Anti-Obesity Effects of Mixture of Atractylodes macrocephala and Amomum villosum Extracts (사인과 백출 추출 혼합물의 항비만 효과)

  • In this study, we investigated the anti-obesity effects of various mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts on high-fat diet (HFD) induced mouse model. We classified five groups as follows; control, HFD, HFD + AM extracts : AV extracts (100mg/kg) (1:1), HFD + AM extracts : AV extracts (100mg/kg) (2:1), HFD + AM extracts : AV extracts (100mg/kg) (3:1). Oral administration of various mixtures of AM and AV extracts for 6 weeks inhibited HFD-induced increases of body, liver and epididymal fat weights. Also, lipid profiles including LDL cholesterol were improved by various mixtures of AM and AV extracts treatment compared with HFD-fed group. Lipogenesis-related genes such as acetyl coA carboxylase (ACC) and fatty acid synthase (FAS) in liver changed in a favorable way for lipid biosynthesis by HFD compared to control, but various mixtures of AM and AV extracts-treated groups did not. Our results show that various mixtures of AM and AV extracts can prevent HFD-induced obesity in mice and suggests that the mechanisms are involved in expressions and modifications of lipogenesis-related genes such as ACC and FAS in liver.

Protective Effect of Atractylodes macrocephala and Taraxacum spp. Combination Treatment in Balb/c Mice with Dextran Sulfate Sodium-Induced Ulcerative Colitis

  • Objectives : This study aimed to investigate the protective effects of an herbal mixture of Atractylodes macrocephala and Taraxacum spp. (ATC) on ulcerative colitis. We have previously screened traditional medicinal herbs to discover the effective candidate by the animal model. A. macrocephala and T. spp were identified as one of the effective herbs in the screening process. Methods : Experimental colitis was induced in male Balb/c mice by administering drinking water containing dextran sulfate sodium, which mimics the clinical and histological features of ulcerative colitis in human. ATC at doses of 30, 100 or 300 mg/kg were orally administered to mice twice per day for 10 consecutive days. To evaluate the damage from experimentla ulcerative colitis, body weight, colon length, disease activity index, myeloperoxidase and histological changes were measured and analyzed. Results : The administration of dextran sulfate sodium with drinking water resulted in markedly reduced colon length, severe body weight loss, increased levels of myeloperoxidase activity and histological damages in mice. ATC treatment significantly ameliorated the colon shortening, histological damage, body weight loss and disease activity index score in a dose-dependent manner. ATC also attenuated the colonic myeloperoxidase activity which reflects the severity and extent of inflammatory damage of colon. Conclusions : ATC exerts protective effects against inflammatory colonic structural damage induced by epithelial barrier integrity impairment. ATC also inhibits weight loss and related symptoms of UC which can be considered as the functional recovery of colon.

Effects of Young Sprouts Cutting Times and Nitrogen Split Application on Growth and Yield of Atractylodes japonica KOIDZ (삽주의 어린순 채취(採取)회수 및 질소분시(窒素分施)가 생육(生育)과 수양(收量)에 미치는 영향)

  • This Experiment was conducted to know effect of cutting times and nitrogen split application rates on shoot and root-related characters of Atractylodes japonica. The cutting time was forced on none, once and twice along with three levels of nitrogen split application, all basic 50 -30 -20% and 40 -30 -30%. The plant height declined with increased cutting time and dry matter weight was increased with nitrogen split application under none and once cutting. Average yield of cutted shoot as edible wild plant was 257kg/10a at once cutting condition, but twice cutting condition was little and unfavorable to use edible wild plant because shoot was coarse. Yield 0 dry rhizome was the highest at once shoot cutting und­er 50 -30 -20% nitrogen split application by 471kg/10a.Especially, culture of Atractylodes japonka needed once shoot cutting one the 10th-30th of May.

Effects of herbal acupuncture (Atratylodes japonica Koizumi and Coix lachrymajobi var. mayeun Stapf) at Pungnyung(ST40) and Umnungchon(SP9) in obese Rats induced by high fat diet (풍륭(風隆)·음릉천(陰陵泉)에 대한 약침(藥鍼)(창출(蒼朮), 의이인(薏苡仁))이 고지방식이(高脂肪食餌)로 유발(誘發)된 비만백서(肥滿白鼠)에 미치는 영향(影響))

  • Objectives : This research was performed to investigate the effect of herbal acupuncture (Atratylodes japonica Koizumi and Coix lachrymajobi var. mayeun Stapf) at Pungnyung(ST40) and Umnungchon(SP9) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into normal group(Sham), high fat diet group(Control), high fat diet and Atractylodes japonica-herbal acupuncture in ST40 SP9 treated group (AJ), high fat diet and Coix lachrymajobi var. mayeun Stapf-herbal acupuncture in ST40 SP9 treated group(CL), high fat diet and Atractylodes japonica+Coix lachrymajobi var. mayeun Stapf-herbal acupuncture in ST40 SP9 treated group(AJ+CL). Herbal acupuncture was bilaterally treated at the level of 132.5mg/kg body weight per day. Results : Body weight and food efficiency were decreased in AJ, AJ+CL. Feed intake was decreased in AJ, CL. The level of serum HDL-cholesterol was increased in AJ, CL. The level of serum total cholesterol, LDL-cholesterol and phospholipid were not significant. The level of serum triglyceride, free fatty acid were increased in AJ. The level of serum AST was decreased in AJ, AJ+CL. The level of serum ALT was decreased in AJ, CL, AJ+CL. The level of serum ALP was decreased in AJ+CL. The HDL to Total cholesterol ratio was increased in AJ. Conclusions : Atractylodes japonica-herbal acupuncture in ST40 SP9 can control the body weight, feed intake and feed efficiency ratio, HDL-cholesterol and herbal acupuncture combined Atractylodes japonica with Coix lachrymajobi var. mayeun Stapf is effective on Body weight, feed intake, feed efficiency ratio, protection of liver function and prevention cardiovascular risk by obesity.

Comparison of Anti-oxidative and Anti-inflammatory Effect of Atractylodes Interspecific Hybrid Cultivar Roots (삽주 육성품종 뿌리의 항산화 및 항염증 효과 비교)

  • Background: Atractylodes radix is a well-known medicinal crop having many physiological effects. This study was conducted to select useful Atractylodes japonica ${\times}$ Atractylodes macrocephala (AJM) cultivars by comparing anti-oxidative and anti-inflammatory efficacies. Methods and Results: Seven extracts from AJM cultivars were used to treat lipopolysacchride (LPS)-treated BV2 cells, and the effects on cell viability and inhibition on reactive oxygen species (ROS) and nitric oxide (NO) production were analyzed. In vitro scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and peroxynitrite ($NOO^-$) radicals were also investigated. Contents of total phenol, atractylenolide I, and atractylenolide III in the AJM extracts were measured using high performance liquid chromatography (HPLC) or spectrophotometry. The experiments show that none of the seven extracts was cytotoxic above 89.2% at $20-250{\mu}g/m{\ell}$. Extracts of Gowon, Dawon, Sangchul, and Huchul inhibited ROS generation in a dose-dependent manner, and Sangchul extract showed the highest inhibition on ROS production. All the AJM extracts showed effective inhibitory activity after on NO release in the LPS-treated BV2 cells, and Sangchul extract showed the highest activity. Sangchul extract had the most potent scavenging activities for $NOO^-$ and had some DPPH radical scavenging effect. Sangchul extract also had the highest content at total phenol and atractylenolide I content. Atractylenolide III was not detected in the AJM extracts. Conclusions: The results suggested that Sangchul was the most useful anti-oxidative and anti-inflammatory resource among the AJM cultivars.

Bone Marrow Cell Proliferation Activity through Intestinal Immune System by the Components of Atractylodes lancea DC. (창출 성분의 장관면역 자극을 통한 골수세포 증식활성)

  • Of hot-water extracts prepared from 10 herbal components of Sip-Jeon-Dae-Bo-Tang, Atractylodes lancea DC. (ALR) and Panax ginseng C.A. Meyer (PG) showed the most potent bone marrow cell proliferation activity through intestinal immune system whereas other extracts did not have the activity except for Astragalus membranacues Bunge (ASR) and Angelica acutiloba Kitagawa (AR) having low activity. Especially, ALR had the potent activity irrespective of classes of ALR, a place of production and the condition of breeding. In addition, we found that hot-water extract from Atractylodes lancea DC rhizomes (ALR-0) contributed mainly to Peyer's patch cells mediated-hematopoietic response of Sip-Jeon-Dae-Bo-Tang. ALR-0 was further fractionated into MeOH-soluble fraction (ALR-1), MeOH-insoluble and EtOH-soluble fraction (ALR-2), and the crude polysaccharide fraction (ALR-3). Among these fractions, only ALR-3 showed potent stimulating activity for proliferation of bone marrow cells mediated by Peyer's patch cells, dose-dependently. In treatments of ALR-3 with $NaIO_4,\;NaClO_2$ and pronase, all significantly reduced the intestinal immune system modulating activity of ALR-3, and the activity of ALR-3 was much affected by $NaIO_4$ oxidation particularly. These results reveal that macromolecules, such as polysaccharide, rather than low-molecular-weight substances, are the potent intestinal immune system modulating compound of ALR.

Development of LC-MS/MS analytical methods for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai (LC-MS/MS를 이용한 생약 백출 및 우슬 중 Metalaxyl 잔류분석법 개발)

  • A new rapid and simple method for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai has been developed and validated. This study was conducted to develop a method for analyzing metalaxyl by a method based on QuEChERS using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Samples were extracted with acetonitrile and purified using amino-propyl (NH2) Solid Phase Extraction cartridge. The method limit of quantitation (MLOQ) was 0.01 mg/kg. The linearity of matrix-matched calibration curve (r2) was ≥0.99 at the calibration range of 0.001-0.05 mg/kg. For recovery test, Atractylodes macrocephala Koidzumi or Achyranthes japonica Nakai was treated with standard solutions at MLOQ and 10MLOQ levels. Recovery rates were in the range of 88.1-109.1% with <5.5% coefficient of variation. This established analytical method was fully validated. Based on these results, it can contribute to improving the safety of residual pesticides in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai.

Inhibitory effects of ethanol extract of Atractylodis Rhizoma on melanogenesis in B16/F10 melanoma cells

  • The inhibitory effect of Atractylodis Rhizoma alba extract(AM) on melanogenesis was studied by using B16/F10 melanoma in culture. Cells were cultured in the presence of various concentrations of AM for 48 hrs, and the experiment of total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. AM significantly inhibited tyrosinase activity, and melanin content in a dose-dependent manner. These results show that Atractylodes macrocephala extract could be developed as skin whitening components of cosmetics.
