Effects of Seonpye-tang on OVA-Induced Asthmatic Mice (천식모델생쥐에서 선폐탕(宣肺湯)의 면역조절 효과)

  • This study aimed to evaluate the anti-asthmatic effects of Seonpye-tang (SPT) using OVA-induced asthmatic mice model. Scavenging activity of SPT on DPPH free radical and SOD-like activity of SPT were measured at final concentration 62.5, 125, 250, 500 (${\mu}g/m{\ell}$), RBL-2H3 cells were treated with DNP IgE for 24hr, and treated with SPT (1, 10, 100 ${\mu}g/m{\ell}$) for 1hr, followed by treatment with DNP-HSA for 1hr at $37^{\circ}C$. The level of IL-4 and TNF-${\alpha}$ were measured by ELISA. Asthmatic mice model was conducted by repeated challenge of OVA using C57BL/6 mice. Each group was treated with distilled water, SPT (400 mg/kg and 200 mg/kg) extract or cyclosporin A (10 mg/kg) for the later 8 weeks. Immune cells subpopulation, eotaxin, IL-5 and TNF-${\alpha}$ in BALF were analyzed. SPT dose-dependently increased Scavenging activity on DPPH free radical and SOD-like activity. SPT significantly ameliorated the increase of total cells number and eosinophil including of immune cell subpopulation of $CD3^+/CD69^+$, $CCR3^+$, $B220^+/CD22^+$, $B220^+/CD45^+$ and $B220^+/IgE^+$ cells in BALF comparing to control group. Eotaxin and IL-5 level in BALF were significantly decreased by SPT. These results strongly suggest that SPT would be a effective candidate for herbal-originated anti-asthmatic drug. However, this drug should be further studied for characterization of the accurate action and underlying mechanisms using variant disease model in the future.

Inhibitory Effects of Ethanol Extracts of Some Korean Plants on Ultraviolet B-Induced DNA Damage (HaCaT 세포에서 자외선 B에 의해 유도되는 DNA 상해에 대한 국내 수종 자생 식물 에탄올 추출물의 저해효과)

  • In the present study, the effects of extracts from Korean plants on the DNA damage response in HaCaT cells exposed to ultraviolet B (UVB) were investigated. The activity of cells treated for 24 hr with ethanol extracts from Vaccinium spp. (VS), and Vitis vinifera L (VV) alone was similar to that of the non-treated control, but gradually decreased at concentrations above $200{\mu}g/mL$. However, when post-incubation of UVB-exposed cells was carried out for 24 hr in medium containing VS or VV extracts, the cell activity increased in a concentration-dependent manner compared with that in the normal growth medium. The cell viability of UVB-exposed cells also increased when post-incubated in medium containing VS or VV extracts, in a concentration-dependent manner. Nuclear fragmentation analysis showed that post-incubation with VS or VV extracts decreased the UVB-induced apoptosis by about 10 and 13%, respectively, of that in cells post-incubated in growth medium. After 24 hr of post-incubation in medium containing VS or VV extracts, the level of CPD and 8-OHdG decreased in time- and concentration-dependent manners. Overall these results suggest that VS and VV extracts assist the survival of UVB-exposed cells, in accordance with the respective decrease in the levels of UVB-induced DNA damage.

Phenotypic and Genetic Parameters for Inosine Acid in Relation to Carcass and Meat Quality Traits in Pigs

  • A total of 135 F2 finishing pigs (65 barrows and 69 gilts) from resource population (Large White${\times}$Meishan) were slaughtered at about 87.8 kg BW. Contents of inosine acid (IMP) and carnine (HR) in muscle were assayed by HLPC and genetic parameters for IMP content and HR content were estimated using full sibs model. There was significant sex effect on IMP content(p<0.05), $3.561{\pm}0.077mg/g$ for gilt and $3.287{\pm}0.085mg/g$ for barrow. Heritability estimates for IMP and HR content were 0.127 and 0.357, respectively. The phenotypic correlation between IMP content and HR was 0.335, pH (A) 0.024, water lose rate (WLR) -0.069, intramuscular fat (IMF) -0.214, average marbling score (MARB) -0.143, average backfat measurements (AVBF) -0.084 and average color value (CV) -0.156, respectively. The result indicated that inosine acid content in meat might be retained or slightly improved by reducing backfat depth in pig breeding.

  • Human papillomaviruses (HPVs) are small, non-enveloped, double-stranded DNA viruses that infect epithelial tissues. Specific genotypes of human papillomavirus are the single most common etiological agents of cervical intraepithelial lesions and cervical cancer. Cervical cancer usually arises at squamous metaplastic epithelium of transformation zone (TZ) of the cervix featuring infection with one or more oncogenic or high-risk HPV (HR-HPV) types. A hospital-based study in a rural set up was carried out to understand the association of HR-HPV with squamous intraepithelial lesions (SILs) and cervical cancer. In the present study, HR-HPV was detected in 65.7% of low-grade squamous intraepithelial lesions (LSILs), 84.6% of high-grade squamous intraepithelial lesions (HSILs) and 94% of cervical cancer as compared to 10.7% of controls. The association of HPV infection with SIL and cervical cancer was analyzed with Chi square test (p<0.001). The significant association found confirmed that detection of HR-HPV is a suitable candidate for early identification of cervical precancerous lesions and in the prevention of cervical cancer in India.

Efficacy of Korean Multipurpose Contact Lens Disinfecting Solutions against Acanthamoeba castellanii

  • Acanthamoeba keratitis has been increasing in recent years. Main risk factors are contact lens wear and their cleaning solutions. Most contact lens wearers use multipurpose disinfecting solutions (MPDS) for cleansing and disinfecting microorganisms because of its convenience. We determined amoebicidal effects of MPDS made in Korea and their cytotoxicity on human corneal epithelium cells. Fifteen commercial MPDS (A to O) were tested for their amoebicidal effects on Acanthamoeba castellanii trophozoites and cysts by using a most probable number (MPN) technique. Among them, 7 kinds of MPDS showed little or no amoebicidal effects for 24 hr exposure. Solutions A, B, G, H, L, and O showed positive amoebicidal effects, and solutions M and N killed almost all trophozoites and cysts after 24 hr exposure. However, 50%-N solution showed 56% cytotoxicity on human corneal epithelial cells within 4 hr exposure, and 50%-O solution also showed 62% cytotoxicity on human cells within 4 hr exposure. Solution A did not show any cytotoxicity on human cells. These results revealed that most MPDS made in Korea were ineffective to kill Acanthamoeba. The solutions having amoebicidal activity also showed high levels of cytotoxicity on human corneal epithelial cells. New formulations for improved MPDS that are amoebicidal but safe for host cells are needed to prevent Acanthamoeba keratitis.

Incidence, Risk Factors, and Outcomes of Febrile Neutropenia in Thai Hematologic Malignancy Patients Receiving Chemotherapy: A 6-year Retrospective Cohort Study

  • A 6-year retrospective cohort study was conducted among Thai hematologic malignancy (HM) patients receiving intensive chemotherapy. Of the 145 eligible patients receiving 893 chemotherapy sessions, 46.9% were female, median age was 52 years, and the most common HM diagnosis was diffuse large B-cell lymphoma (46.2%). Febrile neutropenia (FN) occurred in 14.9% of chemotherapy sessions with an incidence of 24.8 per 1,000 chemotherapy cycles per year. Independent factors associated with FN were receiving the first chemotherapy cycle [adjusted hazard ratio (aHR) 4.1], having hemoglobin ${\leq}100g/L$ (aHR 3.7) and platelet ${\leq}140,000/{\mu}L$ (aHR 2.7) on chemotherapy day and receiving acute myeloid leukemia regimens (aHR 20.8). Granulocyte colony stimulating factor was significantly associated with reduced rate of FN when given in those receiving CHOP regimen. With the median follow-up time of 16 months, the overall survival time was significantly longer in patients without FN than those with FN (61.7 vs. 20.8 months; p<0.001).

Effect of Cytochalasin B in Activation Medium on the Development of Rat Somatic Cell Nuclear Transfer Embryos

  • This study was conducted to evaluate the effect of cytochalasin B (CB) treatment in the activation medium on the development of somatic cell nuclear transfer (SCNT) rat embryos. Fetal fibroblast cells were isolated from a Day 14.5 fetus, and the oocytes for recipient cytoplasm were recovered from 4-week old Sprague Dawley rats. After enucleation and nuclear injection, the reconstructed oocytes were immediately exposed to activation medium consisting of 10 mM $SrCl_2$ with or without CB for 4 hr, and formation of pseudo-pronucleus (PPN) was checked at 18 hr after activation. Then, they were transferred into day 1 pseudopregnant recipients (Hooded Wistar) or cultured for 5 days to check their developmental competence in vivo or in vitro. The number of PPN was not affected by CB treatment during the activation. However, CB treatment supported pre-implantation development of rat SCNT embryos. Embryos generated by the procedures of SCNT were also capable of implanting, with 1 implantation scar found from a recipient following the transfer of 87 SCNT embryos to four foster mothers. The result of the present study shows that rat SCNT embryo can develop to post-implantation stage following treatment with CB.

Effects of Cu, Cd, Fenbendazole and Sulfathiazole on the Survival of the Korean Fairy Shrimp Branchinella kugenumaensis (구리, 카드뮴, 펜벤다졸, 설파티아졸이 국내산 풍년새우 생존에 미치는 영향)

  • The short term (24-hr) and long term (21 days) effects of copper, cadmium, fenbendazole and sulfathiazole on the survival of the Korean fairy shrimp Branchinella kugenumaensis were evaluated. The 24-hr median lethal concentrations ($LC_{50}$) of copper, cadmium, fenbendazole, and sulfathiazole were 39, 512, 182, and 31,818 ${\mu}g/L$, respectively. The toxicity of copper is highest among 4 chemicals used in this study, while sulfathazole the lowest. After the long term (21 days) exposure experiment, the $LC_{50}$ copper, cadmium, fenbendazole, and sulfathiazole were 1.12, 2.1, 0.1, 6.6 ${\mu}g/L$, respectively. The long term effects of antibiotics were highly enhanced while the short-term effects were not strong. The sensitivities of B. kugenumaensis to copper and cadmium were higher than or comparable to those of other freshwater branchiopods (Streptocephalus spp., Thamnocephalus sp.), and far higher than the marine species (Artemia sp.). There were significant effects on the survival of B. kugenumaensis after long term exposure to relatively lower concentrations of copper, cadmium, fenbendazole and sulfathiazole. Therefore, B. kugenumaensis seems quite a good candidate species for the ecotoxicological assessments of freshwater environments.

Production of 5-Aminolevulinic Acid (ALA) by Bacillus cereus 1-1 (Bacillus cereus 1-1 균주의 5-Aminolevulinic Acid (ALA) 생산)

  • Bacillus cereus 1-1 strain produced 2 mM of ALA in the aerobic dark condition without any inhibitor like levulinic acid. The optimum culture conditions for the ALA production were that preculture and main culture were continued for 18 hr in TCY medium, and 16 mM of organic acids like acetic acid were added at the late log phase when the pH was 6.8. And the addition of 0.3% glucose was effective at the beginning of the main culture. ALA production was continued for more than 8 hr by the addition of glutamic acid instead of acetic acid, and was inhibited by addition of $40\;{\mu}M$ gabaculine seriously. These results confirmed that B. cereus 1-1 strain produced ALA through C-5 pathway.

The Microwave Dielectric Properties of Low-Temperature Sintered $ZnNb_2O_6$ Ceramics with Addition (첨가물에 따른 저온소결형 $ZnNb_2O_6$ 세라믹스의 마이크로파 유전특성)

  • The $ZnNb_2O_6$ ceramics with 3wt% CuO and $B_2O_2$(1,3,5wt%) were prepared by the conventional mixed oxide method. The ceramics were sintered at the temperature of $1000^{\circ}C\sim1050^{\circ}C$ for 3hr. in air. The structural properties were investigated with sintering temperature by XRD and SEM. Also, the microwave dielectric properties were investigated with sintering temperature. Increasing the sintering temperature, the peak of second phase ($Cu_3Nb_2O_8$) was increased. But no significant difference was observed as sintering temperature. In the $ZnNb_2O_6$ ceramics with 3wt% CuO and 5wt% $B_2O_3$ sintered at $1025^{\circ}C$ for 3hr, the dielectric constant, quality factor, temperature coefficient of the resonant frequency were 22.92, 20,271GHz, -14.27ppm/$^{\circ}C$, respectively.
