To investigate the effect of Feelch on alcohol metabolism, we measured both blood alcohol concentration in human and hepatic alcohol metabolizing enzyme activity in rats. The blood alcohol concentration in Feelch-ingested group was significantly lower than that in water-ingested group at 0, 40, and 80 minute after alcohol intake. The blood alcohol concentration between male and female taken 300ml of $21\%$ alcohol showed the significant differences; the peak value of blood alcohol concentration in male and female were $0.083\pm0.014\%\;and\;0.108\pm0.018\%$, respectively. In alcohol-fed rats, aldehyde dehydrogenase (ALDH) activity was significantly increased, whereas alcohol dehydrogenase (ADH) activity was not changed. In both Feelch-fed group and Feelch plus alcohol-fed group, ADH and ALDH activity were significantly increased as compared with each control group. Feelch decreased phospholipase $A_2$ activity and lipid peroxidation in hepatic tissue and activities of serum aminotransferases as compared with control. These results suggest that Feelch may have a hepatoprotective effects and this is likely due to lower blood alcohol concentration via the increment of hepatic ADH and ALDH activity.
It has been reported that clonidine is $\alpha_2$-adrenergic agonist, potnet new hypotensive drug in human with low dose. The change in blood pressure is implicated in the concentration, release, uptake and metabalism of catecholamine and activity of catecholamine synthesizing enzyme in specific brain areas. Thus the experiment was set up to investigate the effect on the enzyme activity of clonidine alone and that of clonidine pretreated with imipramine or tranylcypromine by measuring activity of the Dopa-forming enzyme, tyrosine hydroxylase (TH) and epinephrine forming enzyme, phenylethanolamine-N-methyl transferase (PNMT) in brain and adrenal gland. The TH activity in brainstem and substantia nigra is decreased by intraperitoneally administered clonidine 0.1mg/kg twice a day for 5 days, but increased in the rats pretreated with imipramine 10mg/kg intraperitoneally given 26 hrs and 5 hrs before decaptitation. However the TH activity in all regions of brain is increased in rats pretreated with tranylcypromine 10mg/kg intraperitoneally twice a day for 5 days. The effect of clonidine on TH activity is due to inhibition release of norepinephrine by activation of presynaptic $\alpha_2$-adrenoreceptor, axon terminal result in the decrease of TH activity in brain. The increasing of TH activity in brain results in attenuation of the role of clonidine by pretreated with imipramine or tranylcypromine in rats. The activity of PNMT was not significantly affected by clonidine, imipramine and tranylcypromine in adrenal gland.
The principal objective of this study was to assess the effects of chitosan oligosaccharide supplementation on the improvement of blood glucose, lipid components and enzyme activities in the serum of streptozotocin(STZ, 55 mg/kg B.W., I.P. injection)-induced hyperglycemic rats fed on experimental diets for 5 weeks. The concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglycerides(TG) and phospholipids(PL) in serum were remarkably higher in the hyperglycemic group(group BSW) and STZ(I.P.)+chitosan oligosaccharide supplementation group(group ECW) than those in the control group(group BW, basal diet+water). However the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, tree cholesterol, cholesteryl ester ratio, TG and PL in serum were lower in the ECW group than in the BSW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the ECW group were higher than in the BSW group. The activities of alkaline phosphatase(ALP) and aminotransferase(AST, ALT) in serum were lower in the ECW group than in the hyperglycemic BSW group. The results shown above suggested that chitosan oligosaccharide supplementation effectively improves blood glucose, lipid composition and enzyme activities in the sera of STZ-induced hyperglycemic rats.

The present study was carried out to evaluate the physiological effects of seaweed supplementation on blood glucose levels, lipid profile, and antioxidant enzyme activities in subjects with type 2 diabetes mellitus. Subjects were randomized into either a control group or a seaweed supplementation group. Pills with equal parts of dry powdered sea tangle and sea mustard were provided to the seaweed supplementation group three times a day for 4 weeks. Total daily consumption of seaweed was 48 g. We found that total dietary fiber intake was 2.5 times higher in subjects receiving seaweed supplementation than in the control group. Accordingly, fasting blood glucose levels (p<0.01) and 2-hour postprandial blood glucose measurements (p<0.05) were decreased significantly in those ingesting seaweed. Furthermore, the serum concentrations of triglycerides were decreased and high-density lipoprotein cholesterol was increased significantly in seaweed supplement group (p<0.05). However, the concentrations of total cholesterol and low-density lipoprotein cholesterol were not affected by seaweed supplementation. The level of thiobarbituric acid reactive substances in erythrocytes was significantly lower with seaweed supplementation compared to controls (p<0.05). Catalase and glutathione peroxidase activities with seaweed supplementation were higher than the controls (p<0.05), but superoxide dismutase activity was not affected. We, therefore, conclude that ingestion of seaweed influences glycemic control, lowers blood lipids, and increases antioxidant enzyme activities.
This study was attempted to investigate the anti-hypertensive activity of Solanum tuberosum, cv. Gui valley. Ethanol extract of Gui vally (EGV) increased free radical scarvenge activity up to 91.6% of control at $200{\mu}g/m{\ell}$. It's anti-oxidant activity is similar to 10 uM of ascorbic acid, well known as antioxidant. EGV inhibited Angiotensin-I-converting enzyme (ACE) activity in vitro. ACE plays a important role in regulation of blood pressure and ACE inhibitors are important for the treatment of hypertension. Anti-hypertensive activity of EGV in vivo was assessed in lead acetate (LAT)-induced hypertensive rats for 8 weeks. Elevated blood pressure in control group was significantly decreased by EGV at 200 mg/kg. Also, ACE activity in blood was also suppressed by EGV treatment. Taken together, these results suggest that EGV has an anti-hypersive activity via inhibition of ACE and can be used for the treatment or prevention of hypertension.

Fibrinolytic enzymes with a direct mechanism of action and safer properties are currently requested for thrombolytic therapy. This paper reports on a new enzyme capable of degrading blood clots directly without impairing blood coagulation. This enzyme is also non-cytotoxic and constitutes an alternative to other thrombolytic enzymes known to cause undesired side effects. Twenty-four Bacillus isolates were screened for production of fibrinolytic enzymes using a fibrin agar plate. Based on produced activity, isolate S127e was selected and identified as B. subtilis using the 16S rDNA gene sequence. This strain is of biotechnological interest for producing high fibrinolytic yield and consequently has potential in the industrial field. The purified fibrinolytic enzyme has a molecular mass of 27.3 kDa, a predicted pI of 6.6, and a maximal affinity for Ala-Ala-Pro-Phe. This enzyme was almost completely inhibited by chymostatin with optimal activity at 48℃ and pH 7. Specific subtilisin features were found in the gene sequence, indicating that this enzyme belongs to the BPN group of the S8 subtilisin family and was assigned as AprE127. This subtilisin increased thromboplastin time by 3.7% (37.6 to 39 s) and prothrombin time by 3.2% (12.6 to 13 s), both within normal ranges. In a whole blood euglobulin assay, this enzyme did not impair coagulation but reduced lysis time significantly. Moreover, in an in vitro assay, AprE127 completely dissolved a thrombus of about 1 cc within 50 min and, in vivo, reduced a thrombus prompted in a rat tail by 11.4% in 24 h compared to non-treated animals.
본 연구의 목적은 혈관계 순환을 촉진시키는 약효제를 발굴하는 것이다. 아사이베리(Acai berry) 에탄올 추출물(ABEE)의 항산화 효과 뿐만 아니라 토끼의 폐에서 유래한 angiotensin converting enzyme (ACE)의 활성 및 대식세포와 혈관내피세포에서 nitric oxide 생성에 대한 효과가 본 연구에서 조사되었다. 먼저 환원력과 지질과산화에 대한 ABEE의 항산화효과가 관찰되었다. 뿐만 아니라, ABEE는 hydroxyl radical에 의하여 유발된 DNA 산화에 대한 보호효과를 보여주었다. 더욱이 ABEE는 0.01%의 농도에서 약 50% 정도의 angiotensin converting enzyme 활성에 대한 억제효과를 발휘하는 것으로 나타났다. 혈관내피세포에서 ABEE는 nitric oxide 의 생성을 증가시켰으나 대식세포에서는 lipopolysaccharide에 의하여 생성된 nitric oxide 생성을 감소시켰다. 게다가 superoxide dismutase (SOD)-2와 -3 발현수준은 ABEE 처리에 의하여 증가되었으나 SOD-1의 발현수준은 일정하였다. 더욱이 nitric oxide synthases-1 (NOS-1)의 발현수준은 ABEE 처리에 의하여 증가되었으나, 유발 효소인 NOS-2의 발현수준은 일정하였다. 또한 SOD의 전사인자인 Nrf-2의 발현수준은 ABEE에 의하여 증가되었다. 그러므로 이러한 결과들은 ABEE가 이상의 작용을 경유하여 혈액순환을 촉진시킬 수 있고, 혈관의 건강을 위한 큰 잠재성을 가지고 있다는 것을 암시하고 있다.
This study aims to investigate the changes in the lipid composition and angiotensin-converting enzyme activity in serum of rats exposed to microwave radiation, and to examine improving effects of green tea catechin to the lipid composition. The microwave-exposed rats received the normal and one of 3 diets: catechin free (MW-0C), 0.25% catechin (MW-0.25C) or 0.5% catechin (MW-0.5C). Rats were sacrificed 6th day after microwave radiation (2.45 ㎓, 15 min). The concentration of serum triglyceridein MW-0C group was increased by 85%, compared with the normal group, but that of MW-0.25C group with 0.25% catechin supplementation was to 17% lower, compared with the MW-0C group. There was no significant difference between normal group and MW-0.5C group. Total-cholesterol and LDL-cholesterol concentrations were increased by 21.4% and 38.6%, respectively, by microwave irradiation. The concentration of HDL-cholesterol in MW-0C group was lower to 29.3%, but it was maintained at the normal level by catechin supplementation. There was no significant difference among four groups in HLD-cholesterol/LDL-cholesterol. Atherogenic index in MW-0C group was increased by 56%, compared with the normal group. Angiotensin-converting enzyme (ACE) activity in lung tissue of MW-0C group was increased by 59 %, while that of MW -0.25C and MW -0.5C group were maintained at the normal level. ACE activity of MW-0C group in serum was increased by 122.8%, compared with the normal group. Catechin supplementation group was significantly reduced, compared with the MW-0C group ACE activity. In conclusion, microwave irradiation increased the serum triglyceride and cholesterol concentrations and ACE activity which are considered as the blood pressure increasing agents. However, catechin supplementation decreased the level of triglyceride, total cholesterol, LDL cholesterol, and ACE activity, which maybe consider catechin as being agent of lowering effect for blood lipid profile for athero-genesis.
The polymorphism (insertion, I or deletion, D) of angiotensin converting enzyme (ACE) gene is designated as the presence of a 287 bp Alu repeat. The D/D homozygote carrier is associated with high ACE activity, and this high activity has been implicated with hypertension, coronary artery disease, or diabetic nephropathy. We studied the clinical candidate marker in ACE gene polymorphism using chemical and hematological analysis. The subjects are divided into normotensive and hypertensive groups and ACE genotype in the group was confirmed by PCR method. Chemical analysis was preceded with Hitachi7060, and hematological analysis was performed using Mythic 22. In 116 targeted people, 17 (38.64%) of 44 I/I genotype group are hypertension, 15 (34.09%) in 44 with D/I, but, D/D type in the 28 cases is 15 patients (53.57%) in hypertension. In hypertension group, biochemical analysis (triglyceride, and alkaline phosphatase) and hematological analysis (white blood cell, platelet) are showed high value in D/D genotype of ACE gene. The relationship between hypertension and ACE genotype is the same results as previously reported and we thought that the high laboratory value of white blood cell, platelet, triglycerides, and alkaline phosphatase are also indicator of hypertension in D/D type of ACE.

This study was conducted to investigate changes of blood metabolites and enzyme activities at different slaughter ages of Hanwoo cattle. Blood samples were taken from six hundred thirty two steers (24~33 months of age) and one hundred twenty eight bulls (17~24 months of age) by venipuncture from caudal vein immediately prior to slaughter. Glucose concentrations were linearly increased (P<0.001) with increases of the slaughter age in steers, and blood urea nitrogen (BUN) concentrations were linearly decreased (P<0.001) with increases of the slaughter age in steers. Likewise, glucose and albumin concentrations in bulls were linearly decreased (P=0.009 and P<0.001, respectively) with increases of the slaughter age. BUN, cholesterol, triglycerides, and total cholesterol concentrations were linearly increased (P<0.001, 0.018 and 0.002, respectively), and creatinine concentrations were quadratically increased (P=0.009) with increases of the slaughter age. Activity of gamma glutamyltransferase (${\gamma}$-GT) and aspartate aminotransferase (AST) was linearly increased (P<0.001) with increases of the slaughter age in both steers and bulls, whereas alanine aminotransferase (ALT) was not affected by increasing slaughter age. In addition, the concentrations of ${\gamma}$-GT and AST were higher (P<0.05) for bulls than those of steers at 24 months of age. Increased levels of ${\gamma}$-GT and AST indicate impaired liver function; it may be associated with increases of concentrate level which is a concomitant of the extended fattening periods in Hanwoo feeding. In conclusion, the results in the present study may be one of the useful information for diagnosis of the metabolic disorder in Hanwoo cattle.
