Allelopathic Potential and Antioxidant Activity of Leaf Extracts from Several Wild Plant Species

  • Several wild plant species are known to contain biologically active substances that are allelopathic to weed species as well as antioxidant to foods. Plant extracts or residues from leaves of 4 species, Achyranthes japonica (speedwell), Cucumis sativus (Cucumber), Trifolium repens (white clover), and Vicia angustifolia (narrowleaf vetch) were bioassayed against Medicago sativa (alfalfa) or Echinochloa crus-galli (barnyard grass) to determine their allelopathic effects, and used for measurement of antioxidant activities. The aqueous extracts applied on filter paper significantly inhibited root growth of alfalfa. Aqueous extracts or residues from V. angustifolia showed the most inhibitory effect on alfalfa or barnyard grass seedling growth and followed by A. japonica and T. repens. Oxidative stability by Rancimat method, antioxidant activity by TBA (2-thiobarbituric acid) method and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity for the ground samples or methanol extracts were the greatest in V. angustifolia, although were less than those of commonly used antioxidants, BHT (butylated hydroxytoluene) and ascorbic acid. These results suggest that the wild plant species had potent allelopathic and antioxidant activities, and that their activities differed depending on plant species.

Polysaccharide Extraction and Comparison of Free Radical Scavenging Activities from Tremella fuciformis and Auricularia auricula Fruit Body (흰목이와 흑목이 버섯의 다당추출 및 유리라디칼 소거활성 비교)

  • The polysaccharides from fruit body of Auricularia auricula and Tremella fuciformis were extracted using hot water, and partially purified through ethanol precipitation and dialysis. Free radical scavenging activities of the crude and purified polysaccharides were examined and compared each other. Free radical scavenging activities of the partially purified polysaccharides were higher than those of crude polysaccharides. DPPH free radical, ABTS radical and SOD-like activities of partially purified polysaccharide at 1 mg/mL of concentration from A. auricula were 61.7, 9.6 and 38.9%, respectively, while those of T. fuciformis were 9.6, 5.7 and 15.3%, respectively. Results of site and non-site specific hydroxyl radical scavenging activities indicated that the partially purified polysaccharide fractions from A. auricula and T. fuciformis exhibited the hydroxyl radical scavenging effect by hydrogen donating ability and iron ion chelating ability. Also, reducing powers of A. auricula and T. fuciformis were 77.1 and 14.7% of BHT (0.1%) as standard, respectively. It was suggested that antioxidant activities of A. auricula were about 1.4~6.4 times higher than those of T. fuciformis due to different levels of polyphenol content.

A Study on the Antimicrobial Activity and the Pharmacological Activities of matrial Isolated from Coptis Radix (황련(Coptis Radix)으로부터 분리된 물질의 항균효능 및 화장품 약리활성에 대한 연구)

  • This study evaluated antimicrobial efficacy and antioxidant effect of fraction isolated from Coptis Radix and confirmed its possibility as a cosmetic material. The extracts of isolated from Coptis Radix conducted an antimicrobial activity against Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans by disc diffusion method and measure clear zone. As a result, it was confirmed that antimicrobial activity against S. aureus and candida. A was observed in all samples except Fr 1. The activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and The activity of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) cation radical scavenging were determined by antioxidant assay according to the concentrations (50, 125, 250) ${\mu}g/mL$ of extracts of isolated from Coptis Radix. As a result, DPPH radical scavenging activity of Fr 1, 2, 3, 4 at $250{\mu}g/mL$ was 11.4%, 30.3%, 42.0% and 53.1%, respectively and $ABTS^+$ radical scavenging activity was 28.6%, 96.2%, 98.6% and 97.1% at the same concentration, respectively. Fr. 3 and 4 showed higher radical scavenging activity than the positive the control group BHT at the same concentration. In the WST assay results of measuring the cytotoxicity of Coptis Radix, except for Fr. 4, Fr. 1, 2 and 3 did not show toxicity. As a result, the fractions isolated from Coptis Radix can be regarded as a cosmetic material having antimicrobial activity and antioxidant ability.

Characterization of Antioxidants Extracted from Leaves of Sanjook(Sasa boreails var. chiisanensis) (산죽잎으로 부터 추출한 항산화물질의 특성)

  • For usage of natural antioxidants, sanjook (Sasa boreails var. chiisanensis) leaves were extracted with methanol and investigated about its antioxidative activities and stability. It showed that the antioxidant activity of methanol extracts from the leaves of sanjook depend on their concentration within range of 0.1 to 0.8 mg/ml. The methanol extracts from the leaves of sanjook represented $583{\mu}g/ml$ for $IC_{50}$ of DPPH radical scavenging ability, $800{\mu}g/ml$for $IC_{50}$ of SOD-like activity and $38{\mu}g/ml$for $IC_{50}$ of $H_{2} O_{2}$ scavenging ability, while BHT, as a compared substance, was $271{\mu}g/ml$ for $IC_{50}$ of DPPH radical scavenging ability, $680{\mu}g/ml$ for $IC_{50}$ of SOD-like activity and $30{\mu}g/ml$ for $IC_{50}$ of $H_{2} O_{2}$ scavenging ability, respectively. The anti-au-toxidation effect for methanol extracts from the leaves of sajook was $55\∼60\%$ within range of 0.1 to 0.8 mg/ml. The pH stability on methanol extracts from the leaves of sanjook was most stable at pH 6. The more acid or akali it became, the more unstable it turned. The thermostability on methanol extracts from the leaves of sanjook remained above $80\%$ of their DPPH activity at range of $0^{\circ}C{\;}to{\;}120^{\circ}$.

Study on the Antioxidative and Physiological Activities of Saururus chinensis Extract (삼백초(Saururus chinensis) 추출물의 항산화능 및 생리활성 연구)

  • Saururus chinensis has long been widely used in oriental folk medicines to treat diseases. In the current study, organic solvent fractions obtained from the main methanolic extract of S chinensis were evaluated for their antioxidative and related physiological activities. The antioxidant activity of the fractions was measured using DPPH free radical scavenging activity, increased in a dose-dependent manner, and the $ED_{50}$ of the ethyl acetate fractions exhibited a value of 12.84 ${\mu}g/ml$ higher than 27.22 ${\mu}g/ml$ compared to the BHT. Also, the cell viability of S. chinensis on $H_2O_2$-induced HDF cell death ($IC_{50}$) showed the highest cell viability of 89.39% in 50 ${\mu}g/ml$ of ethyl acetate fraction and 67.98% of visible cell survival rate in n-butanolic fraction. Meanwhile, all fractions of the S. chinensis extract led to a slight down regulation of the mRNA expression of fibulin-5, which is related to skin elasticity, and the ethyl acetate fraction having high antioxidant activity showed a markedly inhibitory effect on chick embryonic angiogenesis using the CAM assay. These results suggest that the ethyl acetate fraction of S. chinensis extract could be a good material in therapeutic application for antioxidant and related anti-angiogenesis activities.

Evaluation of Biological Activity on Hawthorn Tree (Crataegus pinnatifida) Extracts (산사나무 추출물의 생리활성 평가)

  • The wood and bark of Hawthorn tree (Crataegus pinnatifida Bunge) were immersed with 70% aqueous acetone for 3 days. After filtering, the wood extracts were fractionated with n-hexane, chloroform ($CHCl_3$), n-butanol (n-BuOH) and $H_2O$, and the bark extracts were fractionated with n-hexane, $CHCl_3$, ethylacetate (EtOAc) and $H_2O$. Then antioxidative and anti-inflammatory activities were evaluated on each fraction. Antioxidative activity indicated high activity in the n-butanol soluble fraction of wood and in the EtOAc soluble fraction of bark. Especially, the EtOAc soluble fraction of bark showed much higher antioxidative value compared to the controls, buthylated hydroxytoluene (BHT) and ${\alpha}$-tocopherol. In the anti-inflammatory activity, all of the tested fractions except the $H_2O$ soluble fraction of wood showed high inhibitory effect on nitric oxide (NO) production. Based on the above results, the extracts of hawthorn tree may be applied for one of the natural biomass sources that can be used as an antioxidant and an anti-inflammatory substance.

Evaluation of Biological Activity on The Hydrolyzable Tannins of Katsura Tree (Cercidiphyllum japonicum) (계수나무 가수분해형 탄닌의 생리활성 평가)

  • Eight hydrolyzable tannin compounds, such as gallic acid (1), methyl gallate (2), kurigalin (3), 1,2,3,6-tetra-O-galloyl-${\beta}$-D-(+)-glucose (4), 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-(+)-glucose (5), 6-m-digalloyl-1,2,3,4,6-penta-O-galloyl-${\beta}$-D-(+)-glucose (6), isocorilagin (7), macabarterin (8), were isolated from the EtOAc and $H_2O$ soluble fractions of Katsura tree (Cercidiphyllum japonicum) leaves, wood and bark. Then antioxidative and anti-inflammatory activity were evaluated on the each isolated compound. The antioxidative test was DPPH radical scavenging activity and all of the isolated compounds indicated much higher antioxidative values compare to the controls, BHT and ${\alpha}$-tocopherol. In the anti-inflammatory test measuring nitric oxide (NO) inhibition activity, methyl gallate, 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose and 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucose inhibited NO production, and especially, methyl gallate showed high inhibition activity. However, the anti-inflammatory activity of the hydrolyzable tannins did not show positive effect. Based on the above results, the hydrolyzable tannins of katsura tree may be used as one of the natural biomass sources that can substitute with the synthetic antioxidant.

Antioxidative Effects of Chungkukjang Preparation by Adding Solar Salt (천일염 함유 청국장의 항산화효과)

  • The antioxidant effects of chungkukjang, a popular fermented soybean paste in Korea, prepared with solar salt were investigated in vitro. DJI chungkukjang(DJIC) was prepared using 3%(w/w) solar salt or 3%(w/w) refined salt, and fermented by Bacillus subtilis DJI. All of DJIC with no salt, DJIC with 3%(w/w) solar salt, DJIC containing 3%(w/w) refined salt, and commercial chungkukjang were extracted with ethanol, hexane, and water. Antioxidative activities were measured by 1,1-diphenyl-2-picryl hydrazyl(DPPH) free-radical generation, nitrite scavenging activity, peroxide value in the presence of linoleic acid, and the Rancimat test, in comparison with the commercial antioxidant butylated hydroxytoluene(BHT). Ethanol chungkukjang extracts showed the highest antioxdative capacities, whereas DPPH free-radical generation and scavenging activities, and linoleic acid peroxide values of DJIC extracts prepared using solar salt, and salt-free DJIC, showed the greatest values. In addition, extracts of DJIC prepared using solar salt had the highest antioxidant indices. Antioxidative activities were higher in DJIC extracts than in those of commercial chungkukjang. However, the antioxidative capacities of DJIC prepared using solar salt and DJIC with no added salt were less than those of BHT. In conclusion, these results suggest that extracts of chungkukjang prepared using solar salt are useful nutritional antioxidants and that dietary supplementation with such materials would decrease oxidative stress.

Physiological Activity and Effects on Lipid Peroxidation of Hot Water-Extract Obtained from Euonymus alatus in Cultured Rat Hepatocyte (Rat의 hepatocyte에서 $amyloid-{\beta$}$로 유발된 세포사, 지질과산화 및 세포산화에 대한 귀전우 열수 추출물의 보호효과)

  • This study was carried out to investigate the physiological activityof the water extract from EA. The present study was done to investigate the effects of EA on cultured hepatocyte cell system and lipid peroxidation in $A{\beta}$ treatment conditions. Pretreatment of EA attenuated in cell cytotoxicity enhanced by increasing concentrations of $A{\beta}$. MDA level induced by $A{\beta}$ treatment was significantly increased and the level was slightly reduced by pretreatment of EA. The ability of EA to reduce cell death and MDA level induced by $A{\beta}$ suggest that EA may be a protective agent against free radical generating compounds such as $A{\beta}$. EA exhibited anti oxidative activity at all concentration tested.The extract was as good as antioxidative activity of the synthetic antioxidants, butylated hydroxy toluene and ascorbic acid. Furthermore, this was superior to that of natural antioxidant, a-tocopherol. In the presence of heavy metal ions ($Fe^{2+},{\;}Zn^{2+}$), EA showed strong antioxidative activity. The extracts showed about 3075% in the nitrite scavenging effect under pH 1.2 and $37^{\circ}C$ for 1 hr. There was significant difference among concentration of extracts.

A Study on the Catechins Contents and Antioxidative Effect of Various Solvent Extracts of Green, Oolong and Black Tea (녹차, 우롱차 및 홍차의 용매별 추출물의 카테킨류 함량 및 항산화효과에 관한 연구)

  • The extraction yield of MeOH extract of green teas, oolong tea and black tea were 3 to 5, 4 to 5, and 5 to 7 fold higher than those of EtOH and EtAC extract, respectively. The amount of total catechins of EtAC extract of the black tea, and of the green teas and oolong tea were three- and two-fold higher than that of EtOH or MeOH extract of the corresponding teas, respectively. The antioxidative activities of EtOH, MeOH and EtAc extract were considerably higher than that of BHT and $dl-{\alpha}-tocopherol$ at 200 ppm level. The antioxidative activities of EtOH and MeOH extract at 200 and 500 ppm level, and of EtAc extract at 200 ppm level varied depending on the type of tea as follows : green tea I > green tea II > green tea III > oo-long tea> black tea. The antioxidative activity increased as the content of EGC increased. But the antioxidative activity of MeOH extract at 1000 ppm level, and of EtAc extract at 500 and 1000 ppm level were not affected by the content of EGC and EGCG.
