Whitening and Anti-oxidative Constituents from the Extracts of Hydrangea petiolaris Leaves (등수국 잎 추출물 유래 미백 및 항산화 활성 성분)

  • In this study, the whitening and antioxidant effects of the extracts from Hydrangea petiolaris (H. petiolaris) leaves was confirmed, and the chemical structure was identified by separating the active ingredients. In the whitening tests using α-MSH stimulated B16F10 melanoma cells, the n-hexane (Hex) fraction inhibited the cellular melanogenesis and intracellular tyrosinase activities without causing cell toxicity. In addition, the Hex fraction reduced expression of tyrosinase and TRP-2 protein. Upon the anti-oxidative studies by DPPH and ABTS+ radicals, potent radical scavenging activities were observed in the ethyl acetate (EtOAc) fraction. Also, for the cellular protective effects on HaCat keratinocytes damaged by H2O2, the EtOAc fraction indicated protective effects against oxidative stress. Eight phytochemicals were isolated from the extract of H. petiolaris leaves; ethyl linoleate (1), ethyl linolenate (2), 1-linoleoyl glycerol (3), 1-linolenoyl glycerol (4), epi-catechin (5), afzelin (6), quercitrin (7), hyperin (8). Among the isolates, the compounds 5 - 8 showed DPPH and ABTS+ radical scavenging activities. The contents of quercitirin, a major isolated in this extract, determined by HPLC analysis were confirmed to be about 31.3 mg/g for the 70% ethanol extract and 169.8 mg/g for the EtOAc fraction. Based on these results, it was suggested that the extract from H. petiolaris leaves could be potentially applicable as whitening and anti-oxidative ingredients in cosmetic formulations.

The Anticoagulant and Anticancer Activities of Enteromorpha intestinalis Extracts (파래 추출물의 항응고 활성과 항암 활성에 관한 연구)

  • The study was performed to investigate the biological activity of Enteromorpha intestinalis. In order to examine its blood anti-coagulant effects, Enteromorpha intestinalis was extracted with cold water, methanol, hot water, HCl and NaOH. In general, the alkali extract of Enteromorpha intestinalis was approximately 17 times stronger than the control. The anti-cancer effects of select extracts(methanol, hot water, 0.1 N NaOH, 1 N NaOH) were determined in human melanoma cells(Bl6/F10), fibrosarcoma cells(HTl080) and breast cancer cells(MCF7) by MTT assay. With the treatment of 250 ${\mu}g/m{\ell}$ of methanol extracts. HT1080, B16/F10 and MCF7 cell viabilities significantly decreased to 8.06%, 3.62% and 10.10%, respectively. Thus these results strongly support the possibie use of Enteromorpha intestinalis as a functional materials.

A Study on the Whitening Effect of Erigeron annuus (L.) Pers. Ethanol Extract on Melanoma Cell (B16F10) (멜라노마 세포(B16F10)에서의 개망초 추출물을 이용한 미백효과에 관한 연구)

  • A 70% ethanol extract of Erigeron annuus (L.) Pers. was investigated for its whitening activity for application as a functional ingredient in cosmetic products. At the E. annuus extract concentration of $100{\mu}g/ml$, the electron-donating ability was found to be 67.83%, the tyrosinase inhibitory effect (related to skin-whitening) was 69%, the elastase inhibitory effect (related to skin-wrinkling) was 69%, and the astringent effect was 80%. The $ABTS^+$ radical-scavenging ability was 87% at the $500{\mu}g/ml$ concentration. In the cell viability test measured on melanoma cells, 96% of the cells treated with $100{\mu}g/ml$ of the extract were viable. According to the western blot results, the protein expression of the microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 was decreased by 60.22%, 47.83%, 54.79%, and 67.88%, respectively, at the extract concentration of $100{\mu}g/ml$. The protein expression of phosphorylated extracellular signal regulated kinase (p-ERK) and phosphorylated cAMP response element-binding protein (p-CREB) was decreased with increasing concentrations of the extract. Reverse transcription-polymerase chain reaction of the extract showed that the mRNA expression of MITF, tyrosinase, TRP-1, and TRP-2 was decreased by 86.51%, 85.22%, 74.26%, and 66.66%, respectively, at $100{\mu}g/ml$ extract concentration. The findings suggest that the 70% ethanol extract from E. annuus (L.) Pers. has potential as a cosmeceutical ingredient with whitening effect.

  • In order to find new functional materials for the cosmetics application, we investigated the anti-oxidant and anti-melanogenic properties of Gentiana scabra extracts (GSE), which were prepared by the various extraction methods. Results showed that GSE had high DPPH radical scavenging activity in a dose-dependent manner. Also, GSE inhibited the prodution of melanin in B16F10 melanoma cell as well as tyrosinase activity. We also found that GSE inhibited mRNA expression of tyrosinase-related protein (TRP)-1 and 2. In conclusion we suggest that GSE is applicable to cosmetics as a potential ingredient for their anti-oxidant and whitening effects.

A Study on the Whitening Effect of Mangifera indica L. Peel Extracts through Inhibition of Melanin Synthesis Factor (Melanin 생성 인자 억제 효과를 통한 Mangifera indica L. Peel의 미백효과 연구)

  • The purpose of this study was to investigate the whitening effects of hot water (AMPW) and ethanol (AMPE) extracts of Mangifera indica L. peel. To verify the whitening effects, tyrosinase inhibitory activity was measured. 9.51% inhibitory activity, and 35.98% inhibitory activity at 1,000 ㎍/ml. The effects of AMPW and AMPE on cell viability were measured using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay in B16-F10 melanoma cells. Greater than 95% cell viability was observed at 100 ㎍/ml. Thus, subsequent experiments were performed at concentrations less than 100 ㎍/ml. The whitening effects were confirmed by measuring the protein and mRNA expression levels of microphthalmia-associated transcription factor, tyrosinase, tyrosinase-related protein 1 (TRP-1), and TRP-2, which are factors involved in melanin synthesis. Western blotting and reverse transcription-polymerase chain reaction results confirmed that 100 ㎍/ml AMPW and AMPE showed superior inhibitory effects than the control treatment (alpha-melanocyte stimulating hormone only). Therefore, Mangifera indica L. peel extract had a whitening effect, and thus, has potential as a natural material for use in cosmetics.

Inhibitory Effect of Ginkgo biloba Extracts on Melanin Biosynthesis (은행 열매 추출물의 멜라닌 생성 저해효과)

  • In this study, we investigated the inhibitory effect on melanin synthesis of Ginkgo biloba seed oil. The results showed 9.96% inhibitory effect scavenging activity on DPPH and showed a value of 1.33 mM of $FeSO_4$ at a concentration of 0.06% in DMSO by using FRAP assay. G. biloba seed oil inhibited tyrosinase activity up tp 37.72% and suppressed the biosynthesis melanin up to 48.02% at 0.06% in B16/F10 mouse melanoma cell. In G. biloba seed oil treated group tyrosinase, TRP-1, TRP-2 and MITF gen expression levels significantly decreased compared to the contral group at a concentration of 0.04% and 0.06%. In conclusion, these results indicated that G. biloba seed oil extract have a good antimelanogenetic effects.

Antitumor Effects of Duck′s Egg Oil on the Cancer Cells (청둥오리 압란유의 항암 효과)

  • This study were constructed to investigate effects of duck's egg oil on antitumor agent or a new natural immunomodulator. To obtained the aboved objectives, Duck's egg oil was purified the large scale from Duck. Duck's egg oil was accelerated the increasing reaction of mouse spleen cells, while inhibited to increase the YAC-cells. However, there is no significance the rate of CD4'/CD8'cell. The normal rate of CD4'-T and CD8'-T cells were accelerated the higher rate than that normal mouse group, and Duck's egg oil feeding mice showed a significant enhancement of expression of IL-2 receptors, an increase of numbers of CD4+ T cells, CD8+ T cells. Otherwise, Duck's egg oil stimulated the production of NO from peritoneal macrophages and the production of TNF-a and also significantly accelerated in the spleen mice. On the other hands, lung localization of B16F10 melanoma cells inhibited by Duck's egg oil. These results found that Duck's egg oil is useful new functional materials as antitumor agent or immunomodulator.

Induction and Proliferation of Callus in Rhus chinensis Mill. and Its Effect on Skin Whitening (붉나무 캘러스 유도 및 그 추출물로부터 피부미백 효과에 관한 연구)

  • The objective of this study was to optimize the condition for induction and proliferation of callus from Rhus chinensis Mill. and investigate the skin-brightening effect of Rhus chinensis callus (RCC). It was confirmed that the most proper plant growth regulator (PGR) for callus induction is 1.0 mg/L of 2,4-dichlorophenoxyacetic acid (2,4-D). The most optimal condition of PGR, medium and additives for callus proliferation were 2,4-D (1.0 mg/L), MS medium and citric acid, respectively. Inhibitory activities of tyrosinase were higher at 50 and 100 ㎍/mL of RCC extracts (41.86 and 75.56%, respectively) than arbutin (27.32%). As the results of measuring melanin inhibition in B16F1 melanocyte and B16F10 melanoma cell, RCC extracts increased its inhibitory activities concentration-dependently, and were found to have higher whitening effect than arbutin at a concentration of 100 ㎍/mL. Therefore, it is suggested that RCC can be used as an effective material for skin-brightening cosmetics.

Effects of Lectin-conjugated Ellagitannin on Antitumor Activity (Lectin-conjugated Ellagitannin의 혹색종에 대한 항암활성)

  • Generally, antitumor drugs have strong toxicity and result in damage in normal cells. Previously, lectin has been reported as a tumor cell specific binding protein and tannin as an antitumor substance. In this study, we investigated antitumor activity of lectin-conjugated ellagitannin and used praecoxin A as an ellagitannin source. We injected mouse melanoma cell, B16-F10, on right the femoral region of C57BL/6 mouse. After 10 hours later, first treatment with praecoxin A, lectin-praecoxin A mixiture and lectin-conjugated praecoxin A was carried and followed by injection i.m. every 48 hours. Praecoxin A extended the life of mice up to 14.8% in comparison with the negative control group at 5 mg/kg dose. The life extending ratio of Lectin-praecoxin A mixture was 26.1% at 5 mg/kg dose, and the life extending ratio of lectin-conjugated praecoxin A was 28.7% at 5 mg/kg dose. On the basis of these findings, we suggest that antitumor activities of lectin-praecoxin A mixiture and lectin-conjugated praecoxin A on survival are better than that of praecoxin A.

Antitumor Effects of the Hexane Extract of Stachys Sieboldii MIQ (초석잠 추출물의 항암 및 면역 효과)

  • The present study was designed to investigate the effects of Stachys Sieboldii MIQ as a new natural antitumor agent or immunomodulator. To obtain the above objectives, Stachys sieboldii MIQ was extracted with ethanol. Stachys sieboldii MIQ accelerated mouse spleen cell growth, but inhibited FM3A/S°-cell growth. However, no significant difference was found for CD4+ / CD8+ cells. The growth rates of CD4+ and CD8+ T cells were accelerated more than those normal mouse group. Stachys sieboldii MIQ fed mice showed a significant enhancement of IL-2 receptor expression, increased numbers of CD4+ T cells, and CD8+ T cells. Stachys sieboldii MIQ also stimulated the production of NO by peritoneal macrophages and the production of NO by and the growth of mouse spleen cells. On the other hand, lung localization of Bl6Fl0 melanoma cells was inhibited by ethanol extract of Stachys sieboldii MIQ. These results show that Stachys sieboldii MIQ is a useful new functional antitumor agent or immunomodulator.
