  • This research was performed to investigate the dynamics of microbial community by RBC (Rotating Biological Contactor) using Rhodococcus sp. EL-GT and activated sludge. Cell counts revealed by DAPI were compared with culturable bacterial counts from nutrient agar. Colony counts on nutrient agar gave values 20~25% and 1~15% of cell counts (DAPI). The cell counts for the dynamics of bacterial community were determined by combination of in situ hybridization with fluorescently-labelled oligonyucleotide probes and epifluorescence microscopy. Around 90~80% of total cells visualized DAPI were also detected by the bacteria probe EUB 338. For both reactors proteobacteria belonging to the gamma subclass were dominant in the first stage (1 and 2 stage) and proteobacteria belonging to the gamma subclass were dominant in the last stage (3 and 4 stage).

겨울철 소매점 및 냉장 닭고기의 미생물학적 평가 (Microbiological Evaluations of Retail and Refrigerated Chickens in Winter)

  • Aerobic plate counts(APC) gram-negative bacterial counts (GNC) and sensory evaluations on chic-ken carcasses during retail and refrigerated storages (3$\pm$1$^{\circ}C$ and 1$0^{\circ}C$) were evaluated. APC and GNC on whole chicken in retail store after storage of 7 days at 3$\pm$1$^{\circ}C$ increased to 3.11 and 3.89 log units com-pared to the initial controls. APC and GNC on whole chicken after storage of 7 days at 1$0^{\circ}C$ increased to 5.43 and 5.03 log units. Sensory scores of chicken carcasses obtained from retail store were in the "liked less" category after storage of 7 days compared to fresh controls. These results indicated that chicken carcasses during refrigerated (1$0^{\circ}C$) storages rapidly allowed the growth of aerobic spoilage bacteria dur-ing storage period which cluld not be microbiologically acceptable after of 7 days of 7 days.

성장중인 닭에 있어서 맹장내의 혐기성 미생물수와 맹장 길이와의 관계에 관한 연구 (A Study on the Correlation of Cecal Anaerobic Bacterial Counts with Cecal Length in Growing Chicks)

  • 본 연구는 성장중인 닭에 있어서 맹장내 혐기성 미생물수와 맹장의 길이와의 관계를 구명하기 위해서 실시되었다. 5주령의 160수의 닭을 80수씩 분리하여 케이지내 사육 (대조구)및 염소 방목장내 염소와 같이 사육 (처리구)의 2처리로 구분하여 실시하였으며, 본 시험기간동안의 사료는 닭의 성장단계에 적합한 시판의 사료를 기초사료로 활용하였다. 시험기간 (5∼8주형)까지의 증체량은 8주령 에서는 대조구가 처리구보다 유의하게 놀았으며(P<0.05), 12주령에서는 처리구가 대조구보다도 유의하게 높았다(p<0.05). 맹장의 길이는 6, 9, 10 및 11주령에 처리구가 대조구보다 유의하게 길었다 (P<0.05). 맹장내 총 혐기성미생물수도 6, 9, 10 빛 11주령에 처리구가 대조구보다 유의하게 많았다(P<0.05). 대조구 및 처리구 함께 맹장의 길이와 증체량간에는 r= 0.816의 상관관계가 인정되었다. 맹장의 길이와 맹장내 혐기성 미생물 수는 대조구 및 처리구 함께 r=0.95이상의 높은 상관 관계가 인정되었다(P<0.01). 결론적으로 성장중인 닭에 있어서 섬유소 함량이 높은 염소의 배설물 섭취 등의 영향에 따른 맹장내 혐기성 미생물 수의 증가가 맹장의 길이 증가에 관계되어진다.

도계처리 단계별 도체와 처리수의 세균오염 및 염소처리 효과 (Efficacy of Chlorine for Reducing Bacterial Populations and Bacteriological Contamination on Carcass and Treatment Water at Different Stage of Poultry Processing)

  • This study was carried out to assess the effect of the chlorine treatment into water for processing chicken products in each stage of slaughtering, with a special viewpoint related with reducing the viable number of microorganisms by which the water and the chicken body were contaminated. The mean bacterial number on chicken samples after picking process was log5.37$\pm$0.20~5.84$\pm$0.160CFU/$\textrm{cm}^2$. When assessed by standard plate count method, it was the higher one than any other processing stage in which eviscerating, pinning, packaging, and chilling was followed in order of the mean bacterial number. The coliform bacterial numbers on carcasses after sampling from different processing stages were log2.11$\pm$0.63~2.88$\pm$0.25MPN/$\textrm{cm}^2, which show almost similar numbers in each processing stage. But, after chilling process the number was decreased slightly. The bacterial counts in the water for scalding and chilling showed log3.43 $\pm$ 0.59~5.06$\pm$0.21 and log4.30$\pm$0.21~6.62$\pm$0.33CFU/$m\ell$, respectively. In the coliform counts for the water taken out from the 2nd chilling tank, the number was log1.97$\pm$0.35~2.91$\pm$0.22MPN/$m\ell$ which showed higher than those of the 1st and the 3rd chilling tank water. The effect of chlorination in reducing the bacterial numbers was accepted at the residual chlorine concentration of 1$m\ell$/$\ell$by showing the reduction from $10^8$ to $10^4$CFU level and the numbers were decreased less than 10CFU at the concentration of 5mg/$\ell$, when assessed by viable cell counts. In conclusion, these results suggested that chlorination In chilling water with final concentration of 5mg/$\ell$was strongly recommended to reduce the bacterial numbers on final chicken products.

원유 세균수 검사장비의 정도관리를 위한 표준시료의 개발 및 평가 (Development and evaluation of standard samples for quality control of automated total bacterial counter in raw milk)

  • Standard samples were prepared for this study, and applied for BactoScanTM and BactoCountTM in order for quality control evaluation for total bacterial count in raw milk. Accordingly, the preparation of two lots of standard samples for quality control were lyophilized, which contain Lactobacillus lactis. The standard samples were prepared into three different levels of bacterial counts, those were Low 30,000~40,000, Medium 70,000~90,000, High 150,000~220,000 CFU/mL, respectively. Then, the proficiency tests were performed in total 19 laboratories for measuring total bacterial counts. The total bacterial counts in the standard samples showed 37~42, 82~105, 214~240 CFU/mL in the first lot, and it showed 30~36, 67~75, 131~163 CFU/mL in the second lot in low, medium and high levels, respectively. Based on these results, the absolute values of z-scores of six standard samples in 18 laboratories were ≤2, which means the samples are satisfactory. However, z-score in one laboratory was ≤3, which means the sample is questionable. Using two standard samples, the correlation between BactoScanTM and BactoCountTM was 0.9982, which means the results of total bacterial count measurement of both equipment were equivalent. Therefore, the standard samples manufactured in this study for quality control of total bacterial count using BactoScanTM and BactoCountTM in the raw milk could be applied to proficiency tests.

김밥 세균 오염의 원인 규명을 위한 연구 (Prediction of the Cause of Bacterial Contamination in Kimbab and Its Ingredients)

  • The distribution of bacteria in kimbab and its ingredients have been investigated. The total bacterial counts were over 3$\times$106/g(n=30) when the kimbabs were delivered to restaurant and it exceeded the legal level 1$\times$106/g defined by the Code of Food Standard in 1991. The gram-negative coliforms were also detected 1.9$\times$105. In order to study the cause of bacterial contamination in kimbabs, the ingredients used in kimbabs were examined. The bacterial counts were found 104-108/gfor kim (laver), 104-108/g for sausage, 104-106/g for boiled-spinach, 103-107/g for carrot, and 104-106/g for Danmuji, respectively. From these results it could be concluded that the bacterial contamination in Kimbabs was caused mainly by the ingredients such as kim, boiled-spinach, carrot, and sausage. Therefore, this suggested that the sanitary manufacture of kim, the storage at refrigerator temperature of the ingredients for kimbabs, and the proper treatment of equipments should be maintained in order to reduce the bacterial contamination for kimbabs. Furthermore, it should be required to obtain the basal data for establishment of the sanitary standard of kim and kimbab.

도축처리 단계별 도체 및 환경재료에 대한 미생물학적 분석 (Bacteriological study on carcasses and environmental specimens from different stage of slaughter process)

  • To get the information of sanitary develoment of beef and pork, we get the result of environmental specimens(slaughter house floors, sewage, etc) in laboratory. 1. After examination of bacterial infection on after-bleeding, after-dismemberment and final products at each stage of cattle slaughter process, we got log 3.80~7.48cfu/$\textrm{cm}^2$ of aerobic plate counts and log 2.60~5.23cfu/$\textrm{cm}^2$ of coliform counts or so from the carcasses after bleeding, but these count levels went down little bit after dismemberment but as we continued study to the final products, the count levels kept sililar in mumbers. 2. At the slaughter process of pigs, the aerobic plate counts and the coliform counts reached such high levels of log 5.59~8.80cfu/$\textrm{cm}^2$ and log 3.31~5.67cfu/$\textrm{cm}^2$, respectively, after bleeding, in general, these count diminished in a big way after scalding, but they increased just little bit from dismemberment to final products. And there were few differences in the contamination levels on the final products no matter what seasonal contaminations after bleeding. 3. Test revealed very low levels of cell counts both on the aerobic plate counts of washing water and in the coliform counts, the former was log 1.00~2.69cfu/$\textrm{cm}^2$ and the later was log 3.30~5.67cfu/$\textrm{cm}^2$, but the contamination levels on the beds of transfering vehicles for carcasses were very high as followes : the aerobic counts was log 4.23~7.20cfu/$\textrm{cm}^2$ and coliform counts was log 2.86~5.20cfu/$\textrm{cm}^2$. 4. Study showed the aeroboc plate counts and the coliform counts get to the highest levels in summer, the second highest one is in fall, the third in spring, lowest in winter. Resulting from the test results proven above we reached this kind of conclusion the bacterial contaminations on eatable carcasses were upto hygienic treatment of carcasses and cleaniness of transfering vehicles at the final stop of slaughter processes rather than upto at any stage of slaughter processes. Therefore we have got to establish alternatives immediately to develo sanitary quality of meat and pork.

Survey of Yogurt Powder Storage in Ambient Export Countries A Safety Evaluation Standard Compliance and Comparative Analysis

  • Yogurt powder is fermented milk processed in the form of dry yogurt, and has advantages such as stability, storability, convenience, and portability. China and Vietnam are important export target countries because of the increased demand for dairy products. Therefore, we surveyed dairy product standardization in order to establish an export strategy. Lactic acid bacteria counts are unregulated in Korea and Vietnam. In China, lactic acid bacteria counts are regulated at $1{\times}10^6$ colonyforming units (CFU)/mL and detected at $6.24{\pm}0.33\;Log\;CFU/mL$. All three countries have regulated standards for total bacterial counts. In China, total bacterial counts of milk powder are regulated to n=5, c=2, m=50,000, M=200,000 and detected at $6.02{\pm}0.12\;Log\;CFU/mL$, exceeding the acceptable level. Lactic acid bacterial counts appeared to exceed total bacterial counts. Coliform group counts, Staphylococcus aureus, Listeria monocytogenes, and Salmonella species were not detected. Acidity is not regulated in Korea and Vietnam. In China, acidity was regulated to over $70^{\circ}T$ and detected $352.38{\pm}10.24^{\circ}T$. pH is unregulated in all three countries. pH was compared to that of general fermented milk, which is 4.2, and that of the sample was $4.28{\pm}0.01$. Aflatoxin levels are not regulated in Korea and China. In Vietnam, aflatoxin level is regulated at 0.05 ppb. Therefore, all ingredients of the yogurt powder met the safety standards. This data obtained in this study can be used as the basic data in assessing the export quality of yogurt powder.

Effect of Vacuum Packaging on the Microbial Change of Chicken during Storage

  • To examine the effect of vacuum packaging on the quality of chicken during storage, microbial (total bacterial counts, mold and yeast, E. coli, and Pseudomonas) changes and drip loss were determined. fresh chicken breasts were prepared and packaged using polyethylene film under vacuum and normal atmosphere, respectively. Samples were then stored at 4$^{\circ}C$ for two weeks. At various time intervals during storage, sample was taken homogenized, and diluted with 0.1% sterile peptone water, and followed by microbial counts. Microbial changes of total bacterial counts, mold and yeast, E. coli, and Pseudomonas during storage of chicken were significantly decreased by vacuum packaging. Drip loss was also significantly decreased. These results indicate that vacuum packaging of chicken should be recommended as a suitable storage method in terms of microbial safety as well as quality of chicken.

넙치 자어, 로티퍼와 알테미아의 세균총에 관한 분석 (Studies of Bacterial Flora of Rotifer sp., Artemia sp. and Olive Flounder larvae, Paralichthys olivaceus)

  • The purpose of this research was to investigate the bacterial flora of healthy olive flounder larvae (Paralichthys olivaceus) and live feeds (Rotifer spp. and Artemia spp.). The total bacteria counts were $9.2{\times}10^7$ and $1.2{\times}10^{10}cfu/g$ and Vibrio sp.(82.8%) was dominant in rotifers. The total bacteria counts were $3.8{\times}10^6$ and $9.2{\times}10^6cfu/g$ and Vibrio sp.(73.3%) was dominant in artemia. In olive flounder larvae, the total bacteria counts were $1.4{\times}10^6{\sim}8.3{\times}10^7cfu/g$ and V. harveyi (38.5%) was dominant. It might be potential marker of disease outbreak in olive flounder larvae.
