In vivo Brain-to-blood Efflux Transport of Choline at the Blood-brain Barrier

  • The purpose of this study was to clarify the efflux transport system of choline from brain to blood across the blood-brain barrier (BBB) in rats using the brain efflux index (BEI) method. $[^3H]$Choline was micro-injected into parietal cortex area 2 (Par2) of the rat brain, and was eliminated from the brain with elimination halflife of 45 min. The BBB efflux clearance of $[^3H]$choline was about 124 mL/min/g brain, which was determined from combination of an elimination rate constant $(1.54X10^{-2}min^{-1})$ and the distribution volume in the brain (8.05 mL/g brain). The efflux of $[^3H]$choline was inhibited by unlabeled choline in a dose-dependent manner and was significantly inhibited by cationic substrates, such as hemicholinium-3 and tetraethylammonium (TEA). These results suggest that the BBB may act as an efflux pump for choline to reduce the excessive choline concentration in the brain interstitial fluid.

Fluid-Structure Interaction Analysis of Blood Vessel Considering Internal Diameter Variation (내부직경 변화를 고려한 혈관의 유체-구조 상호작용 해석)

  • A three-dimensional elastic blood vessel model with internal diameter variation is considered to investigate internal flow characteristics and effects of structural deformation. Also, computational analyses for both the rigid model and the fully-coupled FSI model have been conducted in order to compare the shear stress, pressure distribution, and flow velocity in detail. A 70% narrowing area of asymmetric blood vessel model was especially investigated to show the versatility of fluid-structure interaction phenomenon. The results reveal that effect of fluid-structure interaction is very important to accurately investigate the flow characteristics of the blood vessel.

  • This paper reports the numerical results for blood flow of the sac squeezed by moving actuator in the TPLS(Twin Pulse Life Support System). Blood flow in the sac is assumed to be 3-dimensional unsteady newtonian fluid. where the blood flow interacts with the sac, which is activated by the moving actuator. The flow field is simulated numerically by using the FEM code, ADINA. It is well known that hemolysis is closely related to shear stress acted on blood flow. According to this fact, we simulate four models with different speed for moving actuator and examine the distribution of shear stress for each model. Numerical results show that maximum shear stress is strongly dependent on the actuator speed.

Availability of Statistical Quality Control of Nuclear Medicine Blood Test Using Population Distribution (모집단 분포를 이용한 핵의학 혈액검사의 통계적 품질관리의 유용성)

  • Purpose The importance of quality control by the error to a minimum, which for the purpose of enhancing the reliability of the examination is not be emphasized excess. Currently, most nuclear medicine laboratory are conducting the internal and external quality control, and they are applying the Levey-Jennings or Westgard Multi-Rules by using the commercialized quality control materials. The reliability of the nuclear medicine blood test which affects the diagnosis of patients and the treatment policy is being secured through this quality control activity. Therefore, researchers will evaluate the utility of the statistic quality control using the population distribution of the nuclear medicine blood test conducted targeting the checkup examinees by the additional technique of the reliability improvement. Materials and Methods A statistic analysis was performed about 12 items of the nuclear medicine blood test targeting 41,341 peoples who used the health screening and promotion center in Asan Medical Center from January, 2014 to December, 2014. The results of 12 items of the nuclear medicine blood test was divided into the monthly percentage of three groups: within reference values, over reference, and under reference to analyze the average value of the population distribution, standard deviation, and standard deviation index (SDI). Results The standard deviation of the population distribution mostly showed a result within ${\pm}2SD$ in all groups. However, When the standard deviation of the population distribution represented a result over ${\pm}2SD$, it was confirmed SDI was showing a result of SDI > -2 or SDI > 2. As a result of analyzing the population distribution of 12 items(AFP, CEA, CA19-9, CA125, PSA, TSH, FT4, Anti-Tg-Ab, Anti-TPO-Ab, Calcitonin, 25-OH-VitD3, Insulin) of the nuclear medicine blood part basic test, when SDI of the monthly percentage which deviated from the reference values was over ${\pm}2.0$, CA19-9 September was 2.2, Anti-Tg-Ab may was 2.2, Insulin January was 2.3, Insulin March was 2.4. It was confirmed these cases were attributed to the abnormality of the test reagent (maximum combination rate of isotope reagent declined) and the decline of the test response time. Conclusion The population distribution includes the entire attribute which becomes the study object. It is expected the statistic quality management using the population distribution which was conducted targeting the checkup examinees by dividing into three groups: within reference values, over reference, and under reference by means of this characteristics will be able to play a role of complementing the internal quality control program which is being carried out in the laboratory.

Antihyperlipidemic and Antidiabetic Activities of the Ascidian Tunic in Sprague-Dawley Rats (Sprague-Dawley계 쥐에서의 우렁쉥이껍질의 항당뇨.항고지혈증 기능활성)

  • This study examined the antidiabetic and antihyperlipidemic activities of an ascidian (Halocynthia roretzi) in rats in vivo. Rats were fed on experimental diet including dried ascidian powder (200 mg/kg body weight) for 4 weeks, and then the triglyceride and total cholesterol levels in blood were analyzed. On the ascidian tunic powder diet, the triglyceride level decreased by more than 20.9% and the total cholesterol content decreased by more than 24.4%. In comparison, the triglyceride and total cholesterol level in the blood of rats fed ascidian meat powder decreased only slightly. Therefore, the ascidian tunic powder might be a healthy food with antidiabetic and antihyperlipidemic effects.

Lead Levels in Blood and Urine of a Normal Male Person in Korea (일부 남자 정상인의 혈액 및 뇨중 연함량)

  • In order to provide a basic data for the prevention of the adverse effect of lead on health, We examined lead level in the blood and urine of 371 healthy men living in Choongchung-do from May to June, 1997. The results were as follows ; 1. Average lead level of all the subjects was $3.98{\pm}1.02{\mu}g/dl$ in blood, and $3.94{\pm}2.09{\mu}g/L$ in urine, respectively. Lead contents examined in this study were significantly lower than those of other investigators. 2. The lead levels of all the subjects in blood and urine had almost normal distribution. 3. Relation between lead content in blood and urine was a simple linear regression; its equation was "Lead level in blood=36.76+0.77 lead level in urine".

  • This study was conducted to compare the frequency and to find epidemiologic characteristics of the diseases by analyzing the results of routine first health examination for partial healthy public officials. The total number of the examined was 1.128(876 male, 252 female) The results were as follow; 1. The prevalence rate of suspicious disease was liver disease 5.9% hypertension 5.4%, hyperlipemic disease 3.8%, pulmonary tuberculosis 1.2% by order. 2. The mean systolic blood pressure and diastolic blood pressure showed increasing trend by age increasing. The mean of blood pressure in man was rather higher than that in woman. 3. The prevalence rate of suspicious borderline hypertension increased by aging. And the distribution of high cholesterol group and over than overweight group increased by aging. 4. In male, significant relation among age, systolic blood pressure, diastolic blood pressure, relative body weight was found. And significant relation between total cholesterol level and relative body weight was found. In female, age and relative body weight was related to systolic blood pressure, diastolic blood pressure and total cholesterol level. 5. According to the retrospective follow-up for those who had suspicious disease, correspondence rate of results was 5.2% in circulatory disease and 1.5% in liver disease.

Relation between Obesity Pattern Identification and Metabolic Parameters in Overweight and Obese Women (과체중 및 비만 여성에서 한방비만변증에 따른 체성분 및 대사관련 지표의 상관성)

  • Objectives: We conducted this study to analysis obesity pattern and obesity related blood parameters. Methods: A total of 64 overweight and obese (body mass index [BMI] ${\geq}23cm/kg^2$) women who had no other disease was recruited. Body composition and obesity related blood parameters were measured. Also subjects were given and filled out the Obesity pattern identification questionnaire. We analyzed the differences of body composition and blood parameters and measured correlations of BMI and blood parameters in each obesity pattern. Results: The distribution of obesity pattern was liver depression (35.6%), food accumulation (47.5%) and deficiency (pi and yang deficiency, 22.0%), in order. There were no significant differences age, body composition and obesity related blood parameters between obesity patterns. BMI and obesity related blood parameters, however, showed significant correlations depending on obesity patterns. Conclusions: We concluded that correlations between BMI and obesity related blood parameters were differed depending on obesity patterns.

Effects of Citrate-capped Silver Nanoparticles on the Blood Coagulation and Platelet Aggregation in Rats (랫드의 혈액응고 및 혈소판 응집에 미치는 은나노 입자의 영향)

  • Effects of citrate-capped silver nanoparticles (AgNPs) on the blood coagulation and platelet aggregation were investigated using whole blood, platelet rich plasma (PRP) and washed platelet obtained from SD male rats. To confirm the stability of AgNPs in the test, size distribution of the nanoparticles was measured in the vehicles including distilled water, serum, and platelet buffers. The average size of AgNPs was 20 nm in the vehicles, which means that the stability was maintained during the whole experimental period. When blood coagulation was monitored by using whole blood impedance aggregometer, coagulation was not observed at the concentration of 1, 10 and 50 ppm. Platelets in plasma or in buffer were not aggregated by AgNPs at the concentration of 1, 2 and 4 ppm, respectively. The test concentration of AgNPs could not be increased because the dark color of the nanoparticles impeded the transmission of light, which is an indicator of aggregation. Although the blood or platelets were pre-activated by collagen, thrombin, or ADP with sub-threshold level, aggregation was not observed at the test concentration. Microscopic observation also supported the result obtained by the aggregometer.

Gastrointestinal Tissue Blood Volume Affected by Venous Pressure Change (실혈 후 및 혈압상승 후의 소화기 조직 혈액량 및 산소 섭취량 -제 1 편 정맥혈압과 소화기 조직 혈액량-)

  • Changes in gastrointestinal tissue blood volume induced by variations of venous pressure between 6 and 40 mmHg were studied in 32 rabbits. Venous pressure lowering was produced by withdrawal of appropriate volume of blood and venous pressure elevation was obtained by partial occlusion of intra-thoracic vena cava inferior. Estimation of regional tissue blood volume was performed by means of regional distribution of injected $Cr^{51}-labeled$ red blood cells. The following results were obtained. 1. At the normal control venous pressure value of 18 mmHg, spleen showed the highest value of tissue blood volume expressed on weight basis, namely, $111{\mu}l/gm$, Liver tissue blood volume was $95\;{\mu}l/gm$, small intestine 24 and stomach $21\;{\mu}l/gm$, respectively. 2. Linear relationships were observed between venous pressure change and gastrointestinal tissue blood volume. The coefficients of correlation were: in spleen r=0.723; in liver r=0.791; in stomach r=0.704, respectively. In small intestine the relationship was less clear and r=0.358. Tissue blood volume of extrabdominal tissue, such as M. gastrocnemius was not influenced by venous pressure change. 3. The highest change in tissue blood volume expressed on weight basis was observed in spleen. The liver tissue showed the next highest change. Change in total tissue blood volume, however, was greatest in liver and next greatest in small intestine. This was interpreted by the fact that total weight of these two organs was much greater than that of spleen. 4. The mechanism that the change in tissue blood volume lies in the venous system which has a great compliance was discussed.
