Detection Property of Red Blood Cell-Magnetic Beads Using Micro Coil-Channeland GMR-SV Device (마이크로 코일-채널과 GMR-SV 소자를 이용한 적혈구-자성비드 검출 특성연구)

  • The micro device, coil, and channel for the biosensor integrated with the GMR-SV device based on the antiferromagnetic IrMn layer was fabricated by the light lithography process. When RBCs coupled with several magnetic beads with a diameter of $1{\mu}m$ passed on the micro channel, the movement of $RBC+{\mu}Beads$ is controlled by the electrical AC input signal. The $RBC+{\mu}Beads$ having a micro-magnetic field captured above the GMR-SV device is changed as the output signals for detection status. From these results, the GMR-SV device having the width magnitude of a few micron size can be applied as the biosensor for the analysis of a new magnetic property as the membrane's deformation of RBC coupled to magnetic beads.

A Study on the Lap Joint $CO_2$ Laser Welding of Different Gauge Sheets Using ANOVA in Characteristic Zones (특징영역별 분산분석에 의한 이종두께 겹치기 $CO_2$ 레이저 용접에 대한 연구)

  • The laser welding in the automotive industries has been used widely for the butt joint of blank sheets rather than the lap joint of automotive body panels. But as a substitute far the spot welding of automotive body panels, the so called three dimensional laser welding will be important far the body panel engineers. Specially the laser welding of body panels with a smooth weld line is applied increasingly, for example, to the side panels. So far, some criteria of the laser weld quality was suggested by in-house regulations or national standards from experiences and/or rule of thumbs. In the manufacturing places, a go or no-go criterion is adopted because of the simplicity or a lack of rational criteria. It is true specially for the selection of the process parameters, which gives the basic causes for the good quality of laser welds. In this study, the effects of joint combination, gap and welding speed on the lap joint $CO_2$ laser welding of two mild steel sheets with different thicknesses are obtained through a $2{\times}3{\times}7$ factorial experiment. The results of the weld quality are statistically analysed using analysis of variance (ANOVA) and compared between two characteristic zones, which are separated by the type of sectional shapes and the level of input energy per volume. The thickness combinations are 0.8mm/1.2mm, 1.2mm/0.8mm of mild steel sheets. The welding speed covers from the deep penetration to the partial penetration. The gap size has three levels of no-gap, 0.16m, and 0.26mm. The bead width, penetration depth and input energy per volume are measured and used as the weld quality criteria.

Immobilization and Characterization of Rifamycin B Oxidase in Cellulose Acetate Beads (셀룰로오스 아세테이트에 고정화된 리파마이신 B 산화효소의 특성)

  • Rifamycin B oxidase converts rifamycin B to rifamycin S using oxygen as cosubstrate. Humnicola spp. (ATCC 20620) was treated with acetone and the cell powder was immobilized with cellulose acetate. The properties of the immobilized enzyme was examined. The optimum pHs of the immobilized and the free enzymes were 7.2. The optimum temperature of the immobilized enzyme was at 50-55$^{\circ}C$, which was 5$^{\circ}C$ higher than that of the free enzyme. The activities of the immobilized enzyme appeared less sensistive with respect to the changes of temperature and pH as compared to those of the free enzyme. Twenty percent of the enzyme activity was recovered when the enzyme was immobilized in 3mm beads. The storage stability was good below 4$0^{\circ}C$, but the activity decreased very rapidly above 5$0^{\circ}C$. The physical strength of the beads was good and was suitable as packing material in a three-phase enzyme reactor.

Preparation of Ag-impregnated Porous Ceramic Beads and Antibacterial Properties (Ag 담지 다공성 세라믹 비드 제조 및 항균 특성)

  • Porous ceramic beads that had skeleton structure were impregnated with Ag from silver nitrate solution. Ag-impregnated porous ceramic beads were performed to evaluate the antibacterial properties on Escherichia coli and Staphylococcus aureus, also, compared with commercial silver-activated carbon on antibacterial activity. As concentration of silver nitrate solution increased, deposited-Ag contents of outer and inner surface of beads were increased. The size of silver particles supported on porous ceramic bead were range of $0.5{\sim}2.0\;{\mu}m$. The observed effects of the prepared Ag-impregnated beads on antibacterial activity are as follows : i) Antibacterial activity should be directly proportional to silver nitrate solution and reaction time. ii) The antibacterial activity against Escherichia coli was better than that against Staphylococcus aureus.

Densification and Microstructure of Ultrafine-sized AlN Powder Prepared by a High Energy Ball Milling Process (고에너지 볼밀링 방법에 의해 얻어진 초미립 AlN 분말의 치밀화 및 미세구조)

  • In this study, a high energy ball milling process was employed in order to improve the densification of direct nitrided AlN powder. The densification behavior and the sintered microstructure of the milled AlN powder were investigated. Mixture of AlN powder doped with 5 wt.% $Y_2O_3$ as a sintering additive was pulverized and dispersed up to 50 min in a bead mill with very small $ZrO_2$ beads. Ultrafine AlN powder with a particle size of 600 nm and a specific surface area of 9.54 $m^2/g$ was prepared after milling for 50 min. The milled powders were pressureless-sintered at $1700^{\circ}C-1800^{\circ}C$ for 4 h under $N_2$ atmosphere. This powder showed excellent sinterability leading to full densification after sintering at $1700^{\circ}C$ for 4 h. However, the sintered microstructure revealed that the fraction of yitttium aluminate increased with milling time and sintering temperature and the newly-secondary phase of ZrN was observed due to the reaction of AlN with the $ZrO_2$ impurity.

  • A vertical column pneumatic separator was modified to improve its separation performance. A branch column was installed at the center of the main column, which created a bypass flow and changed the flow rate of the main column before and after the branch column. To separate a mixture comprising light and heavy materials, the airflow in main column after the branch column was set to lift the only light materials and the airflow in main column before the branch column was set to prevent the flow of the light materials from flowing downwards. Materials directed into the branch column were separated from the flow and returned to the feeder through the cyclone linked to the branch column. The performances of the straight-type separator and the modified separator were compared using glass and zirconia beads with a narrow size distribution.

  • Structural stress approach is well known as a mesh-size insensitive fatigue assessment method by using finite element analyses. It is, however, difficult to estimate the structural stress (SS) at weld end points due to stress singularities when shell elements are used. In this study, fatigue evaluations with longitudinal load carrying box fillet weldment under out-of-plane bending load have been performed by using virtual node method (VNM) in order to avoid the problem, which is called the weld end effect. Various combinations of virtual node parameters, such as reference point and virtual node locations, are investigated for the estimation of proper structural stress values applying VNM in a systematic manner. The appropriate guidance of virtual node parameter has been offered for the fillet weldment considered in the study. The structural stress values obtained by VNM have also been validated by comparing the result with finite element model including weld bead. Moreover, the fatigue strength of the fillet weldment based on the equivalent structural stress is shown to be consistent with the master S-N curve.

Influence of Substrate Phase and Inclination Angle on Heat Transfer Characteristics in Vicinity of Hastelloy X Regions Deposited on S45C via Directed Energy Deposition (DED 공정을 이용한 S45C 위 Hastelloy X 분말 적층 시 기저부 상과 경사각이 적층부 인근 열전달 특성에 미치는 영향에 관한 연구)

  • The use of additive manufacturing processes for the repair and remanufacturing of mechanical parts has attracted considerable attention because of strict environmental regulations. Directed energy deposition (DED) is widely used to retrofit mechanical parts. In this study, finite element analyses (FEAs) were performed to investigate the influence of the substrate phase and inclination angle on the heat transfer characteristics in the vicinity of Hastelloy X regions deposited via DED. FE models that consider the bead size and hatch distance were designed. A volumetric heat source model with a Gaussian distribution in a plane was adopted as the heat flux model for DED. The substrate and the deposited powder were S45C structural steel and Hastelloy X, respectively. Temperature-dependent thermal properties were considered while performing the FEAs. The effects of the substrate phase and inclination angle on the temperature distributions and depth of the heat-affected zone (HAZ) in the vicinity of the deposited regions were examined. Furthermore, the influence of deposition paths on depths of the HAZ were investigated. The results of the analyses were used to determine the suitable phase and inclination angle of the substrate as well as the appropriate deposition path.

Identification of CNVs and their association with the meat traits of Hanwoo

  • Background: Copy number variation (CNV) can be identified using next-generation sequencing and microarray technologies, the research on the analysis of its association with meat traits in livestock breeding has significantly increased in recent years. Hanwoo is an inherent species raised in the Republic of Korea. It is now considered one of the most economically important species and a major food source mainly used for meat (Hanwoo beef). Methods: In this study, CNVs and the relationship between the obtained CNV regions (CNVRs) can be identified in the Hanwoo steer samples (n = 473) using Illumina Hanwoo SNP 50K bead chip and bioinformatic tools, which were used to locate the required data and meat traits were investigated. The PennCNV software was used for the identification of CNVs, followed by the use of the CNV Ruler software for locating the different CNVRs. Furthermore, bioinformatics analysis was performed. Results: We found a total of 2,575 autosomal CNVs (933 losses, 1,642 gains) and 416 CNVRs (289 gains, 111 losses, and 16 mixed), which were established with ranged in size from 2,183 bp to 983,333 bp and 10,004 bp to 381,836 bp, respectively. Upon analyzing the restriction of minor alleles frequency > 0.05 for meat traits association, 6 CNVRs in the carcass weight, 2 CNVRs in the marbling score, 3 CNVRs in the backfat thickness, and 2 CNVRs in the longissimus muscle area were related to the meat traits. In addition, we identified an overlap of 347 CNVRs. Moreover, 3 CNVRs were determined to have a gene that affects meat quality. Conclusions: Our results confirmed the relationship between Hanwoo CNVR and meat traits, and the possibility of overlapping candidate genes, annotations, and quantitative trait loci that results depended on to contribute to the greater understanding of CNVs in Hanwoo and its role in genetic variation among cattle livestock.

A Genome-Wide Study of Moyamoya-Type Cerebrovascular Disease in the Korean Population

  • Objective : Structural genetic variation, including copy-number variation (CNV), constitutes a substantial fraction of total genetic variability, and the importance of structural variants in modulating susceptibility is increasingly being recognized. CNV can change biological function and contribute to pathophysiological conditions of human disease. Its relationship with common, complex human disease in particular is not fully understood. Here, we searched the human genome to identify copy number variants that predispose to moya-moya type cerebrovascular disease. Methods : We retrospectively analyzed patients who had unilateral or bilateral steno-occlusive lesions at the cerebral artery from March, 2007, to September, 2009. For the 20 subjects, including patients with moyamoya type pathologies and three normal healthy controls, we divided the subjects into 4 groups : typical moyamoya (n=6), unilateral moyamoya (n=9), progression unilateral to typical moyamoya (n=2) and non-moyamoya (n=3). Fragmented DNA was hybridized on Human610Quad v1.0 DNA analysis BeadChips (Illumina). Data analysis was performed with GenomeStudio v2009.1, Genotyping 1.1.9, cnvPartition_v2.3.4 software. Overall call rates were more than 99.8%. Results : In total, 1258 CNVs were identified across the whole genome. The average number of CNV was 45.55 per subject (CNV region was 45.4). The gain/loss of CNV was 52/249, having 4.7 fold higher frequencies in loss calls. The total CNV size was 904,657,868, and average size was 993,038. The largest portion of CNVs (613 calls) were 1M-10M in length. Interestingly, significant association between unilateral moyamoya disease (MMD) and progression of unilateral to typical moyamoya was observed. Conclusion : Significant association between unilateral MMD and progression of unilateral to typical moyamoya was observed. The finding was confirmed again with clustering analysis. These data demonstrate that certain CNV associate with moyamoya-type cerebrovascular disease.
