Phenolic compounds and antioxidant activity of berries produced in South Korea

  • Berries are rich sources of phenolic compounds, which are known to have health-promoting effects. In this study, phenolic compounds of seven popularly consumed berries were analyzed by HPLC-ESI-MS/MS. In addition, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and nitrite scavenging activities were investigated for comparison of the biological activity of the berry extracts. Chlorogenic acid was the major phenolic acid, which had the highest content in aronia, followed by blueberry. The rutin and isoquercetin contents were the highest in mulberry, followed by aronia, black raspberry, and blueberry. The anthocyanin content was the highest in black raspberry. Aronia showed the highest DPPH free radical and nitrite scavenging activities with the highest contents of total phenolics and proanthocyanidins.

  • In the course of our investigation for chemical constituents in the ethyl acetate layer of Korean black raspberry wine, five compounds were isolated and purified by silica gel column chromatography and high-performance liquid chromatography. The isolated compounds were identified as ethyl succinate (1), vanillic acid (2), ethyl 3,4-dihydroxybenzoate (3), furan-2-ol (4), and 4-(4-hydroxyphenyl)butan-2(S)-ol (5) based on the spectroscopic data of electrospray ionization tandem mass spectrometry and nuclear magnetic resonance. The presence of 2 in Korean black raspberry has previously reported. However, 1 and 3-5 in Korean black raspberry and its wine were isolated for the first time.

The Effect of Red Ginseng and Black Raspberry fermented by Lactobacillus acidophilus C in High Fat Diet-fed Mice (김치로부터 분리한 Lactobacillus acidophilus C로 발효한 홍삼 및 복분자 발효물의 비만예방 효과)

  • We investigated the anti-obesity effects of Lactobacillus acidophilus C isolated from kimchi and red ginseng and black raspberry mixture fermented by this strain. Experimental samples were prepared with 10% skim milk, 1% red ginseng extract and 1% immature black raspberry powder and without red ginseng extract and black raspberry fermented by this strain at $37^{\circ}C$ for 24 hour. Male C57BL/6J mice were randomly divided into 4 groups (a normal diet group (N), a high-fat diet group (HF), a high fat diet with L. acidophilus C (C), and a high fat diet with fermented mixture (CEx)) and were orally administered daily for 6 weeks at concentration of 2,000 mg/kg body weight. The results showed that weight gain, weight of periepididymal fat and perirenal fat, adipocyte size, lipid accumulation in liver, serum glucose concentration, serum GOT and GPT concentration and serum leptin were decreased and concentration of HDL-cholesterol and serum adiponectin were increased compared to HF group. These results suggest that fermented mixture (CEx) might be helpful in improvement of high-fat diet-induced obesity than only L. acidophilus C intake.

Comparison of Fatty Acids and Antibacterial Activity against Pathogen of Acne in Different Parts of Ripened Black Raspberry (Rubus coreanus Miquel) (완숙 복분자 부위별 지방산 조성과 여드름 원인균에 대한 항균 활성)

  • The purpose of this study was to obtain the fatty acid composition and the antibacterial activity against pathogen of acne in various parts including seed, flesh and fruit from ripened black raspberry. In fatty acid analysis of lyophilized black raspberry, total fat of dried seed (11.047%) was much higher than in other parts. Moreover, linoleic acid (5.577%) and linolenic acid (2.317%) identified as the major fatty acids in the seed. By contrast, highest fatty acid of dried flesh was palmitic acid (0.323%) but it was remarkably low level compared with fatty acid composition of dried seed. Total polyphenol content in seed extract and flesh extract were 123.64 mg/g and 55.43 mg/g, respectively. DPPH radical scavenging ability of seed extract was higher than flesh and fruit extract. In antibacterial activity experiment against strains related acne such as Propionibacterium acne and Staphylococcus epidermidis, antibacterial activity of flesh extract was stronger than seed extract. However, seed extract showed antibacterial activity of 76.96~89.08% against each strain compared to activity of flesh extract. Based on these results, we concluded that seed of black raspberry can be useful in related field.

Volatile Analysis of Commercial Korean Black Raspberry Wines (Bokbunjaju) Using Headspace Solid-phase Microextraction (Headspace Solid-phase Microextraction을 이용한 시판 복분자주의 휘발성분 분석)

  • In this study, the volatile compounds in 24 commercial Korean black raspberry wines were isolated by headspace solid-phase microextraction and analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 43 volatile components, including 15 esters, 12 terpenes, 7 alcohols, 4 acids, 3 ketones, and 2 aldehydes, were identified. Ethyl esters and alcohols such as ethyl acetate, ethyl octanoate, isoamyl alcohol, and phenethyl alcohol were the most represented groups among the quantified volatiles. In particular, various terpenes such as DL-limonene, linalool, alpha-terpineol, and myrtenol were identified. The differences in volatile components among the 24 black raspberry wines and possible sample grouping were examined by applying principal component analyses to the GC-MS data sets. The first and second principal components explained 43.9% of the total variation across the samples. No apparent sample groupings were observed according to manufacturing locations. The samples KU, BH, SR, and MO showed higher overall levels in the concentrations of terpenes originating from black raspberry, while other samples such as BB and HB, showed higher in ethyl ester and alcohol contents produced by yeast fermentation, respectively.

Physicochemical properties and antioxidant activities of vinegar using black raspberry pomace (복분자 착즙박을 이용한 식초의 이화학적 특성 및 항산화 활성)

  • In this study, vinegar was prepared using black raspberry pomace to increase its utilization capacity. As a result of alcohol fermentation, the final alcohol content was 8.90% and the sugar content was 7.03°Brix. The total acid content after acetic acid fermentation was 4.44%, which was 0.38% higher than that of the juice. Among the organic acid content, the acetic acid content was higher in the pomace than in the juice. The total polyphenol (51.58 TAE mg/mL), total flavonoid (9.55 RUE mg/mL), and total anthocyanin (6.05 CYE mg/mL) contents of vinegar produced using black raspberry residue were higher than those of the juice. In addition, 2,2'-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity, reducing power, and ferric reducing antioxidant power (FRAP) activity were higher than those of the juice, indicating strong antioxidant properties. Thus, black raspberry pomace has excellent acetic acid fermentation ability and high antioxidant activity, indicating that it can be used as a functional fermented vinegar.

Immuno-stimulating Activities of Mannose-rich Polysaccharides Isolated from Korean Black Raspberry Wine (복분자주에서 분리한 Mannan 다당의 면역증진 활성)

  • Polysaccharides isolated from Korean black raspberry wine were examined for their chemical properties and immuno-modulating activities. The molecular mass of RB-1b-I, the main polysaccharide in black raspberry wine, was estimated as 180 kDa and it contained a significant proportion of mannose (76.8%) and 4 different minor component sugars such as galactose (15.8%), arabinose (3.8%), glucose (2.6%) and rhamnose (1.2%). This indicated that RB-1b-I was mainly present as a mannan, which had originated from the cell walls of fermenting yeasts. On the other hand, RB-1b-I induced high levels of macrophage activation as well as mitogenicity regarding murine splenocytes in vitro. The intravenous administration of RB-1b-I significantly augmented NK cytotoxicity against YAC-1 tumor cells. RB-1b-I also showed potent anti-complementary activity in a dose-dependent manner via both alternative and classical pathways. Results indicated that Korean black raspberry wine contains peculiar polysaccharides which provide beneficial immuno-stimulating activities for human health.

  • Common Embedded boards like the Arduino, Raspberry Pi, BeagleBone Black, leverages smart home systems, machine tools and various products in our day to day life. Managing and dealing frequent large scale incidents involving recent reagents and hazardous materials can be dangerous and difficult to detect in advance like in an event of an accidents or fires. In this paper I have done research by utilizing an Embedded (BeagleBone Black) boards sensors and RFID management system to detect a hazardous situation like fire in real time and avoiding it by sending out an alert message to the admin user to minimizing the risk. This system provides immediate information to the administrator of any hazardous situation and prevents any accidents from occurring.

  • The vehicle's black box is not suitable for bicycle black box because they contain sensors that are not suitable for bicycle black box. Therefore, this study implemented a system for viewing a video with a mobile phone as the slope changes. The angle was measured by using the gyro sensor and this angle was saved based on the angle setting on the raspberry pi. It has developed a system that allows users to download saved video files through mobile phone applications via FTP.

Phylogenic Relationships of Rubus Species Revealed by Randomly Amplified Polymorphic DNA Markers

  • Korean cultivated bramble, which is known as Bokbunja-ddal-gi is regarded to be originated from Korea native Rubus coreanus. However, little scientific evidence and significant morphological differences between Korean cultivated bramble(KCB) and R. coreanus throw doubt on the ancestry of KCB. This study was carried out to obtain phylogenetic information on KCB by comparing its nuclear genomic background with those of R. coreanus, black(R. occidentalis) and red(R. idaeus) raspberry, blackberry(R. lanciniatus) and R. crataegifolius. A total of 99 random amplified polymorphic DNA(RAPD) markers were generated and used for phylogenetic analysis of 76 Rubus accessions. Accessions of each species were grouped into each distinct subclade by the RAPD markers at a similarity coefficient of about 0.59. The KCB subclade formed a clade with R. occidentalis and R. crataegifolius subclades at a similarity coefficient of 0.47. The R. coreanus subclade formed a clade with R. idaeus, R. lanciniatus and R. crataegifolius subclades at a similar similarity coefficient. Only one KCB accession from Hoengsung was included in R. coreanus subclade. The accession shows leaf and flower characteristics different from the rest of the KCB accessions. The phylogenetic relationship inferred from the RAPD markers suggests that the nuclear genomic background of KCB accessions which show morphological similarity to black raspberry is more closely related to black raspberry than to R. coreanus. This brings about the need for close scientific evaluations on the ancestry of KCB at both morphological and molecular levels.
