Se-Y treatments were significantly higher (p<0.05) than Se-SMC. No differences were observed for plasma GSH-Px activity between Se-SMC and Se-Y. The Se concentrations in hind leg and liver were significantly different among treatments (p<0.05) and those in both tissues ranked the greatest in Se-Y, followed by Se-SMC, SS, and CON treatments. However, tissue Se concentration for SS was not different from that for CON. These results showed that feeding organic Se sources such as Se-SMC and Se-Y enhanced Se concentration in tissues, while SS, the most common supplement of inorganic Se, was inefficient in Se deposition. Even though Se-Y had a higher Se concentration in tissues than Se-SMC, replacing Se-Y with Se-SMC in diets of beef steers would be an inexpensive way to increase Se concentration in beef.
