Effects of Three-dimensional Scaffolds on Cell Organization and Tissue Development

  • Tissue engineering scaffolds play a critical role in regulating the reconstructed human tissue development. Various types of scaffolds have been developed in recent years, including fibrous matrix and foam-like scaffolds. The design of scaffold materials has been investigated extensively. However, the design of physical structure of the scaffold, especially fibrous matrices, has not received much attention. This paper compares the different characteristics of fibrous and foam-like scaffolds, and reviews regulatory roles of important scaffold properties, including surface geometry, scaffold configuration, pore structure, mechanical property and bioactivity. Tissue regeneration, cell organization, proliferation and differentiation under different microstructures were evaluated. The importance of proper scaffold selection and design is further discussed with the examples of bone tissue engineering and stem cell tissue engineering. This review addresses the importance of scaffold microstructure and provides insights in designing appropriate scaffold structure for different applications of tissue engineering.

  • Recently, microalgae have been focused on potential biomass for bio-diesel and biorefinery process. The aim of this paper is to review the biorefinery process including biodesel using microalgae as a microreactor. The state-of-the-art of biodiesel and biorefinery research such as extraction and reaction process as well as byproducts utilization is described. In addition, we suggest possibility for develop bioactive substances and their industrial products from byproducts of microalgae massively obtained after bio-diesel extraction.

  • A new glycerol ether has been isolated from a marine sponge Stelletta sp. By bioactivity-guided fractionation. The structure was established on the basis of NMR and MS analyses. The compound was evaluated for cytotoxicity against a small panel of five human tumor cell lines and exhibited significant cytotoxicity.

  • The genus Polygonatum species belongs to the family Liliaceae which is widely distributed over areas of the north temperature zone. There are about forty more plants of Polygonatum species in the world widely distributed in eastern Europe and south east Asia. The plants of Polygonatum species have been used not only as ornamental plants but also for their medicinal values. This article is concerned with the specific properties and flavour of the drug and its history as a medicine, showing the main functional components of Polygonatum species of flavonoids, steroidal glycosides, and saccharides.

Tsaokoarylone, a Cytotoxic Diarylheptanoid from Amomum tsao-ko Fruits

  • The crude methanol extract of the fruits of Amomum tsao-ko (Zingiberaceae) showed cytotoxic activity. Bioactivity-guided separation led to the isolation of a diarylheptanoid, tsaokoarylone [7-(4-hydroxy-3-methoxyphenyl)-1-(4-hydroxyphenyl)-hepta-4E,6E-dien-3-one] (2). 2 showed cytotoxicity at 4.9 and 11.4 $\mu$g/mL ($IC_{50}$) against human nonsmall cell lung cancer A549 and human melanoma SK-Mel-2, respectively, determined by SRB colorimetric method. During purification-(4-hydroxyphenyl)-4-hydroxyhexan-2-one (4) together with three known diarylheptanoids was also isolated. Their structures were determined from interpretation of spectroscopic data (IR, UV, MS, and NMR) and synthesis confirmed the structure of 2.

Cytotoxic Constituents of Saussurea lappa

  • The crude extract of Saussurea lappa displayed significant lethality to brine shrimp larvae. Investigation of the causative components by bioactivity-directed fractionation resulted in the isolation of three $C_17$-polyene alcohols. Based on various nmr spectral data, these compounds were identified as shikokiols which had been previously isolated from Cirsium nipponicum and/or Centaurea aegyptica. These $C_17$-polyene alcohols exhibited moderate cytotoxicities against the human tumor cell lines, A549, SK-OV-3, SK-MEL-2, XF498, and HCT15.

Synthesis and Biological Activities of 8-Arylflavones

  • A number of 8-arylflavones have been synthesized as congeners of wogonin and evaluated for their inhibitory activities of $PGE_2$ production. 8-Arylflavones were obtained from commercially available chrysin via two different synthetic pathways. Most 8-arylflavones exhibited much reduced inhibitory activities against COX-2 catalyzed $PGE_2$ production compared to that of wogonin. Functional group replacement at the 8-position of wogonin from methoxy to aryl group caused loss of inhibitory activity. Our present results imply that the functional group at the 8-position of flavones seems to play very important roles for bioactivity.

Polyketides from a Sponge-Derived Fungus, Aspergillus versicolor

  • Bioactivity guided fractionation of the cultured filtrates of Aspergillus versicolor, which was derived from a marine sponge Petrosia sp., yielded three polyketides: decumbenones A (1),B (2), and versiol (3). These compounds were identified on the basis of 1D and 2D NMR spectroscopic and MS analysis. The absolute configuration was defined by the modified Mosher's method. The isolated compounds were tested for cytotoxicity against a panel of five human solid tumor cell lines and antibacterial activity against twenty clinically isolated methicillin-resistant strains. This is the first report on the isolation of these compounds from a marine source.

The Synergistic Antibacterial Activity of 1-Acetyl-$\beta$-Carboline and $\beta$-Lactams Against Methicillin-Resistant Staphylococcus aureus (MRSA)

  • 1-Acetyl-$\beta$-carboline was isolated as an anti-MRSA agent from the fermentation broth of a marine actinomycete isolated from marine sediment. The producing strain was identified to be Streptomyces sp. by phylogenetic analysis of the 16S rRNA gene sequence. The anti-MRSA agent was isolated by bioactivity-guided fractionation of the culture extract by solvent partitioning, ODS open flash chromatography, and purification with a reversed-phase HPLC. Its structure was elucidated by extensive 2D NMR and mass spectral analyses. Combination of 1-acetyl-$\beta$-carboline with ampicillin exhibited synergistic antibacterial activity against MRSA.

  • Calcium phosphate ceramics have been applied intensively to orthopaedic and dental implants by virtue of their osteoconductive nature. In an attempt to enhance the bone implant intergrity and Eta utility, these ceramics are deposited onto the porous surface of metallic implants. The coating procedure and the ensuing phase transformations of the ceramic alter the mechanical properties and surface chemistry of the ceramic layers as well as those of the substrate. These structural and compositional differences are directly related to the interaction mechanisms at the surface-active ceramicbone interface. Material and processing induced influences on dissolution, electrokinetic behavior, ceramic-metallic substrate interface and boRe growth enhancement are presented.
