Antifungal and Insecticidal Activity of Essential Oil from Asarum sieboldii against Wood Contaminant Fungi and Lasioderma serricorne L. (세신 정유추출물의 목재부후균과 궐련벌레에 대한 항진균 및 살충활성)

  • To investigate bioactive materials for development of natural conservative agent on organic cultural heritage, essential oil from Asarum sieboldii was screened for antifungal and insecticidal activity against 4 wood rotting fungi and adult of Lasioderma serricorne. Antifungal activity of essential oil was tested by using paper disc soaking method. Antifungal activity expressed as $IC_{50}$ value showed $1.50{\sim}2.84{\mu}l/disc$ range and the most significant antifungal activity was observed in Lentinus lepideus. The insecticidal activity of essential oil was examined by topical application method against L. serricorne adults. 50% and 100% of essential oil gave 98.3% and 100% mortality for 24 hours, respectively. The major components of the essential oil were methyl eugenol (56.32%), eucarvone (11.53%), safrole (5.79%), ${\delta}$-3-carene (2.09%), which were identified by gas chromatography-mass spectrometry. From these results, essential oil from A. sieboldii could be useful for conservation of organic cultural heritage against biological deterioration by insect and wood rotting fungi.

  • Objectives : The purpose of this study was to investigate the anti-inflammatory effects of extract Asarum sieboldii(AS) on the peritoneal macrophage. Methods : To evaluate of anti-inflammatory of AS, we examined cytokines production in lipopolysacchride (LPS)-induced macrophages. Furthermore, we checked molecular mechanism using western blot. Results : 1. Extract from AS reduced LPS-induced Nitric oxide (NO), tumor necrosis factor-a (TNF-a), interleukin (IL)-6 and IL-12 production in peritoneal macrophages 2. Extract from AS itself does not have any cytotoxic effect. AS inhibited the activation of mitogen-activated protein kinases (MAPKs) such as p38, extracelluar signal-regulated kinase B a (IkBa) in the LPS-stimulated peritoneal macrophages Conclusion : AS down-regulated LPS-induced NO and cytokines production, which could provide a clinical basis for anti-inflammatory properties of AS

Evaluating the Application of Natural Pesticides on Textile Materials of Organic Cultural Heritages (섬유 재질에 대한 천연 살충·살균제의 적용성 평가)

  • Wood vinegar and Asarum sieboldii Miquel were selected as candidate materials showed antimicrobial activity and insect repellent activity. These natural pesticides have its own color and these could cause color difference on fabric materials. In the present study, we investigated the color difference of undyed and dyed fabrics to evaluate negative effect of wood vinegar and A. sieboldii Miquel. Undyed and dyed fabrics were exposed to natural pesticides of various concentrations for six months in relative humidity 70% and temperature $28^{\circ}C$. After exposure of pesticides, color difference (${\Delta}E*$) were investigated at two weeks intervals for six months. As a results, dyed cotton, silk and undyed silk fabrics exposed wood vinegar were not nearly changed in their colors, but color of only undyed cotton fabric was clearly changed by wood vinegar. Especially color difference by wood vinegar on undyed cotton fabric was most distinct as the concentration increased. On the other hand, all of fabrics exposed A. sieboldii Miquel were not nearly changed in their colors for six months. Therefore, this study first suggests that wood vinegar and A. sieboldii Miquel as natural insecticides could be used to conserve for textile cultural properties from insects and microorganism, but wood vinegar couldn't use the high concentration on undyed cotton fabric.

Investigation of anodized titanium implants coated with triterpenoids extracted from black cohosh: an animal study

  • PURPOSE. The purpose of this study was to evaluate bone response to anodized titanium implants coated with the extract of black cohosh, Asarum Sieboldii, and pharbitis semen. MATERIALS AND METHODS. Forty anodized titanium implants were prepared as follows: group 1 was for control; group 2 were implants soaked in a solution containing triterpenoids extracted from black cohosh for 24 hours; group 3 were implants soaked in a solution containing extracts of black cohosh and Asarum Sieboldii for 24 hours; group 4 were implants soaked in a solution containing extracts of pharbitis semen for 24 hours. The implants from these groups were randomly and surgically implanted into the tibiae of ten rabbits. After 1, 2, and 4 weeks of healing, the nondecalcified ground sections were subjected to histological observation, and the percentage of bone-to-implant contact (BIC%) was calculated. RESULTS. All groups exhibited good bone healing with the bone tissue in direct contact with the surface of the implant. Group 2 ($52.44{\pm}10.98$, $25.54{\pm}5.56$) showed a significantly greater BIC% compared to that of group 3 ($45.34{\pm}5.00$, $22.24{\pm}2.20$) with respect to the four consecutive threads and total length, respectively. The BIC% of group 1 ($25.22{\pm}6.00$) was significantly greater than that of group 3 ($22.24{\pm}2.20$) only for total length. CONCLUSION. This study did not show any remarkable effects of the extract of black coshosh and the other natural products on osseointegration of anodized titanium implants as coating agents. Further studies about the application method of the natural products on to the surface of implants are required.

  • This study was carried out to investigate about cultivation and export of major medicinal crops in Korea. The results obtained were summarized as follows'Planted area, harvested area and pro-duction were increase very much, but yield was not increased almost to 1993 from 1988. In spiteof, cultivation area was increased, export was decreased to 1993 from 1988. In cultivation area, major medicinal crops were Angelica gigas, Platycodon grandiflorum, Astragalus membranaceus, Paeonia lactiflora and Codonopsis lanceolata in 1993. In amount of export, major medicinal cropswere Ginkgo biloba, Bupleurum falcatum, Cornus officinalis, Asarum sieboldii in 1993.

Development of an Environmental Friend Additive Using Antibacterial Natural Product for Reducing Enteric Rumen Methane Emission (항균활성 천연물질을 이용한 반추위 메탄저감용 친환경 첨가제 개발)

  • The present study was conducted to investigate effective starter culture to improve biological activity of Asarum sieboldii. Antibacterial activity, antioxidant activity and reduction of enteric rumen methane production were used as criterions for biological activity. Ground A. sieboldii was added in MRS broth at 10% (w/v) and fermented by different starter cultures. Weissella confusa NJ28, Weissella cibaria NJ33, Lactobacillus curvatus NJ40, Lactobacillus brevis NJ42, Lactobacillus plantarum NJ45 and Lactobacillus sakei NJ48 were used for starter culture strains. Each starter culture was inoculated with 1% (v/v) ratio and fermentation was performed at $30^{\circ}C$ with agitation (150 rpm) for 48 h. MRS broth for the control was employed without starter culture. Then the fermentation growth was dried and extracted using ethyl alcohol. The growth of starter culture was detected at NJ40, NJ42, NJ45 and NJ48. And the highest cell growth was found in NJ40. Antibacterial activity against to Staphylococcus aureus, Listeria monocytogens, Mannheimia haemolytica and Salmonella gallinarum were observed in the extract fermented by NJ40 and NJ45. All treatments showed antioxidant activities, however, there were no significant differences (p>0.05). In in vitro rumen fermentation, negative control (NC) and positive control (PC) were assigned to without extract and with non-fermented A. sieboldii extract. Significant suppression of gas productions were detected in positive control and treatments compared to negative control (p<0.05). However, total volatile fatty acid production was not suppressed. Significant methane reduction per total volatile fatty acid productions were found in positive control and NJ45 treatment (p<0.05). The present study suggested a fermentation of A. sieboldii using NJ45 strain could improve its biological activity and make possible for its use in bio additive for enteric rumen methane mitigation without suppression of animal productivity.

Effect of Asarum sieboldii Extracts on the Growth, Acid Production, Adhesion, and Insoluble Glucan Synthesis of Streptococcus mutans (세신 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성억제에 미치는 영향)

  • Dental caries are the most commonly occurring chronic diseases in the dental field. Because of increasing sugar consumption and extension of average human life, these diseases are widely found all over the world as the most typical cause for a person to lose a tooth. Therefore, the development of more effective, substantial and safe preventive agents against dental caries is strongly required. Streptococcus mutans is known as the causative bacterial playing the most important role informing plaque and it is being noticed as major causative bacteria of dental caries. The present study was designed to investigate the effect of Asarum sieboldii Miquel(Aristolochiaceae) extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans(S. mutans). Both methanol and aqueous extracts showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 100, 1,000 and 2,000 μg/ml compared to the control group(p<0.05 - p<0.01). The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration of 10 μg/ml and complete inhibition was observed at the concentration of 2,000 μg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol and aqueous extracts showed more than 70% inhibition over the concentration of 1,000 μg/ml. Hence, we conclude that Asarum sieboldii might be a candidate of anticaries agent.

Taxonomy of Korean Asarum (Aristolochiaceae) by the Morphological Characters (한국산 족도리풀속(Asarum)의 외부형태학적 형질에 의한 분류)

  • In seven species of Korean Asarum, 44 qualitative and 28 quantitative characters were reviewed taxonomically. Among 44 qualitative characters observed in vegetative and reproductive organs, the states of 27 characters were common in all the seven species. Among 28 quantitative characters, five characters have the same values in all the seven species; the number of cataphyll (2-4), leaf (2), calyx lobe (5), stamen (12) and pistil (6). White spots on adaxial surface of leaf (effectively) distinguished A. maculatum and A. versicolor from the other species. Many long soft hairs on abaxial surface of leaf distinguished A. mandshuricum from other species with short hairs. The glandular hairs on inner surface of calyx tube were divided into 2 types, reflecting the taxonomic relationships between the species: Type 1 has non-stalked glandular hairs (A. maculatum, A. sieboldii and A. sp.), and Type 2 has stalked glandular hairs (A. patens, A. versicolor, A. mandshuricum and A. misandrum). The length, shape, colour of calyx lobe and the extent to which the calyx lobe recurves, as well as the diameter and colour of calyx tube throat, were confirmed to be taxonomically valuable. In the case of glandular hair density on the surface of calyx lobe, A. patens with low density and A. sp. with high density were clearly distinct from all the other species with intermediate hair density. Meanwhile, the stylar protuberance length of A. patens (2.9-3.3 mm) was strikingly different from other species (1.4-2.5 mm). Additionally, further examination of the identities of A. yeonbyeonense and A. yeonbyeonense var. viridiluteolum is required.

Toxicity Evaluation of Asarum Sieboldii Extract for Human's Safety (인체안전성을 위한 족두리풀 천연추출물의 독성평가)

  • Chemically derived pesticides have been used to prevent biological damage to domestic cultural property. However, their use is gradually being restricted due to the harmful effects on the human body and environment. Therefore, there is a growing interest in the search for new antifungal biopharmaceuticals whose safety has been confirmed by toxicity evaluation through animal experiments. This paper presents methods of toxicity evaluation of natural biocides using Sprague-Dawley rats and New Zealand White (NZW) rabbits. Safety of the natural biocide extract of Asarum sieboldii was evaluated using single-dose oral and dermal toxicity tests in Sprague-Dawley rats, and eye and skin irritation tests in NZW rabbits. The extract has proven antimicrobial and insecticidal activities against wood-rotting fungi and termites. After single oral administration to rats, the $LD_{50}$ values were determined to be over 4,000 and 2,000 mg/kg for males and females, respectively. After single dermal administration to rats, the $LD_{50}$ values exceeded 10,000 mg/kg for both males and females. The extract was identified to be non-irritant to the rabbit eye, and only slightly irritant to the rabbit skin. In this study, we confirmed the safety of the A sieboldii extract through animal testing. Due to the harmfulness of humidifier disinfectants, focus is on the safety of chemical pesticides, and toxicity evaluation is suggested as the basic method for hazard evaluation.

A study of 'Asari Herba should not exceed Jeon(錢, Qian, 3.0 gram)' ('세신불가전(細辛不過錢)'에 관한 연구)

  • Objectives: Asari Herba comes from the dried perennial herbaceous plant, Asarum sieboldii $M_{IQ}$., A. heterotropides $F_R$. SCHMIDT var. mandshuricum($M_{AXIM}$) $K_{ITAG}$. and A sieboldii $M_{IQ}$ var. seoulense $N_{AKAI}$., family Aristolochiaceae. This medicine is pungent in flavor(味辛), warm in nature(性溫), mildly toxic and affects the lung, kidney, and heart meridians. It is able to repel wind dispel cold(祛風散寒), remove the obstruction of the nose(宣通鼻竅), relieve pain(止痛) and warm the lungs to remove the retention of fluids(溫肺化飮). In a clinic it is prescribed for exterior syndrome of wine-cold type(風寒表證) manifested as headache and general pain. It is indicated for headaches, toothaches, bisyndrome of wind-cold-dampness type(風寒濕痺), etc. For many generations medical books have warned doctors that the dosage of Asari Herba should not be excessive because the pungent flavor has the effect of dispersing exopathogens from superficies of the body. The principle, 'Asari Herba should not exceed Jeon(錢, Qian, 3.0 gram)' is still considered to be relevant. Methods : Extensive research of several ancient medical texts has lead to the following conclusion. Results : The statement 'Asari Herba should not exceed Jeon' is a dosage which is used to prepare single recipe and powder. Conclusion : The definition of Jeon is not 1 Jeon(3.0 gram), but 1 Jeonbi(錢匕, Qianbi, 1.0 gram), moreover, the usage, collection. and refinement process are not in accordance with the contents of the ancient medical texts. A deeper study of Asari Herba's dosage in the future is expected to have interesting results.
