VALUE OF BARLEY GRAIN AND COTTONSEED CAKE AS SUPPLEMENTS TO FOURWING SALTBUSH, AND THE LIVE WEIGHT GAINS AND WATER CONSUMPTION OF SHEEP FED THE DIETS

  • Fifteen sheep were used in a trial which compared the feeding value of whole barley gain and cottonseed cake as supplements to a basal diet of leaves of fourwing saltbush (Atriplex canescens). Diet SB contained 700 g (air-dry matter) of saltbush alone, diet SB + BG contained 700 g SB with 400 g whole barley grain and diet SB + CS contained 700 g SB with 400 g cottonseed cake. The digestibility of the dry matter of diets SB (69%) and SB + CS (70%) were lower (p < 0.001) than of diet SB + BG (76%). Sheep offered SB alone daily lost 80g whereas those fed the other diets gained 11 g (SB + CS) or 17 g (SB + BG) per day. Daily water consumption of the sheep offered the three diets was similar (p > 0.05), but their water consumption was higher (p < 0.001) than that of sheep offered daily 700 g wheat straw and 200 g barley grain. The results indicate that, at the levels of feeding used, barley grain and cottonseed cake had similar value as supplements to fourwing saltbush harvested in summer. The addition of the supplement allowed the sheep to gain some live weight. However, the presence of saltbush leaves in the diet resulted in higher water intakes by the sheep.

Studies on Feed Resources of Non-Conventional Agriculture Roughage (농업부산물 사료화 이용 연구)

  • This study was conducted to investigate the effects of feed resources and collection system of nonconventional agricultural roughages, and reduction of environment pollution by turning non-conventional agricultural roughages to feed resources and diversification of feed supply. And a series of experiment were conducted to determine the effect of moisture control material on quality of sweet potato stalk, peanut stalk, barley straw, the experiments were conducted at Grassland and Forage Crops Division, National Livestock Research Institute, RDA, Suwon in 1999 and 2000. The results obtained from these experiments are summarized as follows; Acid detergent fiber(ADF), neutral detergent fiber(NDF) and total digestible nutrient(TDN) of sweet potato were 38.8, 50.7 and 58.2% in the control silage, but that of rice straw added silage for moisture control were decreased to 41.5, 50.8 and 56.1%. ADF, NDF, CP(crude protein) and TDN of peanut stalk were 42.2, 49.6, 10.5 and 55.6% on the rice straw added silage for moisture control, and there were not affected by barely straw added silage for moisture control. And ADF, NDF and TDN content of barely straw increased 42.8, 65.8 and 55.1% on the water+inoculants treated silage. The results of this study indicate that non-conventional agricultural roughages to feed resources should be make for silage at silo after dry matter percentage(ab0ut 35%) control. Also, rice straw added silage for moisture control will improve the silage fermentation and conservation.   • This study was investigate to evaluate the phosphate uptake rate of green algae in relation to diurnal rhythm and algae control method. The phosphate uptake rates of Chlorella vulgaris and Ankistrodesmus convolutus increased in light period and decreased in dark period. On the contrary, those of Chlamydomonas sp. showed a peak in the late dark period. The differences among species in phosphate uptake in relation to diurnal rhythm were due to the severe competition among species and seemed to alleviate the competition for nutrient supplies. The compound of CellCaSi, Ca and Fe showed the effective removal of the phosphorus. The extracts from rice and barley straw exhibited a significant effect on the growth inhibition of Microcystis aeruginosa.

  • The production media and the enzymatic charateristics of laccase from Flammulina velutipes were investigated. The activity of laccase during incubation reached to the maximum at the 40 days of incubation in the case of Barley straw medium. The maximum laccase activity in Barley straw medium was 5 and 16 times higher than those in Onion basic and Sawdust media, respectively. The laccase from Flammulina velutipes has the optimum pH of 6.6 and showed to be stable at relatively broad pH range. 4.5-9.5. Temperature stability showed that above 96% activity could be preserved after holding at 40$^{\circ}C$ for 40 minutes. At the above 70$^{\circ}C$, the laccase activity decreased very rapidly. The Km value of the laccase was estimated to be 28.0 mM which is much higher than that of the laccase from Pleurotus ostreatus. Organic solvents for precipitiation of the enzyme did not inactivation the laccase. Sodium azide which was added for preventing microbial deterioration affected significantly the inactivation of laccase, but this activity was recovered completely by precipitating the enzyme with acetone.

Development of Thermostable Fusant, CHY1612 for Lignocellulosic Simultaneous Saccharification and Fermentation (섬유질계 동시당화발효를 위한 내열성 융합 효모, Kluyveromyces marxianus CHY1612의 개발)

  • To develop thermostable ethanol fermentative yeast strain for lignocellulosic simultaneous saccharification and fermentation, high ethanol producing yeast, Saccharomyces cerevisiae CHY1012 and thermostable yeast, Kluyveromyces marxianus CHY1703 were fused by protoplast fusion. The thermostable fusant, CHY1612 was identified as a Kluyveromyces marxianus by phenotypic and physiological characteristics, as well as molecular analysis based on the D1/D2 domains of the large subunit (26S) rDNA gene and the internally transcribed spacer (ITS) 1 + 2 regions. For lignocellulosic ethanol production, AFEX pretreated barley straw at $150^{\circ}C$ for 90 min was used in a simultaneous saccharification and fermentation (SSF) process using thermotolerant CHY1612. The SSF from 16% pretreated barley straw at $43^{\circ}C$ gave a saccharification ratio of 90.5%, a final ethanol concentration of 38.5 g/L, and a theoretical yield of 91.2%. These results show that K. marxianus CHY1612 has potential for lignocellulosic ethanol production through simultaneous saccharification and fermentation with further development of process.

  • This study compares the efficacy of soaking and percolation pretreatments with alkaline solutions for lignocellulosic biomass. Various biomass such as rice straw and barley were pretreated by soaking processes in various alkaline solutions including sodium hydroxide, potassium hydroxide, aqueous ammonia and sodium carbonate. The enzymatic digestibility of rice straw and barley that had been pretreated by soaking in aqueous ammonia was over 80%. Eucalyptus residue, Larix leptolepis and Pinus rigida exhibited relatively low enzymatic digestibility. Nevertheless, the enzymatic digestibility of pretreated eucalyptus residue was increased by five times compared to that of the initial biomass. And, the enzymatic digestibility of the percolation pretreated eucalyptus residue was increased 12 times.

Influences of Rice and Barley straw Application in the Rice Rhizosphere (수도근권(水稻根圈) 환경(環境)에 미치는 볏짚과 보릿집 시용(施用)의 영향(影響))

  • This study has been made to investigate the influences of organic matter on the soil composition, nitrogen fixing organism, soil enzyme activity and nitrogen fixing activity in the paddy rice rhizosphere when rice and barley straw were applied. The results are summarized as follows: 1. The pH in the submerged soil was increased from ear formation stage to harvesting. 2. In the rhizosphere, $Fe^{{+}{+}}$ content was decreased according to the growing stage, while increased in the nonrhizosphere. 3. In the initial stage, rhizosphere was higher than nonrhizosphere but in the late stage nonrhizosphere was higher than rhizosphere on the $NH_4-N$ content. 4. In the submerged soil, rhizosphere was higher than nonrhizosphere, on the concentration of glucose and pentose. 5. Changes of the number of nitrogen fixing organism in whole soil was not high. 6. Generally, rhizosphere was higher than nonrhizosphere on the soil enzyme activity such as phosphatase, ${\beta}$-glucosidase, and protease. 7. Acetylene-reducing activity was the highest in the tillering stage, and rhizosphere, Samgang (high-yielding variety) were higher than nonrhizosphere. Dongjin (general variety) respectively. 8. In the submerged soil applied barley straw, acetylene-reducing activity was slightly higher than rice straw in the initial stage.

Application of Lignocellulosic and Macro-algae Hydrolysates for Culture of Chlorella saccharophila (Chlorella saccharophila 배양을 위한 목질계 및 해조류 바이오매스 가수분해물의 이용)

  • In this study, we investigated the possibility of using hydrolysates of lignocellulosics (rapeseed straw, barley straw, rice straw) and marine macro-algae (Undaria pinnatifida, Laminaria japonica, Enteromorpha intestinalis, and Gracilaria verrucosa) to cultivate Chlorella saccharophila. The growth of C. saccharophila was inhibited by 7 hydrolysates without active carbon treatment. In contrast, hydrolysates treated with active carbon increased the cell growth and product (oil and chlorophyll) formation by C. saccharophila. The oil contents of C. saccharophila treated with each hydrolysate were $41.26{\pm}0.69%$ (glucose), $22.06{\pm}1.21%$ (rapeseed straw), $28.65{\pm}1.08%$ (barley straw), $31.15{\pm}0.76%$ (rice straw), $31.50{\pm}2.12%$ (U. pinnatifida), $31.49{\pm}4.53%$ (L. japonica), $29.63{\pm}3.93%$ (E. intestinalis), and $26.15{\pm}1.99%$ (G. verrucosa), respectively. Lignocellulosics and marine macro-algae may be useful resources for improving the mass cultivation of C. saccharophila.

Evaluation of Feed Values of Korean Straws Using Pressure Transducer (Auto-Pressure Transducer를 이용한 국내산 고간류의 사료가치평가)

  • Accurate and rapid evaluation of the nutritional quality of Korean straws is important because of the recent increase in the use of these feedstuffs in Korean. The aim of the study was to establish with relationships between ruminal fermentation of Korean straws and in vitro gas production using a pressure transducer. The pressure transducer system includes pressure censors, AD board, LED monitor, and the computer with real-time graphics. Both gas production and DM digestibility data were fitted into the exponential equation P = a + b (1-e-$1-e^{-ct}$). The initial rate of gas production was highest for rice straw, followed by barley straw and wheat straw. The gas production rate of constant (c) in gas production for rice straw, wheat straw, and barley straw were 3.8, 2.5, and 2.5 $%h^{-1}$, respectively. Total VFA concentration (mM) produced after 72h incubation was similar among three Korean straws, even though was variable during the early (12h) fermentation. Volume of gas production was related (P> 0.05: r = 0.76 to 0.83) to DM disappearance and also strongly related (p< 0.05: r = 0.91 to 0.98) to VFA concentration at all incubation times. Linear correlation showed between gas production and DM disappearance and VFA by in vitro will be matched in in vivo digestibility.
