This study was conducted to identify transition characteristics of mercury in several selected medicinal plants and to find the appropriate management for production of safety food. Cultivated soils and medicinal plants were collected at 29 sites for Angelica gigas (Korean angelica root), 68 sites for Platycodon grandiflorum (Balloon flower), 35 sites for codonopsis lanceolata (Deoduck), 36 sites for Dioscorea batatas (Chinese yam), 32 sites for Rehmannia glutinosa (Foxglove), 16 sites for Cnidium officinale makino (cnidium), and 26 sites for Astragalus membranaceus (milk vetch root) during the harvest season of 2013. Mercury in the soils and medicinal roots were analyzed with a Direct Mercury Analyzer. Average content of mercury in soils cultivated medicinal plants was $0.023mg\;kg^{-1}$ (range: from 0.003 to $0.074mg\;kg^{-1}$) and average content of mercury in medicinal plants was $0.003mg\;kg^{-1}$ (range: from 0.001 to $0.011mg\;kg^{-1}$), indicating that mercury in the surveyed soils and medicinal plants were not exceeded the Korean regulation.
약용작물(藥用作物)중 재배 면적이 많고 소득성(所得性)이 높은 다년생(多年生)의 초본성 작물인 황기의 $1{\sim}2$년생근(年生根) 재배시 헤어리베치 녹비와 액상분뇨 시용이 황기의 생육특주(生育特注) 및 생산성(生産性)과 토양(土壤) 특성에 미치는 영향을 검토한 시험결과를 요약하면 아래와 같다. 1. 지상부 생육 중 경장(莖長)과 분지수(分枝數)는 헤어리베치 녹비, 액상분뇨 시용구에서 생장량이 컸으며, 생육이 늦게까지 지속되었다. 근장(根長), 근경(根徑), 지근수(支根數) 모두 헤어리베치 녹비와 액상분뇨 시용구에서 증가하는 경향이었으며 건물비율(乾物比率) 재배 1년차 보다 재배 2년차에 증가하였다. 2. 재배 2년차의 건근수양(乾根收量)은 녹비를 피복한 무비구, 화학비료, 액상분뇨 시용구에서 각각 196, 240, 255kg/10a으로서 녹비를 피복하지 않는 무비구, 화학비료, 액상분뇨 시용구에서는 각각 115, 225, 232 kg/10a 보다 증수되었다. 황기 재배시 헤어리베치 녹비 피복은 질소공급에 의한 수량증가와 잡초억제 효과를 나타내었다. 3.뿌리의 질소함양(窒素含量)은 재배 1년차에 $1.33{\sim}2.56%$, 재배 2년차에 $1.9{\sim}2.95%$으로 재배 년차가 높을수록 증가(增加)되었다. 질소함량을 조단백질 함량으로 환산하면 재배 2년차에 $20{\sim}23%$로서 콩 단백질 함량의 $45{\sim}65%$ 수준이었다. 4. 시험 후 토양의 총 질소함량은 녹비 액상분뇨 시용구에서 유의성 있게 증가하였으나 토양 내 염류집적(鹽類集積)은 되지 않았다.
In this study, we prepared CheonJeongKiBo-Dan(7 oriental medicinal plants, 7OMP: Astragalus Membranaceus root, Panax Ginseng root, Glycyrrhiza Glabra (licorice) root, Schizandra Chinensis fruit, Polygonatum Odoratum, Rehmannia Glutinosa root, Paeonia Albiflora root) by extracting them in one reactor and studied its efficacies on skin. UV irradiation has been suggested as a major cause of photoaging in skin. In order to investigate protective effects against UV-B induced cellular damage, 7OMP was extracted with 70% ethanol and dissolved in DMSO. The protective effect was detected by MTT assay, reactive oxygen species (ROS) generation, phosphorylation of ATR and p53 in human dermal fibroblast cell system after UV-B irradiation. 7OMP reduced UV-B-induced cellular damage in HDFs cells, and inhibited ROS generation. UV-B-induced toxicity accompanying ROS production and the resultant DNA damage are responsible for activation of ATR, p53 and Bad. In this study, 7OMP hampered phosphorylations of ATR and p53 in human dermal fibroblasts. Therefore, 7OMP may be protective against UV-induced skin photoaging.
The root of Astragalus membranaceus Bunge (Leguminosae), which has been used for the treatment of hypertension, chronic hepatitis, duodenal ulcers, chronic nephritis and promotion of immunity in folk remedies. Cyclooxygenase (COX-2) is responsible for the production of large amounts of proinflammatory prostaglandins (PGs) at the inflammatory site. Thus, a logical approach to the treatment of inflammatory disease should involve the inhibitors of COX-2. To develop new COX-2 inhibitors from natural products, Astragali Radix was screened by inhibiting prostaglandin $E_2(PGE_2)$ generation in the culture medium using enzyme immunometric assay. Two isoflavone glycosides, $7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-{\beta}-D-glucoside$ and $calycosin-7-O-{\beta}-D-glucoside$ isolated from Astragali Radix inhibited COX-2 activity.
국내에 자생하는 당귀, 삽주, 쇠무릎, 지모, 황기의 근권토양내 EPS 생성균주의 분포율을 조사한 결과 당귀로부터 분리된 균주의 56%가 EPS 생성 균주로 가장 높은 분포율을 나타내었다. 또한, 당귀 근계 (근권, 근면, 근 내부) 내 EPS 생성 세균의 밀도를 측정한 결과, 근권 토양 내에는 $9.0{\times}10^6$ CFU/$g{\cdot}soil$, 근면에는 $7.0{\times}10^6$ CFU/$g{\cdot}soil$, 그리고, 근 내부에는 $1.4{\times}10^3$ CFU/$g{\cdot}soil$로 확인되어, 다수의 EPS 생성 세균이 분포하고 있음이 확인되었다. 당귀 근권으로부터 분리된 EPS 생성세균은 Alphaproteobacteria (4 strains), Betaproteobacteria (6 strains), Firmicutes (2 strains), Actinobacteria (3 strains), 그리고 Bacteroidetes (1 strain) 계통군에 속하는 균주였다. 근면으로 부터 분리된 EPS 생성세균은 Alphaproteobacteria (7 strains), Betaproteobacteria (3 strains), Actinobacteria (2 strains), Bacteroidetes (3 strains), 그리고 Acidobacteria (1 strain) 계통군으로 나타났으며, 근 내부에서 분리된 EPS 생성세균은 모두 Bacteroidetes 계통군 Chitinophaga에 속하는 특징을 나타내었다. 약초 근권토양으로부터 분리된 EPS 생성세균 112균주중에서 Burkholderia caribiensis DR14 (1,547 mpa.s), Terriglobus sp. DRP35균주(2,136 mpa.s), Rhizobium hainanense SAP110균주(1,680 mpa.s)를 최우수 EPS 생성 균주로 선발하였다. 분리 정제된 EPS를 Bio-LC로 분석한 결과 glucose, galactose, mannose의 중성당과, galactosamine, glucosamine의 아미노당이 나타났다. 특히 Rhizobium hainanense SAP110 균주는 주요 중성당으로 glucose (60-89%)를 그리고 주요 아미노당으로 glucosamine (8.5%)을 생성하는 특징을 나타내었다.
본 연구에서는 식약청 고시 원료인 레티놀, 레티닐팔미테이트, 아데노신, 폴리에톡실레이티드레틴아마이드 4종류와 항 노화의 효능을 가진 천연 원료인 금송, 하고초, 맨드라미, 브라질린, 여뀌, 황기, 연교, 자근, 대황, 육종용에 대한 연구 동향을 살펴보았다. 현재까지의 항 노화 연구는 주로 콜라겐 생성과 엘라스타아제 합성 억제 기전을 중심으로 이루어지고 있는 것이 확인되었다. 하지만, 피부의 노화 과정은 여러 가지 원인에 의해서 발생하는 것으로 볼 때, 다양한 기전에 맞춰 피부 항노화에 도움을 줄 수 있는 안전하고 효과적인 천연 원료를 활용한 항노화 연구가 지속적으로 이루어져야할 것으로 사료되어 진다.
Objectives : It was investigated new herbal prescription HT005 supported by traditional Korean medicine has a activity on the longitudinal bone growth of rats. HT005 were consisted of the root of Angelica sinensis, the fruit of Alpinia oxyphylla, the root of Astragalus membranaceus, the stem of Eleutherococcus senticosus, and the root of Dioscorea japonica, and Poria cocos. Method : To investigate the effect on longitudinal bone growth in adolescent male Sprague-Dawley rats, the longitudinal length of growth plate was measured directly by the tetracycline fluorescent labeling method and chondrocyte staining method. HT005 administered orally for four days, and the tetracycline was injected twice on the growth plate of the animals for fluorescent dying. The rate of longitudinal bone growth was measured the length between the tetracycline bands which fixed on the 3rd day and 5th day after injection. Cresyl violet was also used to stain the chondrocytes in the growth plate. The length of growth plate after administration was compared. Expression of IGF-1 and BMP-2 in the growth plate was investigated by immunohistochemistry. Results : HT005 showed a significant longitudinal bone growth which was $301.0{\pm}6.1\;{\mu}m/day$ at the dose of 100 mg/kg and $283.8{\pm}1.25\;{\mu}m/day$ (p < 0.001)at the dose of 10 mg/kg of HT005, compared to control group by the tetracycline fluorescent labeling method. HT005 showed a significant chondrocyte length on growth plate which was measured $797.19{\pm}3.31\;{\mu}m$ (p < 0.001) at the dose of 100 mg/kg and $720.14{\pm}2.19$ (p < 0.001) ${\mu}m$ at the dose of 10 mg/kg compared to the control group by cresyl-violet staining method. Both the number and intensity of BMP-2 and IGF-1 positive cells were increased in the hypertrophic zone of growth plate. There was a significant correlation between BMP-2 and IGF-1 expression and heights of chondrocytes in growth plate. Conclusions : Treatment of HT005 on Sprague-Dawley rats markedly increased the longitudinal bone growth. Therefore, HT005 may be an alternative herbal source to growth hormone as it promotes bone growth in children afflicted by growth retardation.
Inhibition of inflammatory response, acceleration of basal cell growth, and balanced synthesis of extracellular matrix (ECM) are important in healing of cutaneous open wounds. In order to evaluate the healing effects of water extracts of Radix Astragali (the root of Astragalus membranaceus (Fisch.)) on open wound at early stage, the experimental open wounds were generated on the dorsal sides of SD rats under anesthesia. The boiled-water extracts of Radix Astragali $(100{\mu}l)$, soaked into an occlusive film dressing were applied once a day for eleven consecutive days. The healing process was assessed by measuring macroscopic appearance and wound areas of the open wounds. The molecular aspects of healing process by Radix Astragali extracts were also investigated by Hematoxylin-Eosin (H-E) double staining and immunohistological staining of collagen type I in the healed skin area, implying cell density and linear alignment of the granulation tissue, and ECM synthesis and its remodeling, respectively. The Astragali radix extracts were found to significantly accelerate the cutaneous wound healing by suppressing the inflammation and stimulating the basal cell growth in wounded area, as compared to epidermal growth factor (EGF).
The safety of plant species used as a source for herbal medicines and dietary supplements has recently been questioned due to poisonings associated with the presence of cadmium (Cd) in these plants. These plants can derive Cd from their presence in the soil. Organic matter (OM) concentrations in soils could affect the availability of Cd for plants. To determine the effect of OM concentration in soil on the concentration of plant available Cd and uptake of this toxic element by medicinal plants, soil and plant samples were collected from 102 fields supporting for 5 species of medicinal plants in 6 province of South Korea. Concentrations of OM and dissolved organic carbon (DOC) in soils affected the phytoavailability of Cd. One M $NH_4OAc$ extractable Cd concentration in soil increased with increasing OM concentrations. There were significantly positive relationships between 1 M $NH_4OAc$ extractable Cd concentration and OM concentration in soil and between 1 M $NH_4OAc$ extractable Cd concentration and DOC concentration. Likewise, OM and DOC concentrations significantly affected Cd concentration in medicinal plant soils. Cadmium concentration in medicinal plants increased with increasing OM concentration in soil [Cd concentration $(mg\;kg^{-1})= 0.179+1.424{\times}10^{-3}$ OM concentrations, $R^2=0.042*$] and with DOC concentration [Cd concentration $(mg\;kg^{-1})= 0.150+5.870{\times}10^{-4}$ DOC concentrations, $R^2=0.124***$]. These results might result from Cd-DOC complex which is easily absorbed Cd form by plant root. Dissolved organic carbon concentration had more positive relationship with Cd concentration in medicinal plants and 1 M $NH_4OAc$ extractable Cd concentration in soils than OM. Cadmium concentration in all 5 species of medicinal plant (Atractylodes macrocephala Koidzumi, Astragalus membranaceus, Codonopsis lanceolata, Platycodon grandiflorum, and Rehmannia glutinosa) significantly increased with increasing DOC concentration in soil. From the above results, formation of Cd-DOC complex caused by OM application might be mainly attributed to increase in Cd concentration in medicinal plants.
