백출에서 분리된 Atractylenolide II의 RAW264.7 대식세포와 BV2 미세아교세포에서의 항염증 효과 (Atractylenoide II Isolated from Atractylodes macrocephala Inhibited Inflammatory Responses in Lipopolysaccharide-induced RAW264.7 Macrophages and BV2 Microglial Cells)

  • Atractylodes macrocephala is a perennial herb and is a member of the Compositae family. This plant is known to contain various bioactive constituents indicating anti-inflammatory, neuroprotective, anti-oxidant, immunological enhancement, and gastroprotective effects. In this investigation, we isolated four compounds with similar chemical structures from A. macrocephala, and evaluated their anti-inflammatory effects. Among the four compounds, compound 2(atractylenolide II) showed the second-best inhibitory effect on the lipopolysaccharide(LPS)-induced production of nitric oxide in RAW264.7 macrophages and BV2 microglial cells. Compound 2 also inhibited the LPS-induced the production of prostaglandin E2(PGE2), and the expression of inducible nitric oxide synthase(iNOS) and cyclooxygenase(COX)-2 proteins in both cells. In addition, compound 2 suppressed the production of pro-inflammatory cytokines including interleukin(IL)-1β, IL-6, and tumor necrosis factor(TNF)-α. These inhibitory effects were contributed by inactivation of nuclear factor kappa B(NF-κB) and mitogen-activated protein kinases(MAPKs) pathways by treatment with compound 2. This compound did not induce the expression of heme oxygenase(HO)-1 protein indicating that the anti-inflammatory effect of compound 2 was independent with HO-1 protein. Taken together, these results suggested that atractylenolide II can be a candidate material to treat inflammatory diseases.

송이줄기 첨가에 따른 Muscat Bailey A 포도의 발효 중 발효액 및 포도주의 생리활성 물질 함량 (Phenolic compounds of must and wine supplemented with Muscat Bailey A grape fruit stem)

  • 이전 연구에서 포도 품종별 및 부위별 생리활성 물질을 분석한 결과 MBA 품종의 송이줄기 부위에서 trans-resveratrol 함량이 다른 품종에 비해 수십배 높았다. 이를 바탕으로 포도주 제조 시 버려지는 비가식 부위인 송이줄기를 농도별로 첨가해 발효 중 발효액 및 포도주의 폴리페놀 함량을 조사하였다. 송이줄기 첨가량에 따른 발효액의 발효 중 적색도, 총안토시안 함량, 총폴리페놀 및 탄닌 함량은 발효후기로 갈수록 송이줄기 첨가에 따라 유의적으로 증가하였고, 포도주에서도 같은 경향을 보였다. 반면 포도주의 휘발산 함량은 송이줄기 첨가량이 증가할수록 감소하는 경향을 보였다. 발효액 및 포도주의 폴리페놀 성분을 분석한 결과 catechin(8.16~23.08 mg/L)함량이 가장 높았으며 gallic acid(2.32~3.28 mg/L), trans-resveratrol(1.38~3.27 mg/L)및 ferulic acid(1.51~1.59 mg/L) 순서로 함량이 높았다. MBA 포도주의 항산화활성을 DPPH $IC_{50}$로 측정한 결과 송이줄기 함량에 비례해 활성이 증가하였으며, 송이줄기 5% 첨가한 포도주(12 mg/L)는 ascorbic acid의 DPPH $IC_{50}$(67 mg/L)보다 낮은 농도에서 높은 항산화활성을 나타내었다.

Potentially Bioactive Two New Natural Sesquiterpenoids from the Rhizomes of Zingiber zerumbet

  • Repeated chromatography of the n-hexane-soluble fraction of the MeOH extract of the rhizomes of Zingiber zerumbet led to the isolation of two isomers of 6-methoxy-2E,9E-humula-dien-8-one (1 and 2) and stigmast-4-en-3-one. The structures of 1 and 2 were determined by spectroscopic methods including 10 and 2D-NMR elucidated by analysis of spectroscopic data as well as by comparison with published values. This is the first report on the isolation of compounds 1 and 2 from the nature. Stigmast-4-en-3-one was first isolated from this plant.

십전대보탕 발효물의 성분 분석 및 뇌신경 세포 보호 활성 (The Study on Compounds of the Fermented Sipjundaebo-tang and its Neuroprotective Activity)

  • Sipjundaebo-tang was a well-known restorative traditional herbal prescription that used to treat anemia, anorexia, fatigue and inflammation. In this study, we examined the bioconversion of compounds in the Sipjundaebo-tang (SJ) and fermented Sipjundaebo-tang with Lactobacillus fermentum KFRI 164 (FSJ) using established HPLC-DAD method. The chromatogram of FSJ has shown that the contents of six bioactive compounds 5-HMF, paeoniflorin, ferulic acid, cinnam aldehyde, decursin, glycyrrhizin in SJ has decreased. And the contents of unknown compounds (1), (2), (3), (4) and (5) in FSJ were higher than each contents of SJ. The antioxidant activities of SJ and FSJ were conducted by DPPH free radical and Hydrogen peroxide ($H_2O_2$) scavenging assay. Electron donating activity (EDA, %) value of FSJ has shown higher than 21.9% and 14.5% at a concentration of 0.5 mg/ml for DPPH radical scavenging activity and $H_2O_2$ scavenging activity, respectively. Also, the neuroprotective activities of SJ and FSJ against glutamate-induced oxidative stress in a mouse hippocampal cell line (HT22) were evaluated by MTT assay. As a result, FSJ has shown higher neuroprotective activity than 56.5% comparing with SJ. In conclusion, the fermented SJ using microorganism could convert compounds in SJ and enhance antioxidant activity and neuroprotective activity.

약용식물 추출물에 의한 면역세포 산화질소 생성 억제 활성 분석 (Inhibitory Effects of Methanolic Extracts of Medicinal Plants on Nitric Oxide Production in Activated Macrophage RAW 264.7 Cells)

  • A variety of herbs and plants have been traditionally used in oriental folk medicine for the treatment of inflammatory diseases. In our attempt to search for anti-inflammatory agents from natural products, we investigated 64 methanol extracts from 42 medicinal plants belonging to 10 families which were evaluated for inhibitory activities of NO production in lipopolysaccharide (LPS)-stimulated macrophage RAW 264.7 cells. Among them, 16 extracts exhibited inhibitory activities of NO production ($IC_{50}$ values ranging from 59.6 to 94.7 ${\mu}g/m{\ell}$). Only the extract from aerial parts of Hosta lancifolia (H. lancifolia) did not exert cytotoxic effects at the concentrations tested. The extract from H. lancifolia decreased the mRNA and protein levels of inducible nitric oxide synthase (iNOS) and pro-inflammatory cytokines in activated macrophage RAW 264.7 cells in dose-dependent manner. The results suggest that the extract may contain bioactive compounds that suppress expression of pro-inflammatory cytokines, which may prove beneficial with regard to the development of natural agents for prevention and treatment of inflammatory diseases.

전탕용수의 종류에 따른 대황 성분의 추출효과 (Effects of the decoction water on the extraction of the bioactive compounds from rhubarb)

  • 본 연구는 제주지역에서 생산되는 세 종류의 물과 서울지역에서 사용되고 있는 수돗물 등 네 종류의 물을 전탕용수로 하여 중요 한약재인 대황으로부터 활성성분을 추출하고 용수 중의 미네랄이 대황활성성분 추출에 미치는 효과에 대하여 비교 분석하였다. 유도결합플라즈마방출분광기와 유도결합플라즈마질량분석기를 이용하여 30여종의 미네랄과 다수의 음이온을 분석하였다. 각 전탕용수로 대황을 추출하고 그 추출액의 일부를 증발시켜 총 추출량을 구하고, 또한 HPLC를 이용하여 추출액으로부터 대황의 생리활성 성분을 분석하였다. 용수 간 총 추출량과 대황 성분함량의 차이 검정을 위하여 Kruskal-Wallis 검정을 시행하였고, 미네랄, 음이온, 총 추출량 및 성분함량과의 관계분석을 위하여 다중요인분석을 시행하였다. 분석결과 용수 중의 미네랄 함량은 대황의 총 추출량 및 성분 추출량과 높은 상관관계가 있는 것으로 나타났다.

지역별, 연근별 가공백삼의 품질과 지표 성분의 변이 (Variation in Content of Marker Compounds and Quality of Processed White Ginseng of Different Cultivation Ages from Different Regions)

  • Background: The ginsenosides Rb1 (G-Rb1) and Rg1 (G-Rg1) are used as marker compounds, and are the principal bioactive compounds assessed in the quality control of white ginseng. This study was conducted to analyze white ginseng samples of different and to obtain useful data for the quality control of white ginseng. Methods and Results: The variation in the content of G-Rb1 and G-Rg1 was evaluated among 35 samples of 4-, 5-, and 6-year-old white ginseng. The content of both G-Rb1 and G-Rg1 did not significantly differ among ages, and the relative ratio of the maximum to the minimum content of these within ginseng of the same ages was more than two. However, the ratio of G-Rb1 to G-Rg1 content in the 5- and 6-year-old ginseng was significantly higher than that in the 4-year-old one. According to the 'Ginseng industrial act', the standard (w/w, %) minimum $G-Rg_1$ and $G-Rb_1$ content is 0.10% and 0.20% or more, respectively. Among the 35 samples examined, the content of $G-Rg_1$ was found to be 0.124 - 0.399% with none being less than the standard level, while that of $G-Rb_1$, was 0.147 - 0.595%, with 4 samples (11.4%) failing to meet the standard levels. The content of $G-Rg_1$ and $G-Rb_1$ did not show a constant relationship with the size of ginseng. Conclusions: In our study, the content of both G-Rg1 and G-Rb1 varied widely, and there was no significant difference among cultivation ages. The results of the present study might provide useful information for the quality control of raw ginseng and processed white ginseng using marker compound.

율무 새싹 추출물의 Riboflavin과 Coixol의 동시 분석법 검증 및 항산화 활성 (Validation of a Method and Evaluation of Antioxidant Activity for the Simultaneous Determination of Riboflavin and Coixol in Coix lacryma-jobi var. ma-yuen Stapf Sprouts)

  • 율무 새싹 추출물의 riboflavin과 coixol의 원료표준화를 위한 동시분석 조건을 검증하고, 율무 새싹의 전초와 잎 추출물의 항산화 활성과 함량의 상관관계에 대해 조사하였다. 1. HPLC 분석을 위한 riboflavin과 coixol의 피크가 각각 분리되어 특이성을 확인하였고, 일내 및 일간 정확성은 모두 회수율이 100.28%에서 103.98% 사이이며 정밀성은 모두 1.93%이하를 나타내므로 본 실험의 분석조건은 높은 정확성과 정밀성을 가지고 있다. 2. Riboflavin과 coixol의 동시분석 조건의 검출한계(LOD) 및 정량한계(LOQ)를 조사한 결과 검출한계는 0.71 및 0.31 ㎍/mL, 정량한계는 2.71 및 0.94 ㎍/mL이므로 확립된 HPLC 분석조건은 riboflavin과 coixol의 동시분석하기에 무리가 없는 분석법으로 조사되었다. 3. 율무 새싹의 전초와 잎의 riboflavin과 coixol의 함량을 본 연구에서 얻어진 동시분석법을 적용하여 잎의 환류 추출물에서 가장 높은 함량이 나타났다. 4. 항산화 활성 측정 시, DPPH와 ABTS+ 소거능에서 모두 율무 새싹의 전초보다 잎에서, 상온 추출보다 환류 추출이 더 낮은 IC50값을 나타내었다. 율무 새싹 추출물의 riboflavin과 coixol 및 항산화 활성과의 상관관계를 실시한 결과, coixol이 증가할수록 DPPH와 ABTS+ 소거능의 IC50값이 유의적으로 감소하는 경향이 나타나므로 원료표준화 지표뿐만 아니라 기능성 지표로 활용이 가능할 것으로 판단되었다.
