Evaluation of Biostability of the Iron Oxide MRI Contrast Agent (산화철 조영제의 생체 안정성 평가)

  • Biostabilities of the iron oxide MRI contrast agent were evaluated using the cytotoxicity test and the animal test. Six experimental groups (35, 17, 5, 3.4, 1.7, 0.9 mg of iron oxide/10ml) and a control group were prepared for the cytotoxicity test. The cell count of higher concentration than 5 mg/10m1 become decrease significantly after 4-days incubation. Iron oxide was injected into the right vein of the two fully grown rabbits with the concentration of 0.5 or 0.9mg/kg of body weight respectively. Blood samples were collected and analyzed in turn after 30min, 1, 2, 4, 8, 16, 30, 60, 90days injection. The body temperature and weight of rabbits were measured simultaneously. The result of blood and serum analyses and the body temperature shows that there was no significant variation with respect to the sampling term and the dosage within this experimental conditions.

  • A new method for the prevention of foreign body-associated infections by controlled release of antibiotic was developed. The polyurethane (PU) matrix containing rifampicin was immobilized with hydrophilic photoreactive polyallylamine (PPA) containing azidophenyl groups. The rifampicin release characteristics and the long-lasting antimicrobial activities of the new material was compared with rifampicin-containing PU matrix without PPA membrane. The release rate of antibiotic from rifampicin-containing PU with PPA membrane significantly decreased as the thickness of PPA membrane was increased. The PPA-immobilized rifampicin-containing PU discs immersed in the PBS for 47 days had an efficient antimicrobial activity against both S. aureus and S. epidermidis.

  • [ $Y_2O_3:Eu$ ] powder was synthesized using a solution-combustion method by dissolving $(CH_3CO_2)_3Y$ and $(CH_3CO_2)_3$ Eu in methyl-alcohol solution. Results from X-ray diffractometery (XRD), thermogravimetry (TG)-differential thermal analysis (DTA) show that $Y_2O_3:Eu$ crystallizes completely when the dry powder is sintered at $500^{\circ}C$. The investigated optical properties were the photoluminescence emission spectra, the excitation spectra and luminescence decay curve. Europium (Eu) concentration had no observable effect on the optical spectrum which depended on the emission intensity. The mean lifetime of synthesized phosphors was $2.3\~2.6 ms$.

  • Cell reprogramming has been considered a powerful technique in the regenerative medicine field. In addition to diverse its strengths, cell reprogramming technology also has several drawbacks generated during the process of reprogramming. Telomere shortening caused by the cell reprogramming process impedes the efficiency of cell reprogramming. Transcription factors used for reprogramming alter genomic contents and result in genetic mutations. Additionally, defective mitochondria functioning such as excessive mitochondrial fission leads to the limitation of pluripotency and ultimately reduces the efficiency of reprogramming. These problems including genomic instability and impaired mitochondrial dynamics should be resolved to apply cell reprograming in clinical research and to address efficiency and safety concerns. Sirtuin (NAD+-dependent histone deacetylase) has been known to control the chromatin state of the telomere and influence mitochondria function in cells. Recently, several studies reported that Sirtuins could control for genomic instability in cell reprogramming. Here, we review recent findings regarding the role of Sirtuins in cell reprogramming. And we propose that the manipulation of Sirtuins may improve defects that result from the steps of cell reprogramming.

A Study on Development of Humeral Intramedullary Fixation Nail based on Korean Cadaver Tests (한국인 Cadaver Test에 대한 상완골 골수정 개발에 관한 연구)

  • The aim of this study is to develop a humeral Intramedullary fixation nail (HIFN) suitable for Korean people. In this study, CT images were obtained from 72 Korean cadaveric humeral bones and 3D Korean humeral bone models were reconstructed based on the CT images to investigate anatomical characteristics. Major design parameters of HIFN were selected using the morphological measurement information of the Korean humeral bone models. Through finite element analysis and mechanical tests, the developed HIFN prototype was compared with the Polarus HIFN ($ACUMED^{(R)}$, USA), and it was found that the HIFN prototype showed similar and/or superior mechanical performance compared to the Polarus HIFN. Also, clinical validation for the HIFN prototype was carried out to check predictable troubles in surgical operations. Finally, optimal design modification was proposed to prevent the possible axillary nerve injury due to the locking screw system of the HIFN prototype.

A simple guide to the structural study on membrane proteins in detergents using solution NMR

  • NMR-based structural studies on membrane proteins are appreciated quite challenging due to various reasons, generally including the narrow dispersion of NMR spectra, the severe peak broadening, and the lack of long range NOEs. In spite of the poor biophysical properties, structural studies on membrane proteins have got to go on, considering their functional importance in biological systems. In this review, we provide a simple overview of the techniques generally used in structural studies of membrane proteins by solution NMR, with experimental examples of a helical membrane protein, caveolin 3. Detergent screening is usually employed as the first step and the selection of appropriate detergent is the most important for successful approach to membrane proteins. Various tools can then be applied as specialized NMR techniques in solution that include sample deteuration, amino-acid selective isotope labeling, residual dipolar coupling, and paramagnetic relaxation enhancement.

Sensor Network-based u-Healthcare System (센서네트워크 기반 u-Healthcare 시스템)

  • Recently, owing to the development of ubiquitous, RFID and local area wireless communication technology, many studies on the system which can measure biomedical signals are being carried out. In this paper, we have designed and implemented an u-Healthcare system based on sensor network using biomedical signal measurement sensors such as ECG, blood pressure, and heartbeats. The biomedical signals from sensor nodes pass through the gateway and are finally transmitted to a healthcare renter. The acquired biomedical signals are processed in the healthcare center and the analyzed results are transmitted to the patients to improve patients' health using either kinesitherapy or dietary treatment.

Increased Association of ${\alpha}$-synuclein to Perturbed Cellular Membranes

  • [ ${\alpha}$ ]synuclein (${\alpha}$-syn) is implicated in the pathogenesis of Parkinson's disease (PD) and other related diseases. We have previously reported that ${\alpha}$-syn binds to the cell membranes in a transient and reversible manner. However, little is known about the physiologic function and/or consequence of this association. Here, we examined whether chemically induced perturbations to the cellular membranes enhance the binding of ${\alpha}$-syn, based on hypothesis that ${\alpha}$-syn may play a role in maintenance of membrane integrity or repair. We induced membrane perturbations or alterations in ${\alpha}$-syn-overexpressing human neuroblastoma cells (SH-SY5Y) by treating the cells with hydrogen peroxide ($H_2O_2$) or oleic acid. In addition, membranes fractionated from these cells were perturbed by treating them with proteinase K or chloroform. Dynamic interaction of ${\alpha}$-syn to the membranes was analyzed by the chemical cross-linking assay that we developed in the previous study. We found that membrane interaction of ${\alpha}$-syn was increased upon treatment with membrane-perturbing reagents in a dose and time dependent manner. These results suggest that perturbations in the cellular membranes cause increased binding of ${\alpha}$-syn, and this may have significant implication in the physiological function of ${\alpha}$-syn in cells.

  • The development of functional polymeric materials for biomedical application has progressed on the basis of functionality, biocompatibility and biodegradability. In this paper we review the functional polymeric biomaterialsbased systems and propose a range of biomedical applications in the near future. These systems include the functional biodegradable polymers synthesized in our research laboratory, biodegradable polymeric materials, thermosensitive polymeric materials, cationic polymeric materials, non-condensing polymeric biomaterials, bio-polymeric DNA matrix for tissue engineering, and polymeric biomaterials for RNA interference (RNAi) technology.

  • As conventional drug delivery system is being improved rapidly by target-based drug delivery system, finding suitable Drug Delivery System (DDS) for new drugs remains a challenge. Although there are many drug delivery vehicles in existence, a significant improvement is required to some DDS such as for local, implant-based treatments used for musculoskeletal infections. Many polymers have been considered for providing the improvement in DDS. Synthetic polymer, for example, has gained popularity for broad-spectrum physicochemical and mechanical properties. This article reviews the biomedical applications of Poly(TriMethylene Carbonate-co-Caprolactone) (PTMCC), which has attracted attention due to its biocompatibility, biodegradability and rubber-like properties. Its synthesis, physical properties, and degradation are also discussed here. Although it is relatively new in biomedical applications, it is readily usable for the fabrication of differing format of DDS of superior mechanical strength and degradation properties. The use of PTMCC is expected to increase in coming years as more is revealed about its potentials.
