Vitis amurensis Ruprecht root inhibited ${\alpha}$-melanocyte stimulating hormone-induced melanogenesis in B16F10 cells

  • BACKGROUND/OBJECTIVES: The root of Vitis amurensis Ruprecht, a sort of wild-growing grape, has been used in oriental medicine for treatment of skin ailments; however, its dermatological activity is not sufficiently understood. The aim of this study was to investigate tyrosinase inhibitory and anti-melanogenic activities of V. amurensis Ruprecht root methanol extract (VARM) in B16F10 mouse melanoma cells and to attempt to isolate and identify the active compound issued from VARM. MATERIALS/METHODS: Anti-melanogenic activity of VARM was analyzed in ${\alpha}$-melanocyte stimulating hormone (MSH)-stimulated B16F10 cells through evaluation of antioxidative activity as well as inhibited tyrosinase activity and melanin contents compared with those of kojic acid and arbutin. After anti-melanogenic analysis of VARM, serial fractionation, nuclear magnetic resonance (NMR), and thin layer chromatorgraphy (TLC) were applied for identification of active compounds contained in VARM. RESULTS: VARM significantly inhibited oxidative stress and tyrosinase activity and attenuated ${\alpha}$-MSH-induced melanin production in B16F10 cells. For isolation of active compounds, VARM was fractionated using a series of organic solvents, including dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), and n-butanol (n-BuOH). Among fractions showing anti-melanogenic activity, the CH2Cl2 fraction induced the most potent attenuation of melanogenesis without cytotoxicity and the major compound in the $CH_2Cl_2$ fraction was identified as betulinic acid. Betulinic acid isolated from the $CH_2Cl_2$ fraction of VARM significantly attenuated ${\alpha}$-MSH-induced melanogenesis in a dose dependent manner, which was stronger than that of arbutin used as a positive control. CONCLUSIONS: These results indicate that VARM inhibits oxidative stress, tyrosinase activity, and ${\alpha}$-MSH-induced melanogenesis in B16F10 cells, due primarily to the active compound, betulinic acid, in the $CH_2Cl_2$ fraction.

Differential Cytotoxic Effects of Jaspine B in Various Cancer Cells (다양한 암세포주에서 Jaspine B의 함암활성 비교)

  • Jaspine B is an anhydrophytosphingosine that is isolated from a marine sponge. Because of its structural similarity to sphingosine, it shows anti-cancer effects in human carcinomas. Therefore, this study aims to investigate its anti-proliferative effect on various cancer cells and to correlate its association with the intracellular accumulation of Jaspine B in relevant cancer cells. The anti-proliferative effect of Jaspine B in various cancer cells was determined by a cell viability test, and the intracellular concentration of Jaspine B in relevant cancer cells was determined using mass spectrometry coupled with liquid chromatography. The correlation coefficient and p value between the cytotoxicity and the cell accumulation of Jaspine B were determined using SPSS 16.1. The cytotoxicity of Jaspine B varied depending on the type of cancer cell when compared the $EC_{50}$ values of Jaspine B. Breast and melanoma cancer cells were susceptible to Jaspine B, whereas renal carcinoma cells were resistant. The intracellular concentrations of Jaspine B had a reciprocal correlation with the $EC_{50}$ values in the same cells (r = 0.838). The results suggested that the anti-proliferative effect of Jaspine B was associated with the cellular accumulation of this compound. However, Jaspine B was not a substrate for P-glycoprotein and breast cancer resistance protein, as major efflux pumps caused multidrug resistance. The maintenance of a high intracellular concentration is crucial for the cytotoxic effect of Jaspine B; however, efflux pumps may not be a controlling factor for Jaspine B-related resistance in cancer cells.

  • To identify inhibitory effect of Bamboo extract on melanogenesis, the effect was compared with arbutin. ascorbic acid, hydroquinone, and kojic acid on the melanin biosynthesis in Bl6 mouse melailoma cells and cultured hunlan melanocytes. The cell viability of the agent was tested on cultured human melanocytes. We also examined. its free radical scavenging activity on 1,1-dipheny1-2-picrylhydrazyl (DPPH). Bamboo extract showed considerable effect against melanin production and did not reduce cell viability at the concentration tested. It also showed potent free radical scavenging activity.

Antitumor Effects of the Hexane Extract of Stachys Sieboldii MIQ (초석잠 추출물의 항암 및 면역 효과)

  • The present study was designed to investigate the effects of Stachys Sieboldii MIQ as a new natural antitumor agent or immunomodulator. To obtain the above objectives, Stachys sieboldii MIQ was extracted with ethanol. Stachys sieboldii MIQ accelerated mouse spleen cell growth, but inhibited FM3A/S°-cell growth. However, no significant difference was found for CD4+ / CD8+ cells. The growth rates of CD4+ and CD8+ T cells were accelerated more than those normal mouse group. Stachys sieboldii MIQ fed mice showed a significant enhancement of IL-2 receptor expression, increased numbers of CD4+ T cells, and CD8+ T cells. Stachys sieboldii MIQ also stimulated the production of NO by peritoneal macrophages and the production of NO by and the growth of mouse spleen cells. On the other hand, lung localization of Bl6Fl0 melanoma cells was inhibited by ethanol extract of Stachys sieboldii MIQ. These results show that Stachys sieboldii MIQ is a useful new functional antitumor agent or immunomodulator.

Antioxidation Activity and Inhibition of Melanin Synthesis of Ethanol Extracts from Morus alba in B16/F10 Melanoma Cells (B16/F10 흑색 종 세포에서 오디(Morus alba) 에탄올 추출물의 멜라닌 생성 저해 작용과 항산화 활성)

  • 본 연구에서는 80% 식물성 알코올을 추출 용매로 사용해 오디를 빛을 차단 후 실온에서 3일 간 추출하였다. 3회 여과한 후 최소 온도($40{\sim}60^{\circ}C$)에서 농축한 뒤 동결 건조하여 파우더 형태로 사용하였다. 오디(Morus alba)의 에탄올 추출물은 B16/F10 세포의 항산화 및 멜라닌 합성 억제 효과를 나타내었다. 멜라닌 함량과 세포 내 tyrosinase 활성을 Western blotting으로 측정 하였다. Tyrosinase와 tyrosinase-related protein (TRP) -1은 tyrosinase-related protein (TRP) -2보다 강력하게 억제되었으며, 이들 결과는 tyrosinase와 TRP-1은 흑갈색을 띠는 eumelanin의 생합성의 억제와 강한 상관관계가 있음을 보여 주었다. ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH) 처리 한 B16/F10 흑색 종 세포에서 M. alba 에탄올 추출물은 멜라닌 생성 연관 단백질의 발현 및 멜라닌 생성이 용량 의존적으로 억제 하였다. 멜라닌 함량과 세포 내 tyrosinase 활성을 Western blotting으로 측정 하였다. 또한 DPPH와 SOD를 사용하여 항산화 활성을 분석하였고 총 폴리 페놀과 총 플라보노이드 함량을 측정 하였다. MTT assay 분석을 사용하여 M. alba 에탄올 추출물의 세포 독성을 측정 하였다. B16/F10 멜라닌 생성 세포의 tyrosinase 저해 활성 및 사멸 효과가 일반적으로 효과적이었다. 따라서 M. alba 에탄올 추출물은 항산화 및 미백 효과를 나타내며, 기능성 화장품의 천연 성분으로서 우수한 것으로 여겨진다.

Peptides-derived from Scales of Branchiostegus japonicus Inhibit Ultraviolet B-induced Oxidative Damage and Photo-aging in Skin Cells (피부세포에서 옥돔 비늘로부터 추출한 펩타이드의 UVB에 대한 산화적 손상 및 광 노화 억제)

  • Collagen peptides, which are found at high concentrations in the human body, are present in animal bones and the skin of marine organisms, namely, fish scales. Collagen is the most abundant structural protein of various connective tissues in animals. Furthermore, it is widely used in biomedical material, pharmaceutical, cosmetic, food, and leather industries. Peptides extracted from scales of various fish protect against ultraviolet B (UVB)-induced skin damage and photo-aging. However, the protective effects of collagen peptides derived from the scales of Branchiostegus japonicus against UVB exposure are unclear. This study investigated the effects of peptides larger than 1 kDa (high-molecular weight peptides [HMP]) and smaller than 1 kDa (low-molecular weight peptides [LMP]), derived from extracts of B. japonicus scales, against UVB-induced skin damage and photo-aging. These peptides scavenged 1,1-diphenyl-2-picrylhydrazyl radicals in a dose-dependent manner. In UVB-exposed HaCaT human keratinocytes, LMP inhibited 8-isoprostane generation, a marker of cellular lipid peroxidation. The peptides also suppressed the UVB-induced increase in tyrosinase activity and melanin content in B16F10 mouse melanoma cells. In addition, the LMP and HMP treatment suppressed UVB-induced elastase and matrix metalloproteinase-1 activities in the HaCaT cells. These results indicate that peptides derived from B. japonicus scales have antioxidant, antiphoto-aging, and skin-whitening effects.

New Whitening Agent From Pimpinella brachycarpa (참나물추출물의 멜라닌 생성저해 효과)

  • To develop a new whitening agent for cosmetics from natural products, Pimpinella brachcarpa was selected for its inhibitory effect on melanogenesis in B16 melanoma cells. Crude ethanolic extract of P. brachycarpa and its four fractions-hexane, ethyl acetate(EtOAc), butanol and aqueous were evaluated for antioxidative effects and tyrosinase inhibitory activity. To elucidate the mechanism of active compounds of P. brachycarpa, we investigated the changes in protein level of tyrosinase, TRP-1 and TRP-2 using Western blotting and the changes in mRNA level of tyrosinase using RT-PCR technique. Following UV irradiation, expression of ET-1 in HaCaT keratinocytes was measured by quantitative enzyme immunoassay(EIA) using human ET-1 antibody. Crude ethanolic extract of P. brachycarpa and its four fractions-hexane, EtOAc, butanol and aqueous had free radical scavenging effect by 87.2, 2.5, 97.2, 80.5, 49.8% at 100 ${\mu}g/mL$ and tyrosinase inhibitory effect by 18.3, 15.1, 55.4, 13.1, 0 % at 100 ${\mu}g/mL$. P. brachycarpa EtOAc fraction significantly inhibited melanin production in B16 melanoma cells. Treatment with P. brachycarpa extract for 72 h suppressed the biosynthesis of melanin up to 58 % at 100 ${\mu}g/mL$. Especially, the EtOAc fraction of P. brachycarpa reduced the tyrosinase activity and tyrosinase expression in B16 melanoma cells in a dose-dependent manner. mRNA levels of tyrosinase and TRP-1 were markedly reduced by the EtOAc fraction of P. brachycarpa. Moreover, at the concentrations of $12.5{\sim}50{\mu}g/mL$ of the fraction, the production of UV-induced ET-1 in HaCaT keratinocytes(24 h after 8 $mJ/cm^2$ UVB irradiation) was reduced about 40%(p<0.05). P. brachycarpa could be used as a new natural skin-whitening agent due to the inhibitory effect of on melanin biosynthesis and endothelin-1 expression.

Inhibitory effect of melanin synthesis using organic solvent extracts from Scolopendra subspinipes mutilans (왕지네의 유기용매 추출물을 이용한 멜라닌 합성 저해효과)

  • This study was performed to determine the whitening effect of organic solvent extracts from the centipede, Scolopendra subspinipes mutilans. We prepared different concentrations (50%, 70% and 100%) of ethanol, methanol, 100% ethyl acetate and water extracts. We tested melanin inhibitory effect and tyrosinase activity using B16/F10 melanoma cell. As a result, treatment of organic solvent extracts is decreased the biosynthesis of melanin and tyrosinase activity to 36 ~ 86%. Especially the 70% ethanol extracts was the most effective in B16/F10 melanoma cells. In the study on melanogenic protein expression, 70% ethanol extracts of Scolopendra subspinipes mutilans blocked glycosylation of tyrosinase. Therefore this result suggests that 70% ethanol extracts could be developed as skin whitening agents.

Anti-aging and Anti-melanogenesis Efficacy by Antioxidative Mineral-bio Water (항산화성 바이오 미네랄 활성수에 의한 항노화 및 미백효과)

  • UV irradiated skin cells produce reactive oxygen species (ROS). ROS is known to be the primary cause of skin inflammation that is eventually leading to skin aging through decrease of collagen in the dermis. In this study, we evaluated basic efficacy of anti-aging, anti-inflammation and anti-melanogenesis using two antioxidative mineral-bio waters (Mineral-bio water 1 (MIBA-W1) and Mineral-bio water 2 (MIBA-W2)). Both antioxidative mineral-bio waters reduced TNF-${\alpha}$ expression which was induced upon UV irradiation. MIBA-W 1 increased collagen synthesis from UVB irradiated fibroblast at 0.01 % concentration but MIBA-W2 shows slight, but linear increase. Stimulation of melanogenesis by ${\alpha}$-MSH treatment in the cultured B16-F1 melanoma was significantly reduced by the treatment of MIBA-W2 in a dose dependent manner. Taken together, antioxidative MIBA-W1 and 2 seem to have potential applications as functional cosmetic materials.
