Environmental Monitoring of Herbicide Tolerant Genetically Modified Zoysiagrass (Zoysia japonica) around Confined Field Trials (제초제저항성 유전자변형 들잔디의 시험 격리포장 주변 환경방출 모니터링)

  • The cultivation area and use of genetically modified (GM) crops have been increased continuously over the world. Seed distribution and transgenes to environmental ecosystem is one of the most important factors in risk assessment and risk management of GM crop. Safe management for the development and commercialization of GM crops is required according to The Act on Transboundary Movements of Living Modified Organisms,etc (LMO Act) in Korea. This study was conducted to setup the environmental monitoring system of GM zoysiagrass (event JG21 and JG21-MS). The monitoring was performed in 4 GMO confined fields, Sungwhan, Ochang, Jeju University and Jeju Namwon. In the result of monitoring, we could not found any gene flow and distribution of GM zoysiagrass in the 3 fields, but one spill of JG21 was found in the Namwon field in 2012. These results suggest that continuous monitoring is necessary to detect the occurrence of GM zoysiagrass for preventing genetic contamination in natural environment.

Morphology Control of Nanostructured Graphene on Dielectric Nanowires

  • Graphene is a sp2-hybridized carbon sheet with an atomic-level thickness and a wide range of graphene applications has been intensely investigated due to its unique electrical, optical, and mechanical properties. In particular, hybrid graphene structures combined with various nanomaterials have been studied in energy- and sensor-based applications due to the high conductivity, large surface area and enhanced reactivity of the nanostructures. Conventional metal-catalytic growth method, however, makes useful applications difficult since a transfer process, used to separate graphene from the metal substrate, should be required. Recently several papers have been published on direct graphene growth on the two dimensional planar substrates, but it is necessary to explore a direct growth of hierarchical nanostructures for the future graphene applications. In this study, uniform graphene layers were successfully synthesized on highly dense dielectric nanowires (NWs) without any external catalysts. We also demonstrated that the graphene morphology on NWs can be controlled by the growth parameters, such as temperature or partial pressure in chemical vapor deposition (CVD) system. This direct growth method can be readily applied to the fabrication of nanoscale graphene electrode with designed structures because a wide range of nanostructured template is available. In addition, we believe that the direct growth growth approach and morphological control of graphene are promising for the advanced graphene applications such as super capacitors or bio-sensors.

Non-gaseous Plasma Immersion Ion Implantation and Its Applications

  • A new plasma process, i.e., the combination of PIII&D and HIPIMS, was developed to implant non-gaseous ions into materials surface. HIPIMS is a special mode of operation of pulsed-DC magnetron sputtering, in which high pulsed DC power exceeding ~1 kW/$cm^2$ of its peak power density is applied to the magnetron sputtering target while the average power density remains manageable to the cooling capacity of the equipment by using a very small duty ratio of operation. Due to the high peak power density applied to the sputtering target, a large fraction of sputtered atoms is ionized. If the negative high voltage pulse applied to the sample stage in PIII&D system is synchronized with the pulsed plasma of sputtered target material by HIPIMS operation, the implantation of non-gaseous ions can be successfully accomplished. The new process has great advantage that thin film deposition and non-gaseous ion implantation along with in-situ film modification can be achieved in a single plasma chamber. Even broader application areas of PIII&D technology are believed to be envisaged by this newly developed process. In one application of non-gaseous plasma immersion ion implantation, Ge ions were implanted into SiO2 thin film at 60 keV to form Ge quantum dots embedded in SiO2 dielectric material. The crystalline Ge quantum dots were shown to be 5~10 nm in size and well dispersed in SiO2 matrix. In another application, Ag ions were implanted into SS-304 substrate to endow the anti-microbial property of the surface. Yet another bio-application was Mg ion implantation into Ti to improve its osteointegration property for bone implants. Catalyst is another promising application field of nongaseous plasma immersion ion implantation because ion implantation results in atomically dispersed catalytic agents with high surface to volume ratio. Pt ions were implanted into the surface of Al2O3 catalytic supporter and its H2 generation property was measured for DME reforming catalyst. In this talk, a newly developed, non-gaseous plasma immersion ion implantation technique and its applications would be shown and discussed.

Study on the Development of the Optimum Feeding Regimen for Pigs Weaned at 21 Days of Age

  • This experiment was conducted to investigate the effect of various feeding regimens on growth performance, nutrient digestibilities, nitrogen retention, blood urea nitrogen (BUN) concentration and feed cost in young pigs weaned at 21 days of age. One hundred twenty crossbred pigs (Landrace${\times}$Large White${\times}$Duroc, average 6.8 kg BW), weaned at 21 days of age, were allotted to 5 treatments in a 5 replicates by a randomized completely block (RCB) design. Treatments were as follow: 1) 1P (1-4 weeks: CP 23% and lysine 1.60%), 2) 2P-I (1 week: CP 23% and lysine 1.60%, 2-4 weeks: CP 21% and lysine 1.45%), 3) 2P-II (1-2 weeks: CP 23% and lysine 1.60%, 3-4 weeks: CP 21% and lysine 1.45%), 4) 2P-III (1-3 weeks: CP 23% and lysine 1.60%, 4 week: CP 21% and lysine 1.45%), 5) 3P (1 week: CP 23% and lysine 1.60%, 2-3 weeks: CP 21% and lysine 1.45%, 4 week: CP 19% and lysine 1.30%). Three different diets were formulated and supplied according to phase feeding programs. Diet 1 contains 23% crude protein and 1.60% lysine, diet 2 contains 21% crude protein and 1.45% lysine and diet 3 contains 19% crude protein and 1.30 lysine, respectively. Although there was no significant difference in growth performances, there was a beneficial effect of 3 phase feeding. The ADG was higher in 3P treatment than other treatments and it was observed clearly in late period (3-4 weeks) than in early period. Also, with increase in age, growth rate of pigs in 3P treatment was higher than that in 1P treatment approximately 37% (p=0.1379). There were no significant differences among all treatments in nutrient digestibility. The concentration of BUN was higher in pigs were fed diet containing 21% crude protein and 1.45% lysine (eg, 2P-1 and 3P) than those supplied diet containing high nutrient value at 2 week. The lowest feed cost/kg weight gain of pigs showed in 3P among treatments (p<0.05) whereas, high feed cost/kg weight gain of pigs was calculated in 1P and 2P-II treatments compared with 2P-I and 2 P-II (p<0.05), because of high milk products were used in those diet.

Diversity of Insect Fauna in Junam Wetland of Korea (주남 습지대의 곤충다양성)

  • A sampling survey was conducted at three reservoirs of Junam wetland ($6.02km^2$) in Korea to identify the wetland insect fauna along with their dominance, diversity, richness and evenness. Methods of monitoring were visual inspection and sweeping in 2010, Malaise trapping in 2011, light trapping and pitfall trapping in 2012. In total, 9,269 individuals (36.3% coleopterans, 21.3% lepidopterans and 13.9% odonates) were collected, belonging to 574 species, 141 families and 14 orders. For the number of species, lepidopterans shared the highest (31.2%), followed by coleopterans (28.0%) and hemipterans (12.9%). Dominant species were Enochrus simulans (Coleoptera) (7.9% of total individuals) followed by Hydaticus grammicus (Coleoptera) (4.3%), Galerucella nipponensis (Coleoptera) (4.1%), Elophila interruptalis (Lepidoptera) (3.1%) and Apis mellifera (Hymenoptera) (2.2%). Total counts of coleopterans, lepidopterans and odonates in the three reservoirs were quite high, but the counts were not significantly different among the reservoirs. Insect diversity index (H') and richness index (RI) of the Junam wetland were 5.04 and 59.10, respectively.

Simultaneous Characterization of Sofalcone and Its Metabolite in Human Plasma by Liquid Chromatography -Tandem Mass Spectrometry

  • A sensitive and selective method for quantitation of sofalcone and its active metabolite in human plasma has been established using liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI/MS/MS). Plasma samples were transferred into 96-well plate using an automated sample handling system and spiked with 10 $\mu$L of 2 $\mu$g/mL $d_3$-sofalcone and $d_3$-sofalcone metabolite solutions (internal standard), respectively. After adding 0.5 mL of acetonitrile to the 96-well plate, the plasma samples were then vortexed for 30 sec. After centrifugation, the supernatant was transferred into another 96-well plate and completely evaporated at 40 ${^{\circ}C}$ under a stream of nitrogen. Dry residues were reconstituted with mobile phase and were injected into a $C_{18}$ reversed-phase column. The limit of quantitation of sofalcone and its metabolite was 2 ng/mL, using a sample volume of 0.2 mL for analysis. The reproducibility of the method was evaluated by analyzing 10 replicates over the concentration range of 2 ng/mL to 1000 ng/mL. The validation experiments of the method have shown that the assay has good precision and accuracy. Sofalcone and its metabolite produced a protonated precursor ion ([M+H]$^+$) of m/z 451 and 453, and a corresponding product ion of m/z 315 and 317, respectively. Internal standard ($d_3$-sofalcone and $d_3$-sofalcone metabolite) produced a protonated precursor ion ([M+H]$^+$) of m/z 454 and 456 and a corresponding product ion of m/z 315 and 317, respectively. The method has been successfully applied to a pharmacokinetic study of sofalcone and its active metabolite in human plasma.

Detection of Mycobacterium Tuberculosis by Loop-Mediated Isothermal Amplification Assay (등온 증폭법을 이용한 결핵균의 빠른 검출 시스템 개발)

  • Mycobacterium tuberculosis (MTB) remains a major worldwide public health problem. In recent years, the incidence of MTB has been rising. Rapid and reliable diagnosis of Mycobacterium tuberculosis is essential to initiate correct treatment, avoid severe complications, and prevent transmission. LAMP was used to develop a rapid and sensitive laboratory diagnostic system for the MTB. In this research, the loop-mediated isothermal amplification method (LAMP) that amplifies DNA with high specificity and rapidity at an isothermal condition was evaluated for rapid detection of MTB. Undiluted DNA (2.10 × 106 copy/mL), 10-1, 10-2, 10-3, 10-4, 10-5 and 10-6 (copy/mL) of MTB DNA were amplified by PCR and LAMP to determine the sensitivity of the assay. At results, the LAMP assay reported here has the advantages of rapid amplification, high sensitivity, and high specificity and will be useful for rapid and reliable clinical diagnosis of MTB in hospital clinical laboratory.

A Study on the Improvement Scheme of University's Software Education

  • In this paper, we propose an effective software education scheme for universities. The key idea of this software education scheme is to analyze software curriculum of QS world university rankings Top 10, SW-oriented university, and regional main national university. And based on the results, we propose five improvements for the effective SW education method of universities. The first is to enhance the adaptability of the industry by developing courses based on the SW developer's job analysis in the curriculum development process. Second, it is necessary to strengthen the curriculum of the 4th industrial revolution core technologies(cloud computing, big data, virtual/augmented reality, Internet of things, etc.) and integrate them with various fields such as medical, bio, sensor, human, and cognitive science. Third, programming language education should be included in software convergence course after basic syntax education to implement projects in various fields. In addition, the curriculum for developing system programming developers and back-end developers should be strengthened rather than application program developers. Fourth, it offers opportunities to participate in industrial projects by reinforcing courses such as capstone design and comprehensive design, which enables product-based self-directed learning. Fifth, it is necessary to develop university-specific curriculum based on local industry by reinforcing internship or industry-academic program that can acquire skills in local industry field.

A Survey on the Public Awareness of the Bioinformation Utilization (바이오정보 활용에 대한 시민 인식 조사 연구)

  • In Korea, currently, only the growth possibility of bioinformation industry has received the public attention, and social discussion on bioinformation management is meager. Based on this critical perspective, this study investigates the current status of public awareness of the bioinformation which can be a basis for the social discussion for bioinformation management. The most significant characteristics identified in the survey is that many citizens, while expressing their agreement on the use of biometric technology by purpose and taking an open stance on national fingerprint information collection, have considerable concern for the identity authentication system using the current biometric technology. In particular, the survey pointed out that it is hard to trust the institutions that collect the bioinformation. Also, the public expressed the particular concern for the health data including the body information and showed overwhelming consent for the necessity of more stringent regulation compared with other personal information. In regards to the medical information gathered from medical institutions, nearly 80% of the survey respondents did not agree on providing the information to commercial enterprises, and even 60% of them disagreed on using it for academic and statistical research. This result well demonstrates that thorough management of the bioinformation is required for its utilization.

Emotion Recognition Method Using Heart-Respiration Connectivity (심장과 호흡의 연결성을 이용한 감성인식 방법)

  • Physiological responses have been measured to recognize emotion. Although physiological responses have been interrelated between organs, their connectivities have been less considered for emotion recognizing. The connectivities have been assumed to enhance emotion recognition. Specially, autonomic nervous system is physiologically modulated by the interrelated functioning. Therefore, this study has been tried to analyze connectivities between heart and respiration and to find the significantly connected variables for emotion recognition. The eighteen subjects(10 male, age $24.72{\pm}2.47$) participated in the experiment. The participants were asked to listen to predetermined sound stimuli (arousal, relaxation, negative, positive) for evoking emotion. The bio-signals of heart and respiration were measured according to sound stimuli. HRV (heart rate variability) and BRV (breathing rate variability) spectrum were obtained from spectrum analysis of ECG (electrocardiogram) and RSP (respiration). The synchronization of HRV and BRV spectrum was analyzed according to each emotion. Statistical significance of relationship between them was tested by one-way ANOVA. There were significant relation of synchronization between HRV and BRV spectrum (synchronization of HF: F(3, 68) = 3.605, p = 0.018, ${\eta}^2_p=0.1372$, synchronization of LF: F(3, 68) = 5.075, p = 0.003, ${\eta}^2_p=0.1823$). HF difference of synchronization between ECG and RSP has been able to classify arousal from relaxation (p = 0.008, d = 1.4274) and LF's has negative from positive (p = 0.002, d = 1.7377). Therefore, it was confirmed that the heart and respiration to recognize the dimensional emotion by connectivity.
