Anti-inflammatory Effects of Aster glehni Water Extracts in LPS-stimulated RAW 264.7 Macrophages (산백국(山白菊) 열수추출물이 RAW 264.7 대식세포에 미치는 항염증 효과)

  • Objectives : This study was conducted to confirm the anti-inflammatory effects of Aster glehni Water extracts. Methods : In this study, MTT assay was performed to detect cell viability. To evaluate the anti-inflammatory effects of Aster glehni Water extracts, we examined NO production in LPS-induced macrophages. Expressions of iNOS, COX-2, ERK, p38, JNK were also investigated by using western blot assay. Results : Aster glehni Water extracts have no cytotoxicity at 15.625-1,000㎍/㎖ in RAW 264.7 cells. Aster glehni Extracts inhibited the NO production in a dose-dependent manner in RAW 264.7 cells treated with LPS. Pretreated 250, 500, 1,000㎍/㎖ of Aster glehni water extracts had significantly suppressed expression levels of iNOS, COX-2, p-ERK, p-p38, p-JNK. Conclusions : These results suggest that Aster glehni Water extracts have anti-inflammatory effects and can be used for various inflammatory skin diseases.

  • The chromatographic separation of MeOH extract of the aerial parts of Aster glehni (Compositae) led to the isolation of three sesquiterpenes, two sterols and four terpenes. Their structures were determined to be $\beta$-amyrin acetate (1), phytol (2), alismol (3), $\alpha$-tocopheryl quinone (4), $\alpha$-spinasterol (5), 10-O-methyl alismoxide (6), alismoxide (7), and 3-O-(6'-O-palmitoyl-$\beta$-D-glucosyl)-spinasta 7,22-diene (8) by physicochemical and spectroscopic methods . These compounds (1-8) were first isolated from the Aster glehni.

  • Aster glehni Franchet et Schmidt is a compositae plant and has been known as a native specie in Ulleung Island, Korea. It is officially recognized as a regional specialty that grows only in this region. In 2014, brown and dark spots were observed on aster leaves in a forest research field, Jinju, Korea. A causal agent was isolated from the disease symptomatic leaves and identified as fungus Alternaria solani. Fungal morphological characteristics and molecular identification with internal transcribed spacer sequences were synchronized as A. solani. The isolated fungi reproduced the same disease symptoms when the fungus was artificially inoculated on healthy aster leaves. This is the first report that A. solani caused leaf blight disease in Aster glehni in Korea.

Physicochemical and sensory characteristics of Aster glehni Kimchi during storage at different fermentation temperatures (숙성온도를 달리한 섬쑥부쟁이 김치의 이화학적 및 관능적 특성)

  • This study was conducted to increase the value of Aster glehni as a useful food resource. The Hunter L, a and b Values of Aster glehni leaves Were 34.23${\pm}$2.80, -10.59${\pm}$1.80 and 13.29${\pm}$2.51, respectively. The Shearing force and contents of tannin and dietary fiber were 4701.2g, 100.9ppm and 37.1%, respectively. The minerals identified in Aster glehni were Ca (6.93mg/kg), K (45.36mg/kg), Mg (1.70mg/kg), Fe (0.36mg/kg) and Na (1.26mg/kg). Aster glehni Kimchi was packed in polyethylene film (200g) and fermented at 20 and 4$^{\circ}C$. With regards to the color changes, the Aster glehni Kimchi fermented at 20$^{\circ}C$ showed greater increases in the Hunter L, a and b values than a 4$^{\circ}C$. The pH of the Kimchi decreased and acidity increased with storage time at both temperatures. The ascorbic acid contents decreased sharply with storage time and by about 85% at 20$^{\circ}C$ after 5 days, and 73% at 4$^{\circ}C$ after 30 days. The reducing sugar content also decreased with storage time at both 20 and 4$^{\circ}C$. The results of the sensory evaluation showed the optimum ripening times of the Aster glehni Kimchi to be 1∼3 days at 20$^{\circ}C$ and less than 20 days at 4$^{\circ}C$.

Analysis of Volatile Flavor Components of Aster glehni (섬쑥부쟁이의 휘발성 향미성분 분석)

  • Essential oils in leaf of fresh Aster glehni were extracted by SDE (simultaneous steam distillation and extraction) method using diethyl ether as solvent. The yield of the essential oils was 0.05%. And their volatile flavor components were analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) and identified by the RI of GC and mass spectrum of GC-MS. A total of 31 components, including 17 hydrocarbons, 2 aldehydes, 7 alcohols, 3 esters, 1 acid and 1 oxide were identified. The major volatile flavor components of fresh Aster glehni were ${\alpha}$-pinene, limonene, $\delta$-elemene, ${\beta}$-pinene, cis-3-hexenol and myrcene. Volatile flavor patterns of fresh ind dried Aster glehni were analyzed using an electronic nose. Sensor PA2 that was sensitive to alcohols showed the highest resistance for fresh and dried Aster glehni. Resistance of six metal oxide sensors was decreased in fresh sample compared with dried one.

Effects of Aster glehni Water Extracts on Skin Inflammation in HaCaT Keratinocytes (산백국(山白菊) 열수추출물이 HaCaT 피부각질세포에서 미치는 항염증 효과)

  • Objectives : This study was conducted to verify the anti-inflammatory effects of Aster glehni water extracts in HaCaT keratinocytes. Methods : In this study, cell viability was confirmed by MTT assay. Production of TNF-α and IL-6 was determined by ELISA. mRNA expression of TARC and MDC were measusred by qRT-PCR. Also, expressions of p-JNK, JNK, p-ERK, ERK, p-p38, and p-38 were investigated by using western blot assay. Results : Aster glehni water extracts were not shown any significant cytotoxicity at 15.625-500㎍/㎖ in HaCaT keratinocytes. Aster glehni extracts inhibited the TNF-α and IL-6 production in HaCaT keratinocytes treated with TNF-α and IFN-γ. Also, expression of TARC, MDC, p-ERK, and p-STAT1 was decreased. Conclusions : These results suggest that Aster glehni water extracts have anti-inflammatory effects in HaCaT keratinocytes and can be applied to the development of anti-inflammatory treatment substances.

Analysis of Vegetation and Vegetation-Environment Relationships in Main Wild Vegetables of Ulleungdo in Korea -Vegetation of herb layer of the Aster glehni, Allium ochotense, and Aruncus sylvester - (울릉도 주요 산채류 자생지의 식생 및 환경과의 상관관계 분석 -섬쑥부쟁이, 울릉산마늘, 눈개승마의 초본층 식생을 중심으로-)

  • This study was conducted to provide ecological basic data that use to establish environmental conditions for cultivation of wild vegetables in 2016-2018. Therefore, we investigated the vegetation structure and the correlation between the community structure and the environmental factors for natural habitats of wild vegetables(Aster glehni, Allium ochotense, and Aruncus sylvester) distributed in Ulleungdo. As a result of population and gradient analysis, the vegetation was classified into Aster glehni community, Allium ochotense community, and Aruncus sylvester community. We confirmed that the classification by population analysis was consistent with that by TWINSPAN method, suggesting that they were complemented each other. The importance value of Aster glehni was the highest in all communities, followed by Aruncus sylvester, Allium ochotense, Hydrangea petiolaris, Dryopteris crassirhizoma, Asperula ldorata, Phryma leptostachya var. asiatica, Disporum viridrescens, Hedera rhombea, Anthriscus sylvestris, and Hepatica maxima. According to the results of DCCA ordination analysis, among those communities, the Aster glehni community was distributed in soil where the nutrition including T-N and O.M. were intermediate. The Allium ochotense community was distributed on the a little high northern slope at the highest altitude where the CEC and O.M. were the highest, and other nutrition and pH were low. The Aruncus sylvester was distributed on high slope and altitude on which the amount of exchangeable cation such as $Ca^{{+}{+}}$, $Mg^{{+}{+}}$ and pH were high, and the CEC, $P_2O_5$, and O.M. were the lowest.

  • In an attempt to find the activity of Aster glehni (Compositae) leaves on the obesity in vivo, 30% ethanolic extract rich in caffeoylquinic acids was orally treated with 100 and 200 mg/kg for consecutive four weeks during feeding high-fat diet in rats for 6 weeks. This extract prevented the increase of atherogenic index and body weight and oxidative stress from dietinduced obese rats probably due to the pharmacological mechanism of the CQ complex.

The Effect of Aster glehni Fr. Schm. Extracts on Whitening and Anti-Wrinkle (섬쑥부쟁이(Aster glehni Fr. Schm.) 분획물의 미백 및 주름 개선 효과)

  • Plants and their extracts can be utilized as inexpensive and rich resources of active constituents in the cosmetic field, as well as the food, pharmaceutical and medicinal fields. Until now, Aster glehni Fr. Schm. had no known active effect, except on anti-oxidation, that was found during investigations for application in the cosmetic field. In this study, we examined the inhibition of enzymatic reactions to protein levels in inclusive B16F10 melanoma cell lines. Significant inhibition of enzymatic reactions was observed in the EtOAc extract, which advanced to tyrosinase protein and TRP-1 in the B16F10 melanoma cell line. These results indicated that the effect of EtOAc extract inhibited expressions of tyrosinase protein and TRP-1 in the B16F10 melanoma cell line by 30.5% and 41.5% at 100ug/ml respectively. On the other hand, antimicrobial activity was evaluated to the four fractions in normal flora of the skin. Hexane extract was only exhibited in the higher clear zone in all strains. In conclusion, any cosmeceutical effects of Aster glehni Fr. Schm. may have a potential meaning, as well as possible value for further studies regarding the effects of chemical constituents of Aster glehni Fr. Schm.

Occurrence of Stem Rot of Aster glehni Caused by Sclerotium rolfsii (Sclerotium rolfsii 에 의한 섬쑥부쟁이 흰비단병 발생)

  • In July 2003, a destructive stem rot occurred sporadically in an exhibition farm on Aster glehni of Hamyanggun, Gyeongsangnam-do Agricultural Research and Extension Services, Korea. The typical symptoms of the disease were water-soaking, stem rot, wilt or blight. The infected plants were mostly died. White mycelial mats were spread over lesions and sclerotia were formed on stems and near soil line. The sclerotia were globoid or irregular in shape, 1~3 mm in size, and white to brown in color. The optimum temperature for fungal growth was about 30oC. Clamp connections were observed in the hyphae of the fungus grown on potato dextrose agar, and hyphal diameter was 3~8 ${\mu}m$. On the basis of mycological characteristics and test of pathogenicity to host plants, the fungus was identified as Sclerotium rolfsii. This is the first report on the stem rot of Aster glehni caused by S. rolfsii in Korea.
