The Manufacturing and Physiological Evaluation of Mulberry Fruit Tea (오디 분말차 제조 및 생리활성 평가)

  • In this study we manufactured mulberry fruit tea(MFT) to be used as an anti-inflammatory agent. Here, MFT was consumed by thirty middle-aged women with symptoms of rheumatoid arthritis for 4 weeks. The anthropometric measurements and serum inflammatory factors were measured before and after consumption of the MFT. For the anthropometric measurements, BMI and body fat decreased after 4 weeks of tea consumption although there were no significant differences. The serum inflammatory factors(uric acid and homocysteine) as well as serum amino transferase activity(aspartate trans-aminase and alanine transaminase) showed a decreasing trend. In addition, serum carbon reactive protein(CRP) decreased significantly decreased after MFT consumption 4 weeks. It's concluded that this MFT is helpful to for rheumatyroid arthritis patients in order to decrease serum inflammatory factors such as CRP.

  • Hepatic encephalophthy was diagnosed with serum chemistry, abdominal radiography and ultrasonography in a 2.6kg, 4 year-old maltese dog showing signs of hypersalivation, involutary spasm of facial muscles, ataxia, behavior abnormalities like dullness, sleep disorder, restlessness. In serum chemistry, the level of alanine transferase and aspartate trasferase was mildly elevated, ammonia was severely increased. On abdominal radio-graphs, the size of liver was mildly decreased. In ultrasonographic examination, diffuse lesion with hyperechoic change and decreased vasculature were seen in the hepatic region. But vascular abnormalities of liver were not observed. Drug and dietary therapy were undertaken and severities of clinical sign were alleviated.

  • This study was done to investigate the protective effect of novel 9-amino-1, 2, 3, 4-tetrahydroacridine derivatives on the hepatoprotective effect intoxicated rats induced by carbon tetrachloride ($CCl_4$). A series of currently derivatives of 9-amino-1, 2, 3, 4-tetrahydroacridine have been prepared through the alkly substitution or the ring expansion for the treatment of the Alzheimer's disease. The activities of aminotransferase (aspartate and alanine) and contents of alkaline phosphatase, triglyceride and glutathione S-transferase in 9-amino-1, 2, 3, 4-tetrahydroacridine derivatives pretreated rats were significantly decreased compared to the only carbon tetrachloride treated rats but the contents of cholesterol were increased compared to the only $CCl_4$ treated rats. The result indicated that 9-amino-1, 2, 3, 4-tetrahydroacridine derivatives showed hepatoprotective effect in $CCl_4$ treated rats.

The Effect of 5-Oxohexyl-3,7-dimethylxanthine on Carbon Tretrachlroride-Induced Hepatotoxicity in Rats (5-Oxohexyl-3,7-dimethylxanthine이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • A series of new derivatives of 1-(5-oxohexyl)-3,7-dimethylxanthine has been prepared for the treatment of the vascular dementia. To investigate hepatoprotective effect of these derivatives, the serum biochemical activity and the histological change of liver tissue were evaluated in carbon tetrachloride ($CCl_4$) treated rats. The activities of aspartate aminotransferase and alanine aminotransferase and the activities of total cholesterol, triglyceride and total bilirubin in 5-oxohexyl-3,7-dimethylxanthine derivatives pretreated rats were significantly decreased compared to the $CCl_4$ only treated rats but the content of glutathione S-transferase was increased compared to the $CCl_4$ only treated rats. These results indicated that 5-oxohexyl-3,7-dimethylxanthine derivatives have hepatoprotective effect in $CCl_4$ intoxicated rats.

Protective Effect of Sachungwhan against CCl4-induced Hepatotoxicity

  • Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine(CRE), total cholesterol(TCHO), triglyceride(TG), low density lipoprotein cholesterol(LDL-CHO), high density lipoprotein cholesterol(HDL-CHO), total protein(TP), albumin(ALB) and total bilirubin(BIL) in serum. Hepatic parameters monitored were levels of cholesterol(CHO), triglyceride(TG), malondialdehyde(MDA), content of cytochrome P450(CYP), level of glutathione(GSH), and activities of NADPH-CYP reductase, superoxide dismutase(SOD), catalase(CAT), glutathione S-transferase(GST), glutathione reductase(GR), glutathione peroxidase(GPx). The histopathological examination showed that the treatment of Sachungwhan relieved the ballooning degeneration of hepatocytes which had been generated by $CCl_4$. The results suggested that hepatoprotective effects of Sachungwhan possibly are due to their promising antioxidative activity.

Effects of Fructus Foeniculi extract on recovering liver function (회향의 간장기능 회복효과)

  • Objectives : Effects of Fructus Foeniculi extract on liver function were investigated in carbon tetrachloride(CCl4) intoxicated rats. Methods : Thirty two male Sprague-Dawley rats with mean weight of $227.28{\pm}7.92g$ were used in these experiments and housed with food and water ad libitum. Fructus Foeniculi extract was administerd at dose 100mg/kg/day and 200mg/kg/day p.o. for 2 weeks after that CCl4 was treated 3 times at dose of 2.5ml/kg, p.o. in alternate day basis. Then serum AFP(${\alpha}$-Fetoprotein), Total protein, Albumin, Triglyceride, Total cholesterol concentrations and ALP (Alkaline phosphatase), AST(Aspartate Aminotransferase), ALT(Alanine Aminotransferase), ${\gamma}$-GT( ${\gamma}$-Glutamyl transferase), LDH(Lactate Dehydrogenase) activities were determined with commercial kit by autoanalyzer. Results : Plasma ${\alpha}$-fetoprotein and total protein concentration showed a tendency to decrease in Fructus Foeniculi extract-treated groups. However, plasma albumin concentration showed no significant differences in all treatment groups. Activity of plasma aspartate aminotransferase and alanine aminotransferase in Fructus Foeniculi extract-treated groups showed a lower value than that of control group. Alkaline phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Fructus Foeniculi treated groups. However, ${\gamma}$-glutamyl transferase activity showed no significant difference in all treated groups. Concentration of plasma triglyceride and total cholesterol showed a high level in CCl4 intoxicated rats but not in Fructus Foeniculi treated groups. Conclusion : Reviewing these experimental results, it appears that Fructus Foeniculi extract have recovering effect against liver injury.

Effects of IgY Supplementation on Hanwoo Calves Fed withHigh Quality Roughage (양질 조사료 급여에 따른 한우 송아지의 IgY 처리효과)

  • This study was conducted to examine the effect of IgY supplementation on growth performance, blood metabolism, and disease occurrence in the weaned calves at 3 or 4 month of age. Average daily gain (ADG) and feed intakes were not affected by IgY supplementation, regardless of weaning months. White blood cell (WBC) count of the control group was higher in the final stage than in the initial stage (p<0.05), while that of the IgY supplemented group was lower in the final stage compared to the initial stage (p<0.05). regardless of treatments. Hematocrit (HCT) and red blood cell (RBC) counts were lower in the final stage than in the initial stage (p<0.05), but mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) counts were higher in the final stage than in the initial stage, regardless of treatments. In the control group, concentrations of albumin and globulin were higher in the final stage compared to the initial stage in the weaned calves at 3 month of age (p<0.05), and concentration of blood urea nitrogen (BUN) was increased in the IgY supplemented group. In the control group, concentrations of aspartate aminotransferase (AST) and gamma (${\gamma}$)-glutamyl transferase (GGT) were higher in the final stage compared to the initial stage (p<0.05). In contrast, concentrations of aspartate aminotransferase (AST) and gamma(${\gamma}$)-glutamyl transferase (GGT) were lower in the final stage compared to the initial stage for the IgY supplemented group. A diarrhea calf was only found in the control group. Manure score was higher in the control group compared to IgY supplemented group (p<0.05). Thus, these results indicates that IgY supplementation had positive effects on some blood metabolites, fecal condition score and diarrhea without negative effect on growth performance of the weaned Hanwoo calves.

Investigation of an Acceptable Hemolysis Index Using Re-collected Samples (재채혈된 검체를 이용한 허용 Hemolysis Index에 대한 연구)

  • This study compared the results of hemolyzed samples and re-collected samples to investigate a hemolysis influence and an acceptable hemolysis index (HI). Before and after hemolysis, alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), amylase (Amy), direct bilirubin (D-bil), total bilirubin (T-bil), creatine phosphokinase (CK), gamma glutamyl transferase (GGT), iron, potassium (K), lactate dehydrogenase (LDH), magnesium (Mg), phosphorus (Phos), total protein (TP), and uric acid (UA) showed significant results in the paired t-test. LDH, K, iron, AST, CK, GGT, TP, Amy and Phos had a high correlation between the degree of hemolysis and the results of samples. When comparing Roche's cut-off HI with HIQChigh obtained using quality control (QC) high standard deviation (SD), AST, D-bil, CK, and LDH were similar, but Amy, GGT, K, iron, Phos, and TP were lower than the cut-off HI of Roche, while ALP and ALT were higher. Some analytes which showed no significant results in the paired t-test, were found to have significant results in HI>200. Hence, it is suggested that the hemolyzed sample should be rejected if HI>200. Based on this study that some analytes were affected when HI<100, we recommend to set the standard of hemolysis starting from HI>50.

Kinetic Properties of Wild-type and C117D Mutant UDP-N-Acetylglucosamine Enolpyruvyl Transferase (MurA) from Haemophilus influenzae

  • In this study, the kinetic properties of wild-type and C117D mutant H. influenzae MurA (Hi MurA), which catalyzes the first reaction in the biosynthetic pathway of the cell wall, were characterized. Purified recombinant Hi MurA was active at pH values ranging from pH 5.5 to pH 10, and its $K_m$ (UNAG), $K_m$ (PEP), and $k_{cat}$ values were measured to be 31 ${\mu}M$, 24 ${\mu}M$, and 210 $min^{-1}$, respectively. Hi MurA activity was effectively inhibited by fosfomycin with an $IC_{50}$ value of 60 ${\mu}M$. Hi MurA contains a cysteine residue (C117) at the loop region near the PEP binding, whereas MurA from fosfomycin resistant Mycobaterium tuberculosis or Chlamydia trachomatis contain an aspartate residue instead of the cysteine at the corresponding site. Aspartate substitution of Cys117 in Hi MurA shifted its optimum pH from 7.8 to 6.0. In addition, the $K_m$ values for UNAG and PEP were increased to 160 ${\mu}M$ and 150 ${\mu}M$, respectively, and the $k_{cat}$ value was significantly reduced to 41 $min^{-1}$. Furthermore, the C117D mutant form of Hi MurA was not inhibited by 1 mM fosfomycin. These results indicate that the Cys117 of Hi MurA is the binding site of fosfomycin and plays an important role in the fast turnover of the catalytic reaction.

Effect of Herbal Medicine on Liver Function in Korean Medical Hospital Inpatients: A Retrospective Chart Review (한약복용이 한방병원 입원환자의 간 기능에 미치는 영향에 대한 후향적 관찰)

  • Objectives: The purpose of this study was to investigate liver function test results in admitted patients before and after herbal treatment. Methods: 54 subjects admitted to hospital had liver function tests (aspartate aminotransferase, alanine amino transferase, alkaline phosphatase, total bilirubin) before and after herbal treatment from 1 March 2017 to 30 June 2019. Results: Aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase were not significantly changed (p>0.05). On admission, 11 patients had abnormal liver function, while seven patients had abnormal results in alkaline phosphatase upon discharge. Three of the seven were normal when they were hospitalized. Conclusions: This study suggests that herbal treatment may have no effect on liver injury.
