생체 신호 처리용 칩 기술 동향

  • 유비쿼터스 네트워크(u-network)를 통해 u-health의 개념이 실현됨에 있어 생체 신호를 최초로 측정, 처리하는 부분의 생체 신호 측정용 칩들에 대한 최근 기술 개발 동향을 기술하였다. 이러한 추세에 맞추어 여러 가지 핵심 기술들이 부상하고 있지만, 본 기고에서는 이러한 시스템의 최종 하위 계층, 즉 단말기 등의 부분에 적용되는 bio 관련 시스템 반도체 칩(SoC : system-on-a-chip)에 대해 기술한다. 바이오 칩 중, 기존의 광을 사용하지 않고 값 싸게 구현 할 수 있는 CMOS 기반의 DNA 칩 개발 동향을 살펴보았으며, 신약 개발이나 치료에 사용할 수 있도록 신경 신호 전달을 검출할 수 있는 신경 신호 전달 측정 칩들의 기술 개발도 살펴보았다. 개인의 의료 생체정보를 모니터링 할 수 있도록 심전도, 근전도, 뇌파, 산소포화도, 체지방 등을 측정할 수 있는 의료용 칩들의 개발 현황도 살펴보았다.

Biochemical Application of IgG Fc-binding peptide: From Biochip to Targeted Nano Carrier

  • FcBP consisting of 13 amino acids specifically binds to Immunoglobulin G Fc domain. Initially, we utilized this peptide for preparation of antibody chip as a PEG composite for enhanced solubility. After then, the peptide conjugate was immobilized on agarose resin, resulting in highly efficient affinity column for antibody purification. The efficiency was comparable to commercial Protein A column. Recently, this peptide was conjugated with cell penetratingpeptide (CPP) on a backbone of GFP, affording antibody transducer, which carries antibody into live cells by simple mixing of antibody and the transducer in cell culture media. Antibody transduction into cells was monitored by live cell imaging. More recently, the FcBP was fused to ferritin cage, which consists of 24 ferritin protein molecules. The FcBP-ferritin cage showed greatly increased binding affinity to human IgG. Its binding was analyzed by QCM and SPR analysis. Finally, it was selectively delivered by Herceptin to SKBR3, a breast cancer cell, over MCF10A, non-tumorigenic cells.

In silico target identification of biologically active compounds using an inverse docking simulation

  • Identification of target protein is an important procedure in the course of drug discovery. Because of complexity, action mechanisms of herbal medicine are rather obscure, unlike small-molecular drugs. Inverse docking simulation is a reverse use of molecular docking involving multiple target searches for known chemical structure. This methodology can be applied in the field of target fishing and toxicity prediction for herbal compounds as well as known drug molecules. The aim of this review is to introduce a series of in silico works for predicting potential drug targets and side-effects based on inverse docking simulations.

  • A new flexible electronic packaging technology and its medical applications are presented. Conventional silicon chips and electronic modules can be considered as "mechanically rigid box." which does not bend due to external forces. This mechanically rigid characteristic prohibits its applications to wearable systems or bio-implantable devices. Using current MEMS (Microelectromechanical Systems) technology. a surface micromachined flexible polysilicon sensor array and flexible electrode array fer neural interface were fabricated. A chemical thinning technique has been developed to realize flexible silicon chip. To combine these techniques will result in a realization of truly flexible sensing modules. which are suitable for many medical applications.

  • Recently, high throughput screening for microorganisms with desired characteristics from a large heterogeneous population has become possible. Single cell separation has taken on increasing significance in recent years, and several different methods have been proposed so far. In this paper, we introduce several cell manipulation methods aiming at single cell separation and investigation. At first, methods for the separation of microorganisms are classified. Then, we introduce two different approaches, that is, (1) indirect manipulation using laser trapped microtools and (2) thermal gelation.

  • In the conventional biology, the most of cell studies was carried out by culturing cells in the Petri dish and by investigating cellular behavior under the diverse bio-molecule (cell signalling materials, drugs or etc.) conditions. However, in vivo environments, diverse stimulations including chemical, mechanical and topological environments involved in the proliferation, differentiation and migration of cells and it is almost impossible to provide these conditions with traditional method. We have developed the methods to provide the well defined chemical and mechanical stimulations using microfluidic devices and applied these approaches to the study of environmental effect on cells. In this paper, we will introduce our microfluidic chips to provide microenvironment and its applications using several cells.

Inferring genetic regulatory networks of the inflammatory bowel disease in human peripheral blood mononuclear cells

  • Cell phenotypes are determined by groups of functionally related genes. Microarray profiling of gene expression provides us response of cellular state to its perturbation. Several methods for uncovering a cellular network show reliable network reconstruction. In this study, we present reconstruction of genetic regulatory network of inflammation bowel disease in human peripheral blood mononuclear cell. The microarray based on Affymetrix Gene Chip Human Genome U133 Array Set HG-U133A is processed and applied network reconstruction algorithm, ARACNe. As a result, we will show that inferred network composed of 450 nodes and 2017 edges is roughly scale-free network and hierarchical organization. The major hub, CCNL2 (cyclin A2), in inferred network is shown to be associated with inflammatory function as well as apoptotic function.

  • 마이크로 바이오칩용 미세 무게 감지 소자를 개발하기 위해 통상적인 MEMS 공정에 PZT sol solution을 함침하여 binder로서 적용하는 복합적인 스크린 인쇄 방법을 적용해 $800-850^{\circ}C}$의 비교적 저온에서 높은 소결밀도와 우수한 전기적인 특성을 가지는 PZT-0.12PCW 후막 구동형 캔틸레버 소자를 Pt/$TiO_2$/YSZ/$SiO_2$/Si 기판에 제조하였다. 제조된 PZT-0.12PCW 후막 구동형 캔틸레버 소자의 공진 주파수와 변위를 레이저 미소 변위 측정 시스템을 이용하여 공기 중 및 액체 중에서 측정함으로써 캔틸레버 크기에 따른 공진 특성 변화, 액체 내에서의 댐핑 효과 등을 분석할 수 있었다. 또한 Au를 증착하거나biotin-streptoavidin 반응을 통해 단백질을 고정화시켜 무게변화를 야기시킨 후 소자의 감도를 평가함으로써 PZT-0.12PCW 후막 구동형 캔틸레버를 우수한 성능의 바이오칩용 미세 무게 감지 소자로 응용할 수 있음을 알 수 있었다.

Production of Recombinant Proteins as Immuno-Analytical Markers of Genetically-Modified Organisms (GMO)

  • Marker proteins of genetically-modified organisms (GMO) and their antibodies were prepared and characterized as major components of an analytical system. We selected two GMO markers, neomycin phosphotransferase II and 5- enolpyruvylshikimate-3-phosphate synthase, and produced them from E. coli employing genetic recombination technology. After purification, their structural conformation and binding affinities to the respective antibodies were characterized. The results showed that the recombinant proteins were identical with commercially obtained reference proteins. We further used them as immunogens to raise polyclonal antibodies capable of discriminating GMO containing protein from non-GMO. Well-characterized marker proteins and antibodies will be valuable as immunoreagents in constructing analytical systems such as biosensors and biochips to measure quantities of GMO.

  • The miniaturization and integration are trend of modern blood analyses. Micro-Bio-Fluidics plays an important role in a micro blood analysis system. In this paper, analysis and design technology for blood analysis system is presented. Numerical simulations of a blood flow in micro separator and reservoir are conducted. As a result, we suggest on-chip micro separator, which performed plasma separation from whole blood in micro channels.
