제주마 Transferrin Gene Exon 13, 15 및 16의 다형현상 (Polymorphisms of the Exons 13, 15 and 16 of Transferrin Gene in Cheju Horses)

  • 본 연구는 제주마집단(GroupⅠ, 제주도 축산진흥원 사육, 137두; Group II, 농가사육, 30두)과 더러브렛 품종집단(한국마사회 육성마목장, 43두)을 이용하여 SSCP를 통한 Transferrin exon 13, 15, 16의 다형현상 확인과 각 SSCP 유전자형의 염기서열을 분석하기 위하여 수행하였다. 공시재료에서 SSCP에서 관찰된 band에 의한 분석결과 대립인자는 exon 13, 15 및 16에서 각각 2개(A,B), 3개(A,B,C) 및 3개(A,B,C)가 존재하는 것으로 확인되었다. Transferrin exon 13에서 제주마와 더러브렛 집단 모두 A인자가 매우 높게 분포하고 있음이 확인되었다. exon 15에서는 그룹간의 빈도차를 확인 할 수 있었다. exon 15에서 높게 출현되고 있는 유전자형은 GroupⅠ에서 AB (0.445)형, GroupⅡ에서 AA(0.367)형, 더러브렛 품종에서는 AA(0.767) 유전자형이 가장 높은 빈도로 출현되어 제주마 집단간 또는 품종간에 빈도의 차이를 관찰할 수 있었다. exon 16에서는 GroupⅠ은 A, B, C 인자, GroupⅡ에서는 A 및 B 2종류의 인자형이 확인되었고 더러브렛 품종에서는 A인자형만 검출되었다. exon 16에서도 그룹간에 유전인자의 빈도차를 확인 할 수 있었다. 또한 exon 13, 15 및 16의 조합으로 형성된 개체의 유전자형은 전체 13종류가 출현되었고 이 조합도 그룹간 차이를 확인 할 수 있었다. SSCP 유전자형에 따른 각 인자들에 대한 염기서열을 분석한 결과 exon 13과 16에서 각 1개의 새로운 SNP가 발견되었다. 본 연구결과 제주마 transferrin exon 13, 15, 16은 더러브렛 품종에서와 같이 높은 대립인자의 다형성을 보였으며, 각 Group 간 빈도차를 확인 할 수 있었다.

자하거(紫何車) 약침액(藥針液)이 B16 melanoma 세포주의 멜라닌 합성에 미치는 영향 (The effects of Hominis Placenta extract on Melanin synthesis of B16 melanoma cells)

  • Objective : This research was carried out for the development of medicine for vitiligo treatment and focused on the effect of Hominis Placenta extract on Melanin synthesis of B16 melanoma cells. Methods : Acitivity of tyrosinase playing a vital role in synthesis of Melanin and the quantity of Melanin, which is the final product in cultured B16 melanoma cells, effects of Hominis Placenta extract were measured. Results: The results indicated that Hominis Placenta extract increased both the amount of Melanin and the activity of tyrosinase according to the concentration, and they also supported by western blot analysis. Conclusion : The results suggests that Hominis Placenta extract has an advantageous effect on the promotion of Melanin synthesis and will contribute to the development of vitiligo treatment through further related studies.

산수유 클로로포름 추출물에 의한 B16/F10 melanoma세포의 증식억제효과 (Effect of Proliferation Inhibition on B16/F10 Melanoma Cell by Chloroform Extract from Cornis fructus)

  • Cornis fructus have various biological effects and major chemical components have been tannins, saponins, ursolic acids, gallic acids, linoleic acids, morronisides, cornins and loganins. The main aim of the present study is measurment the effect of chloroform extract from Cornis fructus on proliferation inhibition and Cell death. Cells were cultured in the presence of chloroform extracts from Cornis fructus for 48 h. after 48h treatment of B16/F10 melanoma cells with chloroform extracts, the cells were observed a dose-dependent inhibitions of cell viability with cell death in their proliferation. the cells were estimated cell viability, cell number, total DNA fragmentation and chromatin condensation in a dose-dependent manner. It also caused cell death as measured by cell morphology, DNA fragmentation and nucleus chromatin condensation. therefore, these results suggest that chloroform extracts from C. fructus is inhibitory proliferation and is related to cell death in this cells.

  • GaAs MESFET을 사용한 12GHz저잡음 증폭기를 MIC로 설계하였다. 증폭기의 입출력 결합은 칩 캐패시터와 대칭구조의 DC블록을 포함시켜 각기 실현하였다. 실험을 통하여 칩 캐패시터를 사용하는 경우 11.8-12.1GHz에서 8-11dB의 이득을 얻었으며 DC 블록을 포함하는 경우 12.16-12.19GHz에서 16-18dB의 이득을 나타내는 비교 결과를 얻었다.

The Antimelanogenic Effect of Inularin Isolated from Flowers of Inula britannica on B16F10 Melanoma Cells and Zebrafish Embryos

  • In the search for novel, natural melanogenesis inhibitors, a new sesquiterpene, inularin, was isolated from the flowers of Inula britannica, and the structure was determined using spectroscopic and chemical methods. The antimelanogenic effects of inularin on B16F10 melanoma cells and zebrafish embryos were evaluated. Inularin dose-dependently reduced melanocyte-stimulating hormone-induced melanin production and L-DOPA oxidation in B16F10 cells. Zebrafish embryos were used to confirm the antimelanogenic activity. Inularin significantly decreased the pigmentation of embryos compared with untreated controls.

B16 마우스 흑색 종양 세포에서 빈랑(Areca catechu) 추출물이 티로시나아제 유전자에 미치는 효과 (The Effects of Areca catechu Extracts on Tyrosinase Gene Expression in B16 Mouse Melanoma Cells)

  • In this study, we found that the methanolic extract of Areca catechu repressed expression of the tyrosinase gene in B16 mouse melanoma cells containing a tyrosinase promoter. Extract concentrations of 100 ${\mu}$g/ml and 500 ${\mu}$g/ml exhibited tyrosinase gene expression rates of aproximately 62% and 48%, respectively, compared to the control. The fraction layers consisting of ethyl acetate, butyl alcohol, and water showed repressive effects on the tyrosinase gene. In particular, the butyl alcohol fraction highly repressed at 100 ${\mu}$g/ml and 500 ${\mu}$g/ml. In the MTT assay, the methanolic extracts exhibited very low cytotoxicities at 1 ${\mu}$g/ml, 10 ${\mu}$g/ml, and 100 ${\mu}$g/ml.

  • Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.

B16 Melanoma 세포에서 Phenylprlopanoid 화합물이 Melanin 생성에 미치는 영향 (Effects of Phenylpropanoid Compounds on Melanin Production in B16 Melanoma Cells)

  • To investigate the relationship between structure and biological activity of phenylpropanoids, we measured effects of phenylpropanoids on anti-oxidant and whitening activity, In DPPH radical scavenging activity, caffeic acid analogues showed the significant anti-oxidant activity. Although phenylpropanoids did not inhibit purified-tyrosinase activity, they significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. However, phenylpropanoids did not affect tyrosinase expression in MSH-stimulated B16 melanoma cells, which suggest that inhibition of MSH-induced melanin production was due to tyrosinase inhibition mediated via other signal pathways but not expression of tyrosinase. Phenylpropanoids also significantly inhibited both hyaluronidase and elastase activity, suggesting that phenylpropanoids may be used as whitening, hydration and anti-wrinkling agents. Hydroxyl residue of aromatic ring in phenylpropanoids plays an important role in anti-oxidant and whitening activity.

Relation of Chitosan oligosaccharide-induced Melanin Production to The Activity and Expression of Tyrosinase in B16 Melanoma Cells

  • To investigate the effect of chitosn oligosaccharide on skin care, we measured tyrosinase activity and melanin production in B16 melanoma cells, and elastase and hyaluronidase activity. Chitosan oligosaccharide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-depednetly increased mealnin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. (omitted)
