The purpose of this study is to determine the effect of diabetes education program on Glucose Metabolism(blood sugar, HbA1c) and Lipid Metabolism(total cholesterol, triglyceride, low density lipoprotein, high density lipoprotein), Self-efficacy in non-insulin independent diabetes mellitus. The study design was a non equivalent control group pre-test post-test design. Data for the study were collected from March 12 to June 19, 2001. Sixty-two research subjects were assigned to experimental(36) and control(26) groups. The collected data was analyzed using the Chi-Square test, t-test by spsswin program The results are as follows : 1. Experimental group had higher level of glucose metabolism than control group(FBS ; t=-3.317, p=.002, HbA1c; t=-4.956, p=.000). 2. Level of lipid metabolism were partly a significant different between experimental group and control group(Triglyceride ; t=-2.108, p=.039). 3. Experimental group had higher efficacy score than control group(t=4.651, p=.000). In conclusion, the study supported the effects of diabetes education program to increase metabolism an d self-efficacy. Further study with a longitudinal design is suggested to verify the effect of diabetes education program in NIDDM and standardized diabetes education program.
본 실험은 CRE 80% 에탄올 추출물을 식이에 0.5%, 1% 혼합하여 5주간 섭취시켰을 때 CRE 추출물이 SHR의 혈압에 미치는 영향을 확인함으로써 CRE 추출물이 고혈압 흰쥐의 지질성분과 혈압에 미치는 영향을 조사함으로써 CRE의 기능성식품으로서의 이용 가능성을 확인하였다. 실험기간 중 실험쥐의 일일평균 체중변화량은 0.5% CRE 보충군(II)은 1.63 g/day로 정상식이 급여군보다 일일평균 체중증가량이 유의적으로 감소하는 결과를 나타내었다. 간, 신장, 비장의 무게 변화를 측정한 결과 유의적인 차이를 나타내지 않았으며, 혈청 내 포도당 농도 또한 정상식이 급여군에 비해서 0.5%, 1% CRE 추가 보충군이 감소하는 경향은 보였으나 유의적인 차이는 나타나지 않았다. 본 실험에서 총 cholesterol 함량은 정상식이 급여군보다 0.5%, 1% CRE 보충 급여군에서 유의적으로 증가하는 경향을 보였지만 HTR을 확인한 결과 정상식이 급여군보다 0.5%, 1% CRE 보충 급여군이 각각 11% 증가한 결과를 나타냈기 때문에 혈중 총 cholesterol 함량의 증가는 HDL-cholesterol의 농도 증가에 의한 것으로 생각된다. HDL-cholesterol 농도는 정상식이 급여군과 0.5%, 1% CRE 보충 급여군 모두에서 유의적으로 증가하는 경향을 나타냈다. LDL-cholesterol 역시 유의적으로 증가하는 경향을 나타냈지만 HDL-cholesterol/LDL-cholesterol의 증가 비율로 봤을 때 HDL-cholesterol이 정상식이 급여군보다 0.5% CRE 보충 급여군이 15%, 1% CRE 보충급여군이 36.5%로 유의적으로 증가하는 결과를 나타냈다. 실험기간 동안 SHR의 혈압 변화는 정상식이 급여군(I)의 경우 거의 일정하게 혈압이 유지되는 경향을 보이다가 5주차에는 다시 증가한 반면 CRE 0.5% 보충군(II)은 초기혈압에서 떨어지는 경향을 보이다가 2주째에 혈압이 다시 상승했다. 하지만 그 후 다시 감소하는 경향을 보였고, CRE 1% 보충군(III)은 일정하게 혈압이 감소하는 경향을 나타냈다. 따라서 CRE 추출물이 SHR의 혈압을 낮추고 지질대사 개선에 영향을 주어 혈관계 장애로 인해 발병되는 고혈압, 당뇨, 뇌혈관질환, 동맥경화증과 같은 질환을 예방하는 기능성식품으로서 이용 가능성이 기대된다.
The glycogen-associated protein phosphatase (PP1G/$R_{GL}$) may play a central role in the hormonal control of glycogen metabolism in the skeletal muscle. Here, we investigated the in vivo epinephrine effect of glycogen metabolism in the skeletal muscle of the wild-type and $R_{GL}$ knockout mice. The administration of epinephrine increased blood glucose levels from 200±20 to 325±20 mg/dl in both wild-type and knockout mice. Epinephrine decreased the glycogen synthase -/+ G6P ratio from 0.24±0.04 to 0.10±0.02 in the wild-type, and from 0.17±0.02 to 0.06±0.01 in the knockout mice. Conversely, the glycogen phosphorylase activity ratio increased from 0.21±0.04 to 0.65±0.07 and from 0.30±0.04 to 0.81±0.06 in the epinephrine trated wild-type and knockout mice respectively. The glycogen content of the knockout mice was substantially lower (27%) than that of both wild-type mice; and epinephrine decreased glycogen content in the wild-type and knockout mice. Also, in Western blot analysis there was no compensation of the other glycogen targeting components PTG/R5 and R6 in the knockout mice compared with the wild-type. Therefore, $R_{GL}$ is not required for the epinephrine stimulation of glycogen metabolism, and rather another phosphatase and/or regulatory subunit appears to be involved.
Serotonin, a biogenic amine widely found in many organisms, functions as both a neurotransmitter and hormone. Although serotonin is involved in various physiological processes, this study aimed to review its role in energy metabolism. Given that serotonin cannot cross the blood-brain barrier and is synthesized by two different isoforms of tryptophan hydroxylase in the central nervous system (CNS) and peripheral tissues, it is reasonable to assume that serotonin in the CNS and peripheral tissues functions independently. Recent studies have demonstrated how serotonin influences energy metabolism in metabolic target organs such as the intestines, liver, pancreas, and adipose tissue. In summary, serotonin in the CNS induces satiety and appetite suppression, stimulates thermogenesis, and reduces body weight. Conversely, serotonin in the periphery increases intestinal motility, stimulates gluconeogenesis in the liver, suppresses glucose uptake by hepatocytes, promotes fat uptake by liver cells, stimulates insulin secretion while suppressing glucagon secretion in the pancreatic islets, promotes lipogenesis in white adipose tissue, inhibits lipolysis and browning of white adipose tissue, and suppresses thermogenesis in brown adipose tissue, thereby storing energy and increasing body weight. However, considering that most experimental results were obtained using mice and conducted under specific nutritional conditions, such as high-fat diets, whether serotonin acts in the same way in humans, whether it will act similarly in individuals with normal versus obese weights, and whether its effects vary depending on the type of food consumed, remain unknown.
Objective: An experiment was conducted to evaluate the effects of Lonicera japonica extract (LJE) on milk production, rumen fermentation and blood biomarkers of energy metabolism, inflammation and oxidative stress during the perinatal period of Holstein dairy cows. Methods: Eighteen Holstein dairy cows were used in a complete randomized design experiment with 3 dietary treatments and 6 cows per treatment. All cows received the same basal total mixed ration (TMR) including a prepartal diet (1.35 Mcal of net energy for lactation [NEL]/kg of dry matter [DM], 13.23% crude protein [CP]) from -60 d to calving and a postpartal diet (1.61 Mcal of NEL/kg of DM, 17.39% CP) from calving to 30 days in milk (DIM). The 3 dietary treatments were TMR supplemented with LJE at 0 (control), 1 and 2 g/kg DM, respectively. LJE was offered from 21 d before calving to 30 DIM. Dry matter intake (DMI) and milk production were measured daily after calving. Milk and rumen fluid samples were collected on 29 and 30 d after calving. On -10, 4, 14, and 30 d relative to calving, blood samples were collected to analyze the biomarkers of energy metabolism, inflammation and oxidative stress. Results: Compared with control diet, LJE supplementation at 1 and 2 g/kg DM increased DMI, milk yield and reduced milk somatic cell count. LJE supplementation also decreased the concentrations of blood biomarkers of pro-inflammation (interleukin-1β [IL-1β], IL-6, and haptoglobin), energy metabolism (nonesterified fatty acid and β-hydroxybutyric acid) and oxidative stress (reactive oxygen metabolites), meanwhile increased the total antioxidant capacity and superoxide dismutase concentrations in blood. No differences were observed in rumen pH, volatile fatty acid, and ammonia-N (NH3-N) concentrations between LJE supplemented diets and the control diet. Conclusion: Supplementation with 1 and 2 g LJE/kg DM could increase DMI, improve lactation performance, and enhance anti-inflammatory and antioxidant capacities of dairy cows during perinatal period.
The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. The purpose of this study was to investigate the relationship among calcium intake, blood parameters related with bone metabolism, and serum lipids in healthy adults on self-selected diet. Subjects were consisted of 40 female college students residing in Chungnam. Anthropometric measurements, dietary intake measurements and blood collection were conducted. Serum concentrations of total protein, albumin, alkaline phosphates, leucine amino peptidase, BUN, calcium, inorganic phosphorus, and lipids were measured by biochemical analyzer and ICP spectrometer. The results were as follows. The mean age of subjects was 22.34 years and weight, height and BMI were 52.89kg, 161.29cm and 20.34, respectively. The daily mean energy and calcium intakes were 81.75% and 64.38% of RDA. The mean animal 1:2. The mean serum concentrations were 6.54g/dl(total protein), 4.12g/dl(albumin), 123.24U/(alkaline phosphates), 36.59U/l(leucine amino peptidase), 8.26mg/dl(calcium), 3.29mg/dl(inorganic phosphorus), 60.73mg/dl(triglyceride), 138.49mg/dl(total cholesterol), 65.95mg/dl(HDL-cholesterol), and 60.39mg/dl(LDL-cholesterol). There were no significant differences among calcium intake, bone metabolism parameters, and serum lipids when analyzed by Pearson's correlation coefficient. More systematic studies are required to investigate the roles of calcium in healthy persons on self-selected diets containing different levels of calcium.
The ethanol extraction yield of Meliae toosendan fructus(MT) was about 24.5% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of MT in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in MT treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in MT treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in MT treated group. Also the activities of glucose-6-phosphate dehydrogenase(G-6-PDH) and glucokinase(Gk) were increaed in MT treated group. The content of hepatic glycogen was significantly increaed in MT treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in MT treated group. Also, content of glutathione(GSH)was dereased in MT treated froup. whereas, activity of catalase(CAT) was significantly increaed in MT treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of MT would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.
The purpose of this study was to investigate the effects of the ethanol extract of Cynanchum wilfordii (ECW) on the blood lipid profile of hypercholesterolemic rats. Thirty 7-week-old male Sprague-Dawley rats were allowed free access to either a normal diet (AIN-93 diet), or 1% high-cholesterol diet with or without 0.5% or 1% ECW for 5 weeks. After sacrifice, the rat serum lipid profile was analyzed. The diets containing ECW decreased body weight gains compared to the normal diet. Serum HDL-cholesterol levels of ECW-fed groups were significantly increased in the hypercholesterolemic groups and normal groups (P<0.05). When 1% ECW was fed to the normal group, total cholesterol level was increased. Moreover, treatment of ECW in hypercholesterolemic groups yielded a dose-dependent and highly significant decrease in the atherogenic index as compared to the control. These results suggest that intake of Cynanchum wilfordii may help reduce the risks of hypercholesterolemia by increasing blood HDL-cholesterol and lowering the atherogenic index.
The purpose of this study was to investigate zinc and copper metabolism and risk factors of chronic diseases in 20 normal adults women. The daily intake, excretions in urine and feces, and serum levels of zinc and copper were determined by 24-hr food records and chemical analysis. The results were summarized as follows. mean age, body weight and BMI were 22.88 years, 54.65kg and 21.28kg/$m^2$ respectively. Mean daily intakes of energy and protein were 1578.84㎉(79% of RDA) and 52.05g (87% of RDA). The zinc and copper intake, excrestion in urine and feces were 7093.23$\mu\textrm{g}$(59% of RDA/2093.87$\mu\textrm{g}$, 203.50$\mu\textrm{g}$/39.87$\mu\textrm{g}$ and 3416.41$\mu\textrm{g}$/857.62$\mu\textrm{g}$, respectively. The serum levels of fasting glucose, insulin, total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, zinc and copper were 76.60mg/㎗, 15.66IU/㎗, 160.30mg/㎗, 50.95mg/㎗, 89.80mg/㎗, 89.79mg/㎗, 95.65$\mu\textrm{g}$/㎗ and 73.28$\mu\textrm{g}$/㎗ respectively. Dietary ratio of Zn to Cu was shown to have significant positive correlations with serum insulin, total cholesterol, LDL-cholesterol. The urinary ratio of Zn to Cu was shown to have significant positive correlations with triglyceride. The serum copper level was shown to have significant negative correlations with serum total cholesterol and LDL-cholesterol. In summary, Zn consumption of adult women in their normal diet does not meet the Zn requirement for Koreans. Also, intakes of Zn and Cu may effect the glucose metabolism and cardiovascular diseases. Therefore, to increase the Zn intake and to maintain an appropriate intake ratio of Zn to Cu, nutrition education needs to be implemented.
