Objectives: The purpose of this study was to investigate anti-obesity effect of Scutellariae Radix extract combined with metformin. Methods: Male C57BL/6 mice, 4 weeks of age, were used to set high-fat diet induced obesity model. They were grouped NOR (normal control), HFD (high fat diet control), MET (metformin, 100 mg/kg/day), MH2 (metformin 50 mg/kg/day+Scutellariae Radix 200 mg/kg/day), and HG4 (Scutellariae Radix 400 mg/kg/day). MET, MH2, and HG4 were orally administered for 10 weeks. Body weight was measured every week. Fasting blood glucose, triglyceride, total cholesterol (TC), high density lipoprotein, glutamic oxaloacetic transaminase (GOT), and glutamic-pyruvic transaminase (GPT) were measured before sacrifice. Oral glucose tolerance test (OGTT) was also performed. Organ weight and internal fat weight were measured after sacrifice. Results: MH2 group showed a reduction of body weight when compared with HFD group. MH2 group showed stable blood level control which was calculated areas under the curves by OGTT. TC, GOT, GPT level, internal fat, and organ weight in MH2 group reduced. Conclusions: The combined treatment of Scutellariae Radix and Metformin has impact on treating obesity. Further studies are needed to evaluate the effect of Metformin and herbal medicine combination therapy.
Gastrodia elata Bl. (GE) is a traditional herbal medicine used for the treatment hypertension and cerebrovascular disease. The purpose of this study was to investigate the antioxidant effect and blood pressure control ability of the GABA containing fermented Gastrodia elata Blume. The fermentation strain provided by Dr. Gi in Seoul National University is L. brevis GABA100, excellent strain of GABA production, which is involved in the ability to degrade gastrodin glycosides and activation of brain function. The Gastrodia elata Blume fermented by Lactobacillus brevis GABA 100 showed antioxidant activity (total phenolic contents, DPPH radical and ABTS anion scavenging activities) than unfermented Gastrodia elata Blume. In the spontaneously hypertensive rats (SHR), the concentration of fermented Gastrodia elata Blume was administered at 500, 1,000, and 2,000 mg/kg for 8 weeks. Systolic blood pressure decreased statistically significantly (p<0.05), especially 2 weeks after feeding fermented Gastrodia elata Blume. Also, it showed a significant decrease in low, medium, and high concentrations of fermented Gastrodia elata Blume at 4 weeks and 8 weeks. These results indicated that Gastrodia elata Blume fermented by the excellent strain of GABA production L. brevis GABA100 shows the antioxidant function and the effect of suppressing the increase in blood pressure.
본 논문에서는 microwave 채널 환경에 적합한 OFDM(Orthogonal Frequency Division Multiplexing) 기반의 DMR(Digital Microwave Radio)시스템 구조를 제안하고 시스템 성능을 분석하였다. 기존의 단일 반송파 방식의 DMR 시스템은 High-level QAM변조방식을 사용하며 시스템 구성이 복잡하고, 페이딩 채널에 매우 민감한 특성을 갖기 때문에 전송용량의 한계가 발생한다. 따라서 이러한 문제점을 극복하기 위한 방안으로 페이딩 채널에 강한 OFDM 방식이 연구되어왔다. 본 논문에서는 기존 OFDM에서 사용된 windowing 방식 대신 BL-PSF(Band-Limited Pulse Shaping Filter)를 이용 하므로서 GI/GB(Guard Interval/Guard Band) 등의 잉여 데이터 사용을 지양하여 전송용량을 향상 시킬수 있도록 하였으며, IFFT/FFT 블록의 이용효율을 개선시킬 수 있도록 하였다. 본논문에서는 microwave 채널 환경에 대해 단일 반송파 방식의 DMR 시스템과 OFDM/QPSK-DMR 시스템의 성능을 시뮬레이션하여 비교분석하였다. 그 결과, 제안하는 DMR 시스템은 부 반송파의 수가 많아질수록 단일 반송파 방식의 DMR 시스템보다 우수한 fade margin을 갖는것을 알 수 있었다.
Objective : The aim of the present study is to examine hepatic lipid-lowering and anti-inflammatory effects of silymarin combined with Jakyakgamcho-tang on non-alcoholic fatty liver disease in a high fat diet-induced obese mice model. Methods : C57BL/6 mice were divided into four dietary groups: (1) Normal, (2) Control (60% high-fat diet), (3) Control + silymarin 50 mg/kg/day (Silymarin), (4) Control + Silymarin 50 mg/kg/day + Jakyakgamcho-tang 100 mg/kg/day (SPG). After 12 weeks administration, mice were sacrificed and lipids and inflammation-related biomarkers were analyzed liver and plasma. Results : Silymarin and SPG treatments significantly lowered body and liver weights compared to the Control. Serumlipids (triglyceride (TG), total cholesterol) and pro-inflammatory cytokines (tumor necrosis factor alpha, interleukin 1𝛽, and IL-6) concentrations were significantly lowered in the Silymarin and SPG groups than the Control group. Silymarin and SPG treatments suppressed hepatic TG level and hepatic lipid droplets compared to the Control. Theses two treatments significantly increased hepatic kinase B1 and AMP-activated protein kinase protein levels, and significantly decreased hepatic key lipogenic enzymes (acetyl-CoA carboxylase, fatty acid synthase and stearyl coenzyme A desaturase 1) protein levels than the Control. SPG also significantly increased hepatic fatty acid oxidation-related protein (peroxisome proliferator-activated receptor alpha and uncoupling protein 2) levels than the Control. Conclusions: Silymarin and SPG suppressed hepatic lipid accumulation by regulating hepatic protein expression, and lowered blood pro-inflammatory cytokines concentrations though the synergic effect of silymarin and Jakyakgamchotang was not clear.
The effects of Korean solar salt on an azoxymethane (AOM)/dextran sodium sulfate (DSS)-induced colon cancer C57BL/6 mouse model were studied. Korean solar salt samples (SS-S, solar salt from S salt field; SS-Yb, solar salt from Yb salt field), nine-time-baked bamboo salt (BS-9x, made from SS-Yb), purified salt (PS), and SS-G (solar salt from $Gu\acute{e}rande$, France) were orally administered at a concentration of 1% during AOM/DSS colon cancer induction, and compared for their protective effects during colon carcinogenesis in C57BL/6 mice. SS-S and SS-Yb suppressed colon length shortening and tumor counts in mouse colons. Histological evaluation by hematoxylin and eosin staining also revealed suppression of tumorigenesis by SS-S. Conversely, PS and SS-G did not show a similar suppressive efficacy as Korean solar salt. SS-S and SS-Yb promoted colon mRNA expression of an apoptosis-related factor and cell-cycle-related gene and suppressed pro-inflammatory factor. SS-Yb baked into BS-9x further promoted these anti-carcinogenic efficacies. Taken together, the results indicate that Korean solar salt, especially SS-S and SS-Yb, exhibited anti-cancer activity by modulating apoptosis- and inflammation-related gene expression during colon carcinogenesis in mice, and bamboo salt baked from SS-Yb showed enhanced anti-cancer functionality.
Objective : This study was carried out to investigate effect of korean medicinal herbs-included hair tonic (MHT) and food (MF) on hair growth in an alopecia model of C57BL/6 mice. Methods : Six-weeks old mice were given laboratory rodent chow diet for 1 week. Then, we used a depilatory for hair removal on mice. The next day, mice were randomized and separated in 3 groups of 6 mice; normal group (N, vehicle epidermal application+normal diet-treated group), positive control group (C, 3% minoxidil sulfate epidermal application+normal diet-treated group), and the treatment group (T, HT epidermal application+ MF diet-treated group). The hair regrowth was determined by photograph, which was taken at 7, 14 and 21 days. And scoring indices, hair density and hair thickness, were evaluated by Folliscope at same day. In addition, the hair regrowth was analyzed insulin-like growth factor (IGF-1) in the dorsal skin of mice. Results : As a result, not only external appearance but also hair density and hair thickness on dorsal skin were increased more in C, T groups than in N group at 21 days with mixed treatment. Therefore, distinct regrowth pattern is appeared by treatment of herbal tonic and natural food for 21 days. Also, the expressions of IGF-1 on dorsal skin were higher in C, T groups than in N group. Conclusion : These results suggested that this herbal hair tonic and natural food has hair growth promoting activities and may be useful for treatment of bald or alopecia.
Objective : The objective of this study is to investigate the inhibitor effects of an traditional oriental herb, Sophora flavescens on the hepatic and renal side effects of chemotherapy by using B16-BL6 melanoma-injected C57BL6 mouse tumor model. Methods : In this study, the effects of an traditional oriental herb, Sophora flavescens, on the side effects of chemotherapy were studied using B16 melanoma-injected C57BL6 mouse tumor model. Results : Sophora flavescen has significant effect on the reduction of the side effects of chemotherapy. Sophora flavescen recovered the reduction of WBC and RBC during cisplatin chemotherapy. Water extract of Sophora flavescens significantly inhibited cisplatin-induced increase of serum blood urea nitrogen (BUN) which is a good indicator of renal toxicity. Sophora flavescens extract does not decrease the anti-tumor activity of cisplatin showing that it can selectively inhibit side effects of anticancer drugs preserving beneficial effort. Conclusion : Theses results suggest a possibility that Sophora flavescens extract can be used for cancer patients for the reduction of the side effects and improving the quality of life during chemotherapy of cancer patients.
This study aims at modeling boundary layers (BLs) encountered in sparse and built environments (i.e. open, suburban and urban) at the subsonic Wind Tunnel (WT) at Ryerson University (RU). This WT has an insignificant turbulence intensity and requires a flow-conditioning system consisting of turbulence generating elements (i.e., spires, roughness blocks, barriers) to achieve proper turbulent characteristics. This system was developed and validated in the current study in three phases. In phase I, several Computational Fluid Dynamic (CFD) simulations of the tunnel with generating elements were conducted to understand the effect of each element on the flow. This led to a preliminary design of the system, in which horizontal barriers (slats) are added to the spires to introduce turbulence at higher levels of the tunnel. This design was revisited in phase II, to specify slat dimensions leading to target BLs encountered by tall buildings. It was found that rougher BLs require deeper slats and, therefore, two-layer slats (one fixed and one movable) were implemented to provide the required range of slat depth to model most BLs. This system only involves slat movement to change the BL, which is very useful for automatic wind tunnel testing of tall buildings. The system was validated in phase III by conducting experimental wind tunnel testingof the system and comparing the resulting flow field with the target BL fields considering two length scales typically used for wind tunnel testing. A very good match was obtained for all wind field characteristics which confirms accuracy of the system.
Objective: The objective of this study was to obtain (co) variance components and genetic parameter estimates for post-weaning body measurements such as body length (BL), height at withers (HW), and chest girth (HG) recorded at six (SBL, SHW, and SHG), nine (NBL, NHW, and NHG) and twelve (YBL, YHW, and YHG) months of age along with yearling weight (YW) in Nellore sheep maintained at livestock research station, Palamaner, Andhra Pradesh, India and also the association among body measurements with YW was studied. Methods: Data on 2,076 Nellore sheep (descended from 75 sires and 522 dams) recorded between 2007 and 2016 (10 years) were utilized in the study. Lambing year, sex of lamb, season of lambing and parity of dam were included in the model as fixed effects and ewe weight was kept as a covariate. Analyses were conducted with six animal models with different combinations of direct and maternal genetic effects using restricted maximum likelihood procedure. Best model for each trait was determined based on Akaike's information criterion. Results: Moderate estimates of direct heritability were obtained for the studied traits viz., BL (0.02 to 0.24), HW (0.31 to 0.49), and CG (0.08 to 0.35) and their corresponding maternal heritability estimates were in the range of 0.00 to 0.07 (BL), 0.13 to 0.17 (HW), and 0.07 to 0.13 (CG), respectively. Positive direct genetic and phenotypic correlations among the traits and they ranged from 0.07 (YBL-YW) to 0.99 (SBL-SHG, SHG-YW, and NBL-YBL) and 0.01 (SBL-YBL) to 0.99 (NBL-NHG), respectively. Further, the genetic correlations among all the body measurements and YW were positive and ranged from 0.07 (YW and YBL) to 0.99 (YW and SHG). Conclusion: There was a strong association of chest girth at six months with YW. Further, it is indicated that moderate improvement of post-weaning body measurements in Nellore sheep would be possible through selection.
We present a study of the inexplicit connection between radio jet activity and ${\gamma}$-ray emission of BL Lacertae (BL Lac; 2200+420). We analyze the long-term millimeter activity of BL Lac via interferometric observations with the Korean VLBI Network (KVN) obtained at 22, 43, 86, and 129 GHz simultaneously over three years (from January 2013 to March 2016); during this time, two ${\gamma}$-ray outbursts (in November 2013 and March 2015) can be seen in ${\gamma}$-ray light curves obtained from Fermi observations. The KVN radio core is optically thick at least up to 86 GHz; there is indication that it might be optically thin at higher frequencies. To first order, the radio light curves decay exponentially over the time span covered by our observations, with decay timescales of $411{\pm}85$ days, $352{\pm}79$ days, $310{\pm}57$ days, and $283{\pm}55$ days at 22, 43, 86, and 129 GHz, respectively. Assuming synchrotron cooling, a cooling time of around one year is consistent with magnetic field strengths $B{\sim}2{\mu}T$ and electron Lorentz factors ${\gamma}$ ~ 10 000. Taking into account that our formal measurement errors include intrinsic variability and thus over-estimate the statistical uncertainties, we find that the decay timescale ${\tau}$ scales with frequency ${\nu}$ like ${\tau}{\propto}{\nu}^{-0.2}$. This relation is much shallower than the one expected from opacity effects (core shift), but in agreement with the (sub-)mm radio core being a standing recollimation shock. We do not find convincing radio flux counterparts to the ${\gamma}$-ray outbursts. The spectral evolution is consistent with the 'generalized shock model' of Valtaoja et al. (1992). A temporary increase in the core opacity and the emergence of a knot around the time of the second ${\gamma}$-ray event indicate that this ${\gamma}$-ray outburst might be an 'orphan' flare powered by the 'ring of fire' mechanism.
