Succession and Appearence Species of Marine Benthic Diatoms on Wave Plates of Indoor Culture Tank (Short-term Observation) (실내 수조에 설치한 파판에서의 해양 부착 규조의 단기간 천이 및 출현종)

  • We observed the succession and appearence species of the marine benthic diatoms for the effective seed production of shellfish culture on wave plates in indoor mass culture tanks. Every five days, the existence species and densitiy of diatoms were counted during the 20 culture days. Experimental conditions were 8.5 $\pm$ 0.3$^{\circ}C$ of water temperature, 1.0250$\~$1.0260 of specific gravity, 2,200$\~$13,000 lux of light intensity. The total density of appearence species was rapidly increased with the lapse of time. However, the number of appearence species decreased on the 20th day. The diatoms observed were 10 species; Navicula sp. of, viridula, Cylindrotheca closterium, Licmophora sp., Pleurosisma normnii, Bacillaria Paradoxa Asterionella glacialis, Stephanopuis sp. cf, turris, Entomoneis sp., Odontella aurita and Cocconeis sp, cf, scutellum. Of them, Navicula sp, of, viridula was most dominant throughout the experiment.

Influence of Temperature on the Photosynthetic Responses of Benthic Diatoms: Fluorescence Based Estimates (온도가 저서규조류 광합성 반응에 미치는 영향: 형광을 이용한 추정)

  • Benthic diatoms are very important primary producers in understanding estuary ecosystems and their productions are largely varied by their photo-physiological characteristics. The short-term effects of increased temperature on the photosynthetic and photo-physiological characteristics of cultured different species of benthic diatoms (Navicula sp., Nitzschia sp., Cylindrotheca closterium, and Pleurosigma elongatum) were investigated by measuring their PSII-fluorescence kinetics using a Diving-PAM. Photosynthesis versus irradiance curves were measured every two hours at six different temperatures (10, 15, 20, 25, 30, and $35^{\circ}C$) for twenty-four hour. The effective quantum yield of PSII ($\Phi_{PSII}$) for most of the species showed a decreasing trend with increased temperature. The relative maximum electron transport rate (rETRmax) was significantly increased up to the optimum temperature level and then sharply decreased. Relative to the values of other parameters, the maximum light use coefficient ($\alpha$) was not substantially changed at lower temperature levels (<$30^{\circ}C$) but significantly decreased only at higher temperatures (30 and $35^{\circ}C$). The light saturation coefficient ($E_K$) mirrored the rETRmax temperature response. In regards to the temperature acclimation abilities of the four species with time, Navicula sp. and C. closterium acclimated to short-term changes in temperature through their photo-physiological adjustments.

Effects of Amino Acids on Larval Settlement and Metamorphosis in Haliotis discus hannai

  • The compositions of amino acid in 6 monocultured benthic diatoms used in aquaculture of Haliotis discus hannai were analyzed, and effects of 15 artificial synthetic amino acids on the settlement and metamorphosis of H. discus hannai larvae. Results showed that the content of L-aspartic acid was highest in all diatoms, and that of L-glutamic acid was second high. In experiment using settlement slat without benthic diatom attached, the highest settlement rate (33.3 ${\pm}$ 8.8%) was obtained with L-glutamic acid, and a higher value (16.7 ${\pm}$ 3.3%) was found with L-aspartic acid at 24 h after experimental commencement, compared to that of control (8.6 ${\pm}$ 5.1%). 80 h later the metamorphosis rates of L-glutamic acid (86.7 ${\pm}$ 10.7%) and L-aspartic acid (80.0 ${\pm}$ 3.3%) groups were higher than control group(0) and other amino acids significantly. The response rate of L-glutamic acid was the highest (62.0%), and those of L-aspartic acid (30.0%) and L-threonine (25.3%) groups were also significantly higher than control group. In the experiment using settlement slat with benthic diatom attached, the best effect of various amino acids on induction of larval settlement was obtained with L-glutamic (82.0 ${\pm}$ 6.9%) and L-aspartic acid (78.7 ${\pm}$ 5.1%) at 24 h after experimental commencement. The settlement rates of L-histidine, L-leucine, L-lysine, L-methionine, L-phenylalanine, and L-tyrosine groups were significantly lower than control group. The same differences in the metamorphosis rate at 56 h after experimental commencement and in the response rate were found. It should be noted that after 80 h the metamorphosis rates of L-histidine (74.0 ${\pm}$ 12.0%) and L-lysine (87.0 ${\pm}$ 8.8%) declined rapidly compared to those of 56 h (8.0 ${\pm}$ 12.0%; 7.7 ${\pm}$ 12.0%).

Wetland Habitat Assessement Utilizing TDI(Trophic Diatom Index) (부착돌말영양지수(TDI)를 활용한 습지환경 평가)

  • The purpose of this study was to analyze the habitat status and species diversity of benthic diatoms and estimate the applicability of TDI (Trophic Diatom Index) to obtain the basic data for the identification and management of created wetlands in the Nakdong River. We observed a total of 38 families and 173 species of benthic diatom during the survey period, and spring and autumn showed a similar number of species of 156 and 154, respectively. The result of the SOM (Self-Organizing Map) analysis showed that the distribution of benthic diatom was sensitive to environmental factors such as nutrient concentration and rainfall in each wetland. The cluster 1 was characterized by the survey sites of autumn mostly and consisted of points of high TDI, although the nutrients such as total phosphorus and total nitrogen were low, and the species number and abundance of diatoms were low. Conversely, cluster 4 was characterized by the survey sites of spring mostly and consisted of points of low TDI, even though total nitrogen was high. Considering that most of the created wetlands had the reduced inflow and outflow, the increased flow rate in the summer lowers nutrient values in autumn, and the species number and abundance of benthic diatom decreases due to the increase of turbidity, which reduces the light penetrations to the substrates. On the contrary, the TDI value is low in spring because the low water level causes insufficient substrate surface to the benthic diatoms, and it is too early for the establishment and development of saprophilous species. Although various studies have used TDI as an indicator for evaluating the habitat environment and water quality, it is not a good evaluation indicator in this study since the nutrient concentration in the wetlands mostly high as they have a low flow rate and are close to the stagnant area. Nevertheless, additional periodic surveys that comprehensively reflect the fact that the summer rainfall and inflow/outflow regulating function might affect the species diversity and distribution of benthic diatoms are necessary.

Distribution of Benthic Algae in Tidal Flats of Saemangeum Lake, Korea (새만금호 갯벌의 부착조류 분포)

  • Distributional patterns of benthic microalgae were studied in the tidal flats of Saemangeum lake, Korea, from March 2007 to October 2009. As benthic microalgae of tidal flats of Saemangeum lake, 44 species belong to 5 classes were identified. Diatoms predominated the benthic microalgal flora with 75.0% of total species occurred. Dominant species were Achnanthes lanceolata, Aulacoseira granulata, Cymbella tumida, Fragilaria construens var. ventor, Melosira varians, Navicula cryptocephala, Navicula cryptocephala var. veneta, Nitzschia palea, and Paralia sulcata. The cell density and biomass of benthic microalgae were highest in 2009, and clear tendency of increment was observed at D1 and D2 sites. In vertical profiles, cell density and biomass of the benthic microalgae showed maximum cells and biomass at 0~1 mm depth of sediments from all sampled sites.

New Records of the Diatom Species (Bacillariophyta) from the Seaweed and Tidal Flats in Korea

  • This research was conducted to find the indigenous diatom species unrecorded in Korea from February to December 2016. The samples were taken at 28 sites of the coastal regions on the west coast of Korea and Jeju Island. Diatoms were collected mostly from sand and mud of tidal flats, including gravel, seaweed on the bottom and macrophytes floating in the seawater. Diatom specimens were observed by means of light and scanning electron microscopy. Twenty one species were discovered and added as new specimens to the Korean diatom flora, which are divided into 9 orders, 12 families, and 16 genera. The list and photographs of the species are included with the description of the morphological characteristics and distribution in Korea.

Newly Recorded Diatoms from the Tidal Flats of Korea (갯벌 조간대에서 발견한 국내 미기록 규조류)

  • To discover unrecorded diatoms, seawater, sand, and pebbles were collected from the intertidal zone of five tidal flats from 2016 to 2023. Diatoms were isolated from the collected samples, cultured, and the ultrastructure of the cells was observed using a scanning electron microscope. 7 species of unrecorded diatoms, consisting of 3 orders, 5 families, and 7 genera, were discovered: Gomphoseptatum aestuarii, Rhoiconeis pagoensis, Seminavis exigua, Plagiolemma distortum, Staurotropis seychellensis, Biremis sigmoidea, Liriogramma sarcophagus. Among these, four genera, Rhoiconeis, Plagiolemma, Staurotropis, and Liriogramma, are reported for the first time in Korea. L. sarcophagus was separated from the central diatom Asteromphalus and transferred to Liriogramma, but its phylogenetic position has not yet been clearly established.

Growth Characteristic, Mono-strain Mass Culture and Antioxidant Effects of Two Benthic Diatoms Amphora coffeaeformis and Achnanthes longipes from Korea

  • Amphora coffeaeformis and Achnanthes longipes are commonly found as dominant benthic microalgae in Jeju coastal water throughout the year. In order to investigate pharmaceutical uses of these diatoms, each single species was isolated with micropipette under phase contrast microscope and subcultured with synthetic seawater media which was enriched with F/2 media, trace metal solution and $Na_2SiO_3$). Growth characteristics of these species were also determined with different combination of salinity, nutrients concentration and temperature. Thereafter, mass culture of each species was done based on the maximum growth condition. Biomass was collected after two weeks of mass culture and freeze dried for antioxidant study. The antioxidant properties of different fractions (n-hexane, chloroform and ethylacetate) obtained by solvent fractionation of 80% methanolic extract of two microalgae were investigated for free radical, reactive oxygen species scavenging (Super oxide, Hydrogen peroxide, Hydroxyl radical and Nitric oxide), metal chelating and lipid peroxidation inhibition activities. All fractions of A. longipes showed higher $DPPH^{\cdot}$ (free radical) scavenging activities (n-hexane: 89.0%, Chloroform: 76.0%, Ethylacetate: 66.0%, Methanol: 90.6% and aqueous residue: 63.0%). N-hexane fraction of A. longipes showed significantly higher activity (49.0%) on nitric-oxide. Ethylacetate fraction of A. longipes and aqueous residue of A. coffeaeformis exhibited 64.0% and 75.6% metal chelating activity which was higher than commercial antioxidants (${\alpha}$-tocopherol: 18.0% and BHT: 16.0%). The n-hexane fraction of A. coffeaeformis had 67.5% activity on $DPPH^{\cdot}$. Chloroform and n-hexane fractions of A. coffeaeformis exhibited 46.2% and 47.6% $H_2O_2$ scavenging effects which were closely similar to commercial antioxidants (${\alpha}$-tocopherol: 49.2% and BHT: 58.6%). Chloroform and ethylacetate fractions of A. longipes and fraction of n-hexane and chloroform of A. coffeaeformis showed better lipid peroxidation activities than ${\alpha}$-tocopherol. These data suggest that both organic and aqueous fractions have good antioxidative compounds with different antioxidant properties.

  • Benthic attached diatoms (BADs), a major primary producer in lotic stream and river ecosystems are micro-sized organisms and require a highly magnified microscopic technique in the observation work. Thus, it is often not easy to ensure accuracy and precision in both qualitative and quantitative analyses. This study proposed a new technique applicable to improve quality control of aquatic ecosystem monitoring and assessment using BADs. In order to meet the purpose of quality control, we developed a permanent mounting slide technique which can be used for both qualitative and quantitative analyses simultaneously. We designed specimens with the combination of grid on both cover and slide glasses and compared their efficiency. As a result of observation and counting of BADs, the slide glass designed with the color-lined grid showed the highest efficiency compared to other test conditions. We expect that the method developed in this study could be effectively used to analyze BADs and contributed to improve the quality control in aquatic ecosystem health monitoring and assessment.

Test Application of KOMPSAT-2 to the Detection of Microphytobenthos in Tidal Flats

  • Microphytobenthos bloom from late January to early March in Korean tidal flats. KOMPSAT-2 will provide multi-spectral images with a spatial resolution of 4 m comparable with IKONOS. Using IKONOS and Landsat data, algal mat detection was tested in the Saemangeum area~ Micro-benthic diatoms are abundant and a major primary product in the tidal flats. A linear spectral unmixing (LSU) method was applied to the test data. LSU was effective to detect algal mat and the classified algal mat fraction well correlated with NDVI image. Fine grained upper tidal flats are generally known to be the best environment for algal mat. Algal mat thriving in coarse grained lower tidal flats as well as upper tidal flats were reported in this study. A high resolution multi-spectral sensor in KOMPSAT-2 will provide useful data for long-term monitoring of microphytobenthos in tidal flats.
