Effect of Oligosaccharides and Inulin on the Growth and Viability of Bifidobacteria in Skim Milk

  • The effects of food grade fructooligosaccharide (FOS), isomaltooligosaccharide (MOS), galactooligosaccharide (GOS), and inulin on the growth of five strains of bifidobacteria in fermented milk were investigated. Their effect on culture viability during refrigerated storage was also determined. FOS showed the highest growth-promoting activity for all bifidobacteria except for Bifidobacterium bifidum. Growth rates of B. adolescentis, B. breve, and B. infantis were stimulated by oligosaccharides and inulin, whereas B. longum growth was stimulated by the oligosaccharides but not inulin. In contrast, growth of B. bifidum was enhanced only by inulin. Both acetic and lactic acid production by Bifidobacterium spp. was also enhanced in the presence of 5.0% oligosaccharides. The viability of bifidobacteria cultured with oligosaccharides and inulin, particularly with FOS, was significantly higher than control cultures after 4 weeks of refrigerated storage. The utilization of oligosaccharides is likely to enhance the growth rate, activity, and viability of bifidobacteria.

  • Bifidobacteria are normal inhabitants of the human gastrointestinal tract throughout lift, starting just days after birth, and are one of several predominant species of the colonic microflora, along with Peptostreptococcus, Eubacteria, Clostridia, and Bacteroides. Bifidobacteria differ from lactic acid bacteria in that they produce not only lactic acid but also acetic acid as major fermentation products. The classification of bifidobacteria has changed numerous times since they were discovered in 1899 in the feces of breast-fed infants. Since 1994, three additional species have been included in the list (B. lactis, B. inopinatum, and B. denticolens), with a current total of 32 species. A variety of probiotic effects of bifidobacteria are currently being investigated. Research reports suggests several potential probiotic advantages, in particular antimicrobial effects, immune-modulation reduction of the cancer risk, and modulation of gastrointestinal flora. As technological challenges related to viability and enumeration are being overcome, milks fermented with these anaerobic microorganisms(alone or in combination with lactic acid bacteria) are more able to provide consistently satisfying with large numbers of viable microorganisms. Over 70 products containing bifidobacteria are currently offered around the world, including fermented milks, cheese, buttermilk frozen desserts, candy, and pharmaceutical preparations.

한국형 Bifidobacterium longum SPM1205 혼합제제의 변비 개선 효과 (Efficacy of Mixture of Lactic Acid Bacteria (LAB) and Bifidobacteria Supplement in the Management of Constipation; Demonstration of Functionality in Animal and Clinical Trials)

  • 이 연구의 목적은 동물실험과 임상실험을 통해 변비 치료에 Lactobacillus acidophilus와 Pediococcus pentosaceus, Bifidobacterium longum SPM1205를 포함한 유산균(Lactic Acid Bacteria, LAB) 및 비피더스균 혼합제제의 효과를 평가하기 위한 것이다. 5마리의 ICR 마우스와 4명의 여성 변비 환자들은 2주 동안 유산균 및 비피더스균 혼합제제를 복용하였다. 복용 전과 후에 분변에서 유산균수와 유해 효소 활성을 측정하였으며 그 결과 분변 중 유산균수가 증가하였고, 장내 미생물에 의한 유해 효소 활성은 일반적으로 감소하였다. 또한, 61명의 피실험자들은 유산균 및 비피더스균 혼합제제와 lactose 중의 하나를 2주 동안 하루에 3번 복용한 후, 설문지를 작성하였다. 이를 분석한 결과, 변비환자군에서 매일 유산균 및 비피더스균 혼합제제를 복용한 군(56.3%)은 lactose를 복용한 군(26.7%)보다 변비증상이 개선되었고, 단단한 변의 배변 횟수가 0.22에서 0.03으로 감소하였다. 따라서 L. acidophilus와 P. pentosaceus, B. longum SPM1205를 포함한 유산균 및 비피더스균 혼합제제는 변비를 개선할 수 있을 것으로 예상한다.

유산균 첨가 배양액이 제빵 품질 특성에 미치는 영향 (Effect of prefermented culture on bread quality)

  • The study investigated the effect of prefermentation of Bifidobacteria longum and Lactobacillus plantarum on baking quality. Firstly, two kinds of prefermentation were cultured using two lactic acid bacteria, Bifidobacteria longum and Lactobacillus plantarum. White pan bread baked with dough that had undergone these two prefermentation methods was compared with that baked with a non-prefermented control. The physicochemical properties of the three breads were analyzed, and then the physicochemical and sensory properties of the dough and baked bread were cross-analyzed. The pH prefermentation of Bifidobacteria longum was lower than that of Lactobacillus plantarum, whereas the titratable acidity was higher. Compared to the results from analyzing the prefermentation of Lactobacillus plantarum, the prefermentation of Bifidobacteria longum was expected to give positive effects on enriching the bread flavor by creating acetic acid at a level three- to eight-fold higher than that of Lactobacillus plantarum. According to the mixogram data, the optimum ending time for both Bifidobacteria longum prefermentation and Lactobacillus plantarum was around 4.5 to 5 minutes. The speed of dough materialization decreased with increasing prefermentation culture time. The baked bread with added Bifidobacteria longum had a higher water content. However, the other contents were not influenced by prefermentation, but were by culture time. The specific loaf volume, oven spring and baking lass rate all peaked at 20 hours after culture for both prefermentation cultures. The sensory test results indicated the highest prefermentation for the bread baked with prefermented Bifidobacteria longum doughwith a culture time ranging from 20 to 26 hours. In addition, the bread baked with prefermented Lactobacillus plantarum dough gave the highest preference when cultured for 20 hours.

Effect of Aster scaber extract on the Growth of Bifidobacteria and Clostridium perfringens

  • Growth responses of some intestinal bacteria such as bifidobacteria and Clostridium perfringens to the extracts of certain foodstuffs were investigated in vitro. Among edible mountain herbs, the extracts of several chui-na-muls (Aster tataricus, Ligularia fischeri and Aster scaber) had an inhibitory activity against C. perfringens on the agar plate and the water extract of Aster scaber worked selectively on it among intestinal bacteria. The water extract showed growth-promoting effect toward bifidobacteria such as B. adolescentis, B. animalis, B. bifidum, B. infantis and B. thermophilum in the broth culture. When the faecal inoculum was incubated in the culture with the extract, the population of C. perfringens decreased, whereas that of bifidobacteria increased by $10^3$ scale.$\beta$-glucuronidase activity in the culture with the water extract of Aster scaber digested with pepsin and pancreatin was lower than that in the control culture.

Effect of Bifidobacteria on Production of Allergy-Related Cytokines from Mouse Spleen Cells

  • To study the effect of bifidobacteria on preventing allergy response, levels of IFN-$\gamma$, IgG2a, IL-4, and IgG1 were investigated in splenocytes isolated from ovalbumin (OVA)­sensitized allergic mice and BGN4-administered allergy­suppressed mice in the presence of various bifidobacterial strains. Most of the bifidobacteria, except 2A, increased production of Th I-associated immune markers, IFN -$\gamma$ and IgG2a. In addition, most of the bifidobacteria, except 2A and 19A, decreased production of IL-4, whereas the differences in the production of IgG1 were less pronounced. These results suggest that some strains of bifidobacteria may have the potential to prevent the occurrence of allergy by switching Th1/Th2-type antibodies and/or related cytokines.

Hypocholesterolemic Activity of Bifidobacteria Isolated from a Healthy Korean

  • This study was undertaken to investigate the hypocholesterolemic activity of Bifidobacteria (B. breve K-110, B. breve K-111, and B. infantis K-525) isolated from a healthy Korean. The administration of B. breve K-110 and K-111 with a high cholesterol diet significantly protected the increase of serum total cholesterol and LDL cholesterol relative to that of a high cholesterol diet alone. Such a diet supplemented with 0.5% B. breve K-111 decreased serum total cholesterol and LDL cholesterol to 57 and 55%, respectively. The administration of Bifidobacteria also significantly inhibited the lipid-deposited surface in the aorta. The normalizing activity of serum cholesterol level in cholesterolemic rats was accelerated by Bifidobacteria. The normalizing activity of B. breve K-111 on serum cholesterol level was superior to that of B. breve K-110. These results suggest that Bifidobacteria in the human intestine playa role in the prophylactics of arteriosclerosis.

한국인 분변에서 분리한 Bifidobacteria의 탈지유에서의 배양특성

  • Wild strains of bifidobacteria isolated from Korean feces were tested for their growth and acid production abilities in 10% skim milk. Growth of bifidobacteria was markedly decreased from the second transfer in the skim milk culture. When two strains, BF5 and BF33, were grown in skim milk with various supplements, the growth was enhanced by supplementation of 0.5% yeast extract, and 0.05% cysteine but not by short chain fatty acids. There was no enhancing effect of CO$_{2}$, substitution in the fermentor on growth. The viable cell counts of bifidobacteria, BF5 and BF33, were 9.76 and 9.98 logCFU/ml, respectively, after 30 hr cultivation and were diminished by 3 and 6 logs during storage at 5$\circ$C for 12 days.

유산균의 장내환경개선효과 (Isolation of bifidobacteria inhibiting harmful enzymes of Korean intestinal bacteria)

  • Five hundreds of bifidobacteria were isolated from an healthy Korean and the inhibitory effects of these isolated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli HGU-3 or total human intestinal microflora. In comparison with the results of E. coli or intestinal microflora cultivation, Bifidobacterium breave K-110, B. breve K-111 and B. infantis K-525 effectively inhibited harmful enzymes (${\beta}-glucuronidase$ and tryptophanase) of E. coli and lowered the pH of the culture media. Also they inhibited the harmful enzymes (${\beta}-glucosidase$, ${\beta}-glucuronidase$, tryptophanase and urease) and ammonia production of intestinal microflora, and lowered pH of the culture media by increasing the number of bifidobateria on intestinal microflora. The inhibitory effect of bifidobacteria on Growth of Helicobacter pylori and Rotavirus infection were exammed. Bifidobacterium K-110 and K-111 inhibited effectively them. When these isolated bifidobacteria were administered to mice, the activities of fecal harmful enzymes were inhibited and the AC and ACF formation were suppressed. Among tested bifidobacteria, B. breve K-110 had high inhibitory effect of fecal harmful enzymes and ACF formation.

Assessment of Lipopolysaccharide-binding Activity of Bifidobacterium and Its Relationship with Cell Surface Hydrophobicity, Autoaggregation, and Inhibition of Interleukin-8 Production

  • This study was performed to screen probiotic bifidobacteria for their ability to bind and neutralize lipopolysaccharides (LPS) from Escherichia coli and to verify the relationship between LPS-binding ability, cell surface hydrophobicity (CSH), and inhibition of LPS-induced interleukin-8 (IL-8) secretion by HT-29 cells of the various bifidobacterial strains. Ninety bifidobacteria isolates from human feces were assessed for their ability to bind fluorescein isothiocyanate (FITC)-labeled LPS from E. coli. Isolates showing 30-60% binding were designated LPS-high binding (LPS-H) and those with less than 15% binding were designated LPS-low binding (LPS-L). The CSH, autoaggregation (AA), and inhibition of LPS-induced IL-8 release from HT-29 cells of the LPS-H and LPS-L groups were evaluated. Five bifidobacteria strains showed high levels of LPS binding, CSH, AA, and inhibition of IL-8 release. However, statistically significant correlations between LPS binding, CSH, AA, and reduction of IL-8 release were not found. Although we could isolate bifidobacteria with high LPS-binding ability, CSH, AA, and inhibition of IL-8 release, each characteristic should be considered as strain dependent. Bifidobacteria with high LPS binding and inhibition of IL-8 release may be good agents for preventing inflammation by neutralizing Gram-negative endotoxins and improving intestinal health.
