In this study, we investigated the effects of the flowers of Lespedeza bicolor on melanogenesis in a mouse melanocyte cell line, B16/F10 cells. Our results show that the flowers of Lespedeza bicolor significantly inhibits melanin synthesis in concentration-dependent manner. In addition, it was also found to inhibit the activity of tyrosinase, the rate-limiting melanogenic enzyme. While the flowers of Lespedeza bicolor have no direct inhibitory effect on tyrosinase activity in cell free assay system using mushroom tyrosinase. Moreover, the flowers of Lespedeza bicolor effectively suppressed the ${\alpha}-MSH-stimulated$ melanin formation, tyrosinase activity and dendrite outgrowth. These results suggest that the flowers of Lespedeza bicolor is a potent depigmetation agent.

This study was to investigate the effect of Ponciri Fructus ethanol extract on the melanogenesis. The Ponciri Fructus markedly decreased melanin contents of B16F10 cells at a dose of $80{\mu}g/m{\ell}$. It was observed that the color of cell pellets was totally whitened compared with the control. The treatment of Ponciri Fructus inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. Regarding protein levels of the melanogenic enzymes, the Ponciri Fructus dose-dependently decreased tyrosinase and TRP-1 protein levels, and tyrosinase and TRP-1 were detected in similar manner. These results suggest that the Ponciri Fructus ethanol extract exerts its depigmenting effects through the suppression of tyrosinase and TRP-1.
Endostatin, a proteolytic fragment of collagen XVIII, is a potent inhibitor of angiogenesis and the growth of several primary tumors. However, the opinions on the activity of endostatin derivatives deleted N- or C- terminal are still controversial. In this regard, we produced mouse endostatin and its derivatives in the prokaryotic system, and studied their anti-tumor activity. The [$^3H$]-thymidine incorporation assay demonstrated that N-terminal deleted mouse endostatin, and a C- and N-terminal deleted mutant, effectively inhibited the proliferation of human umbilical vein endothelial cells (HUVECs). The biological activity of endostatin was also shown by its in vivo anti-angiogenic ability on the chorioallantoic membrane (CAM) of a chick embryo. Treatment of $200\;{\mu}g$ of mouse endostatin, or N-terminal deleted mouse endostatin, inhibited capillary formation of CAM 45 to 71%, which is comparative to a 80% effect of positive control, $1\;{\mu}g$ of retinoic acid. An in vivo mouse tumor growth assay showed that N-terminal deleted mouse endostatin, and the N-/C-terminal deleted mutant, significantly repressed the growth of B16F10 melanoma cells in mice as did the full-length mouse endostatin. According to these results, N-and N-/C-terminal deleted mouse endostatins are the potent inhibitors of tumor growth and angiogenesis.
A Pseudomonas sp. strain NGK1 (NCIM 5120) capable of utilizing salicylate was immobilized in alginate and polyurethane foam (PUF). The degradation rate of salicylate by freely suspended cells was compared with the degradation rate by immobilized cells. In an initial 20 and 40 mM salicylate, free cells ($2{\times}10^{11}\;cfu\;ml^{-1}$) degraded to 16 and 14 mM, alginate-entrapped cells degraded to 18 and 26 mM, and PUF-entrapped cells degraded to 20 and 32 mM salicylate, respectively, in batch cultures. The alginate-and PUF-entrapped cells were used in repeated batch and continuous culture systems. The efficiency of both the immobilized systems f3r the degradation of salicylate was compared. It has been observed that the PUF-entrapped cells could be reused for more than 20 cycles whereas alginate-entrapped cells could be reused for a maximum of only 12 cycles, after which a decrease in degradation rat was observed with the initial 20 and 40 mM salicylate. The continuous degradation of sallcylate by freely suspended cells showed a negligible degradation rate of salicylate when compared with immobilized cells. With the immobilized cells in both alginate and polyurethane foam, the degradation rate increased with an increase in the dilution rate up to $2\;h^{-1}$ for 20 mM, and $1.5\;h^{-1}$ for 40 mM salicylate. The results revealed that PUF-entrapped cells were more efficient for the degradation of salicylate than alginate-entrapped cells and freely suspended cells.
Effect of a surfactant Tween 80 on the bacterial growth in the rumen was examined on the in vitro pure cultures of Streptococcus bovis, Selenomonas ruminantium, Butyrivibrio fibrisolvens, Prevotella ruminicola, Megasphaera elsidenni, Fibrobacta succinogenes, Ruminanococcus albus and Ruminococcus flavefaciens. Dry matter degradability (DMD), concentrations and compositions of volatile fatty acids (VFA), and the most probable number (MPN) of cellulolytic bacteria and total number of bacteria in the presence of Tween 80 were also examined on the in vitro rumen mixed culture either with barley grain or orchardgrass hay. The growth of S. bovis, S. ruminantium, B. fibrisolvens, P. ruminicola, M. elsidenni and F. succinogenes were significantly higher (p<0.05) at over 0.05% concentrations of Tween 80 than those of the control cultures, while was not changed with R. albus and R. flavefaciens. With rumen mixed culture the DMD of barley grain and orchardgrass hay was significantly higher (p<0.05) at a 0.2% concentration of Tween 80 than the control, being reflected in the significantly higher (p<0.05) VFA production (mmol $g^{-1}$DDM) with orchardgrass hay. The higher (p<0.05) ratio of propionate to acetate at a 0.2% concentration of Tween 80 was also observed with orchardgrass hay, showing a similar trend with barley grain. No changes in the total bacterial number and MPN of cellulolytic bacteria were observed.
The complement system is a part of the natural immune regulation mechanism against invading pathogens. Complement activation from three different pathways (classical, lectin, and alternative) leads to the formation of C5-convertase, an enzyme for cleavage of C5 into C5a and C5b, followed by C6, C7, C8, and C9 in membrane attack complex. The C9 is the last complement component of the terminal lytic pathway, which plays an important role in lysis of the target cells depending on its self-polymerization to form transmembrane channels. To address the association of C9 with traits related to disease resistance, the complete porcine C9 cDNA was comparatively sequenced to detect single nucleotide polymorphisms (SNPs) in pigs of the breeds Hampshire (HS), Duroc (DU), Berlin miniature pig (BMP), German Landrace (LR), Pietrain (PIE), and Muong Khuong (Vietnamese potbelly pig). Genotyping was performed in 417 $F_2$ animals of a resource population (DUMI: $DU{\times}BMP$) that were vaccinated with Mycoplasma hyopneumoniae, Aujeszky diseases virus and porcine respiratory and reproductive syndrome virus at 6, 14 and 16 weeks of age, respectively. Two SNPs were detected within the third exon. One of them has an amino acid substitution. The European porcine breeds (LR and PIE) show higher allele frequency of these SNPs than Vietnamese porcine breed (MK). Association of the substitution SNP with hemolytic complement activity indicated statistically significant differences between genotypes in the classical pathway but not in the alternative pathway. The interactions between eight time points of measurement of complement activity before and after vaccinations and genotypes were significantly different. The difference in hemolytic complement activity in the both pathways depends on genotype, kind of vaccine, age and the interaction to the other complement components. These results promote the porcine C9 (pC9) as a candidate gene to improve general animal health in the future.

The feasibility of incorporating orange peel powder (OPP) as a value-added food ingredient into convenient food products, was investigated using yanggaeng as a model system. OPP was incorporated into yanggaeng at amounts of 0, 3, 6, 9, and 12% (w/w) based on total weight of cooked white beans and OPP. pH decreased significantly with increasing levels of OPP (P<0.05), and moisture content was ranged from 46.28% to 47.87%. In terms of color, lightness, redness, and yellowness increased significantly (P<0.05) with increasing levels of OPP. Hardness increased significantly when OPP content was higher than 6% in the formulation (P<0.05). Total polyphenol content and 1,1-diphenyl-1-picrylhydrazyl radical scavenging activity were significantly increased by addition of OPP in the formulation (P<0.05). Finally, consumer acceptance test indicated that the highest levels of OPP incorporation (12%, w/w) had an adverse effect on general consumer preferences. In contrast, yanggaeng with moderate levels of OPP (3~6%, w/w) is recommended (with respect to overall preference score) to take advantage of the antioxidant properties of OPP without sacrificing consumer acceptability.
Aerodynamic effects, such as drag force and flow-induced vibration (FIV), on civil engineering structures can be minimized by optimally modifying the structure shape. This work investigates the turbulent wake of a square prism with its faces modified into a sinusoidal wave along the spanwise direction using three-dimensional large eddy simulation (LES) and particle image velocimetry (PIV) techniques at Reynolds number $Re_{Dm}$ = 16,500-22,000, based on the nominal width ($D_m$) of the prism and free-stream velocity ($U_{\infty}$). Two arrangements are considered: (i) the top and bottom faces of the prism are shaped into the sinusoidal waves (termed as WSP-A), and (ii) the front and rear faces are modified into the sinusoidal waves (WSP-B). The sinusoidal waves have a wavelength of $6D_m$ and an amplitude of $0.15D_m$. It has been found that the wavy faces lead to more three-dimensional free shear layers in the near wake than the flat faces (smooth square prism). As a result, the roll-up of shear layers is postponed. Furthermore, the near-wake vortical structures exhibit dominant periodic variations along the spanwise direction; the minimum (i.e., saddle) and maximum (i.e., node) cross-sections of the modified prisms have narrow and wide wakes, respectively. The wake recirculation bubble of the modified prism is wider and longer, compared with its smooth counterpart, thus resulting in a significant drag reduction and fluctuating lift suppression (up to 8.7% and 78.2%, respectively, for the case of WSP-A). Multiple dominant frequencies of vortex shedding, which are distinct from that of the smooth prism, are detected in the near wake of the wavy prisms. The present study may shed light on the understanding of the underlying physical mechanisms of FIV control, in terms of passive modification of the bluff-body shape.
Four Holstein heifers were used in the present study to investigate the effect of standing and lying behaviors on heat production and physiological responses under low (L), medium (M) and high (H) levels of TDN intake. Rectal temperature (RT), respiration rate (RR), heart rate (HR), heat production (HP) and animals behavior (standing time, ST) were measured continuously for 5h (11:00-16:00) periods. There was a tendency for increased RT with TDN intake, and no difference in RT was observed between standing and lying. The RR, HR and HP during standing were significantly higher (p<0.05) than during lying. The difference between standing and lying HP were 4.41, 4.68 and $5.37kJ/kg^{0.75}$ h for L, M and H of TDN intake, respectively. These values showed that standing HP was 18.6-20.9% higher than lying HP. A multiple regression analysis of HP ($kJ/kg^{0.75}$ h) on TDN intake ($g/kg^{0.75}$ d) and ST (min/h) was HP = 7.75 + 0.28 TDN intake + 0.12 ST (R = 0.84). This analysis showed that the total HP not only depend on feed intake levels, but also depends on animal behavior. It was suggested that the change in HP due to the change in feed intake and animal behavior would influence the level of heat loss which was indicated by the changes in the RR and HR.
