Water-soluble sulfobetaine chitosan (SCs) was prepared for a blending film with Bombyx mori silk fibroin (SF) by reacting chitosan with 1,3-propanesultone. A series of SF/SCs blended films were successfully prepared by mixing aqueous solutions of B. mori SF and SCs. The SF/SCs blended films were examined through spectroscopic and thermal analysis to determine the morphological changes of SF in the SCs. The effects of the SF/SCs blend ratios on physical and mechanical properties were investigated to discover the feasibility of using these films as biomedical materials such as artificial skin and wound dressing. X-ray analysis showed good compatibility between the two biopolymers. The in vitro degradation behavior of the SF/SCs blended films was systematically investigated for up to 8 weeks in phosphate buffered saline solution at $37^{\circ}C$ and showed a mass loss of 46.4% after 8 weeks. All films showed no cytotoxicity by MC3T3-E1 assay. After 3 days of culture, the relative cell number on all the SF/SCs films was slightly lower than that of an optimized tissue culture plastic.

Central Sericultural Research and Training Institute, Mysore have evolved several highly productive bivoltine hybrids which can produce international grade raw silk. Among them $CSR2{\times}CSR4,\;CSR2{\times}CSR5,\;CSR3{\times}CSR6,\;CSR17{\times}CSR16,\;CSR18{\times}CSR19$ and $CSR12{\times}CSR6$ are being popularized in the field. There is a minimum difference in their economic characters but they appear to differ in survival. Though they are productive under high input management conditions, they are very susceptible to different diseases under normal rearing practices. No systematic attempts have been made to test their susceptibility status / resistance. Thus the present study is a modest attempt to screen the above six productive bivoltine hybrids to two important pathogens viz., Bombyx mori Nuclear Polyhedrosis Virus (BmNPV) and Bombyx mori Infectious Flacherie Virus (BmIFV) along with existing hybrid, $KA{\times}NB4D2$ to assess their susceptibility / resistance. The results shows that the productive hybrid $CSR2{\times}CSR4$ is the most resistant to BmNPV and it is suggested by its highest $LC_{50}$ value followed by $CSR12{\times}CSR6,\;KA{\times}NB4D2,\;CSR3{\times}CSR6,\;CSR17{\times}CSR16,\;CSR18{\times}CSR19,\;CSR2{\times}CSR5$. Based on the $LC_{50}$ value and $LT_{50}$ values for BmIFV, the hybrid $KA{\times}NB4D2$ was found to be the most resistant (1st position) one followed by $CSR3{\times}CSR6$ (2nd position) $CSR2{\times}CSR$ (3rd position) and $CSR12{\times}CSR6$ (4th position) $CSR17{\times}CSR16$, $CSR18{\times}CSR19$ (5th position) and $CSR2{\times}CSR5$ being the least. The response of 7 bivoltine hybrids to both the pathogens BmNPV and BmIFV indicates that, the hybrids $CSR2{\times}CSR4$, $CSR12{\times}CSR6$ and $KA{\times}NB4D2$ were found to be the most resistant when compared to others. Further, $KA{\times}NB4D2$ being less productive hybrid with a shell ratio of 20.08%, the other two hybrids $CSR2{\times}CSR4$ (Cocoon shell ratio, 21.44%) and $CSR12{\times}CSR6$ (cocoon shell ratio, 23.45%) can be considered to be most productive with superior quality cocoon and resistant to both BmNPV and BmIFV pathogens. The overall study indicated that the hybrid $CSR2{\times}CSR5$ is the most susceptible hybrid to both the pathogens.

It has already been reported that short neuropeptide F (sNPF) stimulates feeding behaviors in a wide variety of insect species. In the present study, we cloned cDNA, encoding a sNPF receptor homologue from a silkworm, Bombyx mori, named BsNPF-R. The amino acid sequence of BsNPF-R was compared with those of sNPF-R thus far reported, which is shared with humans (36%), mice (34%), zebrafish (35%), and fruit flies (51%), respectively. A BsNPF-R protein’s mass was theoretically estimated to be 42,731 Da and it is a putative plasma membrane-penetrating protein. The mRNA expression of BsNPF-R was tested; the results showed that a strong expression was detected at the midgut, post-silk gland, Malpighian, and testis; however, a weak expression was at the fat body, hemocyte, and ovary. In addition, the synthesized sNPF of a silkworm regulated the BsNPF-R mRNA expression through the cell-based functional analysis.

The interest in the use of borate crystals in ultraviolet(UV) nonlinear optics(NLO) has increased because all solid-stste UV lasers obtained with NLO are in highly demand. Much effort has been spent on developing borates series, such as{{{{ { beta -BaB }_{2 } {O }_{4 }(BBO), {LiB }_{ 3}{O }_{5 }(LBO)}}}} and{{{{{ CsKiB}_{6 }{O }_{10 }(CLBO)}}}} in this decade. Recently another new borate crystals, {{{{{ YCa}_{4 }O({BO }_{3 })_{3}}}}} and{{{{{Gd }_{x }{Y }_{1-x }{Ca }_{4 }O({BO }_{ 3})({Gd }_{x }{Y }_{1-x }COB)}}}} have been developed by the present authors. Here, the growth and NLO properties of YCOB and {{{{ {Gd }_{ x} {Y }_{ 1-x} }}}}CO B crystal are reported and their properties discussed in relation to those of other nonlinear optical crystals, such as LBO.

The Bombyx mori Microarray Database (BmMDB; http://silkworm.swu.edu.cn/microarray) provides information for tissue-specific gene expression by using the whole-genome oligonucleotide microarray in the silkworm. We analyzed the tissue-specific expression patterns in the silk gland, fat body, and midgut five days of fifth instar larvae during the development of B. mori. To verify the tissue-specific expression, analysis was conducted using quantitative Real-time RT-PCR and the highly expressed endogenous Actin RNA as an intrinsic reference. Finally, we confirmed five genes, (sw15872, sw00692, sw20990, sw05300,and sw2250), out of 18 candidates expressed in two different tissues, which was consistent with the data published by Dr. Xiang's group, thereby supporting the BmMDB. Further studies for promoter regions of candidate genes can be applied in creating transgenic silkworms as biomedical insects for use in producing biomaterials, and to serve as well-characterized models for understanding the mechanism for the genetic regulation of tissue-specific development.
To investigate the effect of hemolymph of silkworm larvae on the multiplication of Bombyx mori nuclear polyhedrosis virus (BmNPV) in BmN-4 cells, BmN-4 cells were infected with BmNPV, which were sequentially Heated with the hemolymph exracted from B. mori larvae. When the culture media TC-100 containing 3% fetal bovine serum was mixed with 10% hemolymph heated at 65$^{\circ}C$ for 30 minutes, the released polyhedra by multiplication of BmNPV in BmN-4 cells were increased more than those of non-treated. However, multiplication of BmNPV in BmN-4 cells treated with non-heated hemolymph was not effective, since non-heated hemolymph was toxic for the cell growth. The result of plaque assay showed that plaque forming units in BmN-4 cells treated with heated hemolymph are significantly increased, suggesting that efficiency of multiplication of BmNPV in BmN-4 cells is due to increase not of cell growth but of infectivity of BmNPV.

A detailed evaluation of relative toxicity of fluoride to die 5th instar larvae of PM and NB$_4$D$_2$ races of silkworm and the effects of lethal, sublethal and prevailing levels of fluoride in groundwater on the growth, economic characters and fecundity of the silkworm, Bombyx mori (Lepidoptera: Bombycidae) were studied. The feeding of mulberry, Morus alba leaves treated with lethal and sublethal doses of fluoride to PM and NB$_4$D$_2$ races of silkworms from the beginning of the 5th instar to the end of the feeding period resulted in significant reduction in growth, single cocoon weight, single shell weight, silk index, average filament length, and fecundity when compared to controls. These manges were more pronounced on exposure to lethal dose than sublethal dose and in general, the changes induced by fluoride were more striking in NB$_4$D$_2$ than PM, indicating the greater resistance of PM to higher fluoride levels. Groundwater quality in sericulturally important villages of Karnataka and Andhra Pradesh States has been studied with special reference to the presence of fluoride. On exposure to dose of prevailing levels in the waters of sericulturally important areas, die changes observed on growth, economic characters and fecundity were slightly lower when compared to controls and the decrease was found to be insignificant (P$\geq$0.05). It was concluded that, though minute dose (4.0 ppm) of fluoride did not have any toxic impact, it is toxic at higher concentrations to silkworms.

Four thermo-tolerant lines of silkworm, Bombyx mori, (L.) viz., A HT, B HT (Chinese type) and F HT, G HT (Japanese type) were evolved by utilizing the breeding resource material (identified from initial screening at a temperature of 31 ${\pm} 1^{\circ}C$ and relative humidity 85 ${\pm}$ 5%) through conventional breeding. These tolerant lines were crossed with productive breeds and forty four hybrids were evaluated on eight economic traits by the Multiple Trait Evaluation Index Method. Ten hybrids were short-listed based on the average evaluation index value larger than 50 for eight economic traits studied. The identified ten hybrids recorded higher index values (> 50) for most of the traits studied. Single hybrid G ${\times}$ CSR12 indicated average index value larger than 50 for six traits viz., pupation number (58), cocoon weight (67), shell weight (65), average filament length (74), raw silk % (69), reelability % (51) except for shell ratio % (41). The standard deviation of the cocoons in the above hybrid was 8.41 in the hybrid cocoon length and width measurement. However, two selected hybrids viz., A ${\times}$ CSR5 and G ${\times}$ CSR13 recorded average index value larger than 50 for all the traits viz., pupation number (57, 60), cocoon weight (50, 54), shell weight (56, 57), shell ratio percentage (59, 53), average filament length (55, 60), raw silk percentage (63, 67) and reelability percentage (53, 53). The standard deviation of the cocoons in the two selected hybrids viz., A ${\times}$ CSR5 and G ${\times}$ CSR13 was 8.41 and 8.06 respectively in the cocoon length and width measurement.

Infection effect of $Nosema$$bombyics$ on the midgut of silkworm $Bombyx$$mori$ and subsequent appearance of spores and the performance of larvae was studied. Autopsy of larvae showed white pustules on the surface of midgut at 5 days of post infection (pi). At later stage, important organs like midgut, silk gland and gonads reduced in size and all these organs showed white pustules. Light microscope observation of pustules revealed enormous spores. Spore multiplication was at a faster rate in young larvae. Infection of the adult larvae resulted in pebrinized pupa and moths. Larval weight, cocoon weight and cocoon shell ratio reduced as the post infection period increased. Transverse sections of midgut showed $N.$$bombycis$ infection limited to a few columnar cells at 3-5 days of pi. At 7 days pi, cell volume increased, cells were swollen and elongated. Heavily infected cells looked like sacks filled with parasite and the apical region of certain cells were bulging into the gut lumen. Later at 8-9 days of pi, spores or its developing stages leaked into the lumen either freely or enclosed within the globules of host cytoplasm. Besides columnar cells, development of $N.$$bombycis$ was observed in the regenerative cells and rarely in goblet cells. Development of $N.$$bombycis$ was also observed in both longitudinal and circular muscles at the late pi period. The histopathological changes, deformities and spore production time in the host were all influenced by the spore dosage and age of the host.
We tried to identify differentially expressed genes (DEGs) from a silkworm, Bombyx mori, involved in fungal (Aspergillus niger) infection. A total RNA purified from fungal-induced and normal B. mori ($5^{th}$ instar larvae) was used for the cDNA synthesis. Differentially expressed genes were screened by annealing control primer (ACP)-based PCR technique. Comparing the gene expression profiles between fungal infection and control silkworm, we detected 10 genes that were differentially expressed in fungal induction and performed molecular cloning and nucleotide sequencing of the 10 genes. We confirmed the expression patterns of 3 DEGs by RT-PCR. The 3 DEGs over-expressed in fungal infection were identified as lysozyme, enbocin and an unknown gene. They were first identified to be genes induced by fungal infection. Although the detailed functions of 3 genes and their products remain to be determined, the genes will provide insight into the molecular mechanisms of insect-immune systems induced by fungal infection.
