Assessment of Dietary Requirement of Broiler Chicks for Available Methionine during Summer

  • A six week growth performance trial involving 450 birds was conducted to determine the dietary requirement of broiler chicks for available methionine (AM). Body weight gain was significantly (p<0.01) lower on high AM diet (1411 g/bird) than low AM (1470 g/bird) and normal AM (1466 g/bird) diets. The feed intake by birds ranged from $3241{\pm}25.69$ in high AM diet to $3321{\pm}25.69g/bird$ in low AM diet. The feed efficiency for the three diets having low, normal and high level of AM were $2.26{\pm}0.02$, $2.24{\pm}0.02$ and $2.30{\pm}0.02$, respectively. The results indicated that the birds fed high AM diet consumed significantly (p<0.01) more feed per unit body weight gain as compared to birds on low and normal AM diets.

Dietary Chromium-methionine Chelate Supplementation and Animal Performance

  • Chromium has emerged as an essential trace mineral in nutrition. However, it readily causes toxicity because of slightly excessive dose and/or form of chromium supplement. Therefore, developing a noble form of chromium supplement which is capable of not only an increased availability but also a reduced toxicity has been a critical issue in chromium nutrition. Chromium-methionine chelate has been, so far, one of the latest developments in its kind. Although not much information is available for the chromium-methionine chelate, especially in view of animal performances upon dietary supplementation, several studies indicated chromium methionine chelate could be effective to improve meat quality by increasing muscle mass but decreasing body fat. Highly-graded beef was produced by dietary chromium methionine supplementation during fattening stage of Korean native steers. Body muscle was increased in replace of decreasing body fat in both pig and rat that were dietary supplemented with chromium methionine chelate. However, a pig farm study did not show any significant improvement of body gain upon supplementation of chromium methionine. Immune responses of pig and rat were not always dependent upon chromium form but were varied by species. These results suggest there could be a different mode of responses due to species as well as onset time of dietary supplementation of chromium methionine. It is still early to conclude the bio-efficacy of chromium methionine chelate presumably due to its recent appearance into the field. But the chelate is certainly worth more application to animal since it certainly reduced the application level of dietary chromium.

유지의 산화에 대한 Methionine과 Lysine의 항산화 효과 (Antioxidative Effect of Methionine and Lysine on Oil Rancidity)

  • 대두유와 라아드의 항온저장 및 가열시 methionine과 lysine의 첨가농도에 따른 항산화 효과 및 기존 항산화제들의 항산화력에 대한 상승효과를 비교 연구하였다. 유지의 항온(60$\pm$2$^{\circ}C$) 저장시 methionine과 lysine을 각 농도별로 첨가한 경우 모든 시료에서 항산화 효과를 보였으며 그 중에서 methionine을 1% 첨가하였을 때 가장 효과가 좋았고 그 정도는 TBHQ를 첨가한 경우와 유사하였으며 tocopherol 보다 월등히 높은 항산화 효과를 보였다. 그리고 이들의 항산화 효과는 농도증가에 따라 비례적으로 증가하였으나 농도간에 큰 차이는 없었다. 즉 이들의 항산화 효과는 TBHQ>methionine 1%>methione 0.1%> methionine 0.02%> lysine 1%>ly-sine 0.1%>Lysine 0.02%>tocopherol 순이었다. 또한 아미노산, TBHQ, tocopherol 모두 식물성유 보다 동물성유지에서 더 큰 항산화력을 나타내었다. 유지 가열(180$\pm$2$^{\circ}C$)시 methionine과 lysine을 각 농도별로 첨가한 모든 시료에서 항산화 효과가 나타났고 TBHQ는 자동사놔시와는 달리 항산화 효과가 크게 떨어졌으며 이들의 항산화 효과의 크기 순서는 methione 1%>methionine 0.1%>methionine 0.02%> lysine 1%>lysine 0.1%>lysine 0.02%>tocopherol, TBHQ 순이었다. Methionine 및 lysine을 tocopherol, ascorbic acid, cit-ric acid와 혼합하여 대두유에 첨가하였을 때 tocoperol 과의 혼합물이 가장 높은 상승효과를 나타내었으며 asco-rbic acid 및 citric acid와의 혼합물도 약하지만 상승효과를 나타내었다. 따라서 methionine과 lysine은 유지에 대하여 항온저장 또는 가열시 모두 항산화 효과가 있으며 특히 가열 시와 동물성 유지에 대하여 그 효과가 우수하고 식품 항산화제에 대한 상승효과도 크다는 것을 알 수 있었다.

Overexpression, Purification, and Preliminary X-Ray Crystallographic Studies of Methionine Sulfoxide Reductase B from Bacillus subtilis

  • The peptide methionine sulfoxide reductases (Msrs) are enzymes that catalyze the reduction of methionine sulfoxide back to methionine. Because of two enantiomers of methionine sulfoxide (S and R forms), this reduction reaction is carried out by two structurally unrelated classes of enzymes, MsrA (E.C. 1.8.4.11) and MsrB (E.C. 1.8.4.12). Whereas MsrA has been well characterized structurally and functionally, little information on MsrB is available. The recombinant MsrB from Bacillus subtilis has been purified and crystallized by the hanging-drop vapor-diffusion method, and the functional and structural features of MsrB have been elucidated. The crystals belong to the trigonal space group P3, with unit-cell parameters a=b=136.096, $c=61.918{\AA}$, and diffracted to $2.5{\AA}$ resolution using a synchrotron-radiation source at Pohang Light Source. The asymmetric unit contains six subunits of MsrB with a crystal volume per protein mass $(V_M)\;of\;3.37{\AA}^3\;Da^{-1}$ and a solvent content of 63.5%.

Biological Effectiveness of Methionine Hydroxy-analogue Calcium Salt in Relation to DL-Methionine in Broiler Chickens

  • Two feeding trials were conducted to assess the relative bioavailability (RBV) of methionine hydroxy analogue calcium salt (MHA-Ca) in comparison to DL-methionine (DL-Met). Male Ross 308 (1-38 days) and Cobb 500 chickens (1-42 days) were used in studies 1 and 2, respectively. Experimental diets based on wheat and soybean meal or sorghum and soybean meal were fed during three phases. In both experiments graded levels of DL-Met and MHA-Ca were supplemented to Met+Cys deficient basal diets. Additionally, in experiment 1, increasing levels of a DL-Met preparation diluted with corn starch to 65% purity (DL-Met65) were supplemented. Birds were kept in floor pens and feed and water were available ad libitum. Body weights and feed consumption were recorded at the beginning and end of the experimental periods and weight gain and feed efficacy were computed subsequently. At the end of the experiments, a number of birds were slaughtered for carcass evaluation (dressing percentage, breast meat yield). Dose response data were analysed by both ANOVA and nonlinear common plateau asymptotic regression. In both experiments birds responded significantly to increasing levels of either methionine source. However, RBV of MHA-Ca compared to DL-Met was markedly (in many cases significantly) below 84%, the value which would have been expected from MHA-Ca's chemical characteristics. Excluding some extremely low RBV figures of trial 2, RBV of MHA-Ca averaged to about 63% in relation to DL-Met. In addition, supplementation of DL-Met65 allowed confirmation of nonlinear common plateau asymptotic regression to be suitable to determine RBV.

Propionate Attenuates Growth of Oral Streptococci through Enhancing Methionine Biosynthesis

  • Oral streptococci are considered as an opportunistic pathogen associated with initiation and progression of various oral diseases. However, since the currently-available treatments often accompany adverse effects, alternative strategy is demanded to control streptococci. In the current study, we investigated whether short-chain fatty acids (SCFAs), including sodium acetate (NaA), sodium propionate (NaP), and sodium butyrate (NaB), can inhibit the growth of oral streptococci. Among the tested SCFAs, NaP most potently inhibited the growth of laboratory and clinically isolated strains of Streptococcus gordonii under anaerobic culture conditions. However, the growth inhibitory effect of NaP on six different species of other oral streptococci was different depending on their culture conditions. Metabolic changes such as alteration of methionine biosynthesis can affect bacterial growth. Indeed, NaP enhanced intracellular methionine levels of oral streptococci as well as the mRNA expression level of methionine biosynthesis-related genes. Collectively, these results suggest that NaP has an inhibitory effect on the growth of oral streptococci, which might be due to alteration of methionine biosynthesis. Thus, NaP can be used an effective bacteriostatic agent for the prevention of oral infectious diseases caused by oral streptococci.

영양소의 증가가 산란 생산성 및 계란 품질에 미치는 영향

  • 갈색 산란계의 생산특성 및 계란품질 변화를 구명하고자 780수를 공시하여 25∼70주령까지 관행사료와 10 % 증가 사료를 급여하였다. 산란율은 처리간 차이가 없었으나 평균난중은 10 % 증가 사료가 유의적으로 무겁게 나타났다(P<0.05). 사료요구율 역시 10 % 증가 사료가 개선되는 것으로 나타났다(P<0.05). 산란율은 52주령 이후 급격히 감소하였으며 난중은 52주령까지 증가하다가 이후 비슷한 양상을 보였다. 난각강도와 후도는 60주령 근처에서 품질이 약화되는 것으로 나타났으며, Haugh unit는 56주령부터 크게 감소하는 것으로 나타났다. 이상의 결과에서 영양소 함량의 증가는 난중과 사료요구율을 개선시키는 것으로 나타났으며, 산란기 영양소는 52∼56주령 사이에 차별화해야 할 것으로 판단된다.

  • The purpose of this research is that infant artificial feeding products is used in clonic with the study on characteristic, ingredients and indication of geneal and special modified milks. The result is as follows. 1. The main ingredients of four company products-Maeil , Namyang, Pasteur, Aebout is similar but the functional is different 2. General infant formula is divided into 100days, 5-6months, 12months, 24months and 36months out of consideration for growth and development of infant. 3. The indication and sorts of the special infant formula used at a hospital is as follows. PKU-1, PKU-2 formula is available for phenylketonuria. MPA formula is available for propionic acidemia and methylmalonic acidomia. UCD is available for urea cycle disorder Leucine-free formula is available for isovaleric acidemia. Methionine-free formula is available for homocystinuria and hypermethioninemia. Premature infant is available for premature and low birth weight. 4. The special infant formula published in nation is as follows. Maeil soy A, Maeil MF1, Namyang hope doctor and Maeil HA is available for diarrhea. Maeil HA, Maeil HA-21 and Namyang hope allergy is available for hypoallergy. Maeil soy A is available for diarrhea of milk allergy. Maeil MF1 or Namyang hope doctor is available for acute bacterial or viral temporal diarrhea. Maeil HA is available for allergic chronic diarrhea. Maeil HA and Namyang hope allergy as eHP-formula is available for chronic diarrhea for lactose intolerance and milk allergy. Maeil-21 as pHP-formula for neonates with allergy family, allergic symptoms such as atopic dermatitis, asthma except digestive system.

Effect of Enhancers on in vitro and in vivo Skin Permeation and Deposition of S-Methyl-ʟ-Methionine

  • S-methyl-$\small{L}$-methionine (SMM), also known as vitamin U, is commercially available as skin care cosmetic products for its wound healing and photoprotective effects. However, the low skin permeation expected of SMM due to its hydrophilic nature with a log P value of -3.3, has not been thoroughly addressed. The purpose of this study thus was to evaluate the effect of skin permeation enhancers on the skin permeation/deposition of SMM. Among the enhancers tested for the in vitro skin permeation and deposition of SMM, oleic acid showed the most significant enhancing effect. Moreover, the combination of oleic acid and ethanol further enhanced in vitro permeation and deposition of SMM through hairless mouse skin. Furthermore, the combination of oleic acid and ethanol significantly increased the in vivo deposition of SMM in the epidermis/dermis for 12 hr, which was high enough to exert a therapeutic effect. Therefore, based on the in vitro and in vivo studies, the combination of oleic acid and ethanol was shown to be effective in improving the topical skin delivery of SMM, which may be applied in the cosmetic production process for SMM.

검정동부 종피에서의 색소 추출과 추출색소의 저장 안정성 연구 (Research on the Colorants Extraction from Black Cowpea Seed Coats and their Storage Stability)

  • 본 연구에서는 검정 동부 종피에 함유된 천연색소에 대한 연구의 일환으로, 추출방법에 따른 색소 추출 효율과 저장안정제 첨가에 따른 저장 안정성과 특성을 검토하여 다음과 같은 결과를 얻었다. 1) 용액 추출 시 온도가 높을수록, 시간이 경과될수록 색소 추출 효율이 높게 나타났으며, pH 조건에서는 pH 4가 가장 효율적이었다. 2) 착액 추출 시, 단시간 전처리에서는 pH 4가 pH 5에서보다 색소 추출량이 많았지만, 15시간 이상의 장시간 전처리에서는 오히려 pH 5에서 많게 나타났다. 3) 검정동부 색소용액의 pH 변화에 따른 색도는, orange-red색(pH 3.0), orange색(pH 4.0~6.0), blue색(pH 7.0) 그리고 greenish-blue색(pH 9.0~11.0)으로 Antocyanin과 유사한 거동을 보였다. 4) 저장 안정제로 Methionine을 첨가한 결과에서는 첨가에 따른 검정 동부 색소의 안정화 효과에서 저장 기간 18일까지는 변색 지연 효과를 보였으며, Methionine 10 mmol 첨가>20 mmol 첨가>무첨가 순으로 안정효과가 크게 나타났다. 5) 검정 동부 종피로부터 착액 추출한 색소 분말을 paper chromatography한 결과, 하나의 긴 띠 모양으로 전개되었으며 Rf 값은 각각 0.68(BAW), 0.53(30% acetic acid)으로 나타났다. 6) Chromatograph paper상에서 전개속도에 따라 3분획으로 구분하여 취한 색소의 spectrum 분석에서는 552 nm의 shoulder의 존재 유무를 근거로 적어도 2가지 이상의 색소 성분이 혼합되어 있는 것을 명확히 알 수 있었다.
