The effect of Vitamin $B_2$ Deficiency on the Utilization and Recuperation of Fuel in Exercising Rats (비타민 $B_2$ 부족이 운동중 연료의 이용과 회복에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin B2 deficiency on the utilization and recuperation of fuel in exercising rats. Thirthy six rats were fed either a vitamin B2 deficient diet (-B2) or a control diet (+B2) for 3 weeks and then subdivided into 3 groups : non-exercise group(NE), exercise group (ES), exercise and recuperation group (ER). ES group were exercised on treadmill (10$^{\circ}$, 0.5-0.8km/h) for 2 hours and ER group were recuperated three days with the respective diet after exercise. Glucose (GLU), glycogen(GLY), protein(PRO), triglyceride(TG) and free fatty acid(FFA) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. Compared to + B2 rats, in NE group, the level of P-GLU and L-GLU of -B2 rats was lower, L-PRO and M-GLY was higher and there were no differences in P-PRO, P-FFA, L-TG and M-TG. In ES group, the level of P-GLU was lower, P-FFA was higher and there were no differences in P-PRO, P-TG, L-GLU, L-TG, M-GLY, M-TG and M-PRO. In ER groups, the level of P-GLU and L-TG was lower, P-FFA was higher and there were no differences in P-PRO, P-TG, L-GLY, L-PRO, M-GLY, M-TG and M-PRO. These results suggest that a vitamin B2 deficiency may impair the utilization of stored fuel during exercise suggest that a vitamin B2 deficiency may impair the utilization of stored fuel during exercise and may lead a sluggish recuperation related to fuel stores after exercise.

Influential Factor of Life Satisfaction based on Types of Baby Boomer Generation (베이비부머세대의 유형별 삶의 만족 영향 요인)

  • The purpose of this study was to identify the multidimensional factors that affect the satisfaction of life by type of baby boomer generation. The subjects of this study were 1,895 households who were born from 1955 to 1963 among the respondents in the Korea welfare panel (the 10th year). The baby boomer generation was classified into four types such as gender, income status, male and female as socioeconomic status. The male type IV (B=.074, t=1.992, p<.05), health status (B=.233, t=7.268, p<.001), household income (B=.174, t=4.476, p<.001), depression (B=-.182, t=-5.242, p<.001), self-esteem (B=.255, t=7.027, p<.001), trust (B=.064, t=2.036, p<.05), etc. were found to have a significant effect on the life satisfaction of the male baby boomer generation. The female type IV (B=.100, t=2.598, p<.05), health status (B=.239, t=6.790, p<.001) (P<.001), depression (B=-.181, t=-5.142, p<.001), and self-esteem (B=.320, t=8.600, p<.001), etc. were found to have a significant effect on the life satisfaction of the female baby boomer generation.

The Stability of p53 in Ras-mediated Senescent Cells in Response to Nucleolar Stress (Ras에 의해 유도된 노화세포에서 핵인 스트레스에 의한 p53 안정화 연구)

  • B23/nucleophosmin, a nucleolar protein, translocates into the nucleus from the nucleolus when cells are damaged by extracellular stresses. Recently, it was shown that such translocation of B23/nucleophosmin in normal fibroblasts under stress conditions increases both the stability and activation of the p53 protein by disrupting its interaction with MDM2. Senescent cells have a single large nucleolus and a diminished capacity to induce p53 stability upon exposure to various DNA damaging agents. To investigate the role of B23/nucleophosmin in p53 stability in senescent cells, we established a senescence model system by expressing the ras oncogene in IMR90 cells. The stability of p53 was reduced in these cells in response to nucleolar stress, although the level of B23/nucleophosmin protein was not changed. In addition, p53 did not accumulate in the nucleus and B23/nucleophosmin did not translocate into the nucleoplasm. The binding affinity of B23/nucleophosmin with p53 was reduced in senescent cells, whereas the interaction between MDM2 and p53 was stable. Taken together, the stability of p53 in ras-induced senescent cells may be influenced by the ability of B23/nucleophosmin to interact with p53 in response to nucleolar stress.

  • The structure of fresh, salted and heated petiole tissue from Chinese cabbage was examined with optical microscope, and the pectin of the tissues was fractionated by successive extraction. The pectin in Chinese cabbage consisted mainly of high methoxyl pectin(pA) and low methoxyl pectin(pB). The pA content was converted to pB markedly by salting and heating at $60{\sim}70^{\circ}C$ for 30 min while heating the cabbage above 80 decreased pB content. The firmness of heated tissue was highly correlated with pB content(r=0.996). Cytorrhysis test showed that pore size of the tissue became smaller by salting and heating at temperature below $80^{\circ}C$.

Carbon Isotope Analysis for the Climatic Environment Change in South Korea During the Holocene: a Case Study in Yengjong Islands of Yellow Sea (탄소동위원소분석을 이용한 한국 홀로세의 기후환경변화: 서해 영종도지역을 사례로)

  • The landform of the study area is defined as a small-scale alluvial plain in the valley bottom adjacent to the coast. By the aggradation of alluvial materials, this valley bottom plain was formed in dissected parts of low hills. For the purpose of reconstructing the palaeo climate environment in this study, $\delta^{13}C$ analysis and soil organic carbon analysis are therefore employed. Main results of this study are as follows: Section I: the period of $6,600{\pm}60yr$ B.P.$-5,350{\pm}60yr$B.P. was mostly in warm and humid climate environment. A little changes of the humid environment are detected as subdry (or sub-humid)${\rightarrow}$humid in terms of the dryness and wetness. Section II: the period of $5,350{\pm}60yr$ B.P.-2,200 yr B.P. was in warm and humid climate environment, which is similar to the present. However, The sediments between $4,720{\pm}60yr$ B.P. and $4,210{\pm}50 yr$ B.P. experienced the most humid climate environment of all studied sedimentary layers. After $4,210{\pm}50 yr$ B.P., the environment started to change from the humid to the sub-humid (or sub-dry) climate. Section III: the period of 2,200 yr B.P.$-210{\pm}60 yr$ B.P. was distinguish from previous two sections as the environmental changes to sub-humid (or sub-dry) climate was apparent.

The Changes of Benzo(a)pyrene in Sesame Oil Affected by Processing Conditions (가공조건에 따른 참기름의 benzo(a)pyrene 변화)

  • Sesame oil is a simple pressed oil as unrefined oil. During manufacturing process of roasting-expression, benzo(a)pyrene[B(a)P] formed as a strong carcinogenic substance is cause a social problem. In manufacturing process of sesame oil, it had following the forming pathway of benzo(a)pyrene[B(a)P] as well as minimizing plan of B(a)P formation. Suitable roasting condition by roaster was during 15~20min at $220^{\circ}C$, B(a)P content in sesame oil was $1.35{\sim}1.57{\mu}g/kg$. Between roasting temperature and/or roastingtime and forming amount of B(a)P was showed a linear correlation. As a point of view the turbidity and yield of final product, roasting process of the more regular level was required.

Characterization of the recombinant cellulase B from Thermotoga maritima (Thermotoga maritima 유래 내열성 cellulase B 융합단백질의 특성 규명)

  • A gene encoding thermostable cellulase B (TmCelB) was isolated from Thermotoga maritima. The open reading frame (ORF) of TmCelB gene was 825bp long which predicted to encode 274 amino acid residues with a molecular weight of 31,732 Da. The 17 amino acid residues from N-terminal of the TmCelB was known as signal peptides. To analyze the enzymatic activity and biochemical properties, the ORF of TmCelB gene excluding a putative signal sequence encoding 17 amino acids were introduced into the E. coli expression vector, pRSET-B, and overexpressed in E. coli BL21. The optimum temperature of recombinant TmCelB was around 95 ℃, and the optimum pH of recombinant TmCelB was around pH 4.5. The recombinant TmCelB was stable at temperature below 100 ℃.

Autophagy-Dependent Survival of Mutant B-Raf Melanoma Cells Selected for Resistance to Apoptosis Induced by Inhibitors against Oncogenic B-Raf

  • Most patients with mutant B-Raf melanomas respond to inhibitors of oncogenic B-Raf but resistance eventually emerges. To better understand the mechanisms that determine the long-term responses of mutant B-Raf melanoma cells to B-Raf inhibitor, we used chronic selection to establish B-Raf (V600E) melanoma clones with acquired resistance to the new oncogenic B-Raf inhibitor UI-152. Whereas the parental A375P cells were highly sensitive to UI-152 ($IC_{50}$ < $0.5{\mu}M$), the resistant sub-line (A375P/Mdr) displayed strong resistance to UI-152 ($IC_{50}$ < $20{\mu}M$). Immunofluorescence analysis indicated the absence of an increase in the levels of P-glycoprotein multidrug resistance (MDR) transporter in A375P/Mdr cells, suggesting that resistance was not attributable to P-glycoprotein overexpression. In UI-152-sensitive A375P cells, the anti-proliferative activity of UI-152 appeared to be due to cell-cycle arrest at $G_0/G_1$ with the induction of apoptosis. However, we found that A375P/Mdr cells were resistant to the apoptosis induced by UI-152. Interestingly, UI-152 preferentially induced autophagy in A375P/Mdr cells but not in A375P cells, as determined by GFP-LC3 puncta/cell counts. Further, autophagy inhibition with 3-methyladenine (3-MA) partially augmented growth inhibition of A375P/Mdr cells by UI-152, which implies that a high level of autophagy may protect UI-152-treated cells from undergoing growth inhibition. Together, our data implicate high rates of autophagy as a key mechanism of acquired resistance to the oncogenic B-Raf inhibitor, in support of clinical studies in which combination therapy with autophagy targeted drugs is being designed to overcome resistance.

Properties of Promoters from Alkali-tolerant Bacillus sp. (알카리 내성 Bacillus속 Promoter의 특성)

  • The promoters of alkali-tolerant Bacillus sp. had been cloned in the promoter probe vector pPL703 and recombinant plasmid p-12 had been constructed. As a result of subcloning, two different promoters were found to exist in the cloned 2.9 kb promoter fragment and two recombinant plasmids p-l2B1 and p-l2B2, each harboring different promoter, were constructed. The promoter activity, which was expressed in the CAT specific activity, of p-l2B1 was 7 times higher than that of p-l2B2. The promoter activity as a function of growth revealed that both promoters of p-l2B1 and p-l2B2 were expressed after the late logarithmic growth phase and repressed in the presence of 1.0% (w/v) glucose.
