Comparative Analysis of a Putative HLH Transcription Factor Responsible for Conidiation in Aspergillus Species

  • Asexual reproduction or conidiation in aspergilli is a primary mean to produce their progenies that is environmentally and genetically controlled tightly. Previously, intensive researches in the model fungus Aspergillus nidulans disclosed some genes playing important roles in asexual and sexual development. Among them, one gene encoding a putative helix-loop-helix (HLH) transcription factor, named ndrA, has been isolated and characterized as a downstream regulator of developmental master regulator NsdD. By using comparative genome search of A. niduans NdrA protein, its orthologues have been identified in A. fumigatus and A. flavus, respectively (AfudrnA and AfldrnA). Deletion of the ndrA genes in both Aspergillus species made them unable to produce the conidia yet abundant production of sclerotia in A. flavus. Complementation of ndrA deletion strains by intact ndrA ORFs has restored the conidiation as in the control strains. In A. fumigatus, ndrA deletion also resulted in loss of conidiation phenotype. Northern analyses showed that the ndrA genes in both Aspergillus species are highly expressed at the early stage of the conidiation. Interestingly, the ndrA genes were found to be necessary for the proper expression of brlA genes. Antifungal sensitivity test revealed that the ndrA genes might be responsible for the sensitivity or resistance to some antifungal agents. However, ndrA deletion did not greatly influence the growth in both strains. And the A. flavus ndrA gene did not affect the aflatoxin production. Taken together, ndrA genes in Aspergillus species could be an important positive regulator of conidiation under the regulation of the nsdD gene yet upstream of the brlA gene.

Functional Analysis of Aspergillus nidulans Genes Selected by Proteomic Analysis under Conditions Inducing Asexual Development (Aspergillus nidulans 무성분화 촉진 조건의 단백체 및 해당 유전자 기능분석)

  • Despite the significance of external environmental factors in differentiation, putative factors involved in differentiation of Aspergillus nidulans have not yet been fully understood. A sporulation-specific proteome analysis of A. nidulans in the present study revealed that the expression levels of more than 2,400 proteins were affected under conditions inducing sporulation (0.6 M KCl) compared with normal conditions. Among the proteins with predicted functions, two targets, AN1342 and AN9419, were functionally analyzed using targeted deletion strains and phenotypic observations. For AN1342, because the deletion of the corresponding open reading frame caused a reduction in stalk length during asexual development and in pigment production in liquid culture, the gene was designated as sspA ($\underline{s}hort$ $\underline{s}talk$ & $\underline{p}igment$). Deletion of the AN9419 gene, which is predicted to encode alanyl-tRNA synthetase, led to severe growth defects due to alanine auxotrophy and abolishment of asexual reproduction and thus, the gene was designated as alaA.

  • The yeast strain 504D, isolated from salted shrimp soup, showed the best proteolytic activity under alkaline condition. The yeast formed vegetative cells in almost optimal media for yeasts, but formed only pseudohyphae in the MM medium containing citric acid and true hyphae in the MM medium containing N-acetylglucosamin and ${\beta}$-D-glucose. The yeast was classified as hemiascomycetes to form ascospores by sexual reproduction, and formed blastospores and athrospores by asexual reproduction. The yeast strain did not assimilate almost of the carbon sources, nitrate and nitrite, but some organic acids and alcohols. The fatty acids of whole cells were composed of 53.67% unsaturated fatty acids and 14.58% saturated, and, especially, C17:1 was observed in this strain but not in two control yeasts. However, almost of all results were very similar to the morphological and physiological characteristics of Yarrowia lipolytica KCCM 12495 and KCCM 35426, except for a little differences which are the composition of fatty acids and the manner of mycellium formation. Therefore, the isolated yeast strain 504D is identified as a Yarrowia lipolytica.

Isolation and characteristics of yellow-pigment producing mutants of Monascus anka. (Monascus anka로부터 황색소 생성 변이주의 분리 및 특성)

  • To produce yellow pigment selectively, mutants were induced from Monascus anka Nakazawa et Sato IFO 4478 (KCCM 11832 strain), and their characteristics were evaluated. Five kinds of auxotrophic mutants which required amino acids for growth and pigmentation, were isolated through a series of mutagenic treatments. Especially, asparagine auxotroph Y7 produced high ratio of yellow pigment. This mutant showed all the morphological characteristics of Monascuceae but the shape of colony and the diameter of conidia. Mutant Y7 was propagated by sexual reproduction more often than asexual reproduction, which could be effective in production of pigments. Yellow pigment produced extracellularly by the mutant Y7 was more soluble in polar solvents such as ethanol and water than in nonpolar solvents. Its productivity of yellow pigment was 2.2 times higher in the mutant Y7 than in parents. In addition, its yellow pigment showed characteristics of maximum absorption at 373 nm. Moreover, the hue of pigment produced by the mutant Y7 was bright yellow, and it was stable through the subculture over 10 generations.

  • This experiment surveys ecological and morphological phase on Prasiola sp. in the mountain torrent of Chodang-ri, Keunduck-myon, Samchuck-gun, Kangwon-do, Korea from November 1968 through January 1970 and the results obtained were summarized as follows. 1. The temperature of the surface water was 12.4$\pm$2.3$^{\circ}C$, and pH was 7.25$\pm$0.2. 2. The external forms of thallus are very variable, but its basic form likely lanceolated and ovated. 3. The length of thallus is 0.6cm-8.8cm (3.6$\pm$1.72cm), and the width, 0.2cm-5.5cm(1.7$\pm$0.92cm). (The inside of the parentheses the average for 500 individuals). 4. It is observed that thallus at the time of formation of a monospore is a monostratum cell whose thickness is 21$\pm$3.2$\mu$. 5. Thickness of thallus at the transveres section at the time of heteroplanogametes formation is 29.9$\pm$2.2$\mu$, and that of the female gametes cell is 6.3$\pm$1.7$\times$6.3$\pm$0.8$\mu$, and that of the male gametes cell is 2.8$\pm$0.6$\times$28.1$\pm$1.1$\mu$. 6. As for growth, it begins with apical growth, followed by marginal growth. It seems to grow, through cell division when microsopical papillae are produced in growth line cell of marginal rigion thus by fusioning microscopical papillae. 7. The two kinds of thallus, one produced from a monospore and other from a planozygotes, are both haplonts and observed as n-3. 8. It seems that a thallus sprouts in April and increases its number through a asexual reproduction by a monospore and begins a sexual reproduction in November by forming gametes. 9. Female gametes are produced 16 from one thallus cell, whose structure shows 8 cells in the surface view and 4 layers in transverse section. Male gametes are produced 128 from one thallus cell, 16 cells surface, 8 layers in transverse section.

Organ Fossils of Neocalamites carrerei from the Amisan Formation of the Nampo Group, Korea (남포층군 아미산층에서 산출된 Neocalamites carrerei의 기관화석)

  • This study deals with the morphological characteristics and structure of organ fossils of Neocalamites carrerei such as rhizomes, aerial leafy stems, and cones collected in situ from the Amisan Formation of Nampo Group. Judging from the size of some extant Equisetum and paleoclimate of Daedong flora, it is considered that the size of Neocalamites carrerei was probably very large. The cone of Neocalamites takahashii (Kon'no) is closely associated with the leaves of Neocalamites carrerei. It indicates that two organ fossils mentioned above probably belong to the same species. Comparing with very abundant occurrence of aerial leafy stems and rhizomes in the Amisan Formation, cones are very rare. This fact implies that Neocalamites reproduce not only by spore reproduction, but also by asexual reproduction. Neocalamites largely differ in the leaf form and size from Equisetum.

Response of Achlya racemosa, A. proliferoides and Saprolegnia furcata to Sub-lethal Treatments of Amino Acids

  • The effect of four sub-lethal concentrations(400, 800, 1,200 and 1,600 ${\mu}g/ml$) of three amino acids such as isoluecine, aspartic acid and phenylalanine on vegetative growth and sexual and asexual reproduction of Achlya racemosa, A. proliferoides and Saprolegnia furcata was investigated. The density of vegetative growth and diameters of vegetative colonies of species of the Oomycetes fungi decreased with rising the concentration of the applied amino acid. Vegetative hyphae of treated fungi almost appeared branched in case of S. furcata, thick in case of A. racemosa and distorted in case of A. proliferoides as compared with control. The different treatments with amino acids depressed both sporangial formation and discharge, which were dependent on the tested species of zoosporic fungi, the amino acid and its dosage. Phenylalanine was the most effective amino acid in inhibiting sporulation and S. furcata was the most sensitive fungal species. Aspartic acid and isoleucine stimulated germination of discharged spores through the formation of germlings. Gemmae formation by the three fungi was reduced at the low concentrations of amino acids and nearly missed at high concentrations. Sex organs(oogonia and antheridia) were affected partly; rudiment oogonia were observed at low concentrations(400 and 800 ${\mu}g/ml$) and disappeared at higher concentrations, whereas antheridial branch formation was stimulated as the fungi were treated with isoleucine and to some extent phenylalanine.

Genetic Diversity and Population Structure of Glehnia littoralis (Umbelliferae) in Korea

  • Glehnia littoralis Fr. Schmidt (Umbelliferae) is a short-lived herbaceous species that are mostly distributed throughout East Asia. Although G. littoralis has been regarded as ecologically important one, there is no report on population structure in Korea. Starch gel electrophoresis was used to investigate the allozyme variation and genetic structure of Korean populations of this species. A high level of genetic variation was found in G. littoral is populations. Nine enzymes revealed 18 loci, of which 12 were polymorphic (66.7%). Genetic diversity at the species and population levels were 0.159 and 0.129, respectively. The sexual and asexual reproduction, high fecundity, and colonization process are proposed as possible factors contributing to genetic diversity. An indirect estimate of the number of migrants per generation (Nm = 1.45) indicated that gene flow was not extensive among Korean populations of this species. It is suggested that the ability of vegetation and artificial selection may have played roles in shaping the population structure of this species. we recommend that a desirable conservation population should be included at least 30 plants per population and especially those with high variation.

Organism-environment interactions and differential gene expression patterns among open-coastal and estuarine populations of Porphyra umbilicalis Kützing (Rhodophyta) in the Northwest Atlantic

  • Intertidal macroalgae are exposed to many abiotic stress factors, and they must regularly react to changes in their environment. We used RNA-seq to describe how Porphyra umbilicalis (Rhodophyta) changes gene expression patterns to interact with different habitats. Tissue samples were taken from a typical habitat along the open-coast of the Northwest Atlantic, as well as from a rare, atypical habitat in an estuarine tidal rapid environment. Differential gene expression analyses suggest that pathogic bacteria and viruses may be a significant factor influencing the transcriptome in the human-impacted estuarine environment, but the atypical habitat does not necessarily induce more stress in Porphyra umbilicalis growing there. We found genes related to nitrogen transport are over-expressed in tissue from the open-coastal site compared to those from the estuarine site, where environmental N levels approach hypertrophic levels. Low N levels impede growth, but high levels are toxic to cells, and we use qPCR to show this species regulates expression of a putative high-affinity $NH_4{^+}$ transporter under low and high N conditions. Differences in expression of this transporter in these habitats appear to be inherited from parent to offspring and have general implications for adaptation to habitat in other species that are capable of asexual reproduction, as well as more specific implications for this species' use in aquaculture.

Artificial Seed Production and Nursery Culture Conditions Using Regeneration of Isolated Utricles and Medullary Filaments of Codium fragile (Suringar) Hariot (청각, Codium fragile (Suringar) Hariot의 분리수사 재생에 의한 종묘생산과 가이식 조건)

  • Codium fragile is commercially farmed in Korea by natural blooming zygote attachment. Experiments found optimum conditions for artificial seed production and nursery culture of C. fragile by asexual reproduction. Isolated utricles and medullary filaments were regenerated to erect thalli using both indoor and outdoor culture experiments. Under the indoor culture conditions, irradiance was an important factor to control the development of erect thalli. Formation of erect thallus from the isolated medullary filaments in the indoor culture was induced after 30 days under $20^{\circ}C$ and $60{\mu}mol/m^2/sec$. The detachment of isolated utricles and medullary filaments from the substrates of seed strings was reduced by exposure to the air during 2 hrs before the indoor culture of seed strings. The maximum growth and development of erect thalli in the nursery culture was induced at a water depth of 0.5 m. Depending on the substrates of the seed strings the growth of erect thalli was not significantly different (p>0.05).
