  • This review paper serves to describe the composition and activity of biological activated carbon (BAC) biofilm which is considered as a progressive process for water treatment. As well as several physical-chemical, biochemical and microbiological analysis methods for characterizing the composition and activity of BAC biofilm, the ability of the biofilm to remove and biodegrade organic matters and pollutants related to other water treatment processes such as pre-ozonation will be reviewed. In this paper, conversion of GAC into BAC, removal mechanism of pollutants, characteristics and affecting factors of BAC biofilm, and modeling of BAC are described in detail. In addition, strategies to control the growth of the BAC biofilm, such as varying the nutrient loading rate, altering the frequency of BAC filter backwashing and applying oxidative disinfection, will be dwelled on related to their respective process control challenges.

Characterization of Organic Matters Removed by Biological Activated Carbon (생물활성탄처리에서 제거된 유기물 특성)

  • The objective of this study was to clarify the characteristics of the removed micropollutant since the breakthrough of adsorption ability was occurred in biological activated carbon(BAC) process. The removal efficiency of DOC (Dissolved Organic Carbon) was 36 % in the breakthrough of BAC occurred by NOM (Natural Organic Matter). The most of removal DOC was found out the adsorbable and biodegradable DOC (A&BDOC). But it was not clear to remove by any mechanism because A&BDOC have simultaneously the adsorption of activated carbon and biodegradation by microorganism in BAC. The removal of bromophenol was examined with BAC and rapid sand filter, for investigation of DOC removal mechanism in the breakthrough of BAC. In this experiment, BAC filter has been operated for 20 months for the treatment of reservoir water. The BAC filter was already exhausted by NOM. Bromophenol, adsorbable and refractory matter, was completely removed by BAC filter. Therefore, it might be removed by the adsorption in BAC. Adsorption isotherms of bromophenol were compared to two BACs which was preloaded with 500 daltons and 3,000 daltons of NOM. BAC preloaded with 3,000 daltons of NOM was not decreased to the adsorbability of bromophenol but BAC preloaded with 500 daltons of NOM was greatly decreased to it. These result indicated that NOM of low molecular weight can be removed by adsorption after a long period of operation and the breakthrough by NOM in BAC. Therefore, micropollutants might be removed through adsorption by saturated BAC.

Effects of Benzalkonium Chloride on the growth and survival of Human corneal epithelial cells (점안액 보존제 성분 Benzalkonium Chloride에 의해 유도된 각막상피세포의 세포고사 유도)

  • The aim of this study was to investigate the action of benzalkonium chloride (BAC) used as a preservative in most ophthalmic topical solutions, on human corneal epithelial (HCE) cells in vitro. HCE cell line was exposed to BAC solutions at various concentrations (0.01%~0.0001%) for 15 minutes followed by 24 hours of cell recovery. Cell viability was assessed using MTT assay and chromatin condensation with a Hoechst 33342 test. The expression of membrane protein Fas and Fas ligand was examined by western blot and immunocytochemistry, and DNA fragmentation was studied by agarose gel electrophoresis. A significant decrease of membrane integrity with chromatin condensation was observed with BAC tested at concentrations of 0.005% and higher. BAC was cytotoxic preservatives in this study. An apoptotic mechanism appeared to be present at low concentrations of BAC, whereas a necrotic process appeared at higher concentrations. A functional Fas-mediated apoptotic pathway is present in cultured HCE cells and can be activated by upregulation of Fas expression with BAC.

Engineering lacZ Reporter Gene into an ephA8 Bacterial Artificial Chromosome Using a Highly Efficient Bacterial Recombination System

  • In this report, we describe an optimized method for generation of ephA8 BAC transgenic mice expressing the lacZ reporter gene under ephA8 regulatory sequences. First, we constructed a targeting vector that carries a 1.2 kb ephA8 DNA upstream of its first exon, a lacZ expression cassette, a kanamycin cassette, and a 0.7 kb ephA8 DNA downstream of its first exon. Second, the targeting vector was electroporated into cells containing the ephA8 BAC and pKOBEGA, in which recombinases induce a homologous recombination between the ephA8 BAC DNA and the targeting vector. Third, the FLP plasmid expressing the Flipase was electroporated into these bacteria to eliminate a kanamycin cassette from the recombinant BAC DNA. The appropriate structures of the modified ephA8 BAC DNA were confirmed by Southern analysis. Finally, BAC transgenic mouse embryos were generated by pronuclear injection of the recombinant BAC DNA. Whole mount X-gal staining revealed that the lacZ reporter expression is restricted to the anterior region of the developing midbrain in each transgenic embryo. These results indicate that the ephA8 BAC DNA contains most, if not all, regulatory sequences to direct temporal and spatial expression of the lacZ gene in vivo.

Removal of Ammonia Nitrogen and Reduction of THMs in Low Temperature by BAC Pilot Plant (BAC Pilot Plant 를 이용한 겨울철 암모니아성 질소 제거 및 THMs 변화)

  • The raw drinking water quality is getting worse because of the winter drought and the conventional treatment system is'nt suitable to obtain the satisfied quality of water. So, the advanced water system, BAC(Biological Activated Carbon) process is said to be effective to remove dissolved organics and ammonia nitrogen. In our study, the BAC pilot plant using Nak-dong river water is tested in low temperature. Following results are found from the study. The ammonia nitrogen removal rate of BAC system using wood-based carbon (PICABIOL) was 99% in $6^{\circ}C$ temperature. Chlorine dosage in wood-based BAC effluent was reduced to 67% of that in sand filtered wate. It resulted from the removal of ammonia nitrogen. Also, THM formed by chlorine addition in wood-based BAC effluent was decreased to 65% of that in sand filtered water. In the case of dual-filter, the removal efficiency of ammonia nitrogen was increased 30% more than in conventional sand filter. According to this result, the ammonia nitrogen load to BAC system could be lessened by the use of dual-filter.

Purification and Characterization of a Bacteriocin, BacBS2, Produced by Bacillus velezensis BS2 Isolated from Meongge Jeotgal

  • Bacillus velezensis BS2 was isolated from meongge (common sea squirt) jeotgal, a Korean fermented seafood, and produces a bacteriocin, BacBS2, which strongly inhibits Listeria monocytogenes and Bacillus cereus. BacBS2 was partially purified by Q-Sepharose column chromatography after ammonium sulfate precipitation of the culture supernatant, then further purified by Sephadex G-50 column chromatography. Partially purified BacBS2 was estimated to be 6.5 kDa in size by Tricine-SDS PAGE and activity detection by gel-overlay. Enzyme treatment and FT-IR spectrum of partially purified BacBS2 confirmed its proteinaceous nature. BacBS2 was fully stable at pH 4-9, and half of activity was retained at pH 1-3. Full activity was retained after exposure to $80^{\circ}C$ for 15 min, but half of the activity was retained upon exposure to $90^{\circ}C$ for 15 min or $100^{\circ}C$for 10 min. BacBS2 inhibited L. monocytogenes by bactericidal mode of action. B. velezensis BS2 and its BacBS2 seem useful as biopreservatives for fermented foods such as jeotgal.

Evaluation of Influence Factors for Determination of Proper Backwashing Time of Biological Activated Carbon (BAC) Process in Drinking Water Treatment Process (정수처리용 활성탄 공정의 적정 역세척 시점 선정을 위한 영향인자들 평가)

  • In Korea, many drinking water treatment plants (DWTPs) have introduced and are going to introduce biological activated carbon (BAC) process to treated dissolved organic matter (DOM) in water which are difficult to control by conventional water treatment processes. Even though more decade have passed since introduced BAC in Korea, most of BAC operating method was followed to the modified sand filter operating manuals. In case of BAC backwashing, many DWTPs set the periods of backwashing about 3~5 days. In this study, we have collected data to set the proper BAC backwashing periods from both pilot-plant and real DWTPs. We had measured heterotrophic plate count (HPC), turbidity, water temperature, dissolved organic carbon (DOC) and headloss from just after backwashing to the next backwashing time for two years. Considering water quality factors, the BAC run time from backwashing to the next backwashing could extend more 30 days without water quality deterioration if the head loss do not reach the limited level which depends on each BAC facilities' condition. It means the BAC treated water could be saved in the proportion of extended the backwashing period to the existing backwashing period.

Effects of Acid Modification on Pb(II) and Cu(II) Adsorption of Bamboo-based Activated Carbon (대나무 활성탄의 산 개질이 납과 구리 이온의 흡착에 미치는 영향)

  • Effects of acid ($HNO_3$ and HCl) modification on the adsorption properties of Pb(II) and Cu(II) onto bamboo-based activated carbon (BAC) were investigated through a series of batch experiments. The carbon content increased and oxygen content decreased with acid treatment. $HNO_3$ induced carboxylic acids and hydroxyl functional groups while HCl added no functional group onto BAC. The pseudo-second order model better described the kinetics of Pb(II) and Cu(II) adsorption onto experimented adsorbents, indicating that the rate-limiting step of the heavy metal sorption is chemical sorption involving valency forces through sharing or exchange of electrons between the adsorbate and the adsorbent. The equilibrium sorption data followed both Langmuir and Freundlich isotherm models. The adsorption capacities of BAC were affected by the surface functional groups added by acid modification. The adsorption capacities were enhanced up to 36.0% and 27.3% for Pb(II) and Cu(II), respectively by the $HNO_3$ modification, however, negligibly affected by HCl.

Comparison of Immune Responses to the PCV2 Replicase-Capsid and Capsid Virus-Like Particle Vaccines in Mice

  • Porcine circovirus type 2 (PCV2) is the causative agent of postweaning multisystemic wasting syndrome (PMWS) in pigs. Replicase (Rep) proteins are considered essential for viral replication. Capsid (Cap) protein is the primary immunogenic protein that induces protective immunity. Little is known about comparison on the immunogenicity of PCV2 Rep and Cap fusion protein and Cap protein. In the present study, recombinant baculoviruses expressing the Rep-Cap fusion protein (Bac-Rep-Cap) and the Cap protein (Bac-Cap) of PCV2 were constructed and confirmed with western blot and indirect fluorescence assay. Immunogenicities of the two recombinant proteins were tested in mice. The titers of antibodies were determined with a PCV2-specific enzyme-linked immunosorbent assay (ELISA) and a serum neutralization assay. The $IFN-{\gamma}$ response of immunized mice was measured by ELISA. The mice immunized with the Bac-Rep-Cap and Bac-Cap successfully produced Cap-specific immunoreaction. The mice immunized with the Bac-Cap developed higher PCV2-specific neutralizing antibody titers than mice injected with the Bac-Rep-Cap. $IFN-{\gamma}$ in the Bac-Rep-Cap group was increased compared to those in the Bac-Cap group. Vaccination of mice with the Bac-Rep-Cap showed significantly decreased protective efficacy compared to the Bac-Cap. Our findings will indubitably not only lead to a better understanding of the immunogenicity of PCV2, but also improved vaccines.

Korea Brassica Genome Project: Current Status and Prospective (배추 유전체열구의 현황과 전망)

  • Brassica rape is an important species used as a vegetable, oil, and fodder worldwide. It is related phylogenically to Arabidopsis thaliana, which has already been fully sequenced as a model plant. The 'Multinational Brassica Genome Project (MBGP)'was launched by the international Brassica community with the aim of sequencing the whole genome of B. rapa in 2003 on account of its value and the fact that it has the smallest genome among the diploid Brassica. The genome study was carried out not only to know the structure of genome but also to understand the function and the evolution of the genes comprehensively. There are two mapping populations, over 1,000 molecular markers and a genetic map, 2 BAC libraries, physical map, a 22 cDHA libraries as suitable genomic materials for examining the genome of B. rapa ssp. pekinensis Chinese cabbage. As the first step for whole genome analysis, 220,000 BAC-end sequences of the KBrH and KBrB BAC library are achieved by cooperation of six countries. The results of BAC-end sequence analysis will provide a clue in understanding the structure of the genome of Brassica rapa by analyzing the gene sequence, annotation and abundant repetitive DHA. The second stage involves sequencing of the genetically mapped seed BACs and identifying the overlapping BACs for complete genome sequencing. Currently, the second stage is comprises of process genetic anchoring using communal populations and maps to identify more than 1,000 seed BACs based on a BAC-to-BAC strategy. For the initial sequencing, 629 seed BACs corresponding to the minimum tiling path onto Arabidopsis genome were selected and fully sequenced. These BACs are now anchoring to the genetic map using the development of SSR markers. This information will be useful for identifying near BAC clones with the seed BAC on a genome map. From the BAC sequences, it is revealed that the Brassica rapa genome has extensive triplication of the DNA segment coupled with variable gene losses and rearrangements within the segments. This article introduces the current status and prospective of Korea Brassica Genome Project and the bioinformatics tools possessed in each national team. In the near future, data of the genome will contribute to improving Brassicas for their economic use as well as in understanding the evolutional process.
