Phellinus linteus Extract Exerts Anti-asthmatic Effects by Suppressing NF-${\kappa}B$ and p38 MAPK Activity in an OVA-induced Mouse Model of Asthma

  • Phellinus linteus has been used as a traditional herbal medicine in Asian countries and is known to have anti-tumor, immunomodulatory, anti-inflammatory, and anti-allergic activities. However, the protective effects of P. linteus against experimental asthma have not been fully investigated. The objective of this study was to determine whether P. linteus ethanol extract (PLE) suppresses inflammatory response in an OVA-induced asthma model. As expected, the oral administration of PLE significantly inhibited eosinophilic airway inflammation and airway hyperresponsiveness in OVA-challenged BALB/c mice. Supporting these data, the augmentation of Th2 cytokines (IL-4, IL-5, and IL-13), eotaxin, and adhesion molecules in lung tissues and bronchoalveolar lavage fluid after OVA inhalation was markedly attenuated by PLE. Furthermore, PLE reduced OVA-induced activation of NF-${\kappa}B$ and p38 MAPK in lung tissues. Therefore, our results suggest the potential of P. linteus as a therapeutic agent for asthma.

Effects of Oyo-Tang Herbal Acupuncture on Immune Cells In peripheral blood and Serum IgE of Allergic Asthma in the Rats (오요탕(五搖蕩) 약침(藥鍼)이 알레르기 천식(喘息) 흰쥐의 말초혈관내(末梢血管內) T-cell과 혈청(血淸) IgE에 미치는 영향(影響))

  • Purpose : It has been reported that Oyo-Tang has an effect on allergic asthma clinically. In order to investigate the herbal acupuncture solution of the Oyo-Tang on allergic asthma, the Oyo-Tang herbal-acupuncture(OTHA) was applied on BL13 and ST36 of allergic asthma model of the rats. Methods : Then the concentration of serum IgE and the ratio of CD4+ and CD8+ T-cells in peripheral blood were measured and compared with control group. Results : The results obtained as follows; 1. The concentration of serum IgE was decreased significantly in both of the OTHA on BL13 group and on ST36 group as compared to control group. 2. The ratio of CD4+ and CD8+ T-cells in peripheral blood was not different statistically between the normal, the control, the OTHA on BL13 and the OTHA on ST36 group.

Evaluation of an Education Program for Patients with Asthma who use Inhalers (천식 환자를 위한 흡입기 교육 프로그램의 효과)

  • Purpose: This study was done to evaluate the effectiveness of an education program for patients with asthma who use inhalers. Methods: The research design for this study was a non-equivalent control group quasi-experimental study. Participants in this study were 36 patients for the control group, and 43 patients for the experimental group. The experimental group participated in the education program. The control group received the usual care. Data were collected before and 1 month and 2 months after the program finished and were analyzed using the SPSS 12.0 program. Results: The experimental group had significantly higher scores of knowledge of inhalers, and inhalation technique compared to the control group. However, no significant differences were found between two groups for PEFR, asthma instability, and satisfaction with inhalers. Conclusion: According to the results, the education program was effective in improving knowledge of inhalers, and inhalation technique. Therefore, it is recommended that this education program be used in clinical practice as an effective nursing intervention for patients with asthma on inhalers.

The Effects of Oldenlandiae Diffusae Herba Extract on Eosinophil, IgE and IL-4 in Experimental Asthma induced by Ovalbumin (백화사설초(白花蛇舌草)가 ovalbumin으로 유도된 천식동물모델에서 Eosinophil의 수, IgE 및 IL-4에 미치는 영향)

  • Objective : This study was performed to investigate the effect of oral administration of Oldenlandiae Diffusae Herba (ODH) on eosinophil, immunoglobulin-E and interleukin-4 in the experimental asthma induced by ovalbumin. Methods : Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. And, it was observed the change of the concentrations of IL-4 in BALF and splenocyte, IgE in serum by ELISA. Results : 1. The number of eosinophil in BALF was significantly decreased in ODH group copared with control group. 2. Concentration of IL-4 in serum, BALF and splenocyte was significantly decreased in ODH group compared with control group, respectively. 3. Level of IgE in serum was significantly decreased in ODH group compared with control group. Conclusion : We found that the effect of ODH extract in asthma was implicated in reductions of IL-4 released from Th2 cell, and decreases of IgE from plasma cell. These findings suggest that ODH extract can produce anti-asthmatic effect, which may playa role in allergen-induced asthma therapy.

The Clinical Effects of Gamipaimo-tang (Jiaweibeimu-tang) in Asthmatic Patients Based on Severity (가미구모탕의 기관지천식환자의 중증도에 따른 임상효과)

  • Objective : Recently questionnaires that reflect the quality of life (QOL) in asthma patients were developed. We aimed to identify the clinical effects of Gamipaima-tang and the side effects of Gamipaimo-tang in asthmatic patients. Materials and Methods : The subjects are consisted of 34 patients with asthma who had been treated with Gamipaimo-tang for four weeks. Gamipaima-tang is an herbal decoction which has been used as a traditional therapeutic agent for asthma. Pulmonary function test (PFT) was checked before and after 4-week treatments. Quality of Life Questionnaire for Adult Korean Asthmatics (QLQAKA) was checked before and after 2 weeks and 4 weeks of treatment. Results : Treatment with Gamipaima-tang for four weeks resulted in significant increase in FVC%, $FEV_{1.0}%$, PEFR%, and QLQAKA. The total efficacy rate in the patient group was 52.9% after 4 weeks. The total efficacy rate of the step 4 group (n=18, 55.6%) classified by Global Initiative for Asthma (GINA) was higher than that of the step 3 group (n=13, 46.2%) classified by GINA. Conclusion : This study shows that Gamipaima-tang has the effects of improvement of pulmonary function and quality of life in asthmatic patients. Obviously further research concerning this is still necessary.

Clinical Outcome of Educational Program Using Self-monitoring of Peak Expiratory Flow Rate for Asthma Patients (천식 환자에 있어서 최대호기 유속의 자가 모니터링을 이용한 복약지도의 유용성)

  • The effects of pharmacist's intervention for asthma patients using self-monitoring of peak expiratory flow rate in medication teaching model was evaluated for 3 months in improving clinical outcomes including emergency visits, hospitalizations, antibiotics use, symptoms and sleep disturbance. Twenty seven patients were enrolled in study and twenty three patients completed the follow-up schedules. The selected patients were given the pre-designed instruction for medication including appropriate use of medication, metered-dose inhaler(MDI) technique, identifying and controling asthma triggers and recognizing early signs of deterioration. There were significant improvements in clinical outcomes, in terms of emergency visits, hopitalizations, antibiotics use, symptoms and sleep disturbance. There were also significant improvements in the MDI use, environmental control, and medication knowledges. There was a progressive increase in peak expiratory flow rate during the three-month intervention. In conclusion, pharmacist's intervention using self-monitoring of peak expiratory flow rate has a significant impact on improving clinical outcomes in asthma patients.

  • Objectives The purpose of this study is to report the effect of Hominis Placenta Pharmacopuncture(HPP) for patients with asthma. Methods We treated three patients who have asthma with HPP. To check the effect of HPP we used CAMSOM-V. To check the satisfaction of HPP we used Five-point Likert scale. Results After treatment symptoms were improved. CAMSOM-V decreased in all cases. The Likert scale points were checked 5 and 4 and 4. Conclusions In this study, we have found the meaning of the research that HPP, in the future will be efficacious against asthma.

Neovastat(AE-941) inhibits the airway inflammation and hyperresponsiveness in a murine model of asthma

  • Matrix metalloproteinase (MMP)-9 plays an important role in the pathogenesis of bronchial asthma. Neovastat, having significant antitumor and antimetastatic properties, is classified as a naturally occurring multifunctional antiangiogenic agent. We evaluated the therapeutic effect of Neovastat on airway inflammation in a mouse model of asthma. BALB/c mice were immunized subcutaneously with ovalbumin (OVA) on days 0, 7, 14, and 21 and challenged with inhaled OVA on days 26, 29, and 31. Neovastat was administrated by gavage (5 mg/kg body weight) three times with 12 h intervals, beginning 30 min before OVA inhalation. On day 32, mice were challenged with inhaled methacholine, and enhanced pause (Penh) was measured as an index of airway hyperresponsiveness. The severity of airway inflammation was determined by differential cell count of bronchoalveolar lavage (BAL) fluid. The MMP-9 concentration in BAL fluid samples was measured by ELISA, and MMP-9 activity was measured by zymography. The untreated asthma group showed an increased inflammatory cell count in BAL fluid and Penh value compared with the normal control group. Mice treated with Neovastat had significantly reduced Penh values and inflammatory cell counts in BAL fluid compared with untreated asthmatic mice. Furthermore, mice treated with Neovastat showed significantly reduced MMP-9 concentrations and activity in BAL fluid. These results demonstrate that Neovastat might have new therapeutic potential for airway asthmatic inflammation.

Two Cases of Asthma Patients Treated with CheonPae-hwan (천폐환(喘肺丸)으로 호전된 천식환자 2례)

  • Objective: This study reports on the effects of CheonPae-hwan among asthma patients.Method: This case study was conducted with two asthma patients who had visited a traditional Korean medicine clinic. CheonPae-hwan was prescribed to the two patients; we then evaluated the objective symptom changes through forced vital capacity (FVC) and forced expiratory volume at one second (FEV1). Dyspnea-subjective symptom change was evaluated with the visual analog scale (VAS).Results: In case 1, FVC increased from 2.24 to 2.55 after treatment, FEV1 increased from 1.86 to 1.99, and VAS decreased from 2 to 0. In case 2, FVC increased from 1.51 to 2.79 after treatment, FEV1 increased from 1.12 to 2.22, and VAS decreased from 8 to 5.Conclusion: CheonPae-hwan appeared to be effective for treating the two asthma patients in our study.

Magnolol exerts anti-asthmatic effects by regulating Janus kinase-signal transduction and activation of transcription and Notch signaling pathways and modulating Th1/Th2/Th17 cytokines in ovalbumin-sensitized asthmatic mice

  • Allergic asthma, is a common chronic inflammatory disease of the airway presenting with airway hyperresponsiveness and airway remodelling. T helper cells-derived cytokines are critically associated with asthma pathogenesis. Janus kinase-signal transduction and activation of transcription (JAK/STAT) signaling is found to be involved in asthma. Magnolol is a plant-derived bioactive compound with several pharmacological effects. The study aimed to assess the effects of magnolol in ovalbumin (OVA)-induced asthmatic model. BALB/c mice were sensitized and challenged with OVA. Magnolol (12.5, 25, or 50 mg/kg body weight) was administered to separate groups of animals. Dexamethasone was used as the positive control. Cellular infiltration into the bronchoalveolar lavage fluid (BALF) were reduced on magnolol treatment. The levels of Th2 and Th17 cytokines were reduced with noticeably raised levels of interferon gamma. Lung function was improved effectively along with restoration of bronchial tissue architecture. OVA-specific immunoglobulin E levels in serum and BALF were decreased by magnolol. Magnolol reduced Th17 cell population and effectively modulated the JAK-STAT and Notch 1 signaling. The results suggest the promising use of magnolol in therapy for allergic asthma.
