Physiological Effects on Dried Skin in Rat by Water & Ethanol Extracts of Auricularia Auricula U. (흰쥐 건성피부에 대한 목이버섯의 물추출물과 에탄올추출물의 생리적 효능)

  • It has been well known that mushroom polysaccharide extracts have superoxide and hydroxyl radical scavenging activities. So we studied the efficacy and anti-oxidative effect of mushroom(Auricularia auricula U.) in rats treated with water and ethanol extracts of Auricularia auricula U after inducing dryness of skin by the application of detergent. The results of the research for of the efficacy and anti-oxidative effect Auricularia auricula U. are as follows thats. Weight gain and feed intake in animals treated with water extracts and ethanol extracts are significantly higher(p<0.05) than animal treated with detergent. Animals treated with water extracts of Auricularia auricula U. compared to animals treated with ethanol extracts have the higher HDL-cholesterol levels in the blood and the lower LDL-cholesterol levels in the blood. SOD-like activity of the ethanol extracts of Auricularia auricula U. was $27.39{\pm}1.61%$ at 2000ppm.

Inhibition Effects of Auricularia auricula-judae Methanol Extract on Lipid Peroxidation and Liver Damage in Benzo(a)pyrene-Treated Mice (목이버섯 메탄올 추출물이 벤조피렌(B(a)P) 투여한 마우스의 지질과산화 및 간 손상 억제에 미치는 영향)

  • This study was undertaken to investigate the inhibition effects of Auricularia auricula-judae methanol extract in edible mushroom on lipid peroxidation and liver damage in benzo(a)pyrene(B(a)P)-treated mice. The activities of serum aminotransferase, cytochrome P-450, superoxide dismutase, catalase, glutathione peroxidase and the hepatic content of lipid peroxide after B(a)P-treatment was markedly increased than control but those levels were significantly decreased by the treatment of Auricularia auricula-judae methanol extract. Glutathione S-transferase activity and the hepatic glutathione content were decreased by B(a)P-treatment than control, but those were also inhibited by the treament of Auricularia auricula-judae methanol extract. These results suggest that Auricularia auricula-judae methanol extract have a protective effect on liver damage by B(a)P.

Comparative study on nutritional contents of Auricularia spp. (목이버섯 품종간 영양성분 비교)

  • We conducted an analysis by comparing the nutritional contents of Auricularia auricula-judae(black), Auricularia polytricha and Auricularia auricula-judae (brown). In nutritional contents of three strains of Auricularia spp., four free sugars, seven organic acids and 24 amino acids were detected. Auricularia auricula-judae (black) was highly contained free sugar, organic acid and amino acid. There was the most prevalent Vitamin $D_2$ content in Auricularia auricula-judae (black) of Auricularia spp. Dietary fiber of three strains showed contents of about 60% but were not significantly different. ${\beta}$-glucan contents of Auricularia auricula-judae (brown) contained the highest contents with $25.21{\pm}0.37%$ and showed significant differences between Auricularia polytricha and Auricularia auricula-judae (black). Total polyphenol contents of Auricularia polytricha showed the highest contents, followed by Auricularia auricula-judae (brown) and Auricularia auricula-judae (black).

  • The present study investigated the anti-inflammatory effects of dichloromethane extract of Auricularia auricula-judae. Dichloromethane extract of Auricularia auricula-judae inhibited Lipopolysaccharide (LPS)-induced nitric oxide (NO) production significantly in a dose-dependent manner in the concentration ${\geq}\;10\;{\mu}g/ml$ (p < 0.05). Furthermore, RT-PCR results of this study indicated that the extract markedly reduced the expressions of inflammatory cytokines (IL-6, TNF-$\alpha$ and IL-$1{\beta}$) mRNA in LPS-treated murine RAW 264.7 macrophages, which could possibly ameliorate the inflammation. Nevertheless, dichloromethane extract of Auricularia auricula-judae did not show complete inhibition of IL-6 mRNA expression. The inhibition of IL-$1{\beta}$ cytokine at protein level was also observed in a dose dependent manner. In conclusion, the current study revealed the previously unknown effect of dichloromethane ethyl extract of Auricularia auricula-judae inhibitions of the production of NO, IL-6, TNF-$\alpha$ and IL-$1{\beta}$ in LPS-stimulated macrophages.

Genetic Clarification of Auricularia heimuer Strains Bred and Cultivated in Korea Using the ITS and IGS1 rDNA Region Sequences

  • Auricularia is one of the broadly cultivated edible mushrooms in Korea. Most of the Korean Auricularia strains used for cultivation and breeding are known as A. auricula-judae. Recently, this species has been reported to belong to a species complex. Therefore, this study was carried out to genetically clarify the bred and cultivated Korean A. auricula-judae strains. The internal transcribed spacer (ITS) and IGS1 rDNA region sequences were determined from 10 A. auricula-judae strains by PCR and sequencing. Variation in the nucleotide sequence and sequence length of the two rDNA regions were found among the seven A. auricula-judae strains. A maximum-likelihood (ML) phylogenetic tree based on the ITS sequences clearly placed all the 10 Korean A. auricula-judae strains in the A. heimuer clade of the A. auriculajudae complex. A. heimuer is diverged from A. auricula-judae. An ML phylogenetic tree based on the IGS1 sequences revealed the close relationship between Korean A. heimuer strains to Chinese A. heimuer strains. But each strain could be distinguishable by the IGS1 sequence. Furthermore, progeny strains in the seven Korean strains could be differentiated from their parental strains by the IGS1 sequence based phylogenetic tree. Our results are expected to be used to complement the distinction of domestic Auricularia cultivars.

Antidiabetic Effect of Auricularia auricula Mycelia in Streptozotocin-induced Diabetic Rats

  • Auricularia auricula (Heterobasidiae) called 'tree ear' or 'ear mushroom' was investigated to know its antidiabetic effect. Administration of dried powder of Auricularia auricula mycelia (AAM) (0.5 g/kg bw and 1.0 g/kg bw) caused a statistically significant reduction of plasma glucose (35% and 39%, respectively), total cholesterol (18% and 22%, respectively), triglyceride (12% and 13%, respectively), GOT (53% and 56%, respectively) GPT (26% and 37%, respectively), levels and the rate of liver weight against final total body weight (6% and 7%, respectively) compared with diabetic control group. These results support the antidiabetic effect of AAM.

Antimutagenic Effects of Methyl Alcohol Extracts from Auricularia auricula and Gyrophora esculenta (목이 및 석이 메틸 알콜 추출물의 항돌연변이원성)

  • This study was conducted to investigate the antimutagenic effects of methyl alcohol extracts from Auricularia auricula and Gyrophora esculenta on the SOS response induced by 4-nitroquinoline-1-oxide (4NQO), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), mitomycin C (MMC) and 3-amino-1,4- dimethyl-5H-pyrido-[4,3-b]indol (Trp-P-1) in E. coli PQ37/plasmid pKM101. In the mutagenic test on test strain, both methyl alcohol extracts did not show mutagenic activity. In the antimutagenic test, each sample strongly inhibited the mutagenecity induced by 4NQO, MNNG, MMC and Trp-P-1. Methyl alcohol extracts from Auricularia auricula and Gyrophora esculenta showed inhibitory effects of 52% and 59% against 4NQO, 49% and 58% against MNNG, 53% and 64% against MMC, and 61% and 64% against Trp-P-1, respectively. Gyrophora esculenta extracts on the antimutagenicity showed relatively higher inhibitory effects than that of Auricularia auricula.

Fruit-body Production of Auricularia auricula-judae by Sawdust Cultivation (톱밥재배에 의한 목이의 자실체 생산)

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. Present experiments were conducted to determine the possibility of artificial cultivation with oak sawdust of A. auricula-judae. The duration of mycelial growth and days of pinhead formation on oak sawdust bag (1.4 kg) were 29~32 days and 13~17 days, respectively. The yield of fresh fruit-body was 275~350 g.

  • A dark brownish pigment from fruit body of Auricularia auricula was isolated and characterized in this report. The pigment was obtained with a yield of 0.61%(w/w) by alkaline extraction and subsequent purification steps. It showed the positive $FeCl_3$ test which was the indication of phenolic compounds. A synthetic melanin showed a similar spectrometric characteristics to the pigment extract regarding a characteristic UV absorption between 200-250 nm and infrared absorptions profiles in the finger print region including absorption peaks at 1701 and 1624 $cm^{-1}$. Its element analysis indicated that its atomic copmposition is close to that of DOPA melanin (eumelanin). With the result of its antioxidant activity in the TNBT (5-thio-2-nitrobenzoic acid) assay, we concluded that the dark brownish pigment from A. auricula is a melanin-like compound having a powerful antioxidative activity.

The culture conditions for the mycelial growth of Auricularia auricula-judae

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. This study was carried out to obtain the basic information for mycelial culture conditions of Auricularia auriculajudae. According to colony diameter and mycelial density, the media for suitable mycelial growth were PDA and MCM. The optimum temperature for mycelial growth was $25{\sim}30^{\circ}C$. Carbon and nitrogen sources were mannose and malt extract, respectively. The optimum C/N ratio was in the range of 10 to 1 with 2% glucose. Other minor components for the optimal growth were thiamine-HCl and biotin as vitamins, succinic acid and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $KH_2PO_4$ as mineral salts.
